T-shaped droplet microfluidic device was used for preparing 5-Fu-loaded chitosan microspheres. • 5-Fu-loaded chitosan microspheres exhibit pH-responsive release patterns and sustainable release characteristics for up to 60 days. • The average particle size of the microspheres is 3.90 ± 0.57 μm, and the particle size distribution is narrow. • Drug-loaded microspheres of different particle sizes have different drug release rates and cycles. Microsphere formulations are a promising drug delivery system for targeted response and prolonged sustained release. However, conventional emulsification crosslinking methods for microsphere preparation usually result in a broad particle size distribution, large particle size, poor uniformity, and consequently erratic drug release kinetics. In this investigation, chitosan microspheres encapsulating 5-Fluorouracil (5-Fu CS-MS) were fabricated using a T-type droplet microfluidic device, and the formulation was optimized by single-factor experiments. The resulting microspheres had a consistent drug release profile, a smooth surface, high monodispersity, and a uniform particle size distribution with an average particle size of 3.90 ± 0.57 μm. The polymer-drug interaction in 5-Fu CS-MS was analyzed using FTIR and XRD. The XPS and EDS results confirmed the occurrence of a Schiff base reaction between glutaraldehyde (GA) and chitosan (CS), indicating successful encapsulation. TGA results revealed that the cross-linking reaction enhanced the thermal stability of the microspheres. Biocompatibility assessments using Human umbilical vein endothelial cells (HUVEC) line demonstrated the safety and compatibility of the fabricated microspheres. More importantly, the in vitro release results illustrated a pH-responsive drug release profile and sustained release of 5-Fu CS-MS for a period up to 60 days. In summary, the CS sustained-release microspheres produced using the microfluidic technique have the potential for intelligent response, precise drug delivery, and prolonged sustained release of small-molecule drugs. This study developed chitosan-based hydrogels as field-deployable carriers for sustained fungicide delivery to enhance crop disease resistance and yield. Citral and chitosan were reacted at three molar ratios (N1, N1.5, N2) via acid-catalyzed Schiff-base condensation, and the resulting hydrogels were characterized by FTIR, 1 H NMR, SEM and WXRD. Structural analyses confirmed the formation of self-ordered imine clusters with a d -spacing of 20.5 Å stabilized by non-covalent interactions. N1, the highest-cross-linked hydrogel, showed the best thermal stability ( T 10 = 160°C) and the lowest swelling, releasing 7.0% slower than N1.5 and 37.9% slower than N2. Antifungal assays demonstrated that N1 provided the strongest inhibition against five phytopathogens ( Rhizoctonia solani , Fusarium graminearum , Botrytis cinerea , Rhizoctonia cerealis , and Phytophthora capsici ), improving inhibition of B. cinerea , P. capsici , and F. graminearum by 56.8%, 82.9%, and 181.3%, respectively, compared with free citral. These hydrogels combine antifungal activity, water retention, thermal stability and controlled release, providing a promising strategy for green fungicide delivery in agriculture.

Polysaccharide-based injectable hydrogels have several advantages in the context of biomedical use. However, the main obstruction associated with the utilization of these hydrogels in clinical application is their poor mechanical properties. Herein, we describe in situ gelling of nanocomposite hydrogels based on quaternized cellulose (QC) and rigid rod-like cationic cellulose nanocrystals (CCNCs), which can overcome this challenge. In all cases, gelation immediately occurred with an increase of temperature, and the CCNCs were evenly distributed throughout the hydrogels. The nanocomposite hydrogels exhibited increasing orders-of-magnitude in the mechanical strength, high extension in degradation and the sustained release time, because of the strong interaction between CCNCs and QC chains mediated by the cross-linking agent (β-glycerophosphate, β-GP). The results of the in vitro toxicity and in vivo biocompatibility tests revealed that the hydrogels did not show obvious cytotoxicity and inflammatory reaction to cells and tissue. Moreover, DOX-encapsulated hydrogels were injected beside the tumors of mice bearing liver cancer xenografts to assess the potential utility as localized and sustained drug delivery depot systems for anticancer therapy. The results suggested that the QC/CCNC/β-GP nanocomposite hydrogels had great potential for application in subcutaneous and sustained delivery of anticancer drug to increase therapeutic efficacy and improve patient compliance.

Intelligent hydrogels are attractive biomaterials for various applications, however, fabricating a hydrogel with both adequate self-healing ability and mechanical properties remains a challenge. Herein, a series of novel intelligent konjac glucomannan (KGM)/microcrystalline cellulose (MCC) hydrogels were prepared vis the mussel-inspired chemistry. MCC was firstly functionalized by the oxidative polymerization of dopamine, and the intelligent hydrogels were obtained by mixing aqueous solutions of KGM and functionalized MCC (PDMCC). By introducing PDMCC, a more compact interconnected porous structure formed for the resulting hydrogels. The self-healing ability and mechanical properties of intelligent hydrogels were dependence on the PDMCC content. Compared with KGM hydrogels, KGM/PDMCC hydrogels exhibited a more distinct pH sensitivity and a lower initial burst release, which was attributed to the compact structure and strong intermolecular hydrogen bond interaction between PDMCC and KGM. These results suggest that the KGM/PDMCC intelligent hydrogels may be promising carriers for controlled drug delivery.

ObjectivesIcariin, extracted from Epimedium, is a kind of flavonoid and possesses osteogenesis and antioxidant. This study aimed to evaluate the therapeutic effects of icariin liposome on acute blunt skeletal muscle injury in rats.MethodsIcariin liposome was prepared by the thin-film dispersion method. After muscle injury, the corresponding treatment measures were given every day for two weeks. Recovery and mechanism of muscle injury were evaluated by QRT-PCR, HE, immunohistochemistry, malondialdehyde, superoxide dismutase and serological tests.Key findingsThe particle size, polydispersity index, zeta potential, encapsulation efficiency and drug loading of icariin liposomes were 171.37 ± 38.23 nm, 0.27 ± 0.01, −5.59 ± 1.36 mV, 78.15 ± 2.04% and 15.62%, respectively. The QRT-PCR showed that icariin liposome significantly promoted the expression of MHCIIB and vimentin. Through HE, immunohistochemistry, ELISA and serological tests, we found that icariin liposome effectively promoted desmin expression, reduced collagen I expression and inhibited the production of pro-inflammatory factors, including TNF-α and IL-6. Icariin liposome therapy significantly reduced the level of malondialdehyde and increased the activity of superoxide dismutase.ConclusionsIcariin liposome has excellent therapeutic effects on acute blunt muscle injury in rats by improving immunity, repairing cytoskeleton and cellular integrity, anti-inflammation, anti-fibrosis and antioxidant stress.

Development of biocompatible and bioactive drug-loaded sutures is considered as an effective but challenging strategy for the wound healing process by delivering biological drugs (e.g., antibiotics) or growth factors (e.g., bFGF) at the surgical wound sites. Conventional offline suture strategies often lead to fast and uncontrollable release of drugs at wound sites, rendering wound healing to become a longer and painful process for patients. Herein, we propose an online suture strategy to fabricate electrospun polycaprolactone (PCL) fibrous yarns, incorporated with both collagen (COL) and bFGF, to produce bFGF-COL@PCL sutures. Upon demonstrating the well-oriented and aligned fibrous microstructure, high mechanical properties, and controlled release of bFGF from bFGF-COL@PCL sutures in vitro, we then applied bFGF-COL@PCL sutures to an incision wound healing mouse model in vivo. Further in vivo study showed that as compared to the commercialized Vicryl suture, bFGF-COL@PCL sutures significantly promoted the wound healing at different stages by accelerating granulation tissue formation, collagen deposition, and re-epithelialization. The enhanced wound healing efficiency of bFGF-COL@PCL sutures is likely attributed to two synergistic factors: (i) the well-oriented nanofibrous structure reduces tissue drag to minimize their trauma and (ii) the presence of both collagen and bFGF enhances the basement membrane (BM) reconstruction, cell proliferation, and angiogenesis. This work demonstrates an effective suture strategy and system for surgical suture applications.

The feasibility of using unmodified bamboo shoot cellulose (BSC) to produce composite aerogels with sodium alginate (SA) in a fast and green way for sustained release of curcumin was explored for the first time, in which calcium ion-induced SA cross-linking could effectively retain the structural stability of aerogel skeleton. The aerogels were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy and differential scanning calorimetry . The encapsulation and release of curcumin from aerogels were studied while the antioxidant activity of encapsulated curcumin was investigated. Curcumin was evenly encapsulated in the composite aerogels and showed a sustained release behavior, followed the first-order rate expression. Interpenetrating network structures were built between BSC and SA mainly through hydrogen bonding, which could be further reinforced by the cross-linking of CaCO 3 on the SA matrix. The original characteristics of BSC in the composite aerogels were well retained. The thermal stability and mechanical properties of the composite aerogels were improved by Ca 2+ -induced cross-linking, while the uncross-linked composite aerogels exhibited better encapsulation efficiency and in vitro antioxidant activity. Overall, this study was the first to use cellulose from bamboo shoot to develop aerogels for drug delivery purposes. The cellulose/alginate composite aerogels were promising to be used as biocompatible carriers for drug and nutraceutical delivery.

To efficiently deliver stavudine (d4T) for AIDS therapy, chitosan–stavudine conjugate (Cs–d4T) was synthesized. However, its poor water-solubility limited its clinical application. In this study, a sulphonated chitosan–stavudine conjugate (SCs–d4T) was synthesized with a mild SO 3 ·Py complex sulphonation strategy. Chemical characteristics and morphology of Cs–d4T and SCs–d4T were performed by NMR, XRD, FTIR, ICP-AES and SEM. SCs–d4T demonstrated satisfactory solubility (106-bold of Cs–d4T solubility), good anti-HIV activity (6-fold of d4T anti-HIV activity), and well sustained release ability. The major release product O -isopropyl-5′- H -phosphonate of d4T (d4T-P-H) showed higher anti-HIV activity than d4T. For further evaluating the influence of linker and sulphonation strategy on anti-HIV activity, chitosan grafted with d4T by succinyl linker (Cs–sd4T) and SCs–d4T sulphonated by oleum were also prepared. The result showed that the O -isopropyl monophosphate linker of Cs–d4T and SO 3 ·Py complex sulphonation strategy revealed higher anti-HIV activity than succinyl linker of Cs–sd4T and oleum sulphonation strategy, respectively.

This study aims to investigate the solid lipid nanoparticle (SLN) as a novel vehicle for the sustained release and transdermal delivery of piroxicam, as well as to determine the anti-inflammation effect of piroxicam-loaded SLN. SLN formulation was optimized and the particle size, polydispersity index, zeta potential (ZP), encapsulation efficiency, drug release, and morphological properties were characterized. The transdermal efficiency and mechanism of the piroxicam-loaded SLNs were investigated in vitro. With the inflammation induced edema model in rat, the anti-inflammatory efficiency of piroxicam-enriched SLNs (Pir-SLNs) was evaluated. The SLN formulation was optimized as: lecithin 100 mg, glycerin monostearate 200 mg, and Tween (1%, w/w). The particle size is around 102 ± 5.2 nm with a PDI of 0.262. The ZP is 30.21 ± 2.05 mV. The prepared SLNs showed high entrapment efficiency of 87.5% for piroxicam. There is no interaction between piroxicam and the vehicle components. The presence of polymorphic form of lipid with higher drug content in the optimized Pir-SLNs enables the Pir-SLNs to release the drug with a sustained manner. Pir-SLNs with oleic acid as enhancer can radically diffuse into both the stratum corneum and dermal layer, as well as penetrate through the hair follicles and sebaceous glands with significantly higher density than the other control groups. Pir-SLNs promptly inhibited the inflammation since the 3rd hour after the treatment by decreasing the PGE2 level. SLN was demonstrated to be a promising carrier for encapsulation and sustained release of piroxicam. Pir-SLN is a novel topical preparation with great potential for anti-inflammation application.

In order to increase the solubility, stability, and availability of cinnamaldehyde which was widely used in food industry, ethyl cellulose aqueous dispersions encapsulated cinnamaldehyde had been prepared by using an anti-solvent induced method. The particles size of the obtained aqueous dispersions was in the range of 150–200 nm, and it increased slightly with the increasing concentrations of the precursor solution. The prepared dispersions after being dried at ambient conditions had the ability to form a free-standing film, and the film had denser structure and smooth surface. Moreover, the ethyl cellulose dispersions had powerful encapsulation ability to cinnamaldehyde, and the encapsulated cinnamaldehyde could be sustained released, it could prolong the storage time of fresh pork samples for more than 6 days, and an excellent antimicrobial property for food spoilage was observed. It indicated that the prepared dispersions could be a fascinating supporter for increasing the stability and sustained-release of cinnamaldehyde. Practical applications In this work, the ethyl cellulose aqueous dispersions encapsulating with cinnamaldehyde had good stability, and the sustained released of cinnamaldehyde could effectively inhibit the growth of spoilage organism for a long time, which indicated that aqueous dispersions system had potential applications for food preservation.

Implant surface modification that provides local sustained release of osteoinductive therapeutic agents enhances implant stability. We designed a mesoporous TiO2-layered titanium implant (MLT) by modified anodization technique that allowed local sustained release of zoledronic
acid up to 21 days. Mesoporous layer has pore size 15 nm, depth ∼30 μm, volume 0.32 cm3/g, surface area 112.3 m2/g, surface roughness 20 nm and water contact angle 18.3°. Zoledronic acid-loaded MLT (MLT-Z) was biocompatible, showed anabolic effect on
bone forming osteoblasts and catabolic effect on bone resorbing osteoclasts. MLT or MLT-Z implants were implanted in osteoporotic rat-tail vertebrae. Smooth implant in healthy rats were used as a positive control. Histomorphometric analysis showed that bone implant contact of smooth implant
in osteoporotic rats was reduced by 4.1-fold compared to healthy rats and MLT-Z rescued the effect by 53%. Similar effect was observed in implant fixation, mechanical stability, BV/TV ratio, Tb.N, Tb.Th and OI% among the groups. Histological and μ-CT images strongly supported the
above-mentioned findings. In conclusion, a novel surface-fabricated MLT-Z gives local sustained drug release, robustly enhances implant osseointegration and stability in osteoporotic condition, suggesting it as a promising implant model for patients with compromised bone quality.

This study investigates the usage of a spinneret with a Teflon nozzle for fabrication of higher quality drug sustained-release electrospun nanofibers. Ethyl cellulose (EC) and quercetin were used as a filament-forming polymer matrix and an active pharmaceutical ingredient, respectively. The electrospinning was conducted using both a traditional stainless steel spinneret and a spinneret with a Teflon nozzle. Experimental results demonstrated that a Teflon-fluid interface at the spinneret's nozzle provided a better performance for implementing electrospinning than a traditional metal-fluid interface in the following aspects: (1) keeping more electrical energy on the working fluids for an efficacious process; (2) exerting less negative effect on the fluid to draw it back to the tube; and (3) making less possibility of clogging. The resulted nanofibers from the spinneret with a Teflon nozzle exhibited higher quality than those from the traditional spinneret in those: (1) smaller diameter and narrower distribution, 520 ± 70 nm for the former and 750 ± 280 nm for the later, as indicated by the field emission scanning electron microscopic images; and (2) better sustained-release profiles of quercetin from the former than the latter, as demonstrated by the in vitro dissolution tests. The new protocols about usage of Teflon as a spinneret's nozzle and the related knowledge disclosed here should promote the preparation and application of electrospun functional nanofibers.
