Super-Stable Homogeneously Sustained-Release System Mediates Transcatheter Arterial Ionic-Embolization Strategy for Hepatocellular Carcinoma Therapy

Xuqi Peng,Yating Zheng,Yi Xue,Xiaoliu Liang,Pan He,Hu Chen,Peng He,Yisheng Peng,Zhenwen Zhao,Yulun Chen,Xiran Gui,Lei Yang,Yongfu Xiong,Juan Lin,Yesi Shi,Chengchao Chu,Yang Zhang,Gang Liu

The clinical effectiveness of locoregional therapies in treating hepatocellular carcinoma (HCC) is frequently constrained by multi-drug resistance and/or tumor metastasis. To surmount these challenges, a promising approach, transcatheter arterial ionic-embolization (TAIE) is proposed, which can specifically and continuously disrupt the intracellular ionic balance to significantly inhibit tumor activity and invasion. The hydrophilic micro-nanoscale sodium chloride particles (SCPs) are ingeniously intermixed with hydrophobic lipiodol to create a super-stable homogeneous embolic formulation (lipiodol-sodium chloride, LSC). After interventional administration, the LSC selectively deposits in HCC lesions, where lipiodol stably delivers SCPs to disrupt the cell's ionic balance, causing cell death without drug resistance. Notably, it is demonstrated that LSC can significantly hinder tumor cell migration and invasion. The mechanism is through SCP disruption of the ionic balance, which induces cell swelling and subsequent vimentin hydrolysis-mediated cytoskeletal remodeling. In addition, it is found that LSC treatment notably downregulates the expression of MYLK, TLN, and THBS2 genes in the focal adhesion (FA) signaling pathway of HepG2 cells. LSC formulation integrated tumor-specific deposition, intratumoral sustained release, efficient tumoricidal activity, significant metastasis inhibition, and excellent biological safety, thereby demonstrating superior in vivo tumor therapeutic effects via TAIE strategy, and showing a promising cancer therapeutic approach for clinical application.

Enhancing the salt stress resistance of seeds and seedlings via a brassinolide sustained release agent system

Zhong Ximing,Lan Ruopeng,Su Guofeng,Hao Li,Xu Hua,Zhou Hongjun,Zhou Xinhua

Background The growing escalation of soil salinization is tremendously threatening the global food security and the development of sustainable agriculture. To address the worldwide predicament caused by salt stress toward crops, combining nanotechnology with the merits of plant hormone may become an efficient and effective approach. Results In this work, a sustained release agent system (BR@MSN) was developed by loading brassinolide (BR) to mesoporous silica nanoparticles (MSN) to enhance the salt stress resistance of cucumber seeds and seedlings. The obtained BR@MSN agent was about 120 nm. As an endogenous plant hormone, promotion in crop growth was found at low BR concentration. Due to the sustained release property, BR@MSN avoided excessive BR exposure to seeds and seedlings to cause inhibitory effects. After the soil application of BR@MSN, the promotion effect from BR combined with the regulation enhancement from MSN nanocarrier improved the seed germination rate by 11.76% under saline environment. Compared with the same BR concentration (2.0 mg/L), BR@MSN increased the seed germination rate even by 1324.29%. In addition, remarkable wettability on foliar surfaces was found, and the foliar application of BR@MSN significantly enhanced the salt stress resistance of cucumber seedlings by alleviating the accumulation of reactive oxygen species (ROS) and increasing the cell viability along with the improvement in superoxide dismutase (SOD) activity (234.11%), the decrease in malondialdehyde (MDA) content (61.30%), and the increase in chlorophyll content (110.88%). Conclusions The newly developed BR@MSN agents could effectively enhance the salt stress resistance of crop seeds and seedlings, and their applications significantly improved the seed germination rate and seedling growth. The remarkable efficacy makes this BR@MSN agent system potential in agricultural field for enhancing the salt stress resistance of crops and facilitating the development of sustainable agriculture. 3D printed heart valve mediated nitric oxide sustained release reduced potential for calcification and inflammatory capacity

Penghui Wang,Jialing Li,Wenjie Zhang,Yanhan Ren,Juping Ma,Shuang Li,Xiaoyan Tan,Bo Chi

Heart valve replacement have numerous problems, such as calcification, inflammation, and limited endothelialization. Herein, we used bio-degradable arginine-modified polylysine and oxidized hyaluronic acid as the substrate, expanding the advantages of rapid Schiff-base reaction of aldehydes and amino groups under physiological conditions to form dynamic acyl-hydrazone bonds. These were used to design dynamic adaptive hydrogels, and a polysaccharide/protein dual-network system that mimicked the extracellular matrix, realizing the excellent mechanical tunability and shear thinning properties of its hydrogel. The dual antibacterial mechanism effectively avoided the occurrence of infection-like inflammation after valve implantation. The hydrogel has great ability in promoting cellular proliferation, and can also promote cell migration through a potential cell recruitment mechanism. The hydrogel dynamically triggers and sustains NO release in vivo , removing highly expressed ROS in inflammatory site as needed. It further promotes angiogenesis, and provides targeted treatment for calcified aortic valve disease caused by oxidative stress signals. Furthermore, it exhibits exceptional anti-calcification properties and can enhance the upregulation of CD31 while suppressing the expression of IL-10 and TNF-α, thereby significantly promoting angiogenesis and reducing inflammation. Additionally, 3D printing technology enables the individualized customization of valves with diverse heterogeneous structures to achieve a high degree of biomimetic tissue.

Sustained release of brimonidine from polydimethylsiloxane-coating silicone rubber implant to reduce intraocular pressure in glaucoma

Huang Chang,Shen Yuening,Zhao Yujin,Zhang Zhutian,Gao Shunxiang,Hong Jiaxu,Xu Jianjiang,Meng Qingtao,Sun Xinghuai,Sun Jianguo

Glaucoma is the leading cause of irreversible blindness, affecting 111 million people by 2040 worldwide. Intraocular pressure (IOP) is the only controllable risk factor for the disease and current treatment options seek to reduce IOP via daily taking eye drops. However, shortcomings of eye drops, such as poor bioavailability and unsatisfied therapeutic effects, may lead to inadequate patient compliance. In this study, an effective brimonidine (BRI)-loaded silicone rubber (SR) implant coated with polydimethylsiloxane (BRI@SR@PDMS) is designed and fully investigated for IOP reduction treatment. The in vitro BRI release from BRI@SR@PDMS implant reveals a more sustainable trend lasting over 1 month, with a gradually declined immediate drug concentration. The carrier materials show no cytotoxicity on human corneal epithelial cells and mice corneal epithelial cells in vitro. After administrated into rabbit’s conjunctival sac, the BRI@SR@PDMS implant releases BRI in a sustained fashion and effectively reduces IOP for 18 days with great biosafety. In contrast, BRI eye drops only maintain IOP-lowering effect for 6 h. Therefore, as a substitute of eye drops, the BRI@SR@PDMS implant can be applied as a promising non-invasive platform to achieve long-term IOP-lowering in patients suffering from ocular hypertension or glaucoma.

Osmanthus fragrance polyurethane/silk fibroin-based double-shell microcapsules for aromatic leather with sustained release fragrance

Jiaying Gu,Ming Liu,Lin Li,Lulu Zhou,Lei He,Weijun Deng,Jing Hu

The leather with the long-lasting fragrance is critical for functional leather commodities but limited by poor temperature durability, low loading capacity and inferior flexural resistance. Herein, we develop a novel double-shell fragrance release system via both interfacial polymerization and freeze inducement. Osmanthus fragrance (OF) was encapsulated with polyurethane (PU) and silk fibroin (SF), the double-shell microcapsules with remarkable stability and enhanced sustained release property can be prepared. The OF@PU/SF microcapsules had well-defined spherical shape with an average diameter of 5.21 μm measured by Scanning Electron Microscopy (SEM), which was larger than the single-shell microcapsules (OF@SF) with the size of 1.70 μm. The thermogravimetric analysis (TGA) indicated that the OF loading capacity (LC) of OF@PU/SF microcapsules was 51.7%. Furthermore, the sustained release measurements by UV–vis absorption spectra clarified that 22.9% OF released from OF@PU/SF microcapsules during 30 days, lower than 36.7% of OF@SF microcapsules, much less than nearly 100% of free OF. Finally, the OF@PU/SF microcapsules were applied in leather finishing to efficiently improve the odour of the leather. The aromatic leather possessed sustained aroma with great stability enabling high temperature durability and flexural resistance property.

Sustained release of a synthetic structurally-tailored glycopolymer modulates endothelial cells for enhanced endothelialization of materials

Shuaibing Jiang,Jingxian Wu,Yingjie Hang,Qi Liu,Dan Li,Hong Chen,John L. Brash

Glycosaminoglycans (GAG) play a vital role in the regulation of various cellular functions through their interactions with key proteins involved in cell signaling and can be useful as bioactive agents in the design of biomaterials. However available “natural” GAG that might be used for this purpose suffer a number of drawbacks including structural heterogeneity and the presence of impurities. Alternatively, GAG-mimicking polymers have shown promise for the modification of biomaterials to support the behavior and function of adherent cells. In the study reported herein, a series of glycopolymers of tailored structure were synthesized using a novel “unit-recombination” strategy. These were intended as surface modifiers for the promotion of surface endothelialization. Two key structural units, sulfonate and saccharide, from the building blocks of GAG, were “recombined” to give GAG analogues by copolymerization of sodium 4-vinylbenzenesulfonate (as the sulfonate unit) and 2-methacrylamido glucopyranose (as the saccharide unit). The composition and molecular weight of the copolymers were adjusted to obtain maximum endothelial cell (EC) proliferation. The glycopolymer with sulfonate and saccharide units in a ratio of approximately 1.0 and a molecular weight of 9.8 kDa was found to be optimal. Interestingly, the data emphasized the key role of saccharide units in GAG bioactivity, rather than the commonly believed decisive role of sulfonate units. The optimized glycopolymer was incorporated into fibrous polycaprolactone (PCL) materials by coaxial electrospinning. These materials showed controlled release of the GAG-mimicking polymers over prolonged periods. The proliferation, migration and function of EC on the glycopolymer-modified PCL fiber mats were greatly improved compared to those on unmodified PCL mats. A likely mechanism underlying these improvements is that the glycopolymers significantly enhance the binding of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) to their receptors on the cell membrane. These findings demonstrate that these structurally-tailored glycopolymers modulate the functions of EC adherent to materials and may be effective as modifiers for the promotion of endothelialization of blood-contacting implants.

Sustained Release of Zoledronic Acid from Mesoporous TiO2-Layered Implant Enhances Implant Osseointegration in Osteoporotic Condition

Liu Jinsong,Pathak Janak L.,Hu Xiaohui,Jin Yifan,Wu Zuosu,Al-Baadani Mohammed A.,Wu Shuyi,Zhang Hualin,Farkasdi Sandor,Liu Yi,Ma Jianfeng,Wu Gang

Implant surface modification that provides local sustained release of osteoinductive therapeutic agents enhances implant stability. We designed a mesoporous TiO2-layered titanium implant (MLT) by modified anodization technique that allowed local sustained release of zoledronic acid up to 21 days. Mesoporous layer has pore size 15 nm, depth ∼30 μm, volume 0.32 cm3/g, surface area 112.3 m2/g, surface roughness 20 nm and water contact angle 18.3°. Zoledronic acid-loaded MLT (MLT-Z) was biocompatible, showed anabolic effect on bone forming osteoblasts and catabolic effect on bone resorbing osteoclasts. MLT or MLT-Z implants were implanted in osteoporotic rat-tail vertebrae. Smooth implant in healthy rats were used as a positive control. Histomorphometric analysis showed that bone implant contact of smooth implant in osteoporotic rats was reduced by 4.1-fold compared to healthy rats and MLT-Z rescued the effect by 53%. Similar effect was observed in implant fixation, mechanical stability, BV/TV ratio, Tb.N, Tb.Th and OI% among the groups. Histological and μ-CT images strongly supported the above-mentioned findings. In conclusion, a novel surface-fabricated MLT-Z gives local sustained drug release, robustly enhances implant osseointegration and stability in osteoporotic condition, suggesting it as a promising implant model for patients with compromised bone quality.

Dual cross-linked chitosan microspheres formulated with spray-drying technique for the sustained release of levofloxacin

Jing Zhou,Yuanyuan Chen,Mengmeng Luo,Fen Deng,Sen Lin,Wencan Wu,Guqiang Li,Kaihui Nan

Objective: This study was aimed to develop sustained drug release from levofloxacin (LF)-loaded chitosan (CS) microspheres for treating ophthalmic infections.Significance: Dual cross-linked CS microspheres developed by the spray-drying technique displays significantly higher level of sustained drug release compared with non-cross-linked CS microspheres. Methods: LF-loaded CS microspheres were prepared using the spray-drying technique, and then solidified with tripolyphosphate and glutaraldehyde as dual cross-linking agents. The microspheres were characterized by surface morphology, size distribution, zeta potential, encapsulation efficiency, and drug release profiles in vitro. The drug quantification was verified and analyzed by high-performance liquid chromatography (HPLC). The structural interactions of the CS with LF were studied with Fourier transform infrared spectroscopy. The effect of various influencing excipients in the formulation of the dual cross-linked CS microspheres on drug encapsulation efficiency and the drug release profiles were extensively investigated.Result: The microspheres demonstrated high encapsulation efficiency (72.4 ∼ 98.55%) and were uniformly spherical with wrinkled surface. The mean particle size was between 1020.7 ± 101.9 and 2381.2 ± 101.6 nm. All microspheres were positively charged (zeta potential ranged from 31.1 ± 1.32 to 42.81 ± 1.55 mV). The in vitro release profiles showed a sustained release of the drug and it was remarkably influenced by the cross-linking process. Conclusion: This novel spray-drying technique we have developed is suitable for manufacturing LF-loaded CS microspheres, and thus could serve as a potential platform for sustained drug release for effective therapeutic application in ocular infections.

Mussel-inspired fabrication of konjac glucomannan/microcrystalline cellulose intelligent hydrogel with pH-responsive sustained release behavior

Lin Wang,Yu Du,Yi Yuan,Ruo-Jun Mu,Jingni Gong,Yongsheng Ni,Jie Pang,Chunhua Wu

Intelligent hydrogels are attractive biomaterials for various applications, however, fabricating a hydrogel with both adequate self-healing ability and mechanical properties remains a challenge. Herein, a series of novel intelligent konjac glucomannan (KGM)/microcrystalline cellulose (MCC) hydrogels were prepared vis the mussel-inspired chemistry. MCC was firstly functionalized by the oxidative polymerization of dopamine, and the intelligent hydrogels were obtained by mixing aqueous solutions of KGM and functionalized MCC (PDMCC). By introducing PDMCC, a more compact interconnected porous structure formed for the resulting hydrogels. The self-healing ability and mechanical properties of intelligent hydrogels were dependence on the PDMCC content. Compared with KGM hydrogels, KGM/PDMCC hydrogels exhibited a more distinct pH sensitivity and a lower initial burst release, which was attributed to the compact structure and strong intermolecular hydrogen bond interaction between PDMCC and KGM. These results suggest that the KGM/PDMCC intelligent hydrogels may be promising carriers for controlled drug delivery.

Ethyl cellulose aqueous dispersions: A fascinating supporter for increasing the solubility and sustained-release of cinnamaldehyde

Xingzhong Zhang,Xia Wu,Wei Li,Xiaogang Luo,Yan Li,Bin Li,Shilin Liu

In order to increase the solubility, stability, and availability of cinnamaldehyde which was widely used in food industry, ethyl cellulose aqueous dispersions encapsulated cinnamaldehyde had been prepared by using an anti-solvent induced method. The particles size of the obtained aqueous dispersions was in the range of 150–200 nm, and it increased slightly with the increasing concentrations of the precursor solution. The prepared dispersions after being dried at ambient conditions had the ability to form a free-standing film, and the film had denser structure and smooth surface. Moreover, the ethyl cellulose dispersions had powerful encapsulation ability to cinnamaldehyde, and the encapsulated cinnamaldehyde could be sustained released, it could prolong the storage time of fresh pork samples for more than 6 days, and an excellent antimicrobial property for food spoilage was observed. It indicated that the prepared dispersions could be a fascinating supporter for increasing the stability and sustained-release of cinnamaldehyde. Practical applications In this work, the ethyl cellulose aqueous dispersions encapsulating with cinnamaldehyde had good stability, and the sustained released of cinnamaldehyde could effectively inhibit the growth of spoilage organism for a long time, which indicated that aqueous dispersions system had potential applications for food preservation.

Sustained release of hydrogen sulfide from anisotropic ferrofluid hydrogel for the repair of spinal cord injury

Ruofei Wang,Xia-Xiao Wu,Zhenming Tian,Tian Hu,Chaoyang Cai,Guan-Ping Wu,Gang-Biao Jiang,Bin Liu

Spinal cord injury (SCI) results in massive neuronal death, axonal disruption, and cascading inflammatory response, which causes further damage to impaired neurons. The survived neurons with damaged function fail to form effective neuronal circuits. It is mainly caused by the neuroinflammatory microenvironment at injury sites and regenerated axons without guidance. To address this challenge, a ferrofluid hydrogel (FFH) was prepared with Ferric tetrasulfide (Fe 3 S 4 ), carboxymethyl chitosan, and gold. Its internal structural particles can be oriented in a magnetic field to acquire anisotropy. Moreover, Fe 3 S 4 can release hydrogen sulfide (H 2 S) with anti-inflammatory effects under acidic conditions. Regarding in vitro experiments, 0.01g/ml Fe 3 S 4 FFH significantly reduced the inflammatory factors produced by LPS-induced BV2 cells. Oriented and longer axons of the induced neural stem cells loaded on anisotropic FFH were observed. In vivo experiments showed that FFH reduced the activated microglia/macrophage and the expression of pro-inflammatory factors in SCI rats through the NF-κB pathway. Moreover, it significantly promoted directional axonal regrowth and functional recovery after SCI. Given the critical role of inhibition of neuroinflammation and directional axonal growth, anisotropic Fe 3 S 4 FFH is a promising alternative for the treatment of SCI.

Preparation of Ropivacaine Encapsulated by Zeolite Imidazole Framework Microspheres as Sustained-Release System and Efficacy Evaluation

Peng Wang,Guangyu Wang,Hongwen Tang,Siwen Feng,Lichuan Tan,Pu Zhang,Guihua Wei,Cuijuan Wang

Graphical A ropivacaine microspheres encapsulated by zeolite imidazole framework (RVC@ZIF-8) as sustained-release system was prepared for the first time. This novel material exhibits a high drug loading rate and superior biological safety, with a continuously release in vitro for at least 96 h, and the sensory block time was more than 72 h in vivo pharmacodynamic. The management of persistent postoperative pain still remains a clinical challenge currently. Although ropivacaine (RVC) is widely used for postoperative analgesia as a local anesthetic, the short half-life makes it difficult to achieve the desired duration of analgesia. Herein, a RVC sustained-release microspheres encapsulated by zeolite imidazole framework-8 (RVC@ZIF-8) was synthesized for the first time, which prolonged the sustained-release of RVC and decreased the resulting drug toxicity. RVC can continuously release in vitro for at least 96 h with high drug loading of 30.6 % and RVC@ZIF-8 had excellent biocompatibility and low cytotoxicity. In sciatic nerve block model, the sensory block time of RVC@ZIF-8 was significantly prolonged compared with RVC, achieving more than 72 h post injection and no inflammation or lesion were found. Based on high drug loading, ideal sustained-release and superior biological safety, RVC@ZIF-8 will be a novel delivery material for local anesthetic with potential application.
