An iron-doped Cu2O nanozyme enables highly sensitive colorimetric detection of vitamin E in serum

Xiangyu Feng,Haibing Zhu,Chuanli Ren,Fu-an Wang,Zhanjun Yang,Guiling Li

Vitamin E plays a vital role in maintaining human health as well as in the prevention and treatment of related diseases. However, current methods for detecting vitamin E suffer from complex instrumentation, high cost, and cumbersome operation, making it difficult to meet the demand for rapid detection in serum samples. Herein, we designed an iron-doped cuprous oxide (Fe-Cu2O) nanozyme for the colorimetric detection of vitamin E in human serum. Impressively, the Fe-Cu2O nanozyme exhibits a 7.2-fold enhancement in peroxidase (POD)-like activity compared with the pure Cu2O nanozyme. In the presence of hydrogen peroxide (H2O2), the Fe-Cu2O nanozyme catalyzes the decomposition of H2O2 to generate hydroxyl radicals (˙OH), thereby oxidizing colorless 3,3′,5,5′-tetramethylbenzidine (TMB) into a blue product (ox TMB). As a reducing agent, vitamin E can fade the blue ox TMB, enabling the quantitative determination of vitamin E. Under optimized experimental conditions, the as-prepared Fe-Cu2O nanozyme achieves a linear detection range of 2–80 µM toward vitamin E with a limit of detection (LOD) of 1.89 µM. This approach holds promise as a straightforward, economical, and highly effective strategy for clinical diagnosis and health screening of vitamin E and other bioactive molecules.

Skin-mimicking biogel-based iontronic sensor with hierarchical bionic coupling for dexterous tactile e-skin

Yidan Chen,Shu Wang,Xilu Ye,Chenghui Lv,Jialin Wei,Yingxin Zhang,Jianfeng Ping,Yibin Ying,Lingyi Lan

The human fingertip skin, with its interlocked epidermal-dermal architecture and dense tactile receptors, enables unique perception and inspires the design of innovative electronic skin (e-skin). However, most e-skin materials fail to replicate the hierarchical integration of composition, structure, and function found in natural skin. Drawing direct inspiration from the dermal extracellular matrix–a composite hydrogel reinforced by a collagen-hyaluronic acid, here we report an e-skin that mimics key composition and architectural features of human skin. By incorporating sodium lactate and montmorillonite nanosheets into a gelatin matrix, we engineer a biomimic gel with mechanical and hydration properties similar to those of natural skin through dynamic ionic crosslinking and hydrogen bond networks. The microstructure resembling the dermal papillae is designed using a sandpaper-templating strategy. The resulting iontronic sensor achieves high sensitivity (466.3 kPa −1 ), rapid response (47 ms), and a wide pressure detection range (20 Pa–2000 kPa). We further develop a stretchable, ultrathin, hand-shaped iontronic sensor array that integrates seamlessly with a dexterous robotic hand, achieving precise, nondestructive grasping and high-fidelity, multichannel pressure mapping. Hydrogels are promising materials for electronic skins and devices, though it is challenging to mimic the composition and architectural features of human skin. Here the authors use a sandpaper templating strategy for a gelatin based hydrogel electronic skin.

An E-interwoven Therapeutic Contact Lens System for Rapid Drug Delivery and Precision Dose Monitoring

Huan Yang,Hengtian Zhu,Wenyu Teng,Heyu Huang,Zuoxiu Tie,Zixu Li,Zhenning Qi,Songtao Yuan,Zizhong Hu,Fei Xu

Precise intraocular pressure (IOP) management and rapid intervention preserve glaucoma vision, yet current ocular delivery systems lack in situ dosage verification, causing treatment blind spots. Here, we develop a wireless therapeutic smart contact lens (SCL) containing a rapid voltage-triggered drug delivery system with a real-time dosage monitoring sensor. The e-interwoven design integrates drug delivery and sensing electrodes in an ultra-thin (17.4 µm) tri-interdigital configuration, which enables the electric field to interact with the drug-loaded hydrogel in the same spatial region. A frequency-matching strategy boosts voltage coupling to drive a brimonidine tartrate-loaded hydrogel to achieve high concentrations in the aqueous humor in 20 min, significantly faster than topical eye drops. The built-in drug dose sensor has a high accuracy <2.8 µg, benefiting from the great linear correlation between wireless frequency drift and release dosage. In acute glaucoma rabbit models, this SCL suppresses peak IOP elevation by 85.7% compared to eye drops. This SCL presents a highly integrated therapeutic platform demonstrating potential for rapid and precise glaucoma treatment.

Integrating Membrane Permeability Engineering and Enzyme Design for Efficient Production of Ergothioneine in E. coli

Jiahuan Ling,Rui Chen,Minghai Wang,Chun-Xiao Yan,Ruomu Xia,Lihui Zhang

Ergothioneine (EGT) is a powerful and natural antioxidant, which can protect cells in the human body from oxidative damage. Numerous studies have attempted to enhance the heterologous synthesis. However, research on EGT transport out of Escherichia coli cells and the wild-type enzyme from Trichoderma reesei is rare. Here, membrane permeability engineering and protein engineering are combined to improve EGT production in E. coli. After metabolic engineering to enhance precursor supply, we deleted the genes involved in lipopolysaccharide biosynthesis to increase the membrane permeability, which promoted EGT production. Further, a mutant of Tregt2E155C with improved catalytic capacity was created, and the expression level of enzymes involved in EGT synthesis was optimized. Finally, fermentation parameters were systematically tuned to maximize the EGT production. The engineered strain MT9-PET-T1/RSF-T2E155C produced 334.20 ± 6.33 mg/L EGT during 48 h of shake-flask fermentation, corresponding to an 8.4-fold increase compared with wild-type strain MT1. When scaled up to 5 L bioreactors, the strain achieved a final EGT titer of 4.06 g/L within 96 h (42.29 mg/L/h). Our work shows significant implications for EGT synthesis and is also a reference for manipulating E. coli to synthesize other biomolecules.

Selective Recovery of Gold from E-Wastewater Using Poly-m-phenylenediamine Nanoparticles and Assembled Membranes

Yuchao Chen,Li Wang,Yufei Shu,Qi Han,Beizhao Chen,Mengxia Wang,Xun Liu,Danyal Rehman,Bei Liu,Zhongying Wang,John Lienhard

The economic value of recovering gold from electronic waste (e-waste) has generated significant interest, but selective capture of gold from complex acidic electronic leaching solutions remains challenging. Here, we synthesized poly-m-phenylenediamine (PmPD) nanoparticles with a positively charged surface and amino functional groups, resulting in an adsorption capacity of 2063 mg/g for Au(III) in acidic solutions, superior to most traditional adsorbents. Electrostatic adsorption and reduction were identified as the adsorption mechanism for Au(III) by zeta potential, X-ray diffraction, transmission electron microscope, Fourier transform infrared, and X-ray photoelectron spectroscopy analyses. To enable adsorbent recycling, PmPD nanoparticles were assembled into adsorptive membranes and used for gold recovery from e-wastewater via a continuous-flow membrane separation process. The PmPD membrane achieved a dynamic gold recovery capacity of approximately 530 mg/g and could be effectively regenerated after washing with a mixture of thiourea and HCl. We demonstrated the practical application of the adsorptive membranes by recovering about 100% of gold from the leaching solution of waste printed circuit boards of computers. Finally, the recovered gold nanoparticles on the PmPD membrane were used to catalyze the degradation of p-nitrophenol, showcasing the catalytic property of gold. The Au@PmPD membrane loaded with 4 mg gold exhibited a high catalytic reduction performance with an apparent rate constant of 0.59 min–1, one of the highest catalytic degradation rate constants of p-nitrophenol reported to date. Our study presents an effective and economical approach for recovering gold from e-waste, providing a prototype of resource recovery and reuse.

Jiaren Du,Tengyue Wang,Yukun Li,Puxian Xiong,Yao Xiao,Ang Feng,Xiaomeng Wang,Kai Jiang,Hengwei Lin

Mechanoluminescence (ML) offers a novel approach to photon emissions without requiring extra electrical input. In recent decade, a variety of ML materials have afforded advantageous prospect for various potential applications. Nevertheless, the material composition optimization and application progress are limited by the time-consuming and complicated preparation methods. Here, a one-step rapid microwave-assisted solid-state (MASS) synthesis approach is firstly employed for the preparation of CaZnOS-based quaternary ML materials with lanthanide/transition metal dopants and variable compositions. Notably, this method only takes a total synthesis duration of 20-minute microwave radiation, which can greatly facilitate the efficient preparation of a variety of ML materials. Moreover, bright host emission from the CaZnOS matrix is unexpectedly induced, providing a new opportunity for manipulating multicolor luminescence. Finally, we have proposed that the people’s writing characteristics can be analyzed by using these ML materials, which offers a great potential for pressure visualization, individualized E-signature anti-counterfeiting applications and beyond. This work not only provides a facile one-step synthesis method for the preparation of CaZnOS-based quaternary ML materials that featuring unique host emission, but also suggests the ML materials to be applied in advanced E-signature and on-demand information identification applications, which is of significance for integrated optoelectronic devices in future.

He Li,Jun-Mian Tian,Hao-Yu Tang,Shi-Yin Pan,An-Ling Zhang,Jin-Ming Gao

Fifteen aromatic polyketide metabolites, including four new chromones, chaetosemins B–E (2–5), with compound 4 bearing a new skeleton, and two new natural products, chaetosemin A (1) and (+)-(S)-chaetoquadrin J (14), together with nine known ones (6–13, 15), were isolated from the organic extract of a solid fermented culture of an Ascomycete filamentous fungus, Chaetomium seminudum. The structures of 1–5 were determined by spectroscopic analysis, and the absolute configuration of chaetosemin A (1) was confirmed by X-ray crystallography, whereas the stereochemistry of 1–4 and 14 was assigned by comparison of their specific optical rotations with reported data for structurally related compounds. All compounds were isolated for the first time from Chaetomium seminudum. Compounds 1 and 2 consisted of L-cysteine and D-cysteine units, respectively. Most of them were evaluated for in vitro antifungal activities against some phytopathogenic fungi. Among them, chaetosemin B (2) was the most active against Magnaporthe oryzae and Gibberella saubinettii with MICs of 6.25 and 12.5 μM, respectively. In addition, chaetosemin C (3) showed antioxidant activity with 50.7% DPPH free radical scavenging activity at 50 μM, and chaetoquadrin J (14) displayed weak inhibition of soluble epoxide hydrolase (sEH).

A synthetic system for RNA-responsive pyroptosis based on type III-E CRISPR nuclease-protease

He Mingbin,Wang Weiwei,Zhou Haiwu,Liu Cong,Zhao Chunbei,Li Jian,Han Yuewen,Qin Yali,Chen Mingzhou

Pyroptosis plays a crucial role in immune defense against infections and endogenous threats by eliminating harmful cells and modulating the immune response through inflammation. However, the natural activation of pyroptosis involves intricate signaling pathways, posing significant challenges for its artificial manipulation in research and therapies. Here, we present DAMAGE ( D e a th Ma nipulation Ge ne), an innovative system that integrates gasdermins within the type III-E CRISPR framework, enabling the specific recognition of target RNA (tgRNA) and triggering pyroptosis. This mechanism allows DAMAGE to selectively identify and eliminate virus-infected, cancerous, and senescent cells, all of which exhibit altered RNA transcriptomes. Additionally, DAMAGE exhibits considerable promise as a platform for mRNA-LNP therapy. Our study highlights the potential of this CRISPR-based system in the controllable induction of pyroptosis, offering an innovative therapeutic strategy for treating RNA-heterogeneous diseases.

Changes in vitamin C, vitamin E, and carotenoid metabolism during early kernel development in maize

Nan Xiang,Xinbo Guo

BACKGROUND Maize ( Zea mays L.), as the most widely cultivated cereal and a dietary cornerstone, provides not only carbohydrates but also essential micronutrients critical for human health, including vitamin C, vitamin E, and carotenoids. However, conventional maize varieties typically exhibit suboptimal levels of these compounds. This study investigated the dynamic accumulation of vitamin C, vitamin E, and carotenoids, along with their transcriptional regulation, during early kernel development (9–15 days after pollination (DAP)) in maize. RESULTS Combined metabolite and transcriptome analyses showed a 23-fold increase in vitamin C content, associated with enhanced expression of the gene VTC2 . Id1 and AGAMOUS-like MADS-box transcription factors (TFs) are putative regulators, targeting the promoters of GalDH and VTC2 to regulate vitamin C biosynthesis. Vitamin E showed isomer-specific fluctuations during early kernel development, mainly associated with the expression of VTE1 , VTE2 , and VTE4 , and potentially regulated by knotted 1 and the AGAMOUS-like MADS-box TFs via VTE2 and VTE3 , respectively. Total carotenoid content tripled, with zeaxanthin becoming dominant by 15 DAP, driven by upregulated PSY1 and CHYB-1 . ERF003 was identified as a candidate regulator of PSY3 . CONCLUSION The increases in vitamin C and carotenoid content, and the fluctuation of vitamin E isomers during early maize kernel development, were associated with the modulation of key genes and putative TFs. These findings reveal theoretical insights and provide candidate genetic targets for future breeding efforts aimed at enhancing micronutrient levels in maize. © 2026 Society of Chemical Industry.

An amide-linked pyrene-based imine COF for high-performance gold recovery from e-waste

Hao Chen,Donghua Yang,Yunhao Hou,Aiguo Feng,Xiyong Chen,Kai Chen,Xinpeng Wang

This report presents the first application of an amide linkage strategy in pyrene-based imine COFs. • Amide groups enhance the polarity and coordination ability of TFPPY-DA-COF channels. • The TFPPY-DA-COF achieved an Au adsorption capacity of 2040 mg g −1 . Pyrene-based covalent organic frameworks (COFs) have received considerable attention for gold recovery, yet challenges remain in terms of adsorption capacity and economic cost. Herein, we initially employed an amide linkage strategy to construct a pyrene-based imine COF for gold recovery. The introduction of amide groups enhanced the polarity and coordination ability of the pore walls, enabling amide-linked COF to exhibit an Au uptake of 2040 mg g −1 , which is approximately twice that of its non-amide counterpart, and superior to most previously reported pyrene-based COFs. Moreover amide-linked COF also demonstrates excellent selectivity (distribution coefficient for Au exceeding 2.74 × 10 5 ; maximum separation factor > 6000) and outstanding performance in practical applications. Notably, the ligands used to synthesize both pyrene-based COFs are low-cost, and the captured Au particles can reach micrometer-scale sizes, which facilitates their separation and recovery. Further performance tests, combined with characterization and DFT calculations, reveal that the coordination of Au ions by the carbonyl groups in the amide linkages plays a dominant role during adsorption, and the reduction effect of the protonated imine bonds also contributes to improving the adsorption performance. Overall, this work provides a new approach for using COF materials to selectively capture and recover gold, offering potential for practical applications. Chitosan Nanoparticles Loaded with Truncated ORF2 Protein as an Oral Vaccine Candidate against Hepatitis E

Wenjuan Wei,Nouredine Behloul,Weiqi Wang,Sarra Baha,Zhenzhen Liu,Ruihua Shi,Jihong Meng

In a continuous effort to develop effective vaccines against hepatitis E (HE), oral vaccine nanoparticles using the truncated capsid protein p146 (aa460–605) are formulated and characterized. To improve the immunogenicity of p146, chitosan nanoparticles (CSNPs) are used as a mucosal delivery system. Next, the physical-chemical properties, cytotoxic effects in vitro, and immunogenicity in mice of the produced NPs are analyzed. The results show that the produced CS/p146 NPs are stable and well dispersive and display a near-spherical shape with a mean size of 200–300 nm. The findings also demonstrate high encapsulation efficiency (65–73.9%) and loading capacity (27.7–67.5%) of the formulated nanoparticles. Further, the CS/p146 NPs exhibit low cytotoxicity and an obvious sustained-release effect in vitro. Immunogenicity experiments in mice indicate that CS/p146 NPs can induce antigen-specific systemic and mucosal immune responses higher than the purified p146 do. Besides, the expression levels and mRNA transcription of Interleukin (IL)-4 in spleen cells of CS/p146 NPs-immunized mice are higher than those of p146, indicating that a Th2-mediated cellular immune response is activated by the CS/p146 NPs. Overall, the synthesized CS/p146 NPs display promising properties as a potential HE oral vaccine candidate.

Endothelial cell-derived MMP19 promotes pulmonary fibrosis by inducing E(nd)MT and monocyte infiltration

Zhao Weiming,Wang Lan,Yang Juntang,Chen Xinyu,Guo Xiaoshu,Xu Kai,Wang Ningdan,Zhao Wenyu,Xia Cong,Lian Hui,Rosas Ivan,Yu Guoying

Background Matrix metalloproteinases (MMPs) play important roles in remodeling the extracellular matrix and in the pathogenesis of idiopathic pulmonary fibrosis (IPF). MMP19, which is an MMP, was significantly upregulated in hyperplastic alveolar epithelial cells in IPF lung tissues and promoted epithelial-mesenchymal transition (EMT). Recent studies have demonstrated that endothelial-to-mesenchymal transition (E(nd)MT) contributes to pulmonary fibrosis. However, the role of MMP19 in pulmonary vascular injury and repair and E(nd)MT remains unclear. Methods To determine the role of MMP19 in E(nd)MT and pulmonary fibrosis. MMP19 expressions were determined in the lung endothelial cells of IPF patients and bleomycin (BLM)-induced mice. The roles of MMP19 in E(nd)MT and endothelial barrier permeability were studied in the MMP19 cDNA-transfected primary human pulmonary microvascular endothelial cells (HPMECs) and MMP19 adenoassociated virus (MMP19-AAV)-infected mice. The regulatory mechanism of MMP19 in pulmonary fibrosis was elucidated by blocking its interacting proteins SDF1 and ET1 with AMD3100 and Bosentan, respectively. Results In this study, we found that MMP19 expression was significantly increased in the lung endothelial cells of IPF patients and BLM-induced mice compared to the control groups. MMP19 promoted E(nd)MT and the migration and permeability of HPMECs in vitro, stimulated monocyte infiltration into the alveolus, and aggravated BLM-induced pulmonary fibrosis in vivo. SDF1 and Endothelin-1 (ET1) were physically associated with MMP19 in HPMECs and colocalized with MMP19 in endothelial cells in IPF patient lung tissues. AMD3100 and bosentan alleviated the fibrosis induced by MMP19 in the BLM mouse model. Conclusion MMP19 promoted E(nd)MT by interacting with ET1 and stimulated monocyte infiltration into lung tissues via the SDF1/CXCR4 axis, thus aggravating BLM-induced pulmonary fibrosis. Vascular integrity regulated by MMP19 could be a promising therapeutic target for suppressing pulmonary fibrosis. Video abstract
