Abstract Butterflies use visual and olfactory cues to identify conspecifics during courtship. Tirumala limniace , Idea leuconoe , and Danaus chrysippus are three species of butterflies with a sympatric distribution, and T. limniace butterflies are rarely seen to chase I. leuconoe or D. chrysippus during courtship in artificial cultivation gardens, indicating that they have a mechanism for recognizing conspecifics; however, there are no reports on what kind of signals they use to recognize conspecifics. In this study, we investigated the cues used by T. limniace to identify conspecifics and the relative roles of visual and olfactory cues. We found that in the absence of olfactory cues (visual cues only), male and female butterflies were able to identify the same species based on wing color and pattern. By collecting and identifying body volatiles emitted from T. limniace males and females, we found that the highest amounts were β -ocimene and β -pinene, which were sourced from the insects’ bodies (as opposed to sex pheromones). Furthermore, electroantennogram binding behavioral experiments showed that β -ocimene was involved in the process of species recognition and that the relative roles of olfactory cues were greater than those of visual cues at close range. Therefore, T. limniace males and females could utilize both visual and olfactory cues to identify conspecifics during courtship but preferred olfactory cues. The present study highlighted the important role of adult body odor rather than sex pheromones in butterfly species recognition. Significance statement Butterflies use visual (wing color and pattern) and olfactory (sex pheromones) cues to identify conspecifics during courtship. Butterflies also release volatiles emitted from their bodies during courtship. However, there is no clear evidence that body volatiles (as opposed to sex pheromones) emitted from butterflies are involved in species recognition. We studied the functions of body volatiles emitted from Tirumala limniace adults and the relative roles of visual and olfactory cues in species recognition during courtship. We found that male and female adults could identify conspecifics using only visual cues. Meanwhile, we also found that the highest amounts of β -ocimene, which came from the bodies of T. limniace males and females, were also involved in the process of species recognition. The olfactory cues that butterflies use to identify conspecifics are usually sex pheromones, while the present study highlighted the important role of adult body odor rather than sex pheromones in butterfly species recognition.

Overcoming chemoresistance will prevent cancer relapse and contribute to clinical chemotherapy. In order to explore the underlying mechanism of chemoresistance, we firstly incubated cancer cells with a combination of cisplatin + paclitaxel (C + P) or cisplatin + paclitaxel + docetaxel (C + P + D) to mimic the treatment of cancer therapy in the laboratory. We found that polyploidy is a recurring strategy that cells adopt in response to cisplatin-based treatments. RNA-sequencing was performed to identify differentially expressed genes (DEGs) that may contribute to drug resistance. 4830 and 5518 DEGs were discovered in C + P and C + P + D resistant cells, respectively, and 4384 (73.40%) genes were shared. Possible drug resistance genes like Atg14, Abcb1b, Tbx2, Slc2a9, Slc10a3 and Slc22a18 were up-regulated while Foxm1, Bcl2, Brca1, Chek1, Hiatl1 and Abcb9 were down regulated. Genes involved in the pathways of p53 signaling, lysosomes and apoptosis were up-regulated, and in contrast, genes in the cell cycle, DNA replication, and mismatch repair pathways were down-regulated. Moreover, representative proteins relative to DEGs were examined to validate the results of RNA-seq and RT-PCR. Taken together, these results will contribute to revealing the mechanism of chemoresistance and discovering potential prognostic factors for cancer medication.

Organic anion-transporting polypeptides (OATPs) 1B1 and 1B3 are two highly homologous transport proteins. However, OATP1B1- and 1B3-mediated estradiol-17β-glucuronide (E17βG) uptake can be differentially affected by clotrimazole. In this study, by functional characterization on chimeric transporters and single mutants, we find that G45 in transmembrane domain 1 (TM1) and V386 in TM8 are critical for the activation of OATP1B3-mediated E17βG uptake by clotrimazole. However, the effect of clotrimazole on the function of OATP1B3 is substrate-dependent as clotrimazole does not stimulate OATP1B3-mediated uptake of 4′,5′-dibromofluorescein (DBF) and rosuvastatin. In addition, clotrimazole is not transported by OATP1B3, but it can efficiently permeate the plasma membrane due to its lipophilic properties. Homology modeling and molecular docking indicate that E17βG binds in a substrate binding pocket of OATP1B3 through hydrogen bonding and hydrophobic interactions, among which its sterol scaffold forms hydrophobic contacts with V386. In addition, a flexible glycine residue at position 45 is essential for the activation of OATP1B3. Finally, clotrimazole is predicted to bind at an allosteric site, which mainly consists of hydrophobic residues located at the cytoplasmic halves of TMs 4, 5, 10, and 11.

Fire hazards are a primary concern in aircraft accidents, and controlling combustible materials is pivotal in managing cabin fire risks. This work aims to elucidate the impact of widespread measures that introduce phosphorus-based flame retardants (PFRs) on the fire safety of carbon fiber epoxy composites (CF/EP). Firstly, the hazardous indicators for flame retardant CF/EP composites exhibit two categories of optimization and degradation, which are caused by the intrinsic feature, pyrolysis characteristics and chemical valence states of PFRs. Secondly, the fire hazards of composites are assessed by fusing the analytic network process and information weights guided by Nash Equilibrium theory. The method combines the merits of subjective and objective weights from dimensions of intrinsic hazards, interrelationships and data dispersion of indicators. Finally, the assessed fire hazard rankings under 35 and 65 kW/m 2 both are EP > CF/EP16%DOPO-HQ > CF/EP > CF/EP7%DPO. In conclusion, DOPO-HQ (+1) primarily increases the fire hazard of CF/EP due to gas-phase flame-retardant effects, while DPO (+3) predominantly reduces the fire hazard of CF/EP by contributing to condensed-phase effects. This work highlights the contradictory effects of PFRs on the fire safety of CF/EP composites due to structure and thermal stability. The proposed ANP and IW hybridization method provides a theoretical foundation for selecting flame retardants and assessing material fire hazards comprehensively.

An immortalized mouse fetal liver stromal cell line, named KM3, has demonstrated the potential to support the growth and maintenance of human embryonic stem cells (hESCs). In this study, the characteristics of KM3 cells were examined following cryopreservation at -70˚C and in liquid nitrogen for 15, 30 and 60 days following treatment with 10 µg/ml mitomycin C. In addition, whether the KM3 cells were suitable for use as feeder cells to support the growth of hESCs was evaluated. The inhibition of mitosis without cell death was observed when the KM3 cells were treated with 10 µg/ml mitomycin C for 2 h. The morphology of the KM3 cells cryopreserved in liquid nitrogen for 60 days was not markedly changed, and the cell survival rate was 84.60±1.14%. By contrast, the survival rate of the KM3 cells was 66.40±2.88% following cryopreservation at -70˚C for 60 days; the cells readily detached, were maintained for a shorter time, and had a reduced expression level of basic fibroblast growth factor. hESCs cultured on KM3 cells cryopreserved in liquid nitrogen for 60 days showed the typical bird's nest structure, with clear boundaries and a differentiation rate of 16.33±2.08%. The differentiation rate of hESCs cultured on KM3 cells cryopreserved at -70˚C for 60 days was 37.67±3.51%. These results indicate that the cryopreserved KM3 cells treated with mitomycin C may be directly used in the subculture of hESCs, and the effect is relatively good with -70˚C short-term or liquid nitrogen cryopreservation.

The toxicity of engineered nanoparticles remains a concern. The knowledge of biohazards associated with particular nanoparticles is crucial to make this cutting-edge technology more beneficial and safe. Here, we evaluated the toxicity of Ga 2 O 3 nanoparticles (NPs), which are frequently used to enhance the performance of metal catalysts in a variety of catalytic reactions. The potential inflammatory signaling associated with the toxicity of HA/β-Ga 2 O 3 :Cr 3+ NPs in primary cortical neurons was examined. We observed a dose-dependent decrease in cell viability and an increase in apoptosis in neurons following various concentrations (0, 1, 5, 25, 50, 100 µg/ml) of HA/β-Ga 2 O 3 :Cr 3+ NPs treatment. Consistently, constitutively active forms of calcineurin (48 kDa) were significantly elevated in cultured primary cortical neurons, which was consistent with calpain activation indicated by the breakdown products of spectrin. Moreover, HA/β-Ga 2 O 3 :Cr 3+ NPs result in the elevation of LC3-II formation, SQSTM/p62, and Cathepsin B, whereas phosphorylation of CaMKII (Thr286) and Synapsin I (Ser603) were downregulated in the same context. Taken together, these results demonstrate for the first time that calpain activation and a disturbance of autophagy signaling are evoked by exposure to HA/β-Ga 2 O 3 :Cr 3+ NPs, which may contribute to neuronal injury in vitro .

In recent work, deep eutectic solvents (DESs) as ionic liquid analogues have been abundantly used in catalysis. Herein, vanadium phosphorus oxide (VPO) catalysts were synthesized from mono-, bi-, and tri- metallic DES of Nb, Zr, and Mo metal dopants as structure-directing agents and electronic promoters for n-butane selective oxidation towards maleic anhydride. Higher MA selectivity and larger n-butane conversion was successfully obtained using the newly developed catalysts, while oxidation by-product COx(CO, CO2) was minimized. Characterization techniques including FTIR, DSC, XRD, TEM, SEM, EDS, Raman spectroscopy, TGA, XPS, and NH3-TPD were employed to fully characterize the DESs, precursors and catalysts. This work led to an increase of 7.8% in MA mass yield with 16% more n-butane conversion as compared to an unpromoted VPO catalyst. Moreover, the utilization of a low-carbon alkane brought in a green impact on the chemical plant as well as the environment. Keywords:deep eutectic solvent;vanadium phosphorus oxide;n-butane selective oxidation;structure directing agent;electronic promoter

This study was aimed to investigate the protective effects of three different mulberry fruit polysaccharide fractions (MFP-I, MFP-II, and MFP-III) against palmitic acid (PA)-induced hepatocyte lipotoxicity and characterize the functional polysaccharide fraction using gel permeation chromatography, high-performance liquid chromatography, Fourier transform infrared spectroscopy, and nuclear magnetic resonance analyses. MFP-I, MFP-II, and MFP-III were isolated from mulberry fruit by stepwise precipitation with 30, 60, and 90% ethanol, respectively. MFP-II at 0.1 and 0.2 mg/mL dramatically attenuated PA-induced hepatic lipotoxicity, while MFP-I and MFP-III showed weak protection. It was demonstrated that MFP-II not only increased nuclear factor erythroid-2-related factor 2 (Nrf2) phosphorylation and its nuclear translocation, thereby activating the Nrf2/ARE signaling pathway, but also enhanced heme oxygenase 1, NAD(P)H:quinone oxidoreductase 1, and γ-glutamate cysteine ligase gene expressions and promoted catalase and glutathione peroxidase activities, which protected hepatocytes against PA-induced oxidative stress and lipotoxicity. Further investigation indicated that the molecular weight of MFP-II was 115.0 kDa, and MFP-II mainly consisted of galactose (30.5%), arabinose (26.2%), and rhamnose (23.1%). Overall, our research might provide in-depth insight into mulberry fruit polysaccharide in ameliorating lipid metabolic disorders.

Ginsenoside Rb1 (Rb1), as the major bioactive ingredient of Panax ginseng C.A. Meyer, elicited a novel antidepressant-like effect in the forced swim test (FST) in chronic unpredictable mild stress (CUMS) rats in our previous study. To further explore the molecular mechanism of Rb1 on the neurotransmitters such as 5-hydroxytryptamine (5-HT), 5-hydroxyindole-3-acetic acid (5-HIAA), norepinephrine (NE), dopamine (DA), homovanillic acid (HVA), dihydroxyphenylacetic acid (DOPAC), glutamate (Glu) and gamma-aminobutyric acid (GABA) in this antidepressant-like effect, the neurochemical changes in the monoaminergic and aminoacidergic receptors were investigated in the present pharmacological study by using reuptake inhibitors, receptors agonists and antagonists. The results showed that a sub-effective dose of Rb1 (5 mg/kg, p.o.) co-administered with fluoxetine (1 mg/kg, i.p., a selective serotonin reuptake inhibitor), reboxetine (2.5 mg/kg, i.p., a noradrenalin reuptake inhibitor), bupropion (10 mg/kg, i.p., a dopaminergic reuptake inhibitor), Mk-801 (0.05 mg/kg, i.p., an N-methyl- d -aspartic acid (NMDA) receptor antagonist) or baclofen (0.1 mg/kg, i.p., a selective GABA agonist) significantly decreased the immobility time in the FST. In addition, pretreating mice with NAN190 (0.5 mg/kg, i.p., a 5-HT 1A receptor antagonist), ketanserin (5 mg/kg, i.p., a 5-HT 2A/2C receptor antagonist), ondansetron (1 mg/kg, i.p., a 5-HT 3A receptor antagonist), prazosin (1 mg/kg, i.p., an α 1 -adrenoceptor antagonist), yohimbine (1 mg/kg, i.p., an α 2 -adrenoceptor antagonist), SCH23390 (0.05 mg/kg, i.p., a selective D 1 receptor antagonist), haloperidol (0.2 mg/kg, i.p., a non-selective D 2 receptor antagonist), NMDA (75 mg/kg, i.p., an agonist at the glutamate site) or bicuculline (4 mg/kg, i.p., a competitive GABA antagonist) reversed the antidepressant-like effect of Rb1 (10 mg/kg, p.o.) in the FST. The results obtained for the neurotransmitters in the mouse hippocampus (CA3) and prefrontal cortex showed that Rb1 up-regulated the levels of 5-HT, 5-HIAA, NE, DA, and GABA and decreased the level of Glu. However, there were no significant differences in HVA or DOPAC. Furthermore, there were no significant alterations in the total path of spontaneous locomotor activity in all treatments. These results suggest that both monoaminergic (serotonergic, noradrenergic and dopaminergic) and aminoacidergic (glutamatergic and GABAergic) receptors may be involved in the antidepressant-like effect of Rb1.

The kainic acid (KA)-induced epilepsy experimental model is widely used to study the mechanisms underlying this disorder. Recently, the blood-brain barrier (BBB) has become an innovative alternative treatment target for epilepsy patients. KA causes neuronal injury and BBB damage in this experimental epilepsy model but the mechanisms underlying epilepsy-related neuronal injury, autophagy, and BBB damage remain unclear. Therefore, the present study investigated the relationships among neuronal injury, the expressions of autophagy-related proteins, and changes in BBB-related proteins during the acute phase of epilepsy to further understand the mechanisms and pharmacotherapy of epilepsy. NeuN immunohistochemistry and Fluoro-Jade B (FJ-B) staining in the hippocampal CA3 region revealed that neuronal death induced by intraventricular injections of 10 μg/kg KA was greater than that induced by 3 μg/kg KA. In addition, there were transient increases in the levels of microtubule-associated protein light chain 3-II (LC3I/II) and Beclin-1, which are autophagy-related proteins involved in neuronal death, in this region 24 h after the administration of 10 μg/kg KA. There were also morphological changes in BBB-related cells such as astrocytes, endothelial cells (ECs), and tight junctions (TJs). More specifically, there was a significant increase in the activation of astrocytes 72 h after the administration of 10 μg/kg KA as well as continuous increases in the expressions of platelet endothelial cell adhesion molecule-1 (PECAM-1) and BBB-related TJ proteins (Zonula occludens-1 and Claudin-5) until 72 h after KA treatment. These results suggest that the overexpression of autophagy-related proteins and astrocytes and transient increases in the expressions of BBB-related TJ proteins may be closely related to autophagic neuronal injury. These findings provide a basis for the identification of novel therapeutic targets for patients with epilepsy.

Optimized formulations produce RGB structural colors and dual visible‑UV encryption signals from fluorescent PDLC. • Wavelength-selective Bragg reflection enables multi-stage decryption for high-security anti-counterfeiting. • A ternary PSCLC/PDLC/PCL composite is prepared via two‑step polymerization to achieve tunable cholesteric helical pitch. Liquid crystals (LC) are regarded as promising candidates for optical encryption on account of their distinctive stimulus-responsive structural coloration. However, single-component polymer-stabilized cholesteric liquid crystal (PSCLC) systems suffer from limited information encoding dimensions. Polymer-dispersed liquid crystals (PDLC) can only achieve basic light scattering modulation and cannot meet the requirements of complex multi-channel encryption. To circumvent these drawbacks, a synergistic PSCLC/PDLC/polycaprolactone (PCL) composite thin film is fabricated for optical anti-counterfeiting. Laser-patterned PDLC templates are filled with PSCLC precursors, and S5011-loaded PCL thin films laminated on independent glass substrates serve as sustainable chiral donors. Regulated interlayer chiral diffusion enables continuous tuning of structural hues within predefined laser-etched cavities. Incorporation of red fluorescent powder imparts dual visible and ultraviolet imaging capacity to the composite. Tunable color variation in designated regions is realized via precise modulation of diffusion temperature and duration. Benefiting from the wavelength-selective Bragg reflection effect of cholesteric liquid crystals (CLCs), multi-layered information encryption is accomplished. Colored patterned regions can only be visualized under matching monochromatic illumination. Benefiting from facile manufacturing, large information storage capacity and diversified encryption modes, the proposed composite presents great application potential in premium anti-counterfeiting tags, encrypted two-dimensional codes and intelligent display devices. Background Radiation-induced lung injury (RILI) is a common side effect of thoracic tumor radiotherapy, including early-stage radiation-induced lung injury (RP) and late-stage radiation-induced pulmonary fibrosis (RIPF). Currently, it is urgently needed to clarify the pathogenesis of RILI and find safe and effective RILI treatment methods. Irradiation causes DNA damage and oxidative stress in tissues and cells, induces cellular senescence, and promotes the occurrence and development of RILI. In recent years, Anisodamine (654-2) has shown potential therapeutic value in acute lung injury, acute kidney injury, chlamydial pneumonia, and COVID-19. However, there is currently no research on the mechanism of 654-2-mediated cellular senescence and its preventive and therapeutic effects on RILI. Purpose This study aimed to investigate the protective effect and mechanism of 654-2 on X-ray-induced RILI. Methods In vivo experiments involved a mouse RILI model with 18 Gy X-ray irradiation. Mice were divided into control, model, medication (control + 654-2), and treatment (model + 654-2) groups. And mice in medication and treatment groups were intraperitoneal injection of 5 mg/kg 654-2 every other day until being sacrificed at week 6. In vitro experiments used MLE-12 cells irradiated with 16 Gy and divided into control, model, and model + 654-2(2 μM and 10 μM) groups. Various assays were performed to evaluate lung tissue morphology, fibrosis, apoptosis, cytokine expression, cellular senescence, protein expression, and antioxidant capacity. Results 654-2 mitigated pulmonary pathological damage, inflammation, DNA damage, cellular senescence, and apoptosis in RILI mice and MLE-12 cells. It restored epithelial cell proliferation ability and enhanced antioxidant capacity. Additionally, 654-2 activated the Nrf2/ARE pathway, increased Nrf2 phosphorylation, and upregulated antioxidant gene expression. Inhibition of Nrf2 reversed the effects of 654-2 on ROS production, antioxidant capacity, and cell senescence. Conclusion 654-2 can activate the Nrf2/ARE pathway, enhance cellular antioxidant capacity, and inhibit cellular senescence, thereby exerting a protective effect against RILI.
