An effective signal amplifying strategy for copper (II) sensing by using in situ fluorescent proteins as energy donor of FRET

Qianchen Zhang,Duoduo Zhao,Chengwu Zhang,Jinhua Liu,Zhongfu An,Xiaofei Qin,Yongqian Gao,Shiyu Zhang,Lin Li,Wei Huang

Fluorescence resonance energy transfer (FRET) is a reliable, sensitive, and robust assay method for detection of many biological targets. However, it generally needs an externally-introduced label to form the donor-acceptor pair, which could alter the accuracy of the detection. To address this issue, we report herein the FRET-based reactive copper ion sensors by using in situ fluorescent proteins (FP) of human urine or blood serum as the energy donor. Using Bull Serum Albumin (BSA) as model proteins, the sensor exhibits a remarkably fluorescence enhancement when BSA binds to the surface of copper clusters (Cu NCs) base on electrostatic interaction. Conversely, low fluorescence enhancement is observed without using BSA. As proof-of-principle, this positive approach is directly applied to detect Cu 2+ using urine as the energy donor, accompanying with a signal enhancement by two factors and low detection limit of 0.5 μM Cu 2+ . Moreover, the proposed sensor could be applied in other complex environments, such as blood serum or cell culture medium. Consequently, the effectiveness, simplicity and diversity of our proposed strategy enable the development of a class of probes toward complex human environment for rapid, sensitive, and selective detection of targets.

FRET-based colorimetric and ratiometric sensor for visualizing pH change and application for bioimaging in living cells, bacteria and zebrafish

Xiaojun He,Feng Ding,Wei Xu,Chuchu Xu,Yahui Li,Yuna Qian,Shengzhe Zhao,Hong Chen,Jianliang Shen

Acid-alkaline balance plays a crucial role in all biological processes . Accordingly, monitoring pH changes will help us to understand the functional status of these physiological and pathological processes. Though fluorescent probes may be a useful tool for detecting pH changes, and there are many limitations to currently available probes, such as background interference, potential cytotoxicity, and poor cell permeability , which call for a solution urgently. In this work, a rhodamine-derived colorimetric and ratiometric sensor ( Rh–HN ) was fabricated for monitoring pH change via the mechanism of fluorescence resonance energy transfer (FRET). Rh–HN has been shown to possess several advantages over other probes, such as high sensitivity, outstanding permeability, and low toxicity. Besides, the fluorescence intensity ratio (F 526 /F 592 ) of Rh–HN displays a pH-sensitive response from 2.0 to 7.5 (p K a = 5.05) and linear response from pH 3.8 to 6.4, which was desirable for mapping pH change in the biological systems. Besides, the results indicated that Rh–HN generated a pH-dependent response regulated by switchable forms between closed and opened spirolactam ring. Overall, Rh–HN has accomplished sensing and mapping of pH in living cells, bacteria, and zebrafish. Those results demonstrated that the great potential of Rh–HN in sensing and visualizing pH in the living biosystem.

A FRET-based fluorescent probe for hydrogen peroxide based on the use of carbon quantum dots conjugated to gold nanoclusters

Zhao Qi,Zhou Chaohui,Yang Qingye,Chu Zhongyun,Jia Nengqin

In the Fenton reaction, ferrous ion acts as a catalyst and reacts with hydrogen peroxide (H 2 O 2 ) to produce hydroxy radicals (·OH) and hydroperoxy radicals (·OOH). Both have much stronger oxidization ability than H 2 O 2 . A fluorescent probe for H 2 O 2 is described here that was obtained by covalent conjugation of carbon quantum dots to gold nanoclusters (AuNCs). The conjugate, under 360 nm photoexcitation, displays dual (blue and red) emission, with peaks located at 450 and 640 nm. When introducing ·OH radicals via the Fenton reaction, the fluorescence intensities of both the CQDs and the AuNCs are decreased. The ratio of the fluorescence at the two peaks is related to the concentration of H 2 O 2 in the 1.25 nM to 10 μM concentration range, and the detection limit is 0.16 nM. The probe was applied to the determination of H 2 O 2 in milk and toothpaste and to cell imaging. Schematic diagram of the FRET-based fluorescent probe and enhanced performance of hydrogen peroxide detection via Fenton reaction. The fluorescence intensity of CQD-AuNCs nanoaster was decreased as introducing H 2 O 2 to the probe, and can be applied to the determination of milk and toothpaste and cells imaging.

Photoswitchable fluorescent polymeric nanoparticles for rewritable fluorescence patterning and intracellular dual-color imaging with AIE-based fluorogens as FRET donors

Weibang Zhong,Xuyao Zeng,Jian Chen,Yongxiang Hong,Lehui Xiao,Peisheng Zhang

Photoswitchable fluorescent polymer nanoparticles (PFPNs) were constructed from an aggregation-induced emission (AIE) based fluorogen and a photochromic spiropyran-linked amphiphilic copolymer via a simple co-precipitation method. Possessing the reversible photoisomerization property and high fluorescence resonance energy transfer (FRET) efficiency, not only did the designed spiropyran-linked amphiphilic copolymer provide the PFPNs with excellent long-term stability (∼5 weeks) in aqueous media, but also it could both efficiently and reversibly switch on/off the fluorescence emission of the AIE-based fluorogen upon irradiation with UV and visible light. It is worth noting that the high contrast dual-color characteristics written with PFPNs as rewritable fluorescence patterning can be tuned by altering the wavelength of the irradiation light. Moreover, the resultant PFPNs were successfully applied for intracellular dual-color imaging due to their outstanding cell-membrane permeability and rapid photo-responsiveness, as well as excellent repeatability properties.

An optical FRET inhibition sensor for serum ferritin based on Mn2+-doped NaYF4:Yb,Tm NIR luminescence up-conversion nanoparticles

ShaoZhen Wang,Jianguo Zhang,Hongqi Chen,Lun Wang

A new serum ferritin (SF) biosensor based on fluorescence resonance energy transfer (FRET) from Mn 2+ -doped NaYF 4 :Yb,Tm up-converting rare-earth nanophosphors (UCNPs, donor) to gold nanorods (GNRs, acceptor) was developed in the near-infrared (NIR) region. The as-prepared Mn 2+ -doped NaYF 4 :Yb,Tm UCNPs exhibited a strong emission at 807   nm in the NIR region and were modified with poly-(acrylic acid) (PAA) before conjugating with anti-SF (Ab 1 ). The negatively charged donor (Ab 1 -UCNPs) and the positively charged acceptor (GNRs) were mixed in close proximity due to electrostatic interactions, which quenched the fluorescence emission of the Mn 2+ -doped NaYF 4 :Yb,Tm UCNPs. However, added SF combines with the anti-SF because the binding affinity between SF and Ab 1 is stronger than the electrostatic interactions. Thus, the acceptor separated from the donor, and the fluorescence was restored. Under optimal conditions, the restored fluorescence exhibited a linear relationship with SF concentrations ranging from 0.23 to 16.88   ng/mL and a limit of detection as low as 0.12   ng/mL. The proposed method was also used to monitor SF in human serum samples.

Aggregation and Binding-Directed FRET Modulation of Conjugated Polymer Materials for Selective and Point-of-Care Monitoring of Serum Albumins

Sameer Hussain,Xi Chen,Chaofeng Wang,Yi Hao,Xuemeng Tian,Yulian He,Jing Li,M. Shahid,Parameswar Krishnan Iyer,Ruixia Gao

Nonspecific interactions of conjugated polymers (CPs) with various proteins prove to be a major impediment for researchers when designing a suitable CP-based probe for the amplified and selective recognition of particular proteins in complex body fluids. Herein, a new strategy is presented for the precise and specific monitoring of clinically important serum albumin (SA) proteins at the nanomolar level using fluorescence resonance energy transfer (FRET)-modulated CP–surfactant ensembles as superior sensing materials. In brief, the newly designed color-tunable CP PF–DBT–Im undergoes intense aggregation with the surfactant sodium dodecyl sulfate (SDS), enabling drastic change in the emission color from violet to deep red due to intermolecular FRET. The emission of PF–DBT–Im/SDS ensembles then changed from deep red to magenta specifically on addition of SAs owing to the exclusive reverse FRET facilitated by synergistic effects of electrostatic interactions, hydrophobic forces, and the comparatively high intrinsic quantum yield of SAs. Interestingly, PF–DBT–Im itself could not differentiate SAs from other proteins, demonstrating the superiority of the PF–DBT–Im/SDS self-assembly over PF–DBT–Im. Finally, an affordable smartphone-integrated point-of-care (PoC) device is also fabricated as a proof-of-concept for the on-site and rapid monitoring of SAs, validating the potential of the system in long-term clinical applications.

Construction of mitochondria targeted and FRET based ratiometric sensing nanoplatform for sulfur dioxide accurate detection in vitro and in vivo

Wen Liu,Jinyao Li,Zicheng Wang,Yafei Tian,Guodong Ren,Xiaoyu Hou,Lixia Guo,Lihong Li,Chengwu Zhang,Zhifang Wu,Lili Yan,Sijin Li,Haipeng Diao

Sulfur dioxide (SO 2 ) is a key molecule in organisms that is involved in the regulation of different physiological procedures. Aberrant SO 2 causes a variety of diseases, such as cancer and neurodegeneration. Thus, sensitive and selective detection of SO 2 is of great importance. Based on the Förster resonance energy transfer (FRET) between green fluorescence carbon dots (GCDs) donor and amide-linked near-infrared fluorescence emissive organic small molecular dye (CDDBT) acceptor, one ratiometric fluorescent nano platform, Mito-GCDs-CDDBT for mitochondria SO 2 sensing was constructed. In this FRET sensing system, CDDBT served as the receptor for SO 2 , and the presence of SO 2 enhanced GCDs green fluorescence signal and quenched CDDBT near-infrared fluorescence signal due to the disruption of FRET. Mito-GCDs-CDDBT could sensitively detect SO 2 with a detection limit of as low as 0.701 μM. Meanwhile, Mito-GCDs-CDDBT achieved fluorescence imaging to measure the response of cellular exogenous and endogenous SO 2 with remarkable mitochondrial targeting. Moreover, Mito-GCDs-CDDBT also realized SO 2 sensing in vivo including zebrafish and mice. The as-prepared versatile nanoplatform displayed several advantages, such as mitochondria targeting, FRET-based sensitive detection, and sensing capabilities in biological milieu. Potentially, it could be applied in the diagnostics of SO 2 involved diseases.

Europium Doped Silicon Quantum Dot As a Novel FRET Based Dual Detection Probe: Sensitive Detection of Tetracycline, Zinc, and Cadmium

Jialing Zhou,Ruoxi Zhao,Shikai Liu,lili Feng,Wenting Li,Fei He,Shili Gai,Piaoping Yang

The imbalance of Zn 2+ /Cd 2+ in the human body can lead to many serious diseases due to the overuse of antibiotics and deposition in animal products. Developing a functional material for detecting is challenging and in demand. Herein, silicon quantum dots (SiQDs) are designed as a functional platform for the detection of tetracycline and Zn 2+ /Cd 2+ . The  COOH functionalized SiQDs with the emission wavelength of 450 nm are chelated with Eu(NO 3 ) 3 to form SiQDs-Eu 3+ ratio fluorescent probes, which can be used to detect tetracycline (TCs) and Zn 2+ /Cd 2+ by fluorescence resonance energy transfer (FRET) principle sequentially. The fluorescent probe showed good linearity between ion concentration and fluorescence enhancement. The detection limit of TCs and Zn 2+ /Cd 2+ are 0.2 × 10 −6 m and 3 × 10 −6 m , respectively, when the pH of the solution is 7.4. In addition, the synthesized SiQDs-Eu 3+ exhibited good stability (from 94.9% to 103.1%). The relative standard deviations (RSD, n = 10) of human serum and urine were both less than 3%. Therefore, the SiQDs-Eu 3+ ratio fluorescence probe will provide a good application prospect in actual sample detection.

Attomolar-level detection of respiratory virus long-chain oligonucleotides based on FRET biosensor with upconversion nanoparticles and Au–Au dimer

Yingjin Ma,Menglin Song,Lihua Li,Xinyue Lao,Yuan Liu,Man-chung Wong,Mo Yang,Honglin Chen,Jianhua Hao

Upconversion nanoparticles (UCNPs) are promising nanoprobes in DNA/RNA detection, such as respiratory viral RNAs, and siRNA in cancer. However, recent studies have indicated that the sensitivity of UCNP-based biosensors is restricted, ranging from picomolar to femtomolar level. Moreover, most of existing UCNP-based probes are only able to detect short-chain oligonucleotides, which are not suitable for detection of long-chain oligonucleotides in many real applications. In this work, we introduced a new UCNP-based fluorescence resonance energy transfer (FRET) nanoprobe design composed of NaGdF 4 :Yb 3+ , Er 3+ @NaGdF 4 core-shell UCNPs (csUCNPs) linking with Au–Au dimer to detect long-chain oligonucleotides of SARS-CoV-2 N-gene. Compared with typical single gold nanoparticles (AuNPs) in FRET biosensors, our theoretical investigation shows that a stronger electromagnetic field is generated in the Au–Au dimer where the plasmon resonance can enhance FRET efficiency and increase the working distance. Thus, the synergetic effect of plasmonic resonance and FRET enables a greater quenching efficiency (QE) of Au–Au dimer to UCNPs, which leads to more remarkable upconversion luminescence (UCL) recovery for each target gene recognition. Importantly, our design significantly improved the limit of detection (LOD) to attomolar level, with a linear response ranging from 2 aM to 2 fM. Moreover, the clinical detection with inactivated SARS-CoV-2 samples was successfully performed with excellent specificity within 30 min using the developed UCNPs biosensors incorporated with Au–Au dimer. This UCNP biosensor based on Au–Au dimer strategy with ultra-sensitivity and good selectivity opens a new path for clinical diagnosis without target amplification and plays an instructive role in other virus diagnosis.

‌Discrimination of Triphenylmethane Drug in Aquatic Products Using Fluorescence Sensor Array Based on Rare-Earth Complexes‌

Chen Ling,Shao Keman,Xu Jiajia,Wang Xiwen,Huang Zhenwei,Lin Zhengzhong

Four rare-earth complexes were prepared using Eu³⁺ and Tb³⁺ as central rare-earth ions and benzoic acid, furoic acid, α-thenoyltrifluoroacetone, and phenanthroline as organic ligands, and a four-channel fluorescent sensor array was constructed for the identification of triphenylmethane drugs. The four sensing units have different degrees of fluorescence response to six triphenylmethane drugs. The fluorescence quenching of Eu(III) complex by malachite green and Tb(III) complex by methyl violet were confirmed to be the FRET effect. After five parallel experiments, a 4 × 6 × 5 fluorescence data matrix was obtained, and the linear discriminant method were used to distinguish six triphenylmethane drugs at 0.1, 1.0, and 10 µmol L⁻¹ and mixed triphenylmethane drugs at 1.0 µmol L⁻¹ with an accuracy rate of 100%. The array achieved the identification of triphenylmethane drugs in fish samples with accuracy rate of 97.4%.

Artificial light-harvesting cellulosic fabrics: Programmable dye assemblies for on-demand heating, antibacterial, and luminescent applications

Fu Wang,Jiahui Wu,Qinuo Han,Liu Hu,Wo Nie

Light-harvesting systems play a pivotal role in the development of sustainable antimicrobial and self-heating artificial materials. Fabric-based artificial solar light-harvesting systems were constructed by designing the assemblies of a zinc(II) phthalocyanine photosensitizer (CAZnPc) and three different fluorescent dyes on the cotton fabric. The dye-mediated Förster resonance energy transfer (FRET) process and π–π stacking endow the modified fabric with exceptional antibacterial, photothermal, and luminescent performances. Under visible light irradiation (100 mW/cm 2 ), CF-ZP/Rh6G (the optimal heating performer) achieves a temperature of 64.5 °C (ΔT = 43.7 °C) within 100 s with an outstanding photothermal stability (<10 % efficiency decay after 5 cycles). CF-ZP/RhB exhibits a superior 1 O 2 generation capability (7.64-fold higher than that of CF-ZnPc). The synergistic photothermal–photodynamic effect enables CF-ZP/Rh6G to attain a 99.9999 % antibacterial efficacy against Staphylococcus aureus and Escherichia coli after 90 min of irradiation (30 mW/cm 2 ). Mechanistic study reveals that the performance variations observed for these fabrics arise from the different fluorescent dye molecular structures, causing different intermolecular forces, which modulate energy allocation pathways of photosensitizer to govern functionality. The assembly construction strategy offers a material platform for sunlight-powered wearable thermal management and medical protective applications.

Ultrasensitive detection of Listeria monocytogenes using a ratiometric fluorescence aptasensor in food products

Feifei Wang,Haoyi Wu,Jinru Zhou,Huan Li,Shenjia Chen,Yuxiang Ye,Jinlei Gong

Listeria monocytogenes (LM) is a foodborne pathogen known for its potential to cause serious infections, posing a considerable risk to food safety and endangering public health as well as the broader social economy. Here, we developed a novel switch-conversional ratiometric fluorescence aptasensor enabling ultrasensitive detection of LM by utilizing the Förster resonance energy transfer (FRET) phenomenon mediated by amplicon fragments, which were generated from the exponential amplification reaction (EXPAR). This biosensor integrates EXPAR for signal enhancement with aptamer recognition, facilitating ultra-sensitive detection of LM. Under optimized parameters, this biosensor reaches an impressive detection limit of 0.56 × 10° CFU/mL, along with a wide linear interval extending from 1.0 × 10 1 to 1.0 × 10 6 CFU/mL. Moreover, the biosensor demonstrates the capability to detect LM in spiked real food samples, achieving recovery rates that range from 87.1% to 101.9%, thereby underscoring its reliability and accuracy. Overall, this research demonstrates significant promise for the advancement of highly sensitive detection systems for foodborne pathogens.
