The development of novel Janus nanostructure with desired compositions and functionalities is essential for various applications, especially as drug delivery system to deliver multiple payloads with total different solubility. Here, a unidentified flying object (UFO)-like cyclodextrin–palladium nanosheet/zeolitic imidazolate framework-8 Janus nanoparticle (CD–PdNS/ZIF-8 JNP) prepared via a selective coating of polyacrylic acid (PAA) on one of the flat surfaces of 2D PdNS followed by further formation of ZIF-8 on a PAA template, and alternative modification of CDs on exposed PdNS surface is reported. The CD-PdNS and PAA-ZIF-8 parts are designed to load hydrophobic10-hydroxycamptothecin (HCPT) and hydrophilic doxorubicin (Dox) independently for synergistic chemotherapy (combination index = 0.65). Because of the broad near-infrared (NIR) absorption band of PdNSs, the photothermal effect of UFO-like CD-PdNS/ZIF-8 JNPs in NIR-II biowindow is studied. The in vitro and in vivo results show that the dual-drug loaded JNPs under a laser-irradiation group present highest cancer therapeutic efficiency. The good performance is due to the synergistic chemotherapy and photothermal therapy in NIR-II biowindow. This work significantly broadens the application prospects of metal–metal organic framework JNPs by rationally designing their compositions and discovering related physiochemical properties.

Extensive efforts have been devoted to the design of organic photothermal agents (PTAs) that absorb in the second near-infrared (NIR-II) bio-window, which can provide deeper tissue penetration that is significant for phototheranostics of lethal brain tumors. Herein, the first example of NIR-II-absorbing small organic molecule (N1) derived from perylene monoamide (PMI) and its bio-application after nano-encapsulation of N1 to function as a nano-agent for phototheranostics of deep orthotopic glioblastoma (GBM) is reported. By adopting a dual modification strategy of introducing a donor-acceptor unit and extending π-conjugation, the obtained N1 can absorb in 1000–1400 nm region and exhibit high photothermal conversation due to the apparent intramolecular charge transfer (ICT). A choline analogue, 2-methacryloyloxyethyl phosphorylcholine, capable of interacting specifically with receptors on the surface of the blood-brain barrier (BBB), is used to fabricate the amphiphilic copolymer for the nano-encapsulation of N1. The obtained nanoparticles demonstrate efficient BBB-crossing due to the receptor-mediated transcytosis as well as the small nanoparticle size of approximately 26 nm. The prepared nanoparticles exhibit excellent photoacoustic imaging and significant growth inhibition of deep orthotopic GBM. The current study demonstrates the enormous potential of PMI-based NIR-II PTAs and provides an efficient phototheranostic paradigm for deep orthotopic GBM.

Rationale: Cerebrovascular diseases, together with malignancies, still pose a huge threat to human health nowadays. With the advantages of its high spatial resolution and large penetration depth, fluorescence bioimaging in the second near-infrared spectral region (NIR-II, 900-1700 nm) and its related imaging-guided therapy based on biocompatible fluorescence dyes have become a promising theranostics method. Methods: The biocompatibility of IR-820 we used in NIR-II fluorescence bioimaging was verified by long-term observation. The model of the mouse with a cranial window, the mouse model of middle cerebral artery occlusion (MCAO) and a subcutaneous xenograft mouse model of bladder tumor were established. NIR-II fluorescence cerebrovascular functional imaging was carried out by IR-820 assisted NIR-II fluorescence microscopy. Bladder tumor was treated by NIR-II fluorescence imaging-guided photothermal therapy. Results: We have found that IR-820 has considerable NIR-II fluorescence intensity, and shows increased brightness in serum than in water. Herein, we achieved real time and in vivo cerebrovascular functional imaging of mice with high spatial resolution and large penetration depth, based on IR-820 assisted NIR-II fluorescence microscopy. In addition, IR-820 was successfully employed for NIR-II fluorescence imaging and photothermal therapy of tumor in vivo , and the subcutaneous tumors were inhibited obviously or eradicated completely. Conclusion: Due to the considerable fluorescence intensity in NIR-II spectral region and the good photothermal effect, biocompatible and excretable IR-820 holds great potentials for functional angiography and cancer theranostics in clinical practice.

Near-infrared-II (NIR-II) fluorescence imaging-guided photothermal therapy (PTT) represents a cutting-edge approach for precise tumor diagnosis and treatment, providing real-time therapeutic efficacy evaluation. However, a significant challenge lies in the creation of phototheranostic agents that provide robust imaging and efficient photothermal conversion. To address this, a donor- π -acceptor (D- π -A) structure heptamethine cyanine derivative, IR1116, that confers a strong intramolecular charge transfer effect is developed. To enhance its applicability, IR1116 is formulated with DSPE-PEG to create a water-dispersible NIR-II phototheranostic nanoheater, NP IR1116 . This nanoheater benefits from the incorporation of an electron-withdrawing group, leading to reduced photooxidation activity and significantly improved photostability. NP IR1116 exhibits strong NIR-II absorption and fluorescence, peaking at 1004 and 1116 nm, respectively, as well as a photothermal conversion efficiency of 79% under 1064 nm lasers. In vitro and in vivo studies showed the efficacy of NP IR1116 in tumor imaging via NIR-II fluorescence and its ability to effectively induce tumor cell apoptosis under 1064 nm laser irradiation. These findings underscore the potential of NP IR1116 in NIR-II fluorescence imaging-guided PTT for tumor treatment, paving the way for further advancements in NIR-II dye development and bioimaging technologies.

The integration of chemodynamic therapy (CDT) and photothermal therapy (PTT) has played a huge role in improved anticancer treatments. Here, a novel multifunctional nanoplatform based on Cu2−xS conjugated NaYF4:Yb/Er@NaYF4:Yb upconversion nanoparticles (UCNPs) was proposed and designed. In the UCNPs–Cu2−xS nanocomposites, UCNPs with excellent luminescent properties and a high X-ray attenuation coefficient can serve as an upconversion luminescence (UCL) and computer tomography (CT) imaging contrast agent; meanwhile, Cu(II) in the Cu2−xS nanodots enables the nanocomposites to have a magnetic resonance imaging (MRI) ability owing to the presence of unpaired electrons. Moreover, the Cu2−xS nanodots with a strong absorbance in the NIR II biowindow not only could be employed as a stable photothermal agent under NIR laser irradiation, but also could be used as a photothermal-enhanced Fenton nanocatalyst to respond to over-expressed H2O2 in the tumor microenvironment (TME) and generate toxic hydroxyl radicals (˙OH) to effectively kill cancer cells. Furthermore, the UCNPs–Cu2−xS nanocomposites possess negligible cytotoxicity and a high photothermal conversion efficiency (43.8%) in the NIR-II biowindow (1064 nm), indicating that they possess great potential for the UCL/CT/MR multi-modal imaging guided synergistic enhanced CDT/PTT of cancer.

Thermal quenching of luminescence materials poses a major obstacle to the technological application of luminescence thermometry. It still remains challenging to attain thermally enhanced light emissions, especially in the second near-infrared window (NIR-II). Herein, an anomalous thermal dependence of NIR-II luminescence in the negative thermal expansion (NTE) Sc 2 Mo 3 O 12 :Er 3+ /Ho 3+ nanocrystals is reported. Mechanistic investigations affirm that Ho 3+ ion can work as an energy reservoir and back-transfer to Er 3+ ion with the assistance of lattice phonon at elevated temperatures. Moreover, the Ho 3+ -mediated energy feedback is strengthened by the thermal contraction between dopant ions, thereby enabling a remarkable thermal enhancement of NIR-II emission over 11-fold. The opposite thermal response of Er 3+ and Ho 3+ emissions is harnessed for NIR-II ratiometric thermometry, registering exceptional performance in a high-temperature regime ( S r = 1.71% K −1 , δT = 0.2 K at 513 K). These findings may inspire new insights for addressing the thermal quenching of NIR-II luminescence, which also raises exciting opportunities for flexible thermometry in complex settings.

Near-infrared (NIR) phosphor-converted light-emitting diodes (pc-LEDs) are desired for optoelectronic and biomedical applications, while the development of target broadband NIR phosphors still remains a significant challenge. Herein, a kind of Cr 3+ -doped AlNbO 4 phosphors with a broad NIR emission ranging from 650 to 1400 nm under 450 nm excitation are reported. A giant red-shift emission peak from 866 to 1020 nm together with broadened full width at half-maximum of 320 nm is achieved simply by varying the doped Cr 3+ concentrations. Structural and spectroscopy analysis demonstrate that a concentration-dependent site-occupation of Cr 3+ emitters in different Al 3+ sites is responsible for the tunable NIR luminescence. The as-fabricated NIR pc-LED based on optimized AlNbO 4 :Cr 3+ phosphor exhibits great potential in night-vision applications. This work provides a novel design principle on the Cr 3+ -doped AlNbO 4 phosphor with tunable broadband luminescence from NIR-I to NIR-II, and these materials can be employed in NIR spectroscopy applications.

Small organic phototherapeutic molecules of the second near-infrared (NIR-II) window (1000–1700 nm) serve as promising candidates for theranostics. However, developing such versatile agents for fluorescence-guided photodynamic/photothermal therapy remains a demanding task stirred by competitive energy dissipation pathways, including radiative decay, internal conversion, and intersystem crossing. To the best of current knowledge, the current paradigm for addressing the issue has deliberately approached the optimum balance among three deactivation processes through offsetting from each other, possibly leading to a comprehensively compromised theranostic efficacy. Few reports aim to modulate the three deactivation pathways excluding sacrificing any one of them. Herein, a molecular design strategy to construct a phototherapeutic organic fluorophore CCNU-1060 , armed with NIR-II luorescence-guided phototherapeutic properties, is rationally developed. With a flexible motor, tetraphenylethene, bridged to the rigidified coplanar core boron-azadipyrromethene, the desired CCNU-1060 is subsequently encapsulated into an amphiphilic matrix to form CCNU-1060 nanoparticles (NPs), which match or transcend its precursor NJ-1060 NPs in the three energy dissipation processes. CCNU-1060 NPs are utilized to realize high-spatial vessel imaging and effective NIR-II fluorescence-guided phototherapeutic tumor ablation. This study unlocks a viewpoint of molecular engineering that simultaneously regulates multiple energy dissipation pathways for the construction of versatile phototherapy agents.

Pyroptosis resulted from photodynamic therapy (PDT) plays a crucial role in triggering photoimmunotherapy (PIT). However, visible light-triggered PDT may rise phototoxicity, with low penetration into biological tissues. Crucially, the efficacy of PDT in eliciting pyroptosis remains suboptimal. To address these formidable challenges, we developed a O 2 -supplementing photosensitizer AuNCs@TAT that could efficiently induce pyroptosis upon irradiation with second near-infrared (NIR-II) light in hypoxic tumor microenvironment (TME). Replacing GSH with TAT resulted in a gold nanocluster AuNCs@TAT. The ratio of Au (I) to Au (0) shifted from 1: 0.26 in pristine AuNCs to 1: 1.21 in AuNCs@TAT, which contributed to enhanced catalytic activity of AuNCs@TAT. The superior catalase-like activity of AuNCs@TAT produced 1.9-fold higher O 2 than AuNCs. This O 2 self-supplying capability of AuNCs@TAT not only relieved hypoxic condition in TME but also significantly enhanced the efficiency of AuNCs@TAT to induce pyroptosis upon 1064 nm light irradiation. Western blotting indicated both remarkable upregulation of cleft Caspase-3 and GSDME-N, suggesting GSDME mediated pyroptosis. Further, release of pro-inflammatory cytokines and damage-associated molecular patterns (DAMPs) triggered a strong systematic immune response and reprogrammed the immunosuppressive microenvironment into an immunostimulatory state, which led to a significant suppress of tumor metastasis in mice. Overall, this work sets the stage for future advancements in the design of NIR-II photosensitizer with O 2 -supplementing capability and therefore offers an innovative framework and solid foundation for tumor PIT.

Recurrence of solid tumors after surgical resection is a major barrier to tissue regeneration. As an emerging treatment strategy, photo-thermo-electric therapy ablates tumor cells via photothermal effects and generates reactive oxygen species (ROS) via thermoelectric effects to disrupt heat shock proteins, thereby suppressing their protective function in tumor cells. However, conventional materials suffer from low thermoelectric efficiency and weak tissue penetration ability. In this study, we fabricated iodine-doped bismuth sulfide (I-Bi 2 S 3 ) nanorods with bonding heterostructures to improve thermoelectric performance. The approach employed iodine doping to introduce additional electrons, thereby regulating the band structure of Bi 2 S 3 and exploiting the dual low-energy vibration effect of the heterostructures to reduce thermal conductivity. More importantly, controlling the type of heterostructure modulated the bandgap width, thereby expanding the light absorption range to the higher-penetration near-infrared (NIR)-II region for deep tissue treatment. The I-Bi 2 S 3 nanorods were incorporated into poly- l -lactic acid (PLLA) scaffolds to confer antitumor functionality. According to the results, the bonding heterostructures enhanced the conductivity of Bi 2 S 3 and reduced its thermal conductivity, significantly enhancing thermoelectric efficacy. The heterostructures reduced the bandgap of Bi 2 S 3 from 1.23 to 0.88 eV, enabling optical absorption in the NIR-II region. The ROS tests showed that the PLLA/I-Bi 2 S 3 scaffold exhibited good photothermal effects and ROS generation under 1064-nm laser irradiation. The antitumor efficacy of the PLLA/I-Bi 2 S 3 scaffold reached 84.6% against MG-63 cells, demonstrating its exceptional potential in cancer treatment.

Luminescent thermometry in the second near-infrared (NIR-II) window holds significant promise for non-invasive. However, its practical application is severely hindered by thermal quenching at elevated temperatures, which limits the development of highly sensitive NIR-II thermometers capable of thermally enhanced emission. Here, we present a rationally designed integer phonon-matching strategy that enables NIR thermal enhancement specifically in singly Er 3+ -doped Ca 2 YNbO 6 double perovskite hosts. Through combined experimental investigations and rate equation modeling, we reveal that efficient multi-phonon relaxation (MPR) between the Er 3+ : 4 I 11/2 and 4 I 13/2 levels is activated when the electronic energy gap corresponds to an integer multiple ( n ≤ 5) of the dominant phonon energy. This resonant phonon-matching condition facilitates highly efficient MPR with increasing temperature, leading to a pronounced 1.66-fold enhancement in the Er 3+ NIR-II emission. Furthermore, we demonstrate the simultaneous occurrence of NIR thermal enhancement and ratiometric temperature sensing in Ca 2 YNbO 6 : Yb 3+ /Er 3+ /Ho 3+ /Pr 3+ crystals, achieving an exceptional relative sensitivity of 0.87% K −1 at 398 K. These findings not only circumvent the thermal quenching limitations of NIR-II phosphors but also establish phonon engineering as a versatile strategy for designing thermally enhanced luminescence, opening new avenues for flexible monitoring of pipeline integrity and organic compound leakage based on differential NIR-II luminescence absorption.

Purpose Cervical cancer (CC) is still the fourth most common cause of cancer deaths in women. However, current biomedical imaging techniques exhibit inherent limitations in the diagnosis and treatment of CC. This study aims to develop a biomimetic nanoplatform based on tumor cell membranes, loaded with a palladium (Pd)-based computed tomography (CT) contrast agent and the near-infrared (NIR) fluorescent probe indocyanine green (ICG). This multifunctional nanoplatform is designed to integrate multimodal imaging with photothermal therapy (PTT), thereby improving the diagnostic accuracy and therapeutic efficacy against CC.Methods In this study, biomimetic nanoparticles (NPs), designated as M@Pd-ICG NPs, were synthesized by encapsulating Pd and ICG within HeLa cell membranes derived from cell-derived xenograft (CDX) models. Subsequently, the toxicity, biocompatibility, and tumor suppression capability of the M@Pd-ICG NPs were evaluated in vitro. In vivo, the multimodal imaging performance of the M@Pd-ICG NPs and their photothermal therapeutic efficacy under 808-nm laser irradiation were investigated in mouse model bearing subcutaneous cervical tumor. Results The M@Pd-ICG NPs were successfully prepared and exhibited favorable stability, excellent photothermal conversion efficiency (34.04%), and good biocompatibility, enabling homologous targeting and prolonged circulation time. The M@Pd-ICG NPs integrated the complementary advantages of NIR-II fluorescence imaging (900–1700 nm, NIR-II FI), photothermal imaging (PTI), and CT imaging. Both in vitro and in vivo studies demonstrated that, under 808-nm laser irradiation, M@Pd-ICG NPs induced significant photothermal effects and tumor ablation. Conclusion M@Pd-ICG NPs successfully integrate multimodal imaging and PTT, owing to their excellent targeting capability and good biocompatibility, demonstrating potential for further biomedical applications.
