3D-printed BaTiO3-modified PCL/β-TCP scaffolds with multimodal optimization for enhanced critical-size bone regeneration

Liu Xuanhao,Zhang Jing,Xu Biying,Xu Yang,Wang Yuxin,Huang Zhi,Liu Bin,Fuxiang Song

Multimodal optimization improved scaffold structure, mechanics, and bioactivity. • Low-dose BT generated a detectable but mild local piezoelectric response. • PTB4 achieved the best overall balance of printability and biological performance. • PTB4 enhanced osteogenic differentiation and VEGF-related endothelial responses. • PTB4 promoted osteogenesis–angiogenesis coupling and calvarial bone regeneration. Critical-size bone defects (CSDs) remain a major clinical challenge. Although three-dimensional (3D) printing enables precise structural control and defect-specific scaffold fabrication, conventional polymer-based scaffolds often exhibit insufficient mechanical strength and limited osteogenic activity. In this study, poly(ε-caprolactone)/β-tricalcium phosphate (PCL/β-TCP) scaffolds containing 0, 1, 2, 4, or 8 wt% barium titanate (BaTiO 3 , BT) were fabricated by direct ink writing. Their rheology, microstructure, mechanical properties, local electromechanical response, cytocompatibility, angiogenesis-related activity, osteogenic differentiation, and bone regeneration in vivo were evaluated. All formulations exhibited suitable printability and regular porous structures. Moderate BT incorporation increased surface roughness and mechanical strength, with compressive strength remaining within the range of human cancellous bone. Piezoresponse force microscopy confirmed a detectable but mild local piezoelectric response. However, its direct osteogenic contribution was not isolated from other BT-related material effects and requires further verification. All scaffolds exhibited good cytocompatibility. PTB4 showed the most favorable biological performance, including enhanced MC3T3-E1 cell proliferation, increased ALP-positive area, greater mineralized deposition, and more pronounced VEGF-related fluorescence in HUVECs. After 12 weeks in a rat calvarial critical-size defect model, PTB4 exhibited significantly higher bone volume fraction and bone mineral density than the blank and PT groups. Histological and immunohistochemical analyses showed more extensive new bone formation, collagen deposition, and stronger osteogenesis–angiogenesis coupling. Overall, PTB4 achieved the best balance among printability, mechanical compatibility, cytocompatibility, angiogenesis-related activity, and osteogenic performance, supporting low-dose BT modification as a promising strategy for safe, printable scaffolds for critical-size craniofacial and oral bone defect repair. ROS-Responsive mPEG–PCL Nanoparticles for Co-Delivery of Paclitaxel Prodrug and R848 in Synergistic Chemo-Immunotherapy

Qi Zhang,Luyao Wang,Zhaofan Yang,Hao Meng,Shixian Lv,Linlin Liu

Chemo-immunotherapy aims to integrate chemotherapy-induced antigen release with innate immune activation to achieve enhanced antitumor efficacy. However, the pharmacokinetic mismatch and systemic toxicity of free small molecules often compromise the synergistic effects. Herein, we propose a reactive oxygen species (ROS)-responsive nanoplatform based on mPEG–PCL for the co-delivery of a paclitaxel (PTX) prodrug and the TLR7/8 agonist R848. The nanoparticles remain stable under physiological conditions while releasing drug in oxidative tumor microenvironments, thereby achieving synchronous intratumoral delivery and activation. In vitro, the system exhibited efficient intracellular uptake and potent cytotoxicity against tumor cells while maintaining low toxicity toward normal cells due to prodrug design. Moreover, the platform effectively induced immunogenic cell death and promoted dendritic cell maturation, highlighting its capacity for chemo-immunotherapeutic synergy. This co-delivery strategy provides a versatile approach to realize controllable and safe chemo-immunotherapy through stimuli-responsive nanomedicine design.

Synthesis of Poly(methyl/amino)silsesquioxane Microsphere as an Efficient Compatibilizer for Enhanced PHB/PCL Blend Properties

Haiying Chen,Cheng Chen,Min Zhou,Xiang Wu,Tao Wang,Lingjun Wei

To enhance the compatibility of polyhydroxybutyrate/polycaprolactone (PHB/PCL) binary blend system, a novel covalent compatibilizer, poly(methyl/amino)silsesquioxane (PMASQ) was synthesized via the base-catalyzed sol-gel method using Methyltrimethoxysilane (MTMS) and γ-Aminopropyl trimethoxysilane (APTMS). The films were prepared through the solution casting method with the blend of PHB/PCL (25:75, w/w) incorporating with various content of PMASQ (1   wt%, 3   wt%, 5   wt%). As a result, ATR-FTIR analysis confirmed the successful synthesis of PMASQ and the subsequent grafting of PHB and PCL segments onto the PMASQ. At 3   wt% PMASQ, the blend exhibited optimal mechanical enhancement, with tensile strength and elongation at break increasing by 1.23-fold and 6.17-fold, respectively, compared to uncompatibilized blends. Furthermore, the incorporation of PMASQ resulted in a reduction in crystallinity while enhancing thermal stability within the PHB/PCL blend system. An improvement in continuity was also observed in the PHB/PCL/PMASQ blends, characterized by a reduction in the size of the dispersed phase and an emergence of a homogeneous morphology. These results demonstrate PMASQ’s dual role as a reactive compatibilizer and thermal stabilizer, mitigating inherent brittleness and phase incompatibility in PHB/PCL systems. The improved physicochemical properties and processability of the blend highlight PMASQ’s potential to expand the utility of biodegradable PHB/PCL composites in packaging, biomedical, and environmentally sustainable industrial applications.

A Biocompatible, Eco-Friendly 3D-Printed PCL-NIPUA Resin for Macular Bucking Devices in Myopic Tractional Maculopathy

Zi Fu,Yan Zhou,Ziyi Liu,Zhe Chen,Qiang Fu,Shiqun Lin,Rongping Dai,Huade Zheng

Pathological myopia, characterized by excessive axial elongation, posterior staphyloma (PS) and retinal-choroidal degeneration underlies sight-threatening conditions such as myopic traction maculopathy (MTM) and macular atrophy. Macular buckling (MB) surgery has demonstrated significant efficacy in managing MTM-associated pathologies, including macular schisis, macular holes, and macular retinal detachment. However, its widespread clinical adoption remains limited due to suboptimal material properties and geometrical design constraints of existing buckling devices. Advancements in imaging modalities-such as spectral-domain optical coherence tomography (SD-OCT), swept-source OCT (SS-OCT), ultrawidefield fundus imaging, and three-dimensional magnetic resonance imaging (3D-MRI)-have substantially enhanced the understanding and evaluation of MTM biomechanics. The future development of MB may lie in identifying superior biomaterials and creating customized buckling devices tailored to individual ocular morphology. Here, we developed a photo-curable polycaprolactone (PCL)-based non-isocyanate polyurethane acrylate (NIPUA) resin (PCL-NIPUA) for one-piece J-shaped 3D-printed MB devices. Structural characterization via Nuclear Magnetic Resonance (NMR), Fourier Transform Infrared Spectroscopy (FTIR), and Mass Spectrometry (MS) confirmed successful synthesis. Photocuring kinetics under 465 nm visible light achieved an optimal storage modulus within 82 s, coupled with minimal curing shrinkage (7.2%). By tuning flexible PCL segments, the material exhibited a tensile strength of 25 MPa and elongation at break of 32%, balancing strength and elasticity. The resin demonstrated environmental stability (water contact angle: 72°; water absorption: 8.8%; thermal decomposition: 420°C) and exceptional biocompatibility (hemolysis rate <0.5%; cell viability >97%). Liquid crystal display (LCD) 3D printing produced cost-effective MB devices that provided stable scleral indentation in rabbit models, with few complications, and no pathological structural changes in the optic nerve and retina, showing good biocompatibility and safety. This work establishes PCL-NIPUA as a synergistic platform combining green chemistry, rapid photocuring, mechanical adaptability, and biosafety, offering a clinically translatable solution for customized 3D-printed implants in MB surgery.

Enhanced corrosion resistance and photothermal antibacterial properties of MAO/PCL-PDA/NG composite coating on Mg alloy

Yang Liu,Run Liu,Wenxian Nie,Leiting Yu,Shaoyuan Lyu,Qi Han,Yun Zhao,Minfang Chen

In this study, a composite coating of polycaprolactone layer and polydopamine-cyclodextrin (β-CD)/naringenin (NG) was prepared on the surface of a micro-arc oxidized (MAO) pretreated magnesium alloy by a two-step method. The findings demonstrate that the composite coating exhibits favorable corrosion resistance, biocompatibility, photo-thermal antimicrobial, and bone-enhancing characteristics. Compared with the MAO coating, the corrosion rate of the composite coating was significantly reduced from about 0.087 mm/y to 0.066 mm/y. The antimicrobial efficiency of the coating against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) under 808 nm laser irradiation was found to reach 93 % and 99 %, respectively. Furthermore, in vitro cytotoxicity assays, dead and live cell staining, and immunoblotting analysis showed that the coating exhibited favorable biocompatibility and osteogenic activity. This novel coating may have good application prospects in the treatment of osteoporosis-related fractures.

Fabrication of PCL/keratin composite scaffolds for vascular tissue engineering with catalytic generation of nitric oxide potential

Pengfei Li,Yanfang Wang,Xingxing Jin,Jie Dou,Xiao Han,Xiuzhen Wan,Jiang Yuan,Jian Shen

Tissue-engineered vascular grafts (TEVGs) have been proposed as a promising approach to fulfill the need for small-diameter blood vessel substitutes. However, common failure caused by thrombosis and neointimal proliferation after implantation has restricted their use in the clinic. Herein, a NO-generating scaffold for vascular tissue engineering was developed by coelectrospinning poly(ε-caprolactone) (PCL) with keratin. The morphology and surface chemical composition were characterized via SEM, ATR-FTIR spectroscopy and XPS. The biocomposite scaffold selectively enhanced the adhesion and growth of endothelial cells (ECs) while suppressing the proliferation of smooth muscle cells (SMCs) in the presence of GSH and GSNO due to the catalytic generation of NO. In addition, these mats displayed excellent blood compatibility by prolonging the blood-clotting time. In summary, these NO-generating PCL/keratin scaffolds have potential applications in vascular tissue engineering with rapid endothelialization and reduced SMC proliferation.

Ditelluride-Bridged PEG-PCL Copolymer as Folic Acid-Targeted and Redox-Responsive Nanoparticles for Enhanced Cancer Therapy

Pang Zekun,Zhou Jiayan,Sun Chunyang

The development of the nanosized delivery systems with targeting navigation and efficient cargo release for cancer therapy has attracted great attention in recent years. Herein, a folic acid (FA) modified PEGylated polycaprolactone containing ditelluride linkage was synthesized through a facile coupling reaction. The hydrophobic doxorubicin (DOX) can be encapsulated into the polymeric micelles, and such nanoparticles (F-TeNPDOX) exhibited redox-responsive drug release under abundant glutathione (GSH) condition due to the degradation of ditelluride bonds. In addition, flow cytometric analyses showed that the FA ligands on F-TeNPDOX could facilitate their cellular uptake in 4T1 breast cancer cells. Therefore, F-TeNPDOX led to the promoted drug accumulation and enhanced growth inhibition on 4T1 tumor in vivo. The obtained results suggest F-TeNPDOX excellent potential as nanocarriers for anticancer drug delivery.

Control over the Gradient Differentiation of Rat BMSCs on a PCL Membrane with Surface-Immobilized Alendronate Gradient

Yang Zhu,Zhengwei Mao,Changyou Gao

Gradient biomaterials can offer progressively changing signals to specific tissue interface, and thereby modulate the conjunction between different tissues. A linear density gradient of alendronate (Aln), a molecule that is capable of promoting osteogenic differentiation of bone mesenchymal stem cells (BMSCs), was created on an aminolyzed poly(ε-caprolactone) (PCL) membrane. X-ray photoelectron spectroscopy and quartz crystal microbalance with dissipation revealed the linear increase of the Aln amount as a function of the position on the PCL membrane. By contrast, the surface wettability and energy were kept unchanged. The surface-grafted Aln showed a stronger ability to induce the osteogenic differentiation of rat BMSCs than its counterpart in culture medium of the same amount, and the osteo-inductive culture medium. On the Aln-grafted gradient surface, the BMSCs showed gradient osteogenic differentiation as a function of membrane position in terms of cell morphology, alkaline phosphatase activity, calcium deposition, and the expression of osteogenesis marker proteins including collagen type I (COL I), Runt-related transcription factor 2 (Runx2), and osteocalcin (OCN).

Ca ions chelation, collagen I incorporation and 3D bionic PLGA/PCL electrospun architecture to enhance osteogenic differentiation

Xuefeng Zhou,Xi Cheng,Danlei Xing,Qi Ge,Yan Li,Xianghong Luan,Ning Gu,Yunzhu Qian

Bioactive synthetic scaffolds with 3D porous structure are the most attractive materials among various guided bone regeneration membranes. However, its osteogenic potential is still insufficient. This study was aimed to develop a Calcium surface-anchored Collagen I-PLGA/PCL scaffolds (PP/COL I-pDA-Ca) with enhanced osteogenicity. PP/COL I-pDA-Ca was electrospun from a collagen I-blended PLGA/PCL matrix and then modified by the chelation of Ca-ions via mussel-inspired polydopamine coating. PLGA/PCL, PLGA/PCL-polydopamine, PLGA/PCL-pDA chelated by Ca-ions were used as controls. Osteogenic effects of the scaffolds were examined using MC3T3-E1 cell culture. PP/COL I-pDA-Ca maintained 3D porous architecture with interconnected pores formed by randomly-oriented filamentous fibers and MC3T3-E1 cells cultured on it for 12 h or 24 h were more stretched and spread than those on the controls. PP/COL I-pDA-Ca significantly upregulated α10, α11 and β1 integrin expression after 48 h culture. ALP activity, OCN, OSX, BMP2 and RUNX2 expression of MC3T3-E1 cells cultured on PP/COL I-pDA-Ca scaffolds for 7 and 14 days were substantially enhanced when compared to controls ( p  < 0.05). The pDA-based Ca chelation, COL I incorporation and 3D bionic structure promoted cell adhesion and osteogenic differentiation of MC3T3-E1 cells. This novel PP/COL I-pDA-Ca scaffolds is an attractive alternative for guided bone regeneration.

Curcumin-Metal Complex-Loaded PLGA/PCL Electrospun Scaffolds: Enhanced Antioxidation, Osteogenesis and Antibacterial Activity for Bone Regeneration

Qian Yunzhu,Pan Xujie,Zhou Xuefeng,Ma Buyun,Liu Zhaolin,Cai Zhiyan

Bone regeneration in compromised environments, such as infected or inflammatory defects, remains a significant clinical challenge. This study addresses the limitations of curcumin (Cur), including poor solubility, low bioavailability, and limited antibacterial activity, by developing a series of curcumin-metal complexes through chelation reactions. These complexes were encapsulated into nanoliposomes and subsequently incorporated into poly(lactic-co-glycolic acid)/polycaprolactone (PLGA/PCL) electrospun scaffolds to construct multifunctional bone regeneration platforms. The fabricated scaffolds exhibited uniform nanofibrous morphology, enhanced mechanical properties, and improved surface hydrophilicity, which collectively promoted cell adhesion and proliferation. Among the complexes, Cur-Cu demonstrated the highest antioxidant activity and superior osteogenic performance, significantly enhancing alkaline phosphatase (ALP) activity and upregulating integrin β1/α1 gene expression in MC3T3-E1 cells. In contrast, Cur-Zn exhibited broad-spectrum antibacterial efficacy against both Escherichia coli and Staphylococcus aureus , while Cur-Ca showed limited functional benefits due to rapid ion release and potential cellular interference. These findings highlight the tailored functionality of Cur-metal complexes. Cur-Cu scaffolds are promising for repairing bone defects under oxidative stress, whereas Cur-Zn scaffolds are effective in preventing implant-associated infections. This work provides a rational strategy for designing microenvironment-responsive biomaterials with integrated antioxidant, osteogenic, and antibacterial properties for advanced bone tissue engineering.

Thermosensitive in situ hydrogel based on the hybrid of hyaluronic acid and modified PCL/PEG triblock copolymer

Zesheng Lv,Longlong Chang,Xingwen Long,Jianping Liu,Yuzhang Xiang,Jinjian Liu,Jianfeng Liu,Hongzhang Deng,Liandong Deng,Anjie Dong

In this work, a new hydrogel was constructed using poly(ɛ-caprolactone-co-1,4,8-trioxa[4.6]spiro-9-undecanone)–poly(ethylene glycol)–poly(ɛ-caprolactone-co-1,4,8-trioxa[4.6]spiro-9-undecanone) tri-block copolymers (PECT) with hyaluronic acid (HA) in order to expand application scopes of PECT hydrogel. The rheological and sol–gel phase transition behaviors were investigated by rheometer and test tube inversion method, and the interior morphologies of hydrogel systems were observed by scanning electron microscope (SEM). With the introduction of HA, certain properties of PECT hydrogel, such as viscosity and morphology, have present trends with regularity. Furthermore, with the participation of HA, the degradation and release of acetylsalicylic acid was slightly affected, however, the drug release mechanism of hydrogel has not been changed. PECT/HA hydrogel is confirmed to be non-toxic through a test to NIH3T3 cells. In conclusion, blending with HA is a feasible and safe method to tune properties of PECT hydrogel.

Improved mechanical and EMI shielding properties of PLA/PCL composites by controlling distribution of PIL-modified CNTs

Xu Pei,Huang Bincheng,Tang Ruobing,Wang Zhenfeng,Tu Jiaying,Ding Yunsheng

The functionalization and distribution of conductive fillers and the interfacial compatibilization of polymer blends is the basic scientific problem of electromagnetic interference (EMI) shielding composites. Polylactic acid/polycaprolactone (PLA/PCL)-based nanocomposites containing polymerizable ionic liquid copolymer of poly(methoxy poly(ethylene glycol) monomethacrylate-co-1-vinyl-3-ethylimidazolium bromide) (PILs) and multi-walled carbon nanotubes (CNTs) were fabricated by the melt blending. The high affinity of MEPGMA segments of PILs for both phases and the interaction between the imidazole group of PILs and CNTs improved the dispersion and compatibility of CNTs in the matrix. PILs could expand to the interfacial layers as compatibilizers because of the high affinity of MEPGMA groups for polymer phases. The migration of CNTs from PLA to PCL phase was realized by the kinetic control and PIL-modified CNTs were selectively distributed at the co-continuous PLA/PCL interface and PCL phase of the PLA/PCL/4PIL/8CNT-6 m composite with blending time of 6 min. The selective localization of CNTs at interphase significantly can form effective conductive networks at the interface for PLA/PCL/4PIL/8CNT-6 m composite, which increase the electrical conductivity of the composite. The EMI shielding efficiency of the PLA/PCL/4PIL/8CNT-6 m composites, which can reflect and absorb microwaves with multiple times, reaches up to the maximum value 41 dB. Graphical abstract The selective localization of CNTs at interphase forms effective conductive network. EMI SE of the PLA/PCL/4PIL/8CNT-6 m composite displays 41 dB.
