Coordination bonding-based Fe3O4@PDA-Zn2+-doxorubicin nanoparticles for tumor chemo-photothermal therapy

Mengzhu Kong,Yaling Huang,Ruilin Yu,Juqun Xi

Multistimuli-responsive vehicles that deliver drugs in temporal-, spatial- and dosage-controlled manners have developed as a potential platform in tumor therapy. However, how to integrate these functions in one single carrier effectively still presents a lot of challenge. Here, we developed a strategy to construct a photothermal and pH responsive platform based on Zn 2+ coordination with doxorubicin (DOX) and polydopamine (PDA) on the surface of magnetic nanoparticles (Fe 3 O 4 @PDA-Zn 2+ -DOX NPs). In this architecture, Fe 3 O 4 @PDA nanoparticles triggered photothermal effects for tumor hyperthermia, and Zn 2+ coordinated DOX was released in responding to the thermal effect to achieve chemotherapy simultaneously. Moreover, DOX release was further promoted at acidic environment due to the pH sensitivity of Zn 2+ coordination. Therefore, our system possesses multiple effects in tumor treatment: photothermal therapy with near infrared lighting (NIR), thermosensitive DOX release for chemotherapy, and pH-responsive DOX release to enhance deeper therapy in tumor acidic microenvironment. Our in vivo results provide the evidence that Fe 3 O 4 @PDA-Zn 2+ -DOX NPs upon laser exposure have effective anti-tumor therapeutic activity, indicating this platform is suitable for enhanced chemo-photothermal synergistic tumor therapy.

Dipolar-hollowed α-Fe2O3@Au/Polydopamine nanospindle for photothermal-photodynamic coupling antibacterial and drug-delivery

Guangjin Yu,Jing Wang,Qingshan Xiong,Yunqi Xu,Shouhu Xuan,Ken Cham-Fai Leung,Qunling Fang

With the prevalence of drug-resistant bacteria and the waning effects of antibiotics, nanoplatform has become an effective strategy for fighting infections. This work reports a dipolar-hollowed α-Fe 2 O 3 @Au/Polydopamine (PDA) nanospindle which possesses both photothermal-photodynamic (PTT-PDT) coupling antibacterial and drug carrying performance. Firstly, the spindle type α-Fe 2 O 3 @Au/PDA particle was prepared by a simple one-step strategy and then the dipolar-hollow structure was obtained by controlling etching the inside α-Fe 2 O 3 core with hydrochloric acid. After further loading the photosensitizer zinc phthalocyanine (ZnPc), the dipolar-hollowed α-Fe 2 O 3 @Au/PDA-ZnPc nanospindles were obtained. Owing to the dipolar-hollowed interior, the nanospindles are also effective in carrying antitumor drug doxorubicin (DOX) and shows a good drug loading-release behavior. The dipolar-hollowed α-Fe 2 O 3 @Au/PDA nanospindles exhibits a high antibacterial performance against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) under near-infrared (NIR) and Xenon lamp irradiation. When α-Fe 2 O 3 @Au/PDA-ZnPc nanospindles concentration was increased to 100 μg/mL, the antibacterial rate was close to 100 %. In comparison to the original α-Fe 2 O 3 @Au/PDA nanospindles, the product achieved a lower effective antibacterial temperature. This triple-mode therapy (PTT/PDT/Drug) provides an interesting design idea for anisotropic therapeutic nanoplatform which can be applied in low-temperature antibacterial and drug delivery.

Chitosan Hydrogel Equipped With Hollow Copper Sulfide/Polydopamine Nanoparticles for Near-Infrared/pH Dual-Responsive Drug Delivery

Jiayao Wang,Jingguo Li,Shaokui Cao,Yingliang Liu,Jun Shi

In recent years, the remotely multi-responsive chitosan (CS) hydrogel has been garnering extensive attention owing to its remarkable potential in biomedical applications. Meanwhile, it presents a wide range of uses in the realm of controlled drug delivery and photothermal chemical combination therapy. In this paper, the CS/β-glycerophosphate disodium salt (β-GP) physical crosslinked network was employed as the framework to encapsulate near-infrared (NIR) responsive hollow copper sulfide (CuS)/polydopamine (PDA) nanoparticles, and the NIR/pH synergistic responsive CS/CuS/PDA hydrogel was fabricated. The negatively charged β-GP, along with the hollow CuS/PDA nanoparticles, played a key role in promoting the creation of abundant electronegative cavities. As a result, the hybrid hydrogel achieved an impressively high DOX loading rate, reaching up to 95.5%. The CS/CuS/PDA hydrogel exhibits excellent pH responsiveness, which could be attributed to the disintegration of the CS/β-GP network under acidic buffer solution. In addition, hollow CuS/PDA nanoparticles embedded in the hydrogel endow the hydrogel with excellent NIR photothermal responsiveness, which significantly promotes the release of DOX. Consequently, the CS/CuS/PDA hydrogel reveals excellent synergistically enhanced NIR, pH, and thermal triple-responsiveness, indicating its tremendous promise in the field of controlled drug release.

Magnetically separable h-Fe3O4@Au/polydopamine nanosphere with a hollow interior: A versatile candidate for nanocatalysis and metal ion adsorption

Kezhu Xu,Jin Wu,Qunling Fang,Linfeng Bai,Jinyu Duan,Jiaxing Li,Huajian Xu,Ailing Hui,Lingyun Hao,Shouhu Xuan

The organic pollutants and heavy metal ions are critical harmful substances in waste water because they destroy the ecological systems and lead diseases to animals and human beings. Unfortunately, the simultaneously remove of them in one system is still a challenge due to their different treating mechanism. Herein, a novel magnetic h-Fe 3 O 4 @Au/Polydopamine (Au/PDA) hybrid hollow nanosphere which can both absorb the potentially toxic U(VI) ions and catalyze the reduction of the 4-nitrophenol is reported. By using a simple in situ redox-oxidizing polymerization method, the h-Fe 3 O 4 hollow nanosphere is well encapsulated by the Au/PDA hybrid shell to form the dual-functional magnetic hollow nanocomposites. Owing to the hollow interior, uniform and functional PDA coating, and high active Au nanocrystals, the final h-Fe 3 O 4 @Au/PDA hollow nanosphere possesses high potential in nanocatalysis, heavy metal ions adsorption, and drug delivery. Because of the PDA protection, the h-Fe 3 O 4 @Au/PDA nanosphere can even keep 90% activity after 7 cycling catalytic reactions on reduction of 4-nitrophenol. Similarly, the h-Fe 3 O 4 @Au/PDA exhibits a good adsorption effect for U(VI) in nuclear waste and its adsorption capability reaches to 82.9 mg·g −1 by 6 successive adsorption–desorption experiments. Due to the facile magnetic separation, the h-Fe 3 O 4 @Au/PDA nanosphere can be easily collected by applying an external magnetic field. In summary, such a multifunctional recyclable h-Fe 3 O 4 @Au/PDA nanosphere with green and facile synthesis, easy manipulation, efficient adsorption performance and good catalytic activity possesses broad application potential in treatment of coexisting toxic water pollution.

Guojuan Zhang,Zhipeng Dai,Yujing Guo,Jinzhong Zhao,Yujie Han

In recent years, increased environmental pollution has become a major societal concern, since quite a few people experienced various diseases caused by heavy metal ion pollution. Rare earth oxide perovskite based adsorbents have been applied in heavy metal ions removal on account of a special affinity. However, perovskite tending to agglomeration promoted an urgent and necessary search for protective agents. Here, the rare earth oxide perovskite PDA@LaCo 0.35 Ni 0.65 O 3 was synthesized by introducing PDA onto LaCo 0.35 Ni 0.65 O 3 structure via hydrothermal method. The PDA providing with large specific surface area and oxygen-containing functional groups can enhance the adsorption capacity of nanomaterials for heavy metal ions in water. The as-obtained PDA@LaCo 0.35 Ni 0.65 O 3 displayed maximum adsorption capacity of 142.49 mg∙g −1 , 96.30 mg∙g −1 , 105.12 mg∙g −1 for Cu 2+ , Hg 2+ , Pb 2+ . The proposed PDA@LaCo 0.35 Ni 0.65 O 3 enabled to achieve excellent removal rates toward three heavy metal ions. This work brings new opportunities for sorbents innovation in the field of sewage treatment.

Corrosion mechanism and resistance of g-C3N5@PDA nanosheets modified epoxy-coated carbon steel

Fujian Tang,Yongde Yao,Shangtong Yang

Nitrogen-rich carbon nitride (g-C 3 N 5 ) nanosheets were modified with polydopamine (PDA) and then incorporated into an epoxy resin (EP) matrix to create a g-C 3 N 5 @PDA modified epoxy (GP/EP) coating. The phase composition, microstructure, and mechanical properties were analyzed using X-ray diffraction, scanning electron microscopy, and nano-indentation. The corrosion behavior was investigated by applying the coating to a steel plate and immersing it in a NaCl solution for 20 days using electrochemical impedance spectroscopy and potentiodynamic polarization tests. Molecular dynamics simulations were also conducted to elucidate the corrosion mechanism. The results indicated that the hardness and elastic modulus of the epoxy coating increased from 0.02 GPa to 0.26 GPa and from 0.92 GPa to 2.90 GPa, respectively, after the modification of g-C 3 N 5 nanosheets with PDA. Additionally, the corrosion current density of the GP/EP-coated steel plate was measured at 0.51 μA/cm 2 , which is approximately 17 times lower than that of the EP-coated plate when immersed in a 3.5 wt% NaCl solution.

Synthesis of PDA-rGO/MoO3 nanohybrids for distinguishable anticorrosion enhancement of waterborne epoxy coating

Ying Zhou,Fengyang Kong,Jiaqing Zhou,Jidan Hu,Guoying Zhu

Waterborne epoxy (WEP) resins have gained considerable attention for their environmentally friendly characteristics. Nevertheless, they are susceptible to substantial failures due to electrochemical corrosion, particularly when the coating is compromised or exposed to harsh corrosion environments. Furthermore, its long-term anticorrosive performance is affected by certain inherent defects. In this study, inspired by the adhesive properties of mussels, polydopamine (PDA) was used as a green binder to integrate graphene oxide (GO), h-M O O 3 , and WEP matrix, creating a uniform three-dimensional (3D) network structure. PDA-modified two-dimensional GO nanosheets can also create a porous carrier capable of loading anticorrosive Mo derivatives, imparting self-healing properties that enhance anti-corrosion performance. The resulting composites exhibited remarkable self-healing ability due to the unique design of the PDA-rGO porous structure infused with anticorrosive MoO 3 . Subsequently, X-ray diffraction, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, and ultraviolet-visible spectroscopy were used to confirm the PDA-rGO/MoO 3 nanostructure. The results confirmed the successful self-polymerization of PDA on the GO surface, converting it into reduced GO (rGO). Then PDA-rGO/MoO 3 composite WEP coatings were systematically evaluated by electrochemical impedance spectroscopy (EIS), microscopy and acceleration corrosion experiment. In addition, electrochemical impedance spectroscopy (EIS) and microscopy were used to assess the PDA-rGO/MoO 3 composite coatings. The EIS results revealed that the composite coating with 0.5 wt% PDA-rGO/MoO 3 substantially enhanced the anti-corrosion performance compared with the control samples. After prolonged immersion for 60 days in a 3.5 wt% NaCl solution, the composite coating exhibited outstanding anti-corrosion performance, surpassing that of the GO/WEP coating by three orders of magnitude. This composite nanocoating provides valuable insights into the design of well-dispersed, 3D network-structured coatings with self-healing properties for corrosion protection, which opens up new possibilities for the practical application of composite nanohybrids in the field of anti-corrosive coatings.

Biofriendly and Regenerable Emotional Monitor from Interfacial Ultrathin 2D PDA/AuNPs Cross-linking Films

Liming Chen,Youju Huang,Liping Song,Wuliang Yin,Linxi Hou,Xuqing Liu,Tao Chen

Well-designed 2D materials with ultrathin structures show great potential for humidity-sensing performance owing to their high surface-volume ratio and a great number of exposed atoms on the surface. However, some sensing elements employed for healthcare applications may be considered as potentially risky, such as inflammation, granuloma formation, and carcinogenesis. Herein, we explored biofriendly humidity-sensing characteristics inspired by the great biocompatibility and conductivity of hyperbranched polyethyleneimine-capped gold nanoparticles and cross-linked with polydopamine from the adhesive proteins in mussels. It was successfully employed into two kinds of wearable devices, sports watches and breathing masks, for real-time recording humidity’s fluctuation in expiration and sweat with changes of individual’s crying, laughing, nervous, sleeping, training, and cold states. The wearable devices allow us to monitor individual’s physical activities and emotional states well, suggesting a promising prospect in safe, reusable, long term, and noncontact human health monitoring applications.

Biotin-streptavidin sandwich integrated PDA-ZnO@Au nanocomposite based SPR sensor for hIgG detection

Haohua Yang,Xueqi Zhao,Ziwei Zhang,Pinyi Ma,Xinghua Wang,Daqian Song,Ying Sun

SPR is a mature optical biosensor technology for detecting biomolecular interactions without fluorescence or enzyme labeling. In this paper, we acquire a sensitive SPR biosensor based on [email protected] nanomaterial , and the classical sandwich strategy using biotin-streptavidin for secondary signal amplification system was used to detect human IgG (hIgG). Nano-zinc oxide (ZnO) has the dual characteristics of nanocomposite and traditional zinc oxide, with large specific surface area and high chemical activity. Besides, the gold-coated ZnO nanocrystals improve the optical properties of ZnO and enlarge the loading capacity with better biocompatibility. Therefore, a sensing platform based on [email protected] nanomaterial was constructed on gold film modified with mercaptan. Meanwhile, the biotin-avidin system in SPR sensor field has been rapidly developed and applied. Due to the highly selection of streptavidin (SA) and biotin interact with each other, GNRs-SA-biotin-Ab 2 (GSAB-Ab 2 ) were constructed to obtain the secondary enhancement of SPR signal. The influences of experimental conditions were also discussed. With optimal experimental conditions, introducing GSAB-Ab 2 conjugate combined with a sandwich format, the resulting SPR biosensor provides a favourable range for hIgG determination of 0.0375–40 μg mL −1 . The minimum detection concentration of hIgG that can be obtained by this method is approximately 67-fold lower than the conventional SPR sensor based on gold film. The sensitivity of SPR biosensor is significantly improved in a certain range.

Engineering PDA-coated CM-CS nanoparticles for photothermo-chemotherapy of osteosarcoma and bone regeneration

Xing Sun,Zhiyuan Meng,Qingwen Yu,Xinyu Wang,Zheng Zhao

After an osteosarcoma excision, recurrence and bone defects are significant challenges for clinicians. Combination therapy based on a nanoparticle-based drug delivery system shows great promise in osteosarcoma therapy. In this study, we designed a multifunctional chitosan (CS) nanoparticle , which combined photothermal therapy (PTT) with chemotherapy to synergistically enhance osteosarcoma therapy and bone repair . Curcumin (CM) loaded CS nanoparticles were prepared through ionotropic gelation method and were further functionalized with polydopamine (PDA) coating to form CS-CM-PDA nanoparticles (CCPNPs). The CCPNPs displayed pH- and near-infrared (NIR)- responsive CM release behavior. In vitro anti-cancer results indicated that CCPNPs possessed a long-term stable anti-cancer effect compared to pure CM. The presence of the NIR irradiation combined with CCPNPs resulted in a dramatic decrease in MG-63 osteosarcoma cell viability in 10 min compared with the pure CCPNPs at the concentration of 500 μg/mL. Meanwhile, in vitro study demonstrated that CCPNPs induced MG-63 cell apoptosis via downregulating the expression of Bcl-2 and upregulating the expression levels of Bax and Caspase-3. Besides, CCPNPs could positively induce the proliferation of MC3T3-E1 osteoblast cells in vitro . These results suggested that CCPNPs with bifunctional osteosarcoma therapy and bone repair may be an excellent candidate for local cancer therapy and bone regeneration .

Preparation of environmentally responsive PDA&DOX@LAC live drug carrier for synergistic tumor therapy

Liu Lu,Zhao Xuefen

The development of intelligent, environmentally responsive and biocompatible photothermal system holds significant importance for the photothermal combined therapy of tumors. In this study, inspired by Lactobacillus (LAC), we prepared a biomimetic nanoplatform PDA&DOX@LAC for tumor photothermal-chemotherapy by integrating the chemotherapeutic drug doxorubicin (DOX) with dopamine through oxidative polymerization to form polydopamine (PDA) on the surface of LAC. The PDA&DOX@LAC nanoplatform not only achieves precise and controlled release of DOX based on the slightly acidic microenvironment of tumor tissues, but also exhibits enzyme-like properties to alleviate tumor hypoxia. Under near-infrared light irradiation, it effectively induces photothermal ablation of tumor cells, enhances cellular uptake of DOX with increasing temperature, and thus efficiently inhibits tumor cell growth. Moreover, it is further confirmed in vivo experiments that photothermal therapy combined with PDA&DOX@LAC induces tumor cells apoptosis, releases tumor-associated antigens, which is engulfed by dendritic cells to activate cytotoxic T lymphocytes, thereby effectively suppressing tumor growth and prolonging the survival period of 4T1 tumor-bearing mice. Therefore, the PDA&DOX@LAC nanoplatform holds immense potential in precise tumor targeting as well as photothermal combined therapy and provides valuable insights and theoretical foundations for the development of novel tumor treatment strategies based on endogenous substances within the body.

GNP/Ag@PDA Incorporated Waterborne Epoxy Coating with Excellent Corrosion Resistance and Antifouling Performance

Xingyu Liu,Canhai Cai,Cansen Liu,Qingmei Chen,Jianyu Xu,Ran Zhang

Corrosion and fouling problems are two main challenges for devices working in the ocean. However, most existing solutions lack the ability to concurrently obtain corrosion resistance and antifouling behavior. Here, a novel waterborne epoxy (WEP) based composite coating that has remarkable resistance to corrosion and fouling is developed. The WEP-based composite coating consists of nanohybrid filler prepared by loading silver (Ag) nanoparticles on graphite nanoplatelets (GNP) surface through a facile in situ hydrothermal synthesis and polydopamine (PDA) modification. The PDA modification effectively improves the dispersion and compatibility of GNP/Ag@PDA (GAP) with the WEP matrix. The synergistic effect of Ag with chemical antifouling behavior and GNP with physical barrier function endows the GAP/WEP composite coating with a low corrosion rate of 1.04 × 10 −7 mm a −1 after immersing in 3.5 wt.% NaCl solution for 28 days, which is about two orders of magnitude lower than that of blank WEP coating, and a high inhibition rate of 90.81% after immersing in Phaeodactylum tricornutum solution for 7 days. This approach has the potential to address the needs of a range of applications around anticorrosion and antifouling in the ocean.
