Osteoporotic bone defects (OBDs) pose significant clinical challenges due to impaired tissue regeneration and osteogenic differentiation. Conventional combination therapies using systemic anti-osteoporosis drugs and graft fillings have yielded unsatisfactory results. Excessive reactive oxygen species (ROS) are a hallmark of OBDs, leading to abnormal inflammatory responses and a hostile osteoimmune microenvironment that impedes repair. In this study, we developed a multifunctional injectable hydrogel, termed PTSr@CP, composed of a strontium (Sr) and tannic acid (TA)-based metal–phenolic network core coated with polydopamine (PDA), integrated into a matrix of carboxymethyl chitosan and poly (γ-glutamic acid). The PTSr@CP hydrogel can be easily injected into bone defect sites, where it undergoes rapid in situ crosslinking and exhibits exceptional ROS-scavenging capabilities. Additionally, the hydrogel mediates mild photothermal therapy (MPTT), promoting local tissue repair. Both in vitro and in vivo studies demonstrated its excellent biosafety and confirmed synergistic bone regeneration through enhanced osteogenesis, angiogenesis, and osteoimmunomodulation. This work offers a compelling strategy for OBD treatment by combining mild hyperthermia-enhanced in situ bone repair with sustained delivery of bioactive agents to improve the osteoimmune microenvironment.

Ultrasound-responsive nanomaterials represent a promising approach for achieving non-invasive and localized drug delivery within tumor microenvironments. In this study, we developed a piezocatalysis-assisted hydrogel system that integrates reactive oxygen species (ROS) generation with stimulus-responsive drug release. The platform combines piezoelectric barium titanate (BTO) nanoparticles with a ROS-sensitive hydrogel matrix, forming an ultrasound-activated dual-function therapeutic system. Upon ultrasound irradiation, the BTO nanoparticles generate ROS—predominantly hydroxyl radicals (•OH) and singlet oxygen (1O2)—through the piezoelectric effect, which triggers hydrogel degradation and facilitates the controlled release of encapsulated therapeutic agents. The composition and kinetics of ROS generation were evaluated using radical scavenging assays and fluorescence probe techniques, while the drug release behavior was validated under simulated oxidative environments and acoustic fields. Structural and compositional characterizations (TEM, XRD, and XPS) confirmed the quality and stability of the nanoparticles, and cytocompatibility was assessed using 3T3 fibroblasts. This synergistic strategy, combining piezocatalytic ROS generation with hydrogel disintegration, demonstrates a feasible approach for designing responsive nanoplatforms in ultrasound-mediated drug delivery systems.

Cu 3 P nanoparticles (NPs) were proposed as an efficient antibacterial agent for impeding bacterial resistance. Both sensitive and resistant strains of E. coli and S. aureus (3 × 10 6 CFU mL −1 ), and the microbial populations from a fishery water were completely inactivated by Cu 3 P NPs (≤ 1.5 µg mL −1 ) within 30 min with no regrowth. The antibacterial mechanism of Cu 3 P NPs relied on their own multiple enzyme-like activities, to elucidate, the action of ROS produced by their oxidase- and peroxidase- like activities was facilitated by their inherent activities for glutathione depletion and the lipid peroxidation. Comparing with that traditional antibiotics induced bacterial resistance (6 out of 7) within 3 passages, Cu 3 P NPs kept their initially inactivated efficiency, demonstrating that they can delay the onset of bacterial resistance. Furthermore, the Cu 3 P NPs shown no obviously toxic effect on aquatic organisms and a negligible side-effect toward mammalian tissues. This strategy is insightful for struggle with resistant bacteria.

Single chemotherapy lacked controllability, while smart-responsive hydrogel could deliver drug to specific sites in response to different external stimuli. Hyaluronic acid–Chitosan–Thioketal@Chlorin e6 (HA-CS-TK@Ce6) hydrogel with dual response of ROS and hyaluronidase (HAase) was synthesized by electrostatic interaction force. HA-CS-TK@Ce6 hydrogel loaded with chemotherapeutic drug doxorubicin (DOX) and photosensitizer Ce6 successfully achieved the combined anticancer effect of chemotherapy and photodynamic therapy. Experimental results demonstrated that releasing efficiency of the hydrogel toward DOX could reach 90.6 % in pH = 5 H 2 O 2 solution with HAase at T = 40 ℃. Moreover, HA-CS-TK@Ce6 hydrogel was capable of stimulating Ce6 to produce massive ROS under 650 nm laser irradiation, thus leading to breakage of ROS-cleavable TK and realizing degradation of hydrogel. It was worth mentioning that laser confocal results indicated that MCF-7 cells could successfully uptake Ce6 and DOX, which further proved that HA-CS-TK@Ce6 hydrogel could achieve efficient photodynamic-chemotherapeutic treatment. Therefore, dual-responsive HA-CS-TK@Ce6 hydrogel with excellent biocompatibility and degradability enabled selective and intelligent release of DOX, which would be prospective to improve therapeutic effect of cancer through chemo-photodynamic therapy.

The presence of bacterial persisters is a key factor contributing to chronic infection. However, no effective treatment methods are currently available. Thus, a platform is developed, called reactive oxygen species (ROS) bomb, based on microenvironment-adaptive hydrogel microspheres, to oxidize the cell membranes of persisters in chronic periprosthetic joint infection (PJI). Fenton reagent hydroxy iron oxide (FeOOH) and a glucose oxidase (GOx)/calcium phosphate (CaP) acid-responsive shell are sequentially induced on the surface of mesoporous polydopamine (PDA) nanoparticles by the PDA-mediated ion precipitation and interfacial adhesion, followed by the coloaded with glucose into microfluidic hyaluronic acid hydrogel microspheres. Hydroxyl radicals are explosively generated through GOx-mediated glucose oxidase, H 2 O 2 production, and its Fenton-like reactions with FeOOH, which also benefit from the weakly acidic microenvironment around persisters, result in the destruction of bacterial cell membrane, and subsequent overflow of cellular contents such as dsDNA, proteins, and K + . The bactericidal rates of methicillin-resistant Staphylococcus aureus and Staphylococcus epidermidis persisters are up to 99.14% and 98.96%, and the bacterial loads in lesion location are significantly decreased after ROS bombs treated, effectively alleviated the inflammation and bone resorption damage. This work provides a new strategy toward persisters clearance and shows great application potential in other chronic infection-related diseases.

Benefiting from treating diseases at the genetic level, gene therapy has been considered a new revolution in the biomedical field. However, the extracellular and intracellular barriers during gene transport such as enzymatic degradation and endo-/lysosomal sequestration significantly compromise the therapeutic efficacy. Though photochemical internalization (PCI) has emerged as a promising approach for causing endo-/lysosomal leakage with translocation of the internalized molecules into the cytosol, its effect is still unsatisfactory due to the insufficient light penetration depth. Here, we develop tumor microenvironment-specific enhanced gene delivery by means of ROS generated from the in situ cascaded catalytic reactions in tumors involving GOx-mediated redox reaction and Mn2+-mediated Fenton-like reaction. The efficient enzymatic protection and successful endo-/lysosomal escape of cargo gene complexes have been demonstrated. Moreover, anti-Twist siRNA-loaded G@MMSNs-P exhibit tumor-specific biodegradation, excellent T1-weighted MR imaging, and significant inhibitory effects against breast cancer growth and pulmonary metastasis.

In this work, we presented a kind of intelligent supramolecular hydrogels (SHGs) based on base-pairing interaction between guanine and cytosine, comprising α-cyclodextrin, thioketals-modified guanine (G-TK-G) and cytosine-terminated PEG (C-PEG-C). The hydrogen bond of nucleic acid between G and C as an additional cross-linker effectively enhanced storage moduli ( G ′s) of the hydrogels. Notably, the obtained hydrogels exhibited excellent reactive oxygen species (ROS) responsive property due to the introduction of thioketals group. Under ROS-rich environment, the hydrogels could control the release of loaded-cargoes, outlining the potential of intelligent materials for biomedical application, such as smart drug delivery for anticancer.

Copper-based agents have been utilized widely in a range of commercial antibacterial products due to their superior release-killing antibacterial properties. However, the environmental concerns associated with the release of Cu2+ ions have restricted their application in textiles. In this study, Cu3P-ZnO nanocomposites with enzyme-like properties were synthesized through the phosphating of CuO-ZnO, which were employed to functionalize the surface of cotton fabric using a dipping–padding–drying method. The physicochemical properties of the functional cotton fabrics were characterized using various techniques. The antibacterial efficacy of Cu3P-ZnO/cotton was assessed against Gram-negative E. coli and Gram-positive S. aureus bacteria. Remarkably, Cu3P-ZnO/cotton demonstrated significant antibacterial activity and stability without necessitating auxiliary conditions; achieving >99% antibacterial effectiveness against both E. coli and S. aureus, while retaining 94.7% efficiency after 50 washing cycles. The underlying antibacterial mechanism was elucidated, revealing that reactive oxygen species (ROS), especially singlet oxygen (1O2), plays a crucial role in mediating the antibacterial action of Cu3P-ZnO/cotton in absence of light, whereas direct contact between Cu3P-ZnO and bacteria serve secondary functions in this process. This research provides valuable insights into the design of ROS-mediated antibacterial agents for applications in antimicrobial textiles.

Bionic mimics using natural cartilage matrix molecules can modulate the corresponding metabolic activity by improving the microenvironment of chondrocytes. A bionic brush polymer, HA/PX, has been found to reverse the loss of cartilage extracellular matrix (ECM) and has promising applications in the clinical treatment of osteoarthritis (OA). However, the unknown bioremediation mechanism of HA/PX severely hinders its clinical translation. In OA, the massive loss of the ECM may be attributed to a decrease in transient receptor potential vanilloid 4 (TRPV4) activity, which affects reactive oxygen species (ROS) clearance and [Ca2+]i signaling, initiating downstream catabolic pathways. In this study, we investigated the bioremediation mechanism of HA/PX in a model of interleukin 1β (IL-1β)-induced inflammation. Through TRPV4, HA/PX reduced ROS accumulation in chondrocytes and enhanced [Ca2+]i signaling, reflecting a short-term protection capacity for chondrocytes. In addition, HA/PX balanced the metabolic homeostasis of chondrocytes via TRPV4, including promoting the secretion of type II collagen (Col-II) and aggrecan, the major components of the ECM, and reducing the expression of matrix metal-degrading enzyme (MMP-13), exerting long-term protective effects on chondrocytes. Molecular dynamics (MD) simulations showed that HA/PX could act as a TRPV4 activator. Our results suggest that HA/PX can regulate chondrocyte homeostasis via ROS/Ca2+/TRPV4, thereby improving cartilage regeneration. Because the ECM is a prevalent feature of various cell types, HA/PX holds promising potential for improving regeneration and disease modification for not only cartilage-related healthcare but many other tissues and diseases.

Selective in situ activation of prodrugs or generation of bioactive drugs is an important approach to reducing the side effects of chemotherapy. Herein, a tailored ROS-activable prodrug nanomedicine ( [email protected] ) was developed as the prodrug activation nanoamplifier for highly selective antitumor therapy. [email protected] was rationally constructed by the diethyldithiocarbamate (DTC) prodrug DTC-PPB and the nanoscale coordinated framework Cu-SK based on copper and the ROS generator shikonin (SK). Cu 2+ , SK and DTC were kept in the inactive state in the fabricated [email protected] In the presence of ROS within tumors, DTC-PPB can be activated to release less cytotoxic DTC, which can rapidly chelate Cu 2+ from the Cu-SK framework to synthesize highly cytotoxic Cu(DTC) 2 and induce SK to release in a cascade. The released SK can generate ROS to increase the intracellular ROS level, further activating DTC-PPB to release more DTC. That is, [email protected] can undergo a self-amplifying positive feedback loop to induce numerous bioactive Cu(DTC) 2 formation and SK release triggered by a small amount of ROS within the tumor microenvironment, which endows the transformation of “less toxic-to-high toxic” and thus significantly improve its selectivity towards tumors. Therefore, this study provides a new strategy of prodrug activation for tumor therapy with high efficiency and low toxicity. Statement of significance Owing to the striking difference in ROS level between cancer cells and normal cells, ROS-responsive prodrugs are regarded as a promising approach for tumor-specific therapy. However, the stability and responsiveness of prodrugs are hard to balance. Preferable sensitivity may cause premature activation while favorable stability may lead to incomplete prodrug activation and insufficient active drug release. This study provides a tailored ROS-responsive prodrug activation nanoamplifier with favorable stability and effective prodrug activation capacity. The nanoamplifier can undergo a self-amplifying positive feedback loop to achieve numerous bioactive drugs generation in situ under ROS triggers within the tumor microenvironment, showing the enhanced antitumor therapeutic effect. Thus, this study provides a new strategy for prodrug activation and tumor-specific therapy.

Probiotics are generally used as therapeutic intervention in inflammatory bowel disease. However, the low survival rate in harsh gastrointestinal environment and limited retention in intestine greatly restrict their health benefits. To address this problem, a ROS-responsive hydrogel based on hyaluronic acid (HA) was developed for encapsulation and targeted delivery of probiotics. The hydrogel was prepared facilely by physiological crosslink with methacrylated HA and thiolated thioketal. As a model probiotic, Lactobacillu reuteri showed a significantly increased survival rate in simulated digestive conditions after encapsulated in hydrogel. The negative properties conferred the hydrogel preferential adhesions to inflammation sites. Meanwhile, the excess reactive oxygen species (ROS) produced by inflamed colon tissues selectively cleaved thioketal linkages resulted in hydrogel degradation and local probiotics release. Furthermore, the hydrogel exerted an appropriate ROS-scavenge capacity and protected HT-29 cells from oxidative damage. Animal experiments indicated that hydrogel-encapsulated L. reuteri could remarkably alleviate the symptoms and improve the survival rate of mice with dextran sulfate sodium (DSS)-induced colitis. These results suggested that the biocompatible hydrogel may be a delivery platform to target inflamed intestines and expand the application of probiotics as pharmaceuticals.

Licochalcone A (LCA) exhibited anticancer activity through modulating reactive oxygen species (ROS) levels in some cancer cells and has been evidenced to suppress colorectal cancer (CRC) formation and progression. However, whether LCA mediates the progression of CRC by regulating ROS production remains unclear. To address this, HCT-116 cells were treated with LCA, resulting in G0/G1 phase arrest, apoptosis, and high ROS generation, which were attenuated by N-acetyl-L-cysteine, a ROS inhibitor. In addition, LCA suppressed the expression of thioredoxin reductase 1 (TrxR1) in HCT-116 cells, leading to high ROS levels and apoptosis. Moreover, LCA administration combined with TrxR1 inhibition further enhanced the production of ROS and apoptosis in HCT-116 cells compared to LCA administration or TrxR1 inhibition alone. These results demonstrated that LCA might enhance the production of ROS by targeting TrxR1, leading to apoptosis in HCT-116 cells, which provides potential insight for the interventional treatment of CRC.
