Microelectrode arrays (MEAs) hold inherent potential for the parallel analysis of foodborne pathogens; however, their miniaturized scale introduces critical challenges for multitarget detection, including cross-talk between adjacent units during functionalization and a severely limited sensing area that restricts probe immobilization and ultimate sensitivity. To overcome these limitations, a MEA-based aptasensor is constructed by integrating site-selective electrodeposition with a directed molecular allocation strategy for highly sensitive and specific simultaneous detection of Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). A graphene oxide-polypyrrole nanocomposite and an AuNPs/chitosan-gold composite were precisely deposited on different microelectrode units, enabling the spatially resolved immobilization of aptamers against S. aureus and E. coli, respectively. Fluorescence and Raman spectroscopy confirmed the selective allocation of the respective aptamers to their targeted electrode units, effectively preventing cross-talk. Using methylene blue as an electroactive probe, the binding of target pathogens to their respective aptamers was quantified by square wave voltammetry. With optimized conditions, the limit of detection for S. aureus and E. coli was as low as 25.4 CFU mL–1 and 8.8 CFU mL–1. Real-sample assays confirmed that the MEA-based aptasensor is a precise and viable tool for high-speed quantification of S. aureus and E. coli, exhibiting considerable potential for applications in environmental and food safety monitoring practices.

The escalating threat of methicillin-resistant Staphylococcus aureus (MRSA) necessitates novel therapeutic strategies. Our previous work suggested that an extract from Cyclocarya paliurus leaves (ECPL) inhibits MRSA by targeting the cell division protein FtsZ. Here, guided by anti-MRSA activity, we isolated three compounds from ECPL: asiatic acid (AA), maslinic acid (MA), and ursolic acid (UA). They exhibited antibacterial activity against MRSA and induced cell elongation, indicative of division arrest. Time-kill assays showed AA and MA are bactericides, while UA is bacteriostatic. Mechanistically, these compounds disrupt cell division by differentially affecting FtsZ dynamics: AA promotes polymerization, whereas MA and UA inhibit it. SPR analysis showed direct FtsZ binding to AA (Kd = 2.4 μM), MA (Kd = 9.8 μM), and UA (Kd = 0.7 μM). Molecular docking predicted a shared FtsZ binding pocket but revealed that AA adopts a distinct conformation driven by unique interactions, including a hydrogen bond with Arg191—an interaction not observed for MA or UA, which instead form hydrogen bonds with Thr265 and Thr309. Despite these divergent effects on polymerization and distinct binding modes, all compounds ultimately disrupted Z-ring assembly and septum formation. In a murine skin infection model, AA, selected for its bactericidal activity and unique FtsZ modulation mechanism, significantly reduced bacterial burden and accelerated wound healing. Collectively, our findings validate these compounds as direct FtsZ-targeting agents and establish AA as a promising anti-MRSA lead compound with a novel mechanism disrupting the bacterial divisome.

To facilitate the capture, inactivation, and detection of viable Staphylococcus aureus ( SA ) in food, a vancomycin (Van)-bacterial imprinted polymers (BIPs) hybrid receptor was fabricated by introducing Van into SA -imprinted polydopamine (PDA) via oriented surface imprinting. Taking advantage of the dual recognition arising from Van's affinity for peptidoglycan and BIPs' imprinting effect on SA , the Van-BIPs hybrid exhibited better capture performance than Van or BIPs alone. Leveraging the photothermal effect of PDA, the captured SA could be in situ lysed within 5 min under near-infrared irradiation, causing the release of adenosine triphosphate (ATP) from SA . Using ATP as biomarker, as low as 13.7 CFU/mL of viable SA was able to be detected within 38 min by integrating Van-BIPs hybrid with ATP-bioluminescence assay. Spiked food samples were also successfully analyzed with the recoveries of 85.71 %–106.11 %. The Van-BIPs hybrid might offer a promising tool to control SA in food industries.

The widespread presence of methicillin-resistant Staphylococcus aureus (MRSA) severely threatens convenient therapeutic options in the postantibiotic era. The use of combinations of existing drugs at this stage may be a viable strategy for dealing with complex drug-resistant MRSA infections. An checkerboard MIC (Minimum Inhibitory Concentration) assay, growth curve assay, bactericidal test and scanning electron microscope (SEM) assays were performed to determine whether crisaborole (AN2728), a PDE4 inhibitor for treating atopic dermatitis (AD), produced bactericidal effect with different antibiotics. Here, we identified (AN2728) produced a significant synergistic bactericidal effect with erythromycin, cefuroxime and rifampicin against different bacterial strains of Staphylococcus aureus (S. aureus), especially MRSA (FIC < 0.5) (p < 0.05). Transcriptome analysis, bacterial biofilm assay and several kit assays revealed that AN2728 could also simultaneously affect the membrane and transporter capacity of MRSA. Moreover, in a mouse skin infection model of MRSA, the combination of AN2728 and erythromycin showed remarkable treatment benefits, as shown by significantly reduced bacterial loading (p < 0.05), pathological lesions of the skin and an obvious anti-inflammatory effect (p < 0.05). To our knowledge, this study is the first to establish that AN2728 can cooperate with antibiotics such as erythromycin to completely kill MRSA and that AN2728 can be used to extend the usage life of different antibiotics to address the inevitability of severe MRSA infection.

1, 8-Cineole (Cin), a cyclic monoterpenoid derived from tea trees and eucalyptus species, exhibits diverse pharmacological properties. Yet, its therapeutic impact and underlying mechanism against Staphylococcus aureus (S. aureus) pneumonia remain to be elucidated. In this study, metabolomics based on UPLC-MS/MS was integrated with network pharmacology, molecular biology, and molecular docking to investigate the effects of Cin. The findings demonstrated that Cin markedly reduced mortality and lung bacterial load, lessened pulmonary damage while suppressing the levels of proinflammatory factors, including tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in the bronchoalveolar lavage fluid (BALF) of infected mice. Additionally, 19 metabolites, primarily involved in tryptophan metabolism and arginine biosynthesis, were notably modified by Cin via suppressing the enzymatic activity of indoleamine 2, 3-dioxygenase 1 (IDO1) and inducible nitric oxide synthase (iNOS), thereby attenuating the inflammatory response. Notably, knockdown of IDO1 or iNOS significantly diminished the anti-inflammation effect of Cin. In conclusion, our study validates the therapeutic potential of Cin against S. aureus pneumonia via anti-inflammation by downregulating IDO1 and iNOS. Our results provide a theoretical basis of natural substances applied in bacterial pneumonia treatment.

Medical implants have opened up more possibilities in clinical medicine, while they may lead to severe complications due to biofilm formed by bacteria. Understanding the bacterial response mechanism to the material's stiffness is critical to preventing biofilm formation. This study used Staphylococcus aureus , one of the most common clinical pathogens, to explore the response mechanism. Based on the results, bacteria adhere more efficiently with lower membrane potential on stiff hydrogels than soft hydrogels. Combining experiments and molecular dynamics simulations, we found that the Arsenical pump membrane protein (ArsB) of S. aureus was upregulated on stiff hydrogels and can modulate the bacterial membrane potential and adhesion. The interface membrane curvature was the main morphology difference between bacteria on soft and stiff hydrogels. With the more significant membrane deformation of the bacteria on stiff hydrogels, we demonstrated that the changes in membrane curvature could open the ArsB ion channel. Finally, we screened the ArsB inhibitory drug Rapamycin and verified that Rapamycin-loaded hydrogels could efficiently prevent bacterial adhesion. It can be a way of inhibiting bacterial adhesion during biofilm formation and provide directions for the design of future medical materials.

The development of novel method for drug-resistant bacteria detection is imperative. A simultaneous dual-gene Test of methicillin-resistant Staphylococcus aureus (MRSA) is developed using an Argonaute-centered portable biosensor (STAR). This is the first report concerning Argonaute-based pathogenic bacteria detection. Simply, the species-specific mec A and nuc gene are isothermally amplified using loop-mediated isothermal amplification (LAMP) technique, followed by Argonaute-based detection enabled by its programmable, guided, sequence-specific recognition and cleavage. With the strategy, the targeted nucleic acid signals gene are dexterously converted into fluorescent signals. STAR is capable of detecting the nuc gene and mecA gene simultaneously in a single reaction. The limit of detection is 10 CFU/mL with a dynamic range from 10 to 10 7 CFU/mL. The sample-to-result time is <65 min. This method is successfully adapted to detect clinical samples, contaminated foods, and MRSA-infected animals. This work broadens the reach of Argonaute-based biosensing and presents a novel bacterial point-of-need (PON) detection platform.

Highly pathogenic bacterial infections are a major threat to human health, and the resulting drug resistance is beginning to pose a major clinical threat. To combat multidrug-resistant bacterial infections, there is an urgent need to develop antimicrobial drugs that are highly effective, environmentally friendly, and able to prevent the emergence of bacterial resistance. In this paper, Co4N nanozymes were successfully synthesized by pyrolysis of a presynthesized Co-containing metal organic framework (Co-MOF). Its peroxide and lactate oxidase activity and antimicrobial properties were investigated. Without the addition of exogenous H2O2, the nanozymes can initially generate H2O2 by catalyzing lactic acid and then produce highly active •OH from H2O2. This approach overcomes the obstacle of insufficient H2O2 to achieve a satisfactory antimicrobial effect. Furthermore, the local high temperature generated by Co4N under 808 nm laser irradiation further enhanced the antibacterial effect. These results showed that the nanozyme possesses broad-spectrum antimicrobial activity against Escherichia coli and Staphylococcus aureus, with an inhibition rate as high as 99% within 2 h. Meanwhile, the enzyme has good biocompatibility, paving a way for the application of MOF materials in biomedical fields.

Hydrogel-based delivery systems have now emerged as a pivotal platform for addressing chronic tissue defects, leveraging their innate capacity to suppress pathogenic infections and facilitate expedited tissue regeneration. In this work, an injectable hydrogel dressing, termed AgNPs-dermal matrix hydrogel (Ag@ADMH), has been designed to expedite the healing process of wounds afflicted with methicillin-resistant Staphylococcus aureus (MRSA), featuring sustained antibacterial efficacy. The synthesis of the hydrogel dressing entailed a self-assembly process of collagen fibers within an acellular dermal matrix to construct a three-dimensional scaffold, encapsulated with plant polyphenol-functionalized silver nanoparticles (AgNPs). The Ag@ADMH demonstrated exceptional biocompatibility, and enables a sustained release of AgNPs, ensuring prolonged antimicrobial activity. Moreover, the in vitro RT-qPCR analysis revealed that compared with ADMH, Ag@ADMH diminish the expression of iNOS while augmenting CD206 expression, thereby mitigating the inflammatory response and fostering wound healing. Especially, the Ag@ADMH facilitated a reduction in M1 macrophage polarization, as evidenced by a significant decrement in the M1 polarization trend and an enhanced M2/M1 ratio in dermal matrix hydrogels laden with AgNPs, corroborated by confocal microscopy and flow cytometry analyses of macrophage phenotypes. The in vivo assessments indicated that Ag@ADMH minimized fibrous capsule formation. In a full-thickness skin defect model of MRSA infection, the formulation significantly attenuated the inflammatory response by reducing MPO and CD68 expression levels, concurrently promoting collagen synthesis and CD34 expression, pivotal for vasculogenesis, thereby accelerating the resolution of MRSA-infected wounds. These attributes underscore the injectable extracellular matrix hydrogel as a formidable strategy for the remediation and regeneration of infected wounds. Graphical Abstract

Staphylococcus aureus contamination and prevention has always been a major concern for food industry. This work investigated the antibacterial activity and mechanisms of lauric acid (LA) against S. aureus. Results revealed 156 μg/mL was the minimum inhibitory concentration (MIC) for LA and it retarded growth rate of S. aureus. The inhibitory effect was enhanced with LA concentration. After being treated with 2 MIC LA for 24 h, the number of S. aureus decreased by 3.56 log colony-forming unit (CFU)/mL. Scanning electron microscopy profiling revealed that LA resulted in altered morphology of S. aureus cells. In addition, propidium iodide staining of flow cytometry suggested that LA treatment disrupted the cell membrane integrity. Changes in 8-anilino-1-naphthalenesulfonic acid fluorescence indicated a depolarization change in cell membrane fluidity. For practical applications, LA also displayed an antimicrobial potential in cooked chicken food model system, with 1.25–5 g/L of LA prolonging shelf life by 2 days at 4°C. Moreover, it had no adverse effect on pH values, color in cooked chicken meat, and even reduced lipid oxidation. To sum up, LA has great antimicrobial properties and is a candidate preservative for cooked meat food.

Methicillin-resistant Staphylococcus aureus (MRSA) is an opportunistic pathogen that can cause severe bacterial pneumonia. Amygdalin is the main active pharmaceutical ingredient of bitter almond, which has broad-spectrum antibacterial, anti-inflammatory, anti-oxidation and immunomodulatory effects. It is also the main ingredient of Yinhua Pinggan granule, which is commonly used to moisten the lung and relieve cough. However, little is known about the effects of amygdalin on MRSA. In this study, we found that amygdalin exhibited good antimicrobial activity in vitro against MRSA. Amygdalin has a protective effect on MRSA infected cells, and the effect is better when combined with levofloxacin. It also can reduce the adhesion and invasion of MRSA to cells. Amygdalin has anti-inflammatory and antioxidant effects, which can significantly reduce the increase of inflammatory factors and the production of ROS caused by infection. The protective mechanism of amygdalin on cells may be related to inhibiting the expression of NLRP3, ASC and IL-1β pyroptosis pathways. Taken together, our study suggests that amygdalin exerts antibacterial effects by affecting biofilm formation, the expression of virulence factors, and drug resistance genes. Amygdalin combined with levofloxacin has a protective effect on A549 cells infected with MRSA, and the mechanism may be related to the inhibition of inflammatory response, oxidative damage and pyroptosis.

Staphylococcus aureus contamination continues to be a harmful foodborne pathogen threatening of human health, and there is a growing need for rapid detection technologies. This study proposed a novel paper biosensor based on a polydiacetylene (PDA) polymer functionalized fibrinogen (Fg) for the detection of S. aureus in food sources. The fluorophore was developed based on the high binding ability of fibrinogen-binding proteins on the surface of S. aureus . This binding caused twisting in the PDA backbone, leading to changes in chromatic and fluorescent. The detection limit of this method was 50.1 CFU/mL for S. aureus- contaminated foodstuffs and 65.0 CFU/mL for the pure S. aureus culture, and the novelty came from its rapidity and selectivity for S. aureus compared to other foodborne bacteria . In summary, the present work provides a rapid detection method for S. aureus detection, which will help in addressing food safety-related issues.
