Carbon quantum dots with good biocompatibility can be used as nanomaterial carriers to improve the bioavailability of curcumin. In this paper, green tea was used as raw material to provide a green synthesis method for preparing carbon dots nanoparticles, and Curcumin was loaded to prepare carbon quantum dots/Curcumin nanocomposites (CQDs/Cur). A series of characterization methods were used to study the morphology and chemical structure of composite nanoparticles. The tea carbon dots had the characteristics of excitation wavelength dependence, salt solution stability and light stability. Finally, the antioxidant activities of CQDs/Cur nanocomposites were investigated by DPPH, ABTS •+ , hydroxyl radical, metal ion chelating and reducing power assays. The compound system not only improved the antioxidant performance of curcumin, but also retained the antibacterial activity of curcumin. These results suggested that tea carbon quantum dots loaded with curcumin had the potential to be developed as natural antioxidants and antibacterial agents.

Curcumin-loaded alginate beads, which contain different food emulsifiers, have been prepared using CaCl2 as the cross-linking agent. The controlled release of the curcumin from the beads was investigated at room temperature. For calcium alginate/Span-80/Tween-80 (A/S/T) formulations, almost all of the curcumin loaded in the beads was released into the medium within about 20 h, and the release rates could be regulated by changing the concentration of both Tween-80 and Span-80. However, for the systems of calcium alginate/Q-12A/F-18A (A/Q/F), about 60% of the curcumin loaded in the beads was released at the end of experiments. The studies of scanning electron microscopy indicated that the microstructure of the walls of beads could significantly vary with the concentration or type of emulsifiers. The Fourier transform infrared spectral measurements confirmed that the interactions between calcium alginate and polyglycerol fatty acid esters were stronger than that between calcium alginate and Tween-80/Span-80. The results of swelling studies demonstrated that the initial rates of water uptake for A/Q/F beads were higher than that for A/S/T beads. Moreover, the data of release rates were fitted by an empirical equation, which showed that the release mechanism of curcumin from the alginate gels varied with the composition of emulsifiers for the A/S/T systems. This work provides an important insight into the effect of food emulsifiers on the release rates of the curcumin from calcium alginate beads and will be helpful for the application of the systems in controlled release of other hydrophobic drug.

Disordered reactive oxygen/nitrogen species are a common occurrence in various diseases, which usually cause cellular oxidative damage and inflammation. Despite the wide range of applications for biomimetic nanoparticles with antioxidant or anti-inflammatory properties, designs that seamlessly integrate these two abilities with a synergistic effect in a simple manner are seldom reported. In this study, we developed a novel PEI-Mn composite nanoparticle (PM NP) using a chelation method, and the curcumin was loaded onto PM NPs via metal–phenol coordination to form PEI-Mn@curcumin nanoparticles (PMC NPs). PMC NPs possessed excellent dispersibility and cytocompatibility, was engineered to serve as an effective nanozyme, and exhibited specific SOD-like and CAT-like activities. In addition, the incorporation of curcumin granted PMC NPs the ability to effectively suppress the expression of inflammatory cytokines in microglia induced by LPS. As curcumin also has antioxidant properties, it further amplified the synergistic efficiency of ROS scavenging. Significantly, PMC NPs effectively scavenged ROS triggered by H2O2in SIM-A9 microglia cells and Neuro-2a cells. PMC NPs also considerably mitigated DNA and lipid oxidation in Neuro-2a cells and demonstrated an increase in cell viability under various H2O2concentrations. These properties suggest that PMC NPs have significant potential in addressing excessive ROS and inflammation related to neural diseases. Keywords:nanozyme;ROS scavenging;anti-inflammatory;neuroprotective

This study investigated the chitosan (CS)-octenyl succinic anhydride (OSA) starch complex as a stabilizer for Pickering emulsions. The effects of OSA concentration (0.5wt%-2.5wt%) and viscosity (OSA1, OSA2, OSA3) on the stability and physiochemical properties of emulsion were examined. The results showed that CS-OSA with 2.0wt% concentration OSA stabilized emulsion exhibited optimal emulsifying capacity. Contact angle analysis, Fourier transform infrared spectrometer (FTIR) and X-difference diffractometer (XRD) were utilized to explore the stabilized mechanism of CS-OSA on emulsion, results confirmed the CS and OSA interactions, and strong adsorption of CS-OSA particles at the oil-water interface. Furthermore, CS-OSA stabilized emulsion loading curcumin were assessed, results showed that curcumin loaded emulsion exhibited high stability and better antioxidant capacity than free CS during 10-day storage. The overall study offers valuable insights into the utilization of CS-OSA emulsifiers in delivery application for lipophilic bioactive molecules.

A simple and nontoxic fluorescent chemosensor of di- O -methyl curcumin has been prepared from curcumin. The sensor exhibited selective and sensitive fluorescent responses toward Al 3+ over a wide range of metal ions, such as Mn 2+ , Ce 3+ , Pt 2+ , Sn 4+ , Hg + , Sb 3+ , K + , Ca 2+ , Mg 2+ , Ba 2+ , Cu 2+ , Ni 2+ , Na + , NH 4 + , Ag + , Pb 2+ , Zn 2+ , Fe 2+ , Fe 3+ , Hg 2+ and Cr 3+ in ethanol/water. The free ligand showed quite weak fluorescence emission due to the isomerization of CO double bond in the excited state, however, after addition of Al 3+ , fluorescence emission results in a prominent fluorescence enhancement.

Interactions between nanoparticles and viruses have attracted increasing attention due to the antiviral activity of nanoparticles and the resulting possibility to be employed as biomedical interventions. In this contribution, we developed a very simple route to prepare uniform and stable silver nanoparticles (AgNPs) with antiviral properties by using curcumin, which is a member of the ginger family isolated from rhizomes of the perennial herb Curcuma longa and has a wide range of biological activities like antioxidant, antifungal, antibacterial and anti-inflammatory effects, and acts as reducing and capping agents in this synthetic route. The tissue culture infectious dose (TCID50) assay showed that the curcumin modified silver nanoparticles (cAgNPs) have a highly efficient inhibition effect against respiratory syncytial virus (RSV) infection, giving a decrease of viral titers about two orders of magnitude at the concentration of cAgNPs under which no toxicity was found to the host cells. Mechanism investigations showed that cAgNPs could prevent RSV from infecting the host cells by inactivating the virus directly, indicating that cAgNPs are a novel promising efficient virucide for RSV.

Co-Encapsulated angiostatin (AS) and curcumin (Cur) in poly(ethylene glycol)-b-poly(ε-caprolactone) polymersomes (PEG–PCL-PMs) were prepared by film hydration method. The size and zeta potential of the prepared PEG–PCL-PMs were about 153 nm and −0.22 mV, respectively. Transmission electron microscopy was employed to confirm the polymersomal structure of PEG–PCL-PMs. In vitro drug release profiles showed a sustained release of angiostatin and curcumin from the dual-drug loaded polymersomes under physiological conditions (pH 7.4). In addition, confocal laser scanning microscopy and flow cytometry showed that drug-loaded polymersomes were delivered efficiently into the cytoplasm and nuclei of human microvascular endothelial cell line (HMEC-1) and then released active drugs in cells. Dual drug-loaded polymersomes (AS–Cur-PMs) remarkably inhibited proliferation and migration of HMEC-1 cells compared to those treated with the single drug. The chick chorioallantoic membrane assay showed that AS–Cur-PMs inhibited vessel sprouting in the chicken chorioallantoic membrane. Our data demonstrates a synergistic effect of angiostatin and curcumin in inhibiting angiogenesis, both in vitro and in vivo.

The diseases attributable to viruses remain a global burden. The respiratory syncytial virus (RSV), which is considered as the major viral pathogen of the lower respiratory tract of infants, has been implicated in severe lung disease. In this contribution, we developed a β-cyclodextrin (CD) functionalized graphene oxide (GO) composite, which displayed excellent antiviral activity and could load curcumin efficiently. RSV, a negative-sense single-stranded enveloped RNA virus, was employed as a model virus to investigate the antiviral activity of multifunctional GO. Proved by the tissue culture infectious dose assay and immunofluorescence assay, the curcumin loaded functional GO was confirmed with highly efficient inhibition for RSV infection and great biocompatibility to the host cells. The results showed that the composite could prevent RSV from infecting the host cells by directly inactivating the virus and inhibiting the viral attachment, and possessed prophylactic and therapeutic effects towards the virus. Our data indicate that the composite may provide new insights into antiviral therapy for RSV infection.

Quinocetone (QCT), a new quinoxaline 1,4-dioxides, has been used as antimicrobial feed additive in China. Potential genotoxicity of QCT was concerned as a public health problem. This study aimed to investigate the protective effect of curcumin on QCT-induced oxidative stress and genotoxicity in human hepatocyte L02 cells. Cell viability and intracellular reactive oxygen species (ROS), biomarkers of oxidative stress including superoxide dismutase (SOD) activity and glutathione (GSH) level were measured. Meanwhile, comet assay and micronucleus assay were carried out to evaluate genotoxicity. The results showed that, compared to the control group, QCT at the concentration ranges of 2–16 μg/mL significantly decreased L02 cell viability, which was significantly attenuated with curcumin pretreatment (2.5 and 5 μM). In addition, QCT significantly increased cell oxidative stress, characterized by increases of intracellular ROS level, while decreased endogenous antioxidant biomarkers GSH level and SOD activity (all p < 0.05 or 0.01). Curcumin pretreatment significantly attenuated ROS formation, inhibited the decreases of SOD activity and GSH level. Furthermore, curcumin significantly reduced QCT-induced DNA fragments and micronuclei formation. These data suggest that curcumin could attenuate QCT-induced cytotoxicity and genotoxicity in L02 cells, which may be attributed to ROS scavenging and anti-oxidative ability of curcumin. Importantly, consumption of curcumin may be a plausible way to prevent quinoxaline 1,4-dioxides-mediated oxidative stress and genotoxicity in human or animals.

A facile approach to fabricate micro-complex in moderate condition via self-assembly of κ -carrageenan (CRG) and lysozyme (Lys) was investigated for the first time to encapsulate and protect curcumin . The interaction between CRG and Lys, size and morphology were studied by zeta potential, UV–vis spectroscopy, circular dichroism (CD), scanning electron microscope (SEM), transmission electron microscopy (TEM) and X-ray photoelectron spectroscopy (XPS). A tentative interactive model was proposed for polysaccharide and protein. The results indicated that the spherical shaped CRG/Lys complex with 1 μm diameter was spontaneously formed by one spot incubating. Both CRG and Lys were participated in forming the shell of micro-complex synthesized through physical interactions. The encapsulation efficiency (EE) and loading capacity (LC) of curcumin for the CRG/Lys complex prepared with CRG/Lys ratio 2:1 were 71 ± 0.02% and 50.71 ± 0.71 μg ml − 1 respectively. The stability of curcumin loaded in complex was improved about 2.7 fold, 2 fold and 1.7 fold when disposed with 80 °C treatment for 1 min, 60 °C treatment for 30 min and ultraviolet radiation for 2 h. DPPH measurement verified the protection of bioactivity as encapsulated. The worthwhile endeavor elucidated proteins/polysaccharides complex was feasible to solubilize and protect sensitive amphiphilic bioactive compounds and has extensive potential in food and medicinal application with various purposes.

To precisely manipulate the intracellular delivery of pleiotropic curcumin , this work reports two types of acid-responsive polymer–curcumin conjugates with different structures. One or two amphiphilic poly(ethylene glycol)-co-poly(lactic acid) (PEG-PLA) copolymer(s) were linked to curcumin via pH-liable hydrazone, producing a linear or phospholipid-like conjugate, respectively. Both conjugates efficiently self-assembled into micellar nanocarriers with enhanced stability in contrast to the paralleling PEG-PLA micelles. In comparison to the micelles assembled by phospholipid-like conjugates, the linear conjugate micelles exhibited similar size, doubled loading dose, higher release rate, and hence enhanced cellular uptake and cytotoxicity at the cost of increased critical micelle concentration. This work highlighted the importance of precise tuning of polymer–drug conjugate architecture in determining the pharmaceutical properties and thus delivery efficiency of the corresponding conjugate micelles.

Background The current study aimed to investigate whether the novel folate (FT) modified nanoparticles (NPs) co-loaded with docetaxel (DT) and curcumin (CU) (named as FT-NPs) could enhance the delivery efficiency to tumor compared with the NPs without FT (non-targeted NPs). Methods FT-NPs were successfully formulated in this article. In vitro cytotoxic activity against A549 cells and in vivo antitumor activity of FT-NPs in S180 cell bearing mice were conducted. Cellular uptake test was used to evaluate uptake efficiency of FT-NPs. Histological observation was used to determine the lung security. Besides, the physical chemical properties such as stability, particle size, zeta potential , drug encapsulation efficiency, transmission electron microscopy (TEM) were also conducted. Results FT-NPs exhibited stronger growth inhibition effects on A549 cells compared with non-targeted NPs, moreover, the novel FT-NPs indicated more effective antitumor efficacy in inhibiting tumor growth. Confocal laser scanning microscopy indicated that the uptake of FT-NPs was facilitated and effective. Histological observation meant that FT-NPs were biocompatible and appropriate for pulmonary administration. Conclusion These results confirmed that FT-NPs with relatively high drug loading capacity could effectively inhibit tumor growth and reduce toxicity. The novel FT-NPs could produce as an outstanding nanocarrier for the targeted treatment of cancers .
