A synthetic system for RNA-responsive pyroptosis based on type III-E CRISPR nuclease-protease

He Mingbin,Wang Weiwei,Zhou Haiwu,Liu Cong,Zhao Chunbei,Li Jian,Han Yuewen,Qin Yali,Chen Mingzhou

Pyroptosis plays a crucial role in immune defense against infections and endogenous threats by eliminating harmful cells and modulating the immune response through inflammation. However, the natural activation of pyroptosis involves intricate signaling pathways, posing significant challenges for its artificial manipulation in research and therapies. Here, we present DAMAGE ( D e a th Ma nipulation Ge ne), an innovative system that integrates gasdermins within the type III-E CRISPR framework, enabling the specific recognition of target RNA (tgRNA) and triggering pyroptosis. This mechanism allows DAMAGE to selectively identify and eliminate virus-infected, cancerous, and senescent cells, all of which exhibit altered RNA transcriptomes. Additionally, DAMAGE exhibits considerable promise as a platform for mRNA-LNP therapy. Our study highlights the potential of this CRISPR-based system in the controllable induction of pyroptosis, offering an innovative therapeutic strategy for treating RNA-heterogeneous diseases.

An iron-doped Cu2O nanozyme enables highly sensitive colorimetric detection of vitamin E in serum

Xiangyu Feng,Haibing Zhu,Chuanli Ren,Fu-an Wang,Zhanjun Yang,Guiling Li

Vitamin E plays a vital role in maintaining human health as well as in the prevention and treatment of related diseases. However, current methods for detecting vitamin E suffer from complex instrumentation, high cost, and cumbersome operation, making it difficult to meet the demand for rapid detection in serum samples. Herein, we designed an iron-doped cuprous oxide (Fe-Cu2O) nanozyme for the colorimetric detection of vitamin E in human serum. Impressively, the Fe-Cu2O nanozyme exhibits a 7.2-fold enhancement in peroxidase (POD)-like activity compared with the pure Cu2O nanozyme. In the presence of hydrogen peroxide (H2O2), the Fe-Cu2O nanozyme catalyzes the decomposition of H2O2 to generate hydroxyl radicals (˙OH), thereby oxidizing colorless 3,3′,5,5′-tetramethylbenzidine (TMB) into a blue product (ox TMB). As a reducing agent, vitamin E can fade the blue ox TMB, enabling the quantitative determination of vitamin E. Under optimized experimental conditions, the as-prepared Fe-Cu2O nanozyme achieves a linear detection range of 2–80 µM toward vitamin E with a limit of detection (LOD) of 1.89 µM. This approach holds promise as a straightforward, economical, and highly effective strategy for clinical diagnosis and health screening of vitamin E and other bioactive molecules.

Developing an E. coli heterologous expression system for characterizing a marine debrominase from Roseobacter sp.

Xiaofang Li,Sen Yang,Yuping Liu,Hua Huang,Xinshuai Zhang,Yin Zhong,Lorenz Adrian,Ping’an Peng

Reductive dehalogenases (RDases) are a family of enzymes that employ a cobamide cofactor and two iron-sulfur clusters to selectively cleave carbon-halogen bonds. The identification of yet-uncharacterized RDases is crucial for advancing our understanding of dehalogenation processes in both natural and engineered environments, particularly for bioremediation. In this study, we developed a heterologous expression system in Escherichia coli DH5α to produce a chloroform-reducing RDase (TmrA) from Dehalobacter sp. UNSWDHB, using a pTrcHisA vector without requiring an additional vector for vitamin B12 transporters. By co-expressing the pRSFDuet-BtuCEDFB plasmid, we further enhanced active TmrA production without relying on the T7 promoter system. Using this system, we achieved the first heterologous expression of an RDase (the debrominase Ros_A3X954) from a marine heterotrophic Roseobacter strain, without chemical cofactor reconstitution. Ros_A3X954 catalyzes conversion of 2,6-dibromophenol to 2-bromophenol with a specific activity of 21.8 ± 1.0 nmol s−1 mg−1. The enzyme exhibited optimal activity at pH 6.0 and demonstrated significant thermal stability by maintaining full catalytic capacity across a broad temperature range (20–50°C), a key advantage for applications in thermally fluctuating marine environments. These findings expand our understanding of RDase functional diversity in common marine microbiota and highlight the significant application potential of Roseobacter for bioremediation of bromophenol pollution.

Skin-mimicking biogel-based iontronic sensor with hierarchical bionic coupling for dexterous tactile e-skin

Yidan Chen,Shu Wang,Xilu Ye,Chenghui Lv,Jialin Wei,Yingxin Zhang,Jianfeng Ping,Yibin Ying,Lingyi Lan

The human fingertip skin, with its interlocked epidermal-dermal architecture and dense tactile receptors, enables unique perception and inspires the design of innovative electronic skin (e-skin). However, most e-skin materials fail to replicate the hierarchical integration of composition, structure, and function found in natural skin. Drawing direct inspiration from the dermal extracellular matrix–a composite hydrogel reinforced by a collagen-hyaluronic acid, here we report an e-skin that mimics key composition and architectural features of human skin. By incorporating sodium lactate and montmorillonite nanosheets into a gelatin matrix, we engineer a biomimic gel with mechanical and hydration properties similar to those of natural skin through dynamic ionic crosslinking and hydrogen bond networks. The microstructure resembling the dermal papillae is designed using a sandpaper-templating strategy. The resulting iontronic sensor achieves high sensitivity (466.3 kPa −1 ), rapid response (47 ms), and a wide pressure detection range (20 Pa–2000 kPa). We further develop a stretchable, ultrathin, hand-shaped iontronic sensor array that integrates seamlessly with a dexterous robotic hand, achieving precise, nondestructive grasping and high-fidelity, multichannel pressure mapping. Hydrogels are promising materials for electronic skins and devices, though it is challenging to mimic the composition and architectural features of human skin. Here the authors use a sandpaper templating strategy for a gelatin based hydrogel electronic skin.

An E-interwoven Therapeutic Contact Lens System for Rapid Drug Delivery and Precision Dose Monitoring

Huan Yang,Hengtian Zhu,Wenyu Teng,Heyu Huang,Zuoxiu Tie,Zixu Li,Zhenning Qi,Songtao Yuan,Zizhong Hu,Fei Xu

Precise intraocular pressure (IOP) management and rapid intervention preserve glaucoma vision, yet current ocular delivery systems lack in situ dosage verification, causing treatment blind spots. Here, we develop a wireless therapeutic smart contact lens (SCL) containing a rapid voltage-triggered drug delivery system with a real-time dosage monitoring sensor. The e-interwoven design integrates drug delivery and sensing electrodes in an ultra-thin (17.4 µm) tri-interdigital configuration, which enables the electric field to interact with the drug-loaded hydrogel in the same spatial region. A frequency-matching strategy boosts voltage coupling to drive a brimonidine tartrate-loaded hydrogel to achieve high concentrations in the aqueous humor in 20 min, significantly faster than topical eye drops. The built-in drug dose sensor has a high accuracy <2.8 µg, benefiting from the great linear correlation between wireless frequency drift and release dosage. In acute glaucoma rabbit models, this SCL suppresses peak IOP elevation by 85.7% compared to eye drops. This SCL presents a highly integrated therapeutic platform demonstrating potential for rapid and precise glaucoma treatment.

Antibacterial Mechanism of Allicin E Against Aeromonas hydrophila and Therapeutic Effect in Carassius auratus gibelio

Jinlong Li,Liushen Lu,Kai Chen,Ting Qin,Jun Xie,Ping Fang,Bingwen Xi

Background/Objectives: The frequent use of antibiotics has led to increasing drug resistance inAeromonas hydrophila; therefore, there is an urgent need to develop novel antimicrobial agents to prevent and control bacterial diseases in aquaculture. Allicin E (ALE) is derived from garlic (Allium sativumL.), a plant extensively used in traditional medicine for treating infections. This study aimed to evaluate the potential of ALE againstA. hydrophila, a major aquaculture pathogen, by investigating its antibacterial efficacy, mechanisms of action, and in vivo protective effects. Methods: The minimum inhibitory and bactericidal concentrations (MIC/MBC) were determined by broth microdilution. Antibacterial mechanisms were investigated through ROS detection, electron microscopy, fluorescent staining, and content leakage measurement. In vivo efficacy was evaluated inCarassius auratus gibelioby monitoring survival rates and bacterial loads, analyzing immune and antioxidant biomarkers, and histopathological analysis afterA. hydrophilachallenge. Results: ALE exhibited potent antibacterial activity (MIC = MBC = 8 μg/mL), achieving complete bacterial elimination within 1 h and showing a low resistance propensity. Mechanistically, ALE induced ROS accumulation, causing oxidative damage that disrupted membrane integrity and facilitated the leakage of cellular contents. In vivo, ALE significantly enhanced fish survival, reduced bacterial loads, modulated inflammatory cytokines, boosted antioxidant enzyme activities (SOD and CAT), and alleviated tissue damage. Conclusions: ALE possesses potent in vitro antibacterial activity and exerts an inhibitory effect on bacteria-induced inflammatory responses, effectively combatingA. hydrophilathrough a multi-target mechanism and enhancing host resistance.

Fabrication of Beta Zeolite Enriched in Polymorph E for Highly Efficient Etherification of Furfuryl Alcohol

Kun Lu,Liyu Chen,Yaqi Fan,Wenhao Zhang,Qian Liu,Shasha Qu,Xiao Kong,Yanhang Ma

Zeolite Beta, a prototypical intergrowth zeolite with a three-dimensional (3D) system of intersecting 12-member-ring micropore channels, is extensively employed as a versatile solid-acid catalyst. Its structure typically appears as a nanoscale intergrowth of polymorphs arising from the stacking disorder of topologically equivalent layers. Rational regulation of the Beta polymorph composition is, therefore, crucial for optimizing catalytic performance. Here, we report a heterostructure seed-assisted crystallization strategy to synthesize a polymorph E-enriched Beta zeolite (Beta-ER). The 3D electron diffraction, high-resolution transmission electron microscopy imaging, and DIFFaX simulation show that Beta-ER is an intergrown structure of polymorph E/D with polymorph E as the main phase (∼80%). Powder X-ray diffraction, Fourier transform infrared, Raman, and 19F magic-angle spinning nuclear magnetic resonance characterizations suggest that ITQ-1 seeds partially deconstruct in the early stage of crystallization and generate secondary structural building units, which subsequently promote the formation of Beta-ER. In the etherification of FA, Beta-ER with moderate acidity and open pore channels delivers a higher furfuryl ethyl ether (FEE) yield (75.4%) after 30 min at 393 K, outperforming the commercial ZSM-5 (14.2%), Beta (55.9%), and BEC (57.2%) zeolites. Beta-ER maintained stable activity after four regeneration cycles. Density functional theory calculations confirm that substituting framework Al with Ge increases the energy barrier for the ring opening of FEE, thereby suppressing the formation of byproducts. In addition, kinetic analysis and molecular dynamics simulations reveal that the interconnected channel system of Beta-ER facilitates the diffusion of substrates and products.

Fully-Printed Bimodal E-Textiles with Synchronized Temperature-Pressure Sensing for Next-Generation Adaptive Wearables

Ziyi Gong,Ke Zheng,Qian Wang,Ruoyu Lai,Jian Zou,Xin Guo,Xiao Chen,Wei Wu

The growing demand for integrated temperature-pressure sensing in human-machine interaction and health monitoring makes smart textiles an ideal medium due to their comfort, breathability, and flexibility. However, a key challenge is finding a reliable method to integrate high-performance dual-mode sensors into textile substrates while ensuring effective signal decoupling. Herein, a flexible, ultrathin, and signal-decoupled temperature-pressure dual-mode sensing electronic textile is proposed. The resistive temperature sensing unit utilizes a multi-stage phase transition mechanism to achieve an ultrahigh temperature coefficient of resistance (up to 125.9%/°C) and a broad temperature sensing range (0°C–50°C). The capacitive pressure sensing unit exhibits high sensitivity (up to 62.3 kPa −1 ) and rapid pressure response and recovery times (30 ms/60 ms). By integrating machine learning algorithms, precise recognition of different objects being grasped is achieved. Furthermore, through the fabrication of a sensor array, spatial mapping of pressure and temperature distributions on a simulated body surface is successfully realized, offering a promising route toward developing versatile health-monitoring platforms.

Allicin E enhances the antibacterial activity of enrofloxacin against Aeromonas hydrophila in Carassius auratus gibelio

Lu Liushen,Li Jinlong,Qin Ting,Chen Kai,Xie Jun,Ping Fang,Xi Bingwen

Aeromonas spp. are major pathogens severely affecting the Carassius auratus gibelio farming industry. The prolonged use of enrofloxacin (Enr) has led to reduced therapeutic efficacy, drug residues, and the emergence of antibiotic resistance. Simply increasing antibiotic dosage is unsustainable, and the core challenge lies in potentiating antibiotic efficacy without dose escalation. Here, we employed combination therapy using allicin E (ALE) as a potentiator of Enr and investigated its synergistic antibacterial activity, underlying mechanisms, and protective effects against A. hydrophila in C. auratus gibelio . In vitro, the combination of Enr and ALE exhibited broad-spectrum synergistic or additive activity against multiple aquatic pathogens, rapidly eradicating A. hydrophila without regrowth during the observation period. Mechanistically, the combination triggered a burst of bacterial reactive oxygen species (1.67-fold vs. control), leading to irreversible membrane damage and leakage of intracellular contents, and significantly inhibited biofilm formation and disrupted mature biofilms. In vivo, oral administration of the combination in C. auratus gibelio challenged with A. hydrophila significantly improved survival (66.6% vs. 26.6% in model), reduced bacterial loads in liver, spleen and kidney (by 70.9%, 81.3% and 72.9%, respectively, vs. model group), and mitigated oxidative damage by restored antioxidant enzymes (T-SOD, CAT) activity, decrease MDA, as well as attenuated inflammatory infiltration and hemorrhage in spleen and kidney tissues. This is the first report of ALE potentiating Enr against A. hydrophila via a synergistic ROS-mediated membrane damage, validated in C. auratus gibelio , with approximately 50% Enr reduction potential.

Integrating Membrane Permeability Engineering and Enzyme Design for Efficient Production of Ergothioneine in E. coli

Jiahuan Ling,Rui Chen,Minghai Wang,Chun-Xiao Yan,Ruomu Xia,Lihui Zhang

Ergothioneine (EGT) is a powerful and natural antioxidant, which can protect cells in the human body from oxidative damage. Numerous studies have attempted to enhance the heterologous synthesis. However, research on EGT transport out of Escherichia coli cells and the wild-type enzyme from Trichoderma reesei is rare. Here, membrane permeability engineering and protein engineering are combined to improve EGT production in E. coli. After metabolic engineering to enhance precursor supply, we deleted the genes involved in lipopolysaccharide biosynthesis to increase the membrane permeability, which promoted EGT production. Further, a mutant of Tregt2E155C with improved catalytic capacity was created, and the expression level of enzymes involved in EGT synthesis was optimized. Finally, fermentation parameters were systematically tuned to maximize the EGT production. The engineered strain MT9-PET-T1/RSF-T2E155C produced 334.20 ± 6.33 mg/L EGT during 48 h of shake-flask fermentation, corresponding to an 8.4-fold increase compared with wild-type strain MT1. When scaled up to 5 L bioreactors, the strain achieved a final EGT titer of 4.06 g/L within 96 h (42.29 mg/L/h). Our work shows significant implications for EGT synthesis and is also a reference for manipulating E. coli to synthesize other biomolecules.

Chitosan Nanoparticles Loaded with Truncated ORF2 Protein as an Oral Vaccine Candidate against Hepatitis E

Wenjuan Wei,Nouredine Behloul,Weiqi Wang,Sarra Baha,Zhenzhen Liu,Ruihua Shi,Jihong Meng

In a continuous effort to develop effective vaccines against hepatitis E (HE), oral vaccine nanoparticles using the truncated capsid protein p146 (aa460–605) are formulated and characterized. To improve the immunogenicity of p146, chitosan nanoparticles (CSNPs) are used as a mucosal delivery system. Next, the physical-chemical properties, cytotoxic effects in vitro, and immunogenicity in mice of the produced NPs are analyzed. The results show that the produced CS/p146 NPs are stable and well dispersive and display a near-spherical shape with a mean size of 200–300 nm. The findings also demonstrate high encapsulation efficiency (65–73.9%) and loading capacity (27.7–67.5%) of the formulated nanoparticles. Further, the CS/p146 NPs exhibit low cytotoxicity and an obvious sustained-release effect in vitro. Immunogenicity experiments in mice indicate that CS/p146 NPs can induce antigen-specific systemic and mucosal immune responses higher than the purified p146 do. Besides, the expression levels and mRNA transcription of Interleukin (IL)-4 in spleen cells of CS/p146 NPs-immunized mice are higher than those of p146, indicating that a Th2-mediated cellular immune response is activated by the CS/p146 NPs. Overall, the synthesized CS/p146 NPs display promising properties as a potential HE oral vaccine candidate.

Synthesis of sulfonated chitosan and its antibiofilm formation activity against E. coli and S. aureus

Jianying Huang,Yuhong Liu,Lijun Yang,Fengyan Zhou

A sulfonated chitosan (SCS) was prepared via nucleophilic substitution and characterized by Fourier transform infrared spectroscopy, 1 H and 13 C NMR spectra, gel permeation chromatography, elemental analysis, and thermo gravimetric analysis. The inhibition activities of bacterial adhesion and biofilm formation against E. coli and S. aureus of SCS were assessed in comparison with those of unmodified chitosan hydrochloride (WCS) which was commercially available. The metabolic activity and secretion of exopolysaccharide in biofilms of E. coli and S. aureus were significantly decreased after the treatment of SCS. Scanning electron microscopy and confocal laser scanning microscopy also demonstrated that SCS and WCS at 1MIC concentrations could obviously inhibit the formation of biofilm. Upon the experimental data obtained, it can be concluded that the alkylsulfonation of chitosan could significantly improve efficacy in killing biofilm-embedded bacteria, and the inhibition activities against biofilm formation of E. coli and S. aureus . The exploitation of SCS in this study is helpful to extend the understanding to an alternative to antibiotics and chemical preservatives in food and medicine fields.
