The most common in vitro cell culture platform, standard two-dimensional (2D) monolayer cell culture, often fails to mimic the tumor microenvironment, while animal models complicate research on the effect of individual factors on cell behaviors. Both are unsatisfactory in the research of molecular mechanisms of tumor development and progression and the discovery and development of anticancer drugs. In vitro three-dimensional (3D) cell culture can partially simulate in vivo conditions and 3D-cultured cancer cells can recapture many essential features of native tumor tissues. In this study, to mimic the in vivo breast tumor microenvironment, novel reduction-responsive poly(Nε-acryloyl L-lysine) (pLysAAm) hydrogels were synthesized by rapid photo-polymerization of Nε-acryloyl L-lysine and using N,N′-bis(acryloyl)-(L)-cystine as a crosslinker, and their physicochemical properties were characterized systemically. The results showed that the pLysAAm hydrogels were formed within 93 s under UV irradiation and exhibited almost total elastic recovery from compressions as high as 75%. The lyophilized hydrogel samples displayed a highly porous structure with interconnected pores, had an equilibrium swelling ratio of about 20, and were degraded faster in a glutathione-containing solution than in PBS solution. The biological versatility of the pLysAAm hydrogels was demonstrated by both in vitro MCF-7 cell culture and in vivo tumor formation. Compared to cells cultured as 2D monolayers, the 3D-cultured cells presented 3D cell morphology, exhibited better cell viability, expressed higher levels of pro-angiogenic factors, and showed significantly greater migration and invasion abilities. The results from assay of tumorigenicity in nude mice and histologic analysis demonstrated the enhanced tumorigenic and angiogenic capabilities of the MCF-7 cells pre-cultured in pLysAAm hydrogels. These findings suggest that pLysAAm hydrogels may be used to bridge the gap between standard in vitro cell cultures and living tissues, aid breast cancer research, and help researchers to develop novel anticancer therapeutics.

A series of novel pendant polymers, composed of a poly(ε-lysine) (ε-PL) (w: 4,000 g mol−1) main chain and three kinds of amino acid β-cyclodextrin (CD) derivatives, were prepared by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride and N-hydroxysuccinimide-assisted synthesis and characterized by nuclear magnetic resonance, Fourier transform infrared spectroscopy, thermal gravimetric analysis, X-ray power diffraction, and scanning electron microscopy. Cytotoxicity and moisture absorption of the pendant polymers were also tested. Compared with ε-PL, all the pendant polymers exhibited lower moisture absorption and cytotoxicity. The synthesis method of the pendant polymers is simple, feasible, and has good practicability. We suggest these polymers have potential applications in many fields, especially in gene delivery vector and drug delivery systems.

N 6 -acetyl- L - lysine residue is abundant in dietary protein but little is known about its potential influences on the diet-consumers. Herein, it is reported that Lysyl-tRNA synthetase (KARS) mediates co-translational deposition of diet-derived N 6 -acetyl- L -lysine (AcK) in nascent proteins to contribute to the acetylome in cells. Acetylated dietary protein is a direct source of AcK that can widely and substantially regulate the acetylome in multiple organs of mice. By analyzing the mechanisms underlying AcK contributing to the acetylome in mammalian cells, it is found that KARS can utilize AcK as an alternative substrate to produce N 6 -acetyl- l -lysyl-tRNA. The crystal structure of KARS in complex with AcK at 2.26 Å resolution shows that AcK shares the same substrate-binding pocket as L -lysine, allowed by a sidechain flip of Tyr499. The generated N 6 -acetyl- L -lysyl-tRNA introduces AcK into growing nascent polypeptide and results in protein acetylation, including the regions buried inside folded proteins that are post-translational modification (PTM)-inaccessible and functionally important. This undocumented protein modification mechanism is inherently different from PTM and termed as co-translational modification (coTM). It is expected to extend the repertoire of acetylome and improve the understanding of protein modification mechanisms in cells.

Antibacterial hydrogels as burn wound dressings are capable of efficaciously defending against bacterial infection and accelerating burn wound healing. Thus far, a large plethora of antibacterial hydrogels have adopted numerous components and intricate preparation processes, yet restricting their practical industrialization applications. Simple and effective preparation methods of antibacterial hydrogels are hence urgently needed. Herein, an easy but efficacious strategy with the employment of two natural products pullulan and ε-poly- l -lysine (ε-PL) was designed to fabricate composite antibacterial hydrogels for burn wound healing for the first time. The hydrogel crosslinking networks were formed through amidation reactions between carboxylated pullulan derivative (CP) and ε-poly- l -lysine hydrochloride (ε-PL·HCl). The resulting hydrogels possessed high transparency, porous structures, tunable gelation time and gel content, relatively low swelling ratios, appropriate self-degradability, proper mechanical properties, strong in vitro bacteriostatic activities, non-cytotoxicity, capacities of facilitating cell migration and excellent hemocompatibility. In the infected burn model of mice, the hydrogels were observed to display prominent in vivo antibacterial activities and enable the acceleration of burn wound healing. We opine the simply and effectively prepared antibacterial hydrogels as promising dressings for burn wound recovery have broad industrialization prospects.

Lonicera japonica plays a significant role in traditional Chinese medicine and as a food source, making it a focus of studies on protein succinylation and its potential role in regulating secondary metabolism during flower development. This study aimed to clarify the regulatory mechanism of protein succinylation on phenylpropanoid-related phenotypic changes by conducting a global lysine succinylation proteomic analysis across different flowering stages. A total of 586 lysine succinylated peptides in 303 proteins were identified during early and late floral stages. Functional enrichment analysis revealed that succinylated proteins primarily participated in the tricarboxylic acid (TCA) cycle, amino acid metabolism, and secondary metabolism. The abundance of succinylated aspartate transaminase (AT), 4-coumarate-CoA ligase (4CL), and phenylalanine N-hydroxylase (CYP79A2) in phenylpropanoid metabolism varied during flower development. In vitro experiments demonstrated that succinylation increased AT activity while inhibited 4CL activity. Decreased levels of total flavonoids and phenolic acids indicated significant alterations in phenylpropanoid metabolism during later floral stages. These results suggest that succinylation of TCA cycle proteins not only influences flower development but also, together with AT-4CL-CYP79A2 co-succinylation, redirects phenylpropanoid metabolism during flower development in L. japonica .

This study explored the mechanism of l -lysine intervention in wheat gluten protein (WG) gel formation under a microwave (MW) field. The results showed that the MW treatment had higher ζ -potential values at the same heating rate. After adding l -lysine, the solution conductivity and dielectric loss were significantly increased. Moreover, the WG gel strength enhanced 4.40% under the MW treatment. The Fourier spectra showed that the α-helix content was decreased 13.78% with the addition of lysine. The ultraviolet absorption spectra and fluorescence spectra indicated that MW irradiation impacted the interactions between WG molecules more effectively than the water bath heating, promoting the denaturation and unfolding of the protein structure. In addition, scanning electron microscopy analysis showed that the incorporation of lysine promoted an ordered network structure formation of the protein, which enhanced the gel properties. This indicated that the zwitterion of l -lysine played a regulatory role in the aggregation of proteins in the MW field.

In this work, the feasibility of a novel sensitive electrochemiluminescence aptasensor for the detection of lysozyme using Ru(bpy) 3 2+ [email protected] L -lysine-Au ( [email protected] ) nanocomposites labeling as an indicator was demonstrated. The substrate electrode of the aptasensor was prepared by depositing gold nanoparticles (AuNPs) on 3D graphene-modified electrode. The lysozyme binding aptamer (LBA) was attached to the 3D graphene/AuNPs electrode through gold-thiol affinity, hybridized with a complementary single-strand DNA (CDNA) of the lysozyme aptamer labeled by [email protected] as an electrochemiluminescence intensity amplifier. Thanks to the synergistic amplification of the 3D graphene, the AuNPs and [email protected] NPs linked to Ru(bpy) 3 2+ -ECL further enhanced the ECL intensity of the aptasensor. In presence of lysozyme, the CDNA segment of the self-assembled duplex was displaced by the lysozyme, resulting in decreased electrochemiluminescence signal. Under the optimized conditions, the decrease in electrochemiluminescence intensity varied proportionally with the logarithmic concentration of the lysozyme from 2.25 × 10 –12 to 5.0 × 10 −8 mol L −1 , and the detection limit was estimated to 7.5 × 10 –13 mol L −1 . The aptasensor was further tested in real samples and found reliable for the detection of lysozyme, thus holding great potential application in food safety researches and bioassay analysis.

A novel biodegradable and noncytotoxic polycaprolactone-polyethyleneglycol-ε-poly(L-lysine) (PCL-PEG-EPL) triblock copolymer was synthesized via the combination of ring-opening polymerization and facile coupling reactions. The triblock copolymer can self-assemble into vesicles in aqueous solution, and PCL forms the vesicle membrane, whereas the PEG and EPL blocks constitute the vesicle coronas. Atomic force microscopy (AFM) and transmission electron microscopy (TEM) had proved its vesicular structure. The excellent antibacterial ability of the ε-poly(L-lysine) makes the vesicles have good antibacterial activities towards both E. coli (Gram negative) and S. aureus (Gram positive), and the minimal inhibitory concentrations (MICs) were both 62.5 μg mL−1. Moreover, the special bacterial-killing mechanism of antibacterial peptides may avoid producing antibiotic-resistant bacteria. Besides, the cell viability assays showed that the triblock copolymer was noncytotoxic up to a tested concentration of 1000 μg mL−1. Therefore, the copolymer vesicles have high selectivity (1000/62.5). The DOX drug loading and release profile suggested that the triblock copolymer vesicles have drug releasing ability. At the same time, in vitro enzymatic biodegradation experiments proved that the triblock copolymer vesicles could be degraded in the existence of lipid enzyme. All these results indicate that these multifunctional triblock copolymer vesicles with much lower cytotoxicity, showing antibacterial activities (without antibiotic resistance), can be a promising substitute for antibiotics. In addition, it can be used as an “armed” delivery template for anticancer drugs due to its drug delivery capacity.

Drug-resistant bacterial infections and biofilm formation have long posed significant challenges to public health, particularly in the context of chronic wound healing. In this study, the CuHspMOF were derived from copper and hesperitin (Hsp). The photocurable Epsilon-poly-l-lysine (EPLMA) and blueberry anthocyanin (BA) were synthesized through free radical polymerization. The multifunctional EB@CuHsp hydrogel exhibits excellent photothermal effect. One of the key features of the EB@CuHsp hydrogel is its ability to monitor the healing environment through color changes in response to variations in pH. This property allows for real-time assessment of the wound’s pH levels, reflecting the body fluid’s pH range during the healing process (pH 5–9). The hydrogel components work synergistically to facilitate wound healing. The EPLMA, with its positively charged, could captures bacteria and enable copper ions to eradicate bacteria at lower concentrations. This dual action leads to the creation of an optimal healing environment conducive to wound healing. During the inflammatory phase, BA and Hsp could collaborate to combat inflammation, which transited from the inflammatory stage to the proliferative phase soon. Continuous release of BA and Cu 2+ , coupled with thermotherapy, significantly enhances epithelial and blood vessel regeneration, thereby expediting the healing of chronic wounds. Overall, this multifunctional composite hydrogel holds great promise for promoting the regeneration of infected wound tissue, making it a potential candidate for chronic wound dressings.

The aim was to evaluate the protective effects of 0.4% L-lysine (Lys) on the gel strength, water holding capacity and protein structure of frozen silver carp surimi, and to explore the mechanism of Lys delaying the quality deterioration of frozen silver carp surimi. Compared with control groups and commercial antifreeze agents, Lys-treated frozen surimi demonstrated superior rheological properties, gel strength, water holding capacity, and whiteness after 30 d of frozen storage while exhibiting a compact gel network. The mechanism of action was mainly due to the rise of pH value in the freezing process to form myofibrillar protein aggregates that exhibited high resistance to degradation, which prevented the exposure of hydrophobic amino acids and the oxidation of sulfhydryl groups, thus affecting the effective cross-linking of proteins during the gelation process and forming a denser fiber structure. The findings demonstrated the potential of Lys as a cryoprotectant, thereby establishing a theoretical foundation for the development of eco-friendly alternatives to antifreeze and high-gel frozen surimi products.

Denervation-induced muscle atrophy causes a 40–50% reduction in muscle fiber size within 2 weeks. This negatively impacts muscle quality and function. Oral nonsteroidal anti-inflammatory drugs (NSAIDs) can reduce muscle atrophy by 20%. However, their bioavailability and gastrointestinal side effects raise concerns. Furthermore, multiple intramuscular injections can be difficult for patients. This is due to tolerance issues and discomfort from high doses. Muscle-targeted sustained-release delivery offers a solution by avoiding gastrointestinal problems and first-pass effects. Here, we present a self-powered gelatin nanofiber membrane (NFM) designed for the sustained release of aspirin/lysine (one NSAID). This method eliminates the need for repeated injections and directly targets inflammatory factors and superoxide in denervated muscles. As a result, it leads to a 44% increase in muscle weight and improved functional capacity in daily movements. Although the NFM requires implantation, its self-powered stimulation promotes muscle quality and enhances further drug release. Additionally, exercise-induced stimulation can intelligently control drug delivery through the self-powered nanofibers. Transcriptomic studies have confirmed that the NFM regulated muscle angiogenesis via inhibiting renin-angiotensin signal. Furthermore, its anti-inflammatory and antioxidant effects were enhanced with the incorporation of aspirin/lysine.

The recognition of amino acid enantiomers is of great significance in drug research and disease diagnosis. However, the similar physical and chemical properties of enantiomers make chiral recognition challenging. Optical characterization is a promising approach to distinguish enantiomers. Carbon dots (CDs) have attracted widespread attention due to their simple synthesis, high economic benefits, and good biocompatibility. Chiral carbon dots, as fluorescent probes, have gradually received more attraction. Here, a fluorescence probe (CYS-CDs + Cu 2+ ) was constructed using D-Cys as chiral source to synthesize chiral carbon dots (CYS-CDs), and Cu 2+ as inducer to recognize the chirality of lysine enantiomers(L-/D-Lys). And then, based on the ability of Cu 2+ to quench the fluorescence of CYS-CDs and L-Lysine (L-Lys) could restore the fluorescence of the CYS-CDs + Cu 2+ probe, so achieve an “on–off-on” detection mode to detect L-Lys. On the contrary, D-Lys cannot restore the fluorescence of CYS-CDs + Cu 2+ . The probe can distinguish lysine enantiomers not only by fluorescence spectra but also by circular dichroism spectra. And, the probe can be used for the quantitative detection of L-Lys by fluorescence method, of which the detection range is 0–520 μM, and the limit of detection is only 13.70 μM. In addition, a paper-based fluorescent senor was constructed on this basis. This work may be of practical significance to expand the recognition method of amino acid enantiomers.
