A new approach for ionic liquid-based enzymatic hydrolysis in situ trans -resveratrol extraction (ILEHE) from tree peony seed oil extracted residues is presented, in which enzymatic hydrolysis is used in an ionic liquid aqueous medium to enhance the trans -resveratrol yield. Various factors of the ILEHE procedure, including the variety and concentration of enzyme, type and concentration of ionic liquid, liquid–solid ratio and pH of the solution system, and ILEHE temperature and time, were investigated by single-factor experiments, response surface methodology, and first-order kinetic models. A satisfactory yield of trans -resveratrol (5.48 ± 0.14 μmol/g) was obtained by the novel developed approach compared with other conventional techniques. Scanning electronic microscopy of the samples indicated that tree peony seed treated by mixed cellulase and pectinase (1/1, w/w) enzymes in an ionic liquid led to more efficient extraction by reducing mass transfer barrier. The proposed ILEHE method was validated by stability, repeatability, and recovery experiments and shows promising prospects in the extraction and hydrolysis aspects, for enhancing efficiency of target components in natural products.

1-Ethoxy-2,3-difluoro-4-(trans-4-propylcyclohexyl) benzene
(EDPrB),
a highly polluting fluorinated liquid-crystal monomers (FLCMs), accumulates
in the kidneys over the long term. First, our study demonstrates that
exposure to EDPrB can induce inflammatory responses and fibrosis in
human renal cortical proximal tubule epithelial cells (HK-2), posing
a risk of nephrotoxicity. Next, our study investigates the renal injury
induced by EDPrB in male Kunming mice after 70-day exposure at 13,
130, and 1300 μg/kg bw/day, using an adverse outcome pathway
(AOP) framework. Histopathological observations demonstrate that EDPrB
corresponds to an elevated probability of developing chronic kidney
disease (CKD). Proteomics and Western blot analysis revealed that
mTOR signaling may be the mechanism by which EDPrB induces CKD in
mice and causes damage to HK-2 cells. Finally, by treating EDPrB-exposed
mice with RAPA, we demonstrated that activation of the mTOR signaling
is the molecular initiating event driving EDPrB-induced CKD in mice.
This study proposes an AOP framework focused on EDPrB-induced CKD.
The molecular initiating event is identified as mTOR activation, followed
by autophagy inhibition leading to inflammation and fibrosis as key
events, ultimately resulting in CKD as an adverse outcome. This highlights
the renal risks of FLCMs and supports stricter environmental release
regulations.

ESO acts as a multifunctional interfacial modifier in NR/BR/TBIR composites. • ESO improves matrix compatibility and filler dispersion in the composites. • Optimal ESO enhances strength, thermal behavior and wear resistance. • Interfacial regulation reduces DIN abrasion and stabilizes wear mechanism. Epoxidized soybean oil (ESO) was employed as a multifunctional interfacial modifier to regulate matrix compatibility, filler dispersion, and interfacial interactions in natural rubber/cis-1,4-polybutadiene rubber/trans-1,4-poly(isoprene-co-butadiene) rubber (NR/BR/TBIR) composites. The effects of ESO on rubber-rubber compatibility, filler network formation, and rubber-filler interfacial adhesion were systematically investigated, together with the resulting mechanical, thermal, and wear-resistant properties. The incorporation of 2.5 phr ESO, significantly reduced the Mooney viscosity difference between NR and BR phases from 56.42 to 51.85 and suppressed the crystallinity of TBIR, with the relative crystallinity decreasing to 74.6%. Owing to its amphiphilic molecular structure, ESO effectively improved filler dispersion, achieving a dispersion degree of 84.34% and an average aggregate size of 7.73 μm, while simultaneously enhancing rubber-filler interfacial adhesion, as evidenced by increased bound rubber content. These interfacial improvements led to a synergistic enhancement of tensile and tear strength, optimized thermal conductivity, reduced compression fatigue heat generation, and significantly improved wear resistance. Notably, the DIN abrasion volume reached a minimum value of 68.8 mm³ at 2.5 phr ESO, which was attributed to the optimized filler network and strong interfacial bonding that suppressed the transition from abrasive wear to rolling wear. Beyond previous ESO-based rubber modification studies that mainly focused on plasticization or compatibilization, this work establishes a bio-based multi-interface regulation strategy that links rubber-rubber compatibility, filler dispersion, rubber-filler adhesion, and wear-resistant performance in multi-component rubber composites. The identification of spatial structures of drugs and the researches on their interaction mechanism with DNA are always attractive to the researchers. However, their realization is lack of simple and fast method. This paper reports the establishment of multiple-functional detection platform based on the “turn off-on” model of ZnCdSe quantum dots. In this system, ZnCdSe quantum dots work as the fluorescent probe, platinum anti-cancer drugs as the quencher and triplex DNA as the trapping agent. The seemingly similar cisplatin and transplatin exhibited different fluorescent recovery behaviors due to their difference in structure, and thus realized the selective detection of cisplatin and transplatin with the reaction time set at 10 min as well as the quantitation of cisplatin over the range of 2.5 × 10 −8 –100 × 10 −8 M. Based on this, the interactions between platinum anti-cancer drugs and ctDNA as well as polymorphic DNA were further studied, and realized the recognition of triplex DNA. The multiple-functional detection platform integrates the functions of the filtration of high-efficient platinum anti-cancer drugs, the researches on interaction mechanism of drugs, and the recognition of polymorphic DNA, meaningful to the future treatment of viral and cancers based on antisense gene strategy.

Synergistic radiosensitization of cancer cells by metal-based nanomaterials and natural products is a forward-looking alternative for radiotherapy enhancement while inhibiting tumor metastasis. This work explores a phase change material (PCM)-gated Ti 3 C 2 T x nanosheet as a photothermal-responsive drug delivery system for loading natural trans -resveratrol (RSV), aiming at such synergistic radiosensitization in a controlled manner. This system can improve the biodispersity of RSV in the physiological solutions and inhibit the radiation-enhanced enzymatic activity of Ti 3 C 2 T x nanosheets to a large extent. Upon 808 nm near infrared light illimitation, Ti 3 C 2 T x -mediated photothermal heating makes the loaded PCM to be melted, resulting in the controlled release of RSV and simultaneous recovery of radiation-enhanced enzymatic activity of Ti 3 C 2 T x nanosheets. By further irradiating with X-ray, Ti 3 C 2 T x nanosheets is responsible for enhanced catalytic decomposition of H 2 O 2 to highly toxic hydroxyl radical as well as the consumption of intracellular glutathione . Both in vitro and in vivo studies demonstrate that those excess radical oxygen species can synergize with the released RSV to induce cancer cell death by apoptosis/ferroptosis hybrid pathway, fully inhibiting growth and metastasis of orthotopic breast cancer. These results reveal that the combination of PCM-gated Ti 3 C 2 T x nanosheets with RSV is capable of fabricating the photothermal-responsive theranostic systems and aiding the confinement of synergistic radiosensitization exclusively in the cancer cells for radiotherapy enhancement against tumor metastasis.

The preparation of fried meat products is prone to the formation of large amounts of heterocyclic amines (HCAs), polycyclic aromatic hydrocarbons (PAHs), and trans fatty acids (TFAs), which are potential risks to human health. Spices contain natural antioxidants that can inhibit the oxidation of fats and oils and the formation of hazardous substances. In this experiment, the effect of adding different levels (0.25%, 0.75%, 1.25%) of ginger or rosemary during meatball preparation on the formation of HCAs, PAHs and TFAs in fried pork balls was investigated. The results showed that the addition of ginger and rosemary reduced the content of HCAs in fried pork balls compared to the control group (no added spices). The inhibition of total HCAs was 63% when 0.25% ginger was added, while the total HCA content was reduced to 59% when 0.25% rosemary was added. The addition of 0.25% and 0.75% rosemary reduced the PAH content in fried pork balls by 30% and 35%. In addition, ginger and rosemary showed significant inhibition of C20:1 11t in TFAs, with a maximum inhibition rate of 40%. Therefore, adding appropriate levels of ginger or rosemary to fried pork balls could simultaneously inhibit the formation of HCAs, PAHs, and TFAs.Keywords:heterocyclic amines;polycyclic aromatic hydrocarbons;trans fatty acids;inhibitory profile;antioxidant

Sialic acid-binding immunoglobulin (Ig)-like lectins (Siglecs) is a type I transmembrane receptor on the cell surface. Siglec-5, as one of the Siglecs family, play an important role as an inhibitory receptor for leukocytes in the human body. The development of novel siglec-5 assays can help to study the pathogenesis of related diseases as well as to develop novel therapeutic drugs. We use catalytic hairpin assembly (CHA) amplification strategy combined with CRISPR-Cas12a′s side-cutting feature to build a 2D ultra-thin Ti 3 C 2 T x (MXene) based electrochemiluminescence (ECL) biosensor for the detection of Siglec-5. By using this ECL biosensor, the cleavage of CRISPR-Cas12a is reasonably combined with CHA-mediated isothermal amplification, thereby realizing the sensitive amplification assay Siglec-5 with 20.22 fM sensitivity. By introducing pairs of sites that are not in the same double-stranded DNA into the DNA duplex, the hybridization sequence of CRISPR-Cas12a complements the targeting mechanism to enhance indirect Siglec-5 amplification assay. Also, the double-strand DNA (dsDNA) design based on CRISPR-Cas12a amplification allows the same CRISPR RNA (crRNA, also known as guide RNA (gRNA)) to detect the output of DNA duplexes from different intermediate DNAs, which provides a common way for biomarker detection based on the conversion of protein analytes to intermediate DNA strategy. This work extends the application scope of CRISPR-Cas12a to the construction of ECL biosensors, evaluates the role of lectins, which can be used for the biochemical research and clinical diagnosis of protein markers. This is the first investigative work exploring the Trans-Cleavage activity of CRISPR-Cas12a for Mxene-based ECL biosensor establishment to the best of our knowledge.

In this work, we constructed an exonuclease III cleavage reaction-based isothermal amplification of nucleic acids with CRISPR/Cas12a-mediated pH-induced regenerative Electrochemiluminescence (ECL) biosensor for ultrasensitive and specific detection of SARS-CoV-2 nucleic acids for SARS-CoV-2 diagnosis. The triple-stranded nucleic acid in this biosensor has an extreme dependence on pH, which makes our constructed biosensor reproducible. This is essential for effective large-scale screening of SARS-CoV-2 in areas where resources are currently relatively scarce. Using this pH-induced regenerative biosensor, we detected the SARS-CoV-2 RdRp gene with a detection limit of 43.70 aM. In addition, the detection system has good stability and reproducibility, and we expect that this method may provide a potential platform for the diagnosis of COVID-19.

Development of chemo/photothermal/photodynamic therapy with nanoplatforms offers a promising strategy for effective cancer treatment. Recently, all-trans retinoic acid (ATRA), as a potential antitumor drug, has attracted great attention due to its antitumor activity. In this study, a novel coumarin-containing ATRA (AC) and indocyanine green (ICG) dye-loaded nanoparticles with the targeted ligand cyclic (Arg-Gly-Asp-D-Phe-Lys) (cRGD) peptide were fabricated by self-assembly and used as a new theranostic nanoplatform for chemo/photothermal/photodynamic therapy. The as-formed nanoparticles (AC/ICG-TNPs) had a diameter of around 133 nm with uniform monodispersity. Additionally, AC/ICG-TNPs showed marked stability under normal physiological conditions. However, it could rapidly release drugs in a mild acidic microenvironment. Moreover, confocal microscopic observations confirmed that the uptake of AC/ICG-TNPs increased in the breast cancer cells, particularly in MDA-MB-231 cells, probably mediated by cRGD via specific recognition of the overexpressed integrin αvβ3. Moreover, free AC exhibited stronger cytotoxic effects than free ATRA in the MTT assay, and AC/ICG-TNPs were demonstrated to possess excellent antitumor efficacy when exposed to NIR irradiation through the combination therapy. Hence, the therapeutic method designed in this study is a good candidate for improved bioactivity of ATRA and site-specific combinational therapy against breast cancer.

The solubility of cinnamyl alcohol in 11 pure solvents (methanol, ethanol, n-propanol, isopropyl alcohol, n-butyl alcohol, isobutyl alcohol, sec-butyl alcohol, acetone, methyl acetate, ethyl acetate and n-hexane) and two binary solvents of n-hexane + (n-propanol, n-butyl alcohol) was measured by a gravimetric method over temperatures from 253.15 K to 293.15 K at atmosphere pressure. From the experimental results, it was found that the solubility increased with the increasing temperature in all these solvents. The Apelblat model, the λh model, and the nonrandom two-liquid (NRTL) model were used to correlate the solubility data of cinnamyl alcohol in pure and mixed solvents. The thermodynamic properties of the mixing process in binary solvents were calculated by the NRTL model. The result indicates that the mixing process of cinnamyl alcohol is endothermic and spontaneous. The experimental solubility provides good guidance for the subsequent study of crystallization design and crystal morphology of cinnamyl alcohol.

Based on the study of the effect of positional isomerism of coformer functional groups on the cocrystallization and physicochemical properties of the active pharmaceutical ingredients, the impact of cis–trans isomeric butenedioic acid as coformers on the conformation, crystal structure, and its physicochemical properties of marbofloxacin was further explored. In this work, fumaric acid (FA) and maleic acid (MA) with different configurations were chosen as coformers to synthesize the pharmaceutical salt cocrystal (MBF-FA-H2FA) and salt (MBF-MA) of marbofloxacin (MBF), and their structures were fully characterized. Significant differences between the conformations of marbofloxacin in the salt cocrystal and in salt were found. In the salt cocrystal, the N atom of the piperazine group from marbofloxacin is coplanar with the pyridone ring, and the whole is straight like fumaric acid, whereas the marbofloxacin piperazine group in the salt is bent like the maleic acid configuration. Furthermore, the conformational variability of marbofloxacin in the salt cocrystal and the salt resulted in different crystal structures and opposite physicochemical properties. Notably, both multicomponent crystals have a surface hydrophilic intercalation structure. However, the salt cocrystal and salt exhibited different solubility and permeability. Specifically, the MBF-MA salt showed improved solubility and permeability, while the MBF-FA-H2FA salt cocrystal showed a decreased solubility and permeation rate compared to MBF. In addition, in vitro bacterial inhibitory activity assays indicated that the MBF-MA salt has stronger inhibitory activity against Gram-negative and Gram-positive bacterial strains than the MBF-FA-H2FA salt cocrystal and pure MBF.
