Synchronization of material resorption and new bone formation is vital to achieve harmonious bone regeneration in the treatment of large bone defects. To exposit the resorption/osteogenesis properties in the guided bone repairing, rhBMP-2-loaded trimodal macro/micro/nano-porous bioactive glass scaffolds (TMS-rhBMP-2) were set as substrate model. We penetratingly investigated the particular function of hierarchical structure and incorporated rhBMP-2 in the resorption/osteogenesis, and dissected the cellular interplay throughout the regenerative procedure. The results suggested that rhBMP-2 significantly facilitated osteoclastogenesis-mediated scaffold degradation and strikingly up-regulated mesenchymal stem cells (MSCs)-involved osteogenesis in vitro. Further gene microarray and related proteins expression indicated that in the presence of rhBMP-2, MSCs rather than differentiated MSCs could exert synergistic effects on osteoclastogenesis , osteoclasts maturation and resorptive function; meanwhile, rhBMP-2-induced MSCs osteogenesis was also strengthened by the osteoclasts. In vivo micro-CT, X-ray, kinetic and histological analyses qualitatively and quantitively demonstrated the optimized coupling of bioresorption/osteogenesis and the most rapid regeneration in TMS-rhBMP-2. Consequently, with rhBMP-2 acted as ignitor and MSCs/osteoclasts interaction as booster, a harmonious bone regeneration was obtained. Besides, long-term magnetic resonance imaging (MRI) in virtue of Gd 3+ suggested that the degradation products mainly distributed in liver and spleen, verifying the accumulation/discharge profiles and safety application of TMS-rhBMP-2 system in vivo. This study will not merely provide guidance for the design of clinical bone repairing materials, but shed substantial light on the multicell-mediated tissue regeneration .

Astragaloside (AS) has an anti-osteoporotic effect, but its poor water solubility and low bioavailability limit its application. In this study, a novel nano-carrier with bone targeting was prepared by modifying mPEG-PLGA with alendronate (AL) before incorporation into astragaloside nano-micelles (AS-AL-mPEG-PLGA) to enhance the oral bioavailability, bone targeting and anti-osteoporosis effect of AS. The release behavior of AS-AL-mPEG-PLGA in vitro was investigated via dialysis. The pharmacokinetics of AS-AL-mPEG-PLGA was studied in Sprague-Dawley (SD) rats. The cytotoxicity of AS-AL-mPEG-PLGA in vitro (via MTT method), coupled with bone targeting ability in vitro and in vivo were evaluated. The therapeutic effects of free AS and AS-AL-mPEG-PLGA (ELISA, micro-CT, H&E staining) were compared in osteoporotic rats. AS-AL-mPEG-PLGA with smaller particle size (45.3 ± 3.8 nm) and high absolute zeta potential (−23.02 ± 0.51 mV) were successfully prepared, wherein it demonstrated higher entrapment efficiency (96.16 ± 0.18%), a significant sustained-release effect for 96 h and acceptable safety within 10–200 μg/mL. AS-AL-mPEG-PLGA could enhance the hydroxyapatite affinity and bone tissue concentration of AS. The relative bioavailability of AS-AL-mPEG-PLGA was 233.90% compared with free AS. In addition, the effect of AS in reducing serum levels of bone metabolism-related indicators, restoring the bone microarchitecture and improving bone injury could be enhanced by AS-AL-mPEG-PLGA. AS-AL-mPEG-PLGA with small particle size, good stability, remarkable sustained-release effect, safety and bone targeting was successfully constructed in this experiment to potentially improve the oral bioavailability and anti-osteoporosis effect of AS. Thus, AS-AL-mPEG-PLGA may be a promising strategy to prevent and treat osteoporosis.

Purpose MXenes are two-dimensional (2D) materials that are increasingly being applied in biomedical fields. This is ascribed to their good physiochemical properties, unique structure and high biological compatibility. However, the osteogenic activity and suitability of these materials for bone tissue engineering are not clearly understood. Thus, the aim of this study is to evaluate the biocompatibility, osteoinductivity and guided bone regeneration ability of Ti 3 C 2 T x MXene in vitro and in vivo. Methods Multilayered Ti 3 C 2 T x MXene films were prepared and characterized by XRD and SEM. In vitro experiments were performed to evaluate the effect of MXene films on cell adhesion and morphology with SEM and fluorescence microscopy. The cytotoxicity of MXene films was detected with the Live/Dead double-staining tests. The EdU assay was employed to evaluate cell proliferation on MXene films and ALP activity was tested to determine the effect of the films on osteogenic differentiation in vitro. The mRNA expression of osteogenic differentiation-related markers was measured using qRT-PCR. In vivo animal studies were performed in which the MXene films were implanted subcutaneously in rats to evaluate biocompatibility and host tissue response in vivo. In addition, a rat calvarial defect model was established to examine the bone regeneration performance of the Ti 3 C 2 T x MXene films. The specimens were analyzed with micro-CT evaluation and histological tests. Results The XRD and SEM analyses revealed that the Ti 3 C 2 T x MXene film was successfully synthesized. The cellular experiments showed that MXene films were highly cytocompatible and enhanced osteogenic differentiation in vitro. When implanted into subcutaneous sites and calvarial defect sites in rats, MXene films showed good biocompatibility, osteoinductivity and bone regeneration activity in vivo. Conclusion In summary, this study presents new applications of MXenes in bone tissue engineering and in guided bone regeneration therapy.

With the rapid advancement of urbanization, individuals spend over 90 % of their time in indoor environments, where prolonged exposure to indoor air pollutants particularly particulate matters (PMs) poses significant health risks. This highlights the urgent need to develop advanced fiber-based filtration systems that simultaneously exhibit high filtration efficiency and antimicrobial properties. In this study, a multifunctional ZIF-8@PET/CA-PP composite filter material was fabricated using commercial PET membranes as substrates via a combination of electrostatic spraying and self-assembly techniques. The resulting material features a surface-functionalized CA/ZIF-8/PVDF-HFP composite layer. Comprehensive characterization revealed that the optimized filter material achieves exceptional PM2.5 filtration efficiency of 98.52 % (±0.72 %) with a low air resistance of only 57 Pa, while demonstrating antibacterial rates of 93.75 % against Escherichia coli ( E. coli ) and 94.2 % against Staphylococcus aureus ( S. aureus ). The outstanding performance can be attributed to two key factors: (1) the uniform dispersion and dense nanostructure of the functional layer significantly enhances particle capture capacity through combined mechanical and electrostatic filtration mechanisms; (2) the incorporated ZIF-8 nanoparticles provide effective antimicrobial activity by disrupting microbial membranes. Further validation through X-ray micro-CT scanning simulations confirmed the filter material's structural integrity and consistent filtration performance across varying airflow velocities. The ZIF-8@PET/CA-PP composite filter material developed in this work represents a promising candidate for next-generation air filtration applications, offering an optimal combination of low air resistance, high filtration efficiency, and robust antimicrobial properties. These advantages position the material as particularly suitable for improving indoor air quality in urban environments.

Berberine (BBR) has a long history of use in the treatment of Rheumatoid arthritis (RA) and is considered one of the most promising natural product for the treatment of RA. However, oral administration of berberine has low bioavailability and requires frequent administration, resulting in poor patient compliance. In this study, we developed a BBR-loaded phospholipid-based phase separation gel (BBR-PPSG) to achieve sustained drug release and long-term therapeutic effect. The stability of BBR-PPSG was verified and it was found that it can be stored for a long time. The pharmacokinetic study on rats and rabbits showed that BBR-PPSG not only achieved 1-month of sustained release, but also significantly increased the area under the curve (AUC) by nearly 9-fold and prolonged the half-life (t1/2) by 10-fold. By constructing rat and rabbit models of RA, we also proved that BBR-PPSG administration once a month effectively alleviated joint swelling, and significantly reduce TNF-α levels in AIA rats and OIA rabbits. Histopathological analysis of rabbit joint sections revealed that after intra-articular injection of BBR-PPSG, the synovial cell layer remained intact, while in the model group, the synovial cells were significantly reduced and exhibited necrosis. MicroCT data analysis showed that the values of Tb.N and Tb. Sp in the BBR-PPSG group were significantly better than those in the model group (p < 0.05). This study addressed the limitations of frequent administration of BBR by developing a phospholipid-based phase separation gel system for berberine delivery, achieving long-term sustained release. The BBR-PPSG demonstrated good biocompatibility, simple preparation and excellent stability, thus holding potential as a novel pharmaceutical formulation for RA treatment.

Three-dimensional (3D) printing technology has attracted considerable focus for preparing porous bone repair scaffolds to promote bone regeneration. Inspired by organic-inorganic components and the porous structure of natural bone, novel porous degradable scaffolds have been printed using hydroxyapatite (HA), carboxymethyl chitosan (CMCS), and polydopamine (PDA). The well-designed HA/CMCS/PDA scaffolds exhibited a porous structure with 60.5 ± 4.6% porosity and 415 ± 87 μm in mode pore diameter. The weight loss percentage (WL%) of the HA/CMCS/PDA scaffolds reached about 17% during a 10-week degradation in vitro . The degradation process between the CMCS and HA induced the release of calcium ions. Using commercial product as the contrast material, the osteogenic properties of the scaffolds were assessed in vivo . The implantation and degradation of HA/CMCS/PDA scaffolds had no adverse effects on the kidney and liver of rabbits with no inflammatory response in the implantation sites. The micro-CT and histology data suggested that the HA/CMCS/PDA scaffolds could effectively stimulate new bone formation within the femoral lacuna defect region of rabbits versus blank control at 12 weeks after implantation. Surface cortical bone was generated in the defect area in the HA/CMCS/PDA group; the defect in the blank group remained obvious. HA/CMCS/PDA scaffolds had excellent biodegradability matching the formation of new bone during implantation. In conclusion, 3D-printed HA/CMCS/PDA scaffolds have remarkable potential as a new material for repairing bone defects.

Scaffold degradation regulation has become the key aspects in promoting bone regeneration and reconstruction. In the study, a novel stepwise-degraded PLGA/PCL/HA:Yb/Ho/Zn (PPHZ) scaffold was designed to compare with the fast-degradable PLGA/HA:Yb/Ho/Zn (PLHZ) scaffold to investigate the effects of scaffold degradation regulation on osteogenesis and to trace the scaffold degradation process. Micro-CT reconstruction, confocal fluorescence imaging, and histological sections were used for the analyses of the harvested animal samples, which demonstrated that the degradation regulated stepwise-degraded PPHZ scaffold maintaining higher mechanical strength and longer scaffold integrity is more advantageous for in vivo osteogenesis than the fast-degradable PLHZ scaffold. Meanwhile, the multifunctional HZ particles would help the anti-infection and tracking the distribution during scaffold degradation. The PPHZ scaffold may benefit future repair of anti-infective bone defects and in vivo multimodal tracking.

Nest-like nanofiber structures have potential applications in surface modifications of titanium implants. In this study, nest-like nanofiber structures were prepared on a titanium surface at room temperature and pressure by using the nanobowl template-assisted method combined with alkali etching. The characterization and biocompatibility of this material were analyzed by cellular adhesion, death, CCK-8, ALP, and RT-PCR assays in vitro, and osseointegration was evaluated by micro-CT and fluorescent labeling in vivo . The results showed that this nest-like nanofiber structure has a firmer and asperate surface than nanotubes, which leads to better cellular adhesion, proliferation, and differentiation capacity. In a beagle alveolar bone implant model, the nest-like nanofiber structure showed a better osseointegration capacity. In conclusion, this nest-like nanofiber structure has potential applications in dental implantology.

Background Existing implant materials cannot meet the essential multifunctional requirements of repairing infected bone defects, such as antibacterial and osteogenesis abilities. A promising strategy to develop a versatile biomimicry composite of the natural bone structure may be accomplished by combining a multifunctional nanoparticle with an organic scaffold. Methods In this study, a quaternary ammonium silane-modified mesoporous silica containing nano silver (Ag@QHMS) was successfully synthesized and further combined with silk fibroin (SF) to fabricate the multifunctional nano-reinforced scaffold (SF-Ag@QHMS) using the freeze-drying method. Furthermore, the antibacterial and osteogenic effects of this composite were evaluated in vitro and in vivo. Results SF-Ag@QHMS inherited a three-dimensional porous structure (porosity rate: 91.90 ± 0.62%) and better mechanical characteristics (2.11 ± 0.06 kPa) than that of the SF scaffold (porosity rate: 91.62 ± 1.65%; mechanic strength: 2.02 ± 0.01 kPa). Simultaneously, the introduction of versatile nanoparticles has provided the composite with additional antibacterial ability against Porphyromonas gingivalis , which can be maintained for 15 days. Furthermore, the expression of osteogenic-associated factors was up-regulated due to the silver ions eluting from the composite scaffold. The in vivo micro-CT and histological results indicated that the new bone formation was not only localized around the border of the defect but also arose more in the center with the support of the composite. Conclusion The multifunctional silver-loaded mesoporous silica enhanced the mechanical strength of the composite while also ensuring greater and sustained antibacterial and osteogenic properties, allowing the SF-Ag@QHMS composite to be used to repair infected bone defects.

Structural electrolytes with both mechanical and electrochemical properties are of great significance in structural energy storage devices. In this paper, we succeeded to synthesize a novel epoxy resin/hydrogel bicontinuous phase structure electrolyte for the first time by the ingenious design of the hydrophobic curing agent and the substitution of hydrogel for water, and filled the application of bicontinuous electrolyte in aqueous electrochemical energy storage system. The fine structure composition of electrolyte was divided and analyzed in detail by X-ray micro-CT technology. The epoxy resin phase is composed of a reasonable distribution of skeleton structures of various sizes, the largest of which is able to support heavy weights. And a large number of tiny skeletons are distributed to connect the skeletons of different sizes, which improves the three-dimensional interconnection of the epoxy resin phase and makes it excellent mechanical properties (Tensile strength 11.91 Mpa). The large size PVA gel phase constitutes the main pathway for conducting ions. Furthermore, the large-diameter gel phases are coupled in three dimensions with a huge number of micro-diameter gel phases, increasing the number of micro-ion transport channels and decreasing the distance of ion transport with good ionic conductivity (0.15 mS cm −1 ). Using carbon cloth coated in slurry as the electrode, an integrated structural energy storage bearing device was created in situ using layer brush technology, which exhibited exceptional electrochemical performance during in-situ variable load tests. We have successfully solved the construction of aqueous bicontinuous phase structure electrolyte, which made the application of various water chemistry systems in structural energy storage possible.

Aims The main pathological features of osteoarthritis (OA) include the degeneration of articular cartilage and a decrease in matrix synthesis. Chondrocytes, which contribute to matrix synthesis, play a crucial role in the development of OA. Liquiritin, an effective ingredient extracted from Glycyrrhiza uralensis Fisch., has been used for over 1000 years to treat OA. This study aims to investigate the impact of liquiritin on OA and its underlying mechanism. Materials and methods Gait and hot plate tests assessed mouse behavior, while Micro-CT and ABH/OG staining observed joint morphological changes. The TUNEL kit detected chondrocyte apoptosis. Western blot and immunofluorescence techniques determined the expression levels of cartilage metabolism markers COL2 and MMP13, as well as apoptosis markers caspase3, bcl2, P53, and PUMA. KEGG analysis and molecular docking technology were used to verify the relationship between liquiritin and P53. Key findings Liquiritin alleviated pain sensitivity and improved gait impairment in OA mice. Additionally, we found that liquiritin could increase COL2 levels and decrease MMP13 levels both in vivo and in vitro. Importantly, liquiritin reduced chondrocyte apoptosis induced by OA, through decreased expression of caspase3 expression and increased expression of bcl2 expression. Molecular docking revealed a strong binding affinity between liquiritin and P53. Both in vivo and in vitro studies demonstrated that liquiritin suppressed the expression of P53 and PUMA in cartilage. Significance This indicated that liquiritin may alleviate OA progression by inhibiting the P53/PUMA signaling pathway, suggesting that liquiritin is a potential strategy for the treatment of OA.

Aging is a major cause of bone loss and osteoporosis. Diallyl trisulfide (DATS), one of the main organic sulfides in garlic oil, has been shown to alleviate arthritis in mice. However, further research is still needed to determine how DATS affects bone formation and bone loss in aging mice. Here, we established a mouse model of natural aging for dietary DATS intervention. DATS treatment improved bone microstructure, including the disorganized arrangement of bone trabeculae and promoted collagen synthesis, with micro-CT and histological analyses. The abundance of beneficial bacteria for bone formation, such as Clostridiaceae and Erysipelotorichaceae, as well as the microbial diversity and community richness, were all altered by DATS, according to 16S rRNA sequencing data. 14 potential biomarkers and 9 important metabolic pathways were examined using serum metabolomics analysis. Additionally, there has been a significant reduction in sphingosine, which was directly associated with bone metabolism. The level of sphingosine and relative abundance of Clostridium were found to be negatively correlated by correlation analysis, indicating that bacteria may regulate bone reconstruction via influencing metabolites. Furthermore, Runx2 and β-catenin gene expression levels increased in bone, which may be related to the ameliorative mechanism of DATS. Our results suggested that DATS may prevent age-related bone loss by upregulating osteogenic gene expression through altering gut microbes and serum metabolism.
