The overuse of antibiotics in livestock farms and their subsequent accumulation in manures lead to increase antibiotic resistance genes (ARGs) in the compost products. Yet, the function of humic acid (HA) in shaping antibiotics and ARGs abundances, microbial structure, and product quality during composting remains unclear. Here, the efficiency of added HA in reducing antibiotics and ARGs abundances during swine manure (SM) composting was studied using five rates of HA: 0%, 1%, 2%, 4%, and 6%. The findings indicated that the abundance of ARGs types such as LSM, bacitracin, macrolide, sulfonamide, tetracycline , and vancomycin and ARGs subtypes including MacB , MexD , OleB , SrmB , Sul 1, VanHB , VanSB , pbp2b , tetPB , and tetX decreased significantly under 6% HA amount compared with CK compost ( P < 0.05). Besides, the added 6% HA shaped the abundance of the bacterial community (notably Acinetobacter , Pseudoxanthomonas , and Luteimonas ), as well as the level of carbohydrate metabolism and amino acid metabolism (6.9% and 5.4%, respectively), which can be taken as an indicator for microbial activity , thereby accelerating antibiotics decrease. RDA analysis revealed that TOC , moisture content, and pH considerably influenced microbial activity and ARGs reduction ( P < 0.05). The estimation of compost quality indicated that 6% HA significantly improved the total content of humic substance and HA to 120.7 ± 1.02 g/kg and 62.3 ± 0.92 g/kg, and reduced the content of fulvic acid to 12.8 ± 1.22 g/kg, thus promoting antibiotics and ARGs reduction. Overall, this study offers a promising option for improving compost quality and safety and mitigating ARGs circulation in the natural environment.

The dissemination of antimicrobial resistance (AMR) in wastewater treatment poses a severe threat to the global ecological environment. This study explored the effectiveness of photocatalysis in inactivating antibiotic resistant bacteria (ARB) and quantitatively clarified the inhibiting rate of the transfer of antibiotics resistance genes (ARGs). Herein, the magnetic heterojunction as UiO-66-NH 2 @CuFe LDH-Fe 3 O 4 (UN-66@LDH-Fe) effectively facilitated the electron-hole separation and accelerated the photogenerated charge transfer, thereby guaranteeing the stable practical application in aeration tanks. Notably, the internal electric field of heterogeneous photocatalyst resulted in significant increase of ARGs inactivation, achieving 5.63 log of ARB, 3.66 log of tet A and 3.57 log of Ampr genes were photodegraded under optimal reaction conditions within 6 h. Based on the complex microbial and molecular mechanism of multiple-ARB communities inactivation in photo-treatment, the photogenerated reactive oxygen species (ROSs, ·OH and ·O 2 − ) effectively destroyed bacterial membrane protein, thereby the intracellular ROSs and redox cycles further induced oxidative stress, attributing to the abundance reduction of ARGs and their host bacteria. Moreover, long-term (7 days) continuous operation preliminarily verified the practical potential in reducing AMR spread and developing wastewater treatment efficacy. Overall, this study presented an advantageous synergistic strategy for mitigating the AMR-associated environmental risk in wastewater treatment.

There is growing concern about the transfer of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs) in airborne particulate matter. In this study, we investigated the effects of various types of carbonaceous particulate matter (CPM) on the transfer of ARGs in vitro. The results showed that CPM promoted the transfer of ARGs, which was related to the concentration and particle size. Compared with the control group, the transfer frequency was 95.5, 74.7, 65.4, 14.7, and 3.8 times higher in G (graphene), CB (carbon black), NGP (nanographite powder), GP1.6 (graphite powder 1.6 micron), and GP45 (graphite powder 45 micron) groups, respectively. Moreover, the transfer frequency gradually increased with the increase in CPM concentration, while there was a negative relationship between the CPM particle size and conjugative transfer frequency. In addition, the results showed that CPM could promote the transfer of ARGs by increasing ROS, as well as activating the SOS response and expression of conjugative transfer-related genes (trbBp, trfAp, korA, kroB, and trbA). These findings are indicative of the potential risk of CPM for the transfer of ARGs in the environment, enriching our understanding of environmental pollution and further raising awareness of environmental protection.

The present study aims to investigate the mechanism by which triclosan influences the dissemination of antibiotic resistance genes (ARGs) during the whole anaerobic digestion process. qPCR and metagenomic analyses revealed that triclosan facilitated ARGs dissemination in a dose- and time-dependent manner. Furthermore, integrons exhibited a significant correlation with the majority of quantified ARGs, and various ARGs were frequently linked on integron gene cassettes. Microbial community and redundancy analyses indicated that triclosan altered the components of dominant ARGs hosts Firmicutes, Synergistetes and Bacteroidetes. Path modeling analysis confirmed integrons were the main driving force for facilitating ARGs dissemination. The promoted ARGs dissemination may be associated with the increased reactive oxygen species generation, cell membrane permeability, close-connected the ARGs transfer related regulatory proteins induced by triclosan. This study broadens the understanding of triclosan facilitates ARGs dissemination through anaerobic treatment, the strategies for preventing potential risks should be proposed in practice.

Cutibacterium acnes is the most abundant bacterium of the human skin microbiome since adolescence, participating in both skin homeostasis and diseases. Here, we demonstrate individual and niche heterogeneity of C. acnes from 1,234 isolate genomes. Skin disease (atopic dermatitis and acne) and body site shape genomic differences of C. acnes , stemming from horizontal gene transfer and selection pressure. C. acnes harbors characteristic metabolic functions, fewer antibiotic resistance genes and virulence factors, and a more stable genome compared with Staphylococcus epidermidis . Integrated genome, transcriptome, and metabolome analysis at the strain level unveils the functional characteristics of C. acnes . Consistent with the transcriptome signature, C. acnes in a sebum-rich environment induces toxic and pro-inflammatory effects on keratinocytes. L-carnosine, an anti-oxidative stress metabolite, is up-regulated in the C. acnes metabolome from atopic dermatitis and attenuates skin inflammation. Collectively, our study reveals the joint impact of genes and the microenvironment on C. acnes function.

It is unadvisable to discuss the antibiotic resistance genes (ARGs) reduction in anaerobic digestion (AD) system neglecting its main purpose-methane production. The methane production improvement coupling with antibiotic resistance genes (ARGs) reduction in anaerobic digestion (AD) by zero valent iron (ZVI) were simultaneously investigated. Whether the role of ZVI on the ARGs fate was random or specific was clarified through the high-throughput qPCR (HT-qPCR). Results indicated that ZVI improved methane production and ARGs reduction by 23.9% and 25.0%, respectively. The improved methane production was associated with chemical reaction and variances of microbial community caused by ZVI, where DIET between Petrimonas , Clostridium and Syntrophomonas , Methanosarcina was established along with ACAS being enriched. ZVI specifically, not randomly, facilitated the reduction of aminoglycoside resistance genes of antibiotic inactivation and tetracycline resistance genes of ribosomal protection proteins. The specifical reduction could be attributed to enzyme activity inhibition and intracellular ionic disturbance caused by higher amounts of ZVI, although most of ARGs fate could be well explained by microbial community which contributed the most to ARGs dynamics as a whole. ZVI-based AD was a promising way for the improvement of methane production coupling ARGs reduction.

In this study, the spatial location effect of the incorporated metal atoms on the hydrolysis activity of metal organic framework-based nanomaterials (MOFs) was investigated. UiO-type MOFs incorporated with single-atom Cu at different spatial positions were prepared and their hydrolysis activity towards phosphoester bonds was found to be highly depended on the location of the atomically dispersed Cu atoms. Especially, the attachment of Cu atoms to ligand (UiO-67-Cu N ) resulted in the most significant increase of their hydrolysis activity, which can be attributed to the elevated oxygen vacancies and increased Lewis acidity of MOFs. As a result, UiO-67-Cu N efficiently catalyzed the cleavage of DNA acting as DNA hydrolases, thereby effectively inhibiting the horizontal gene transfer of ARGs and eliminating bacterial resistance to antibiotic. This study opens up new prospects for the rational design of high-performance MOF-based DNA hydrolases and developing new treatment strategies for the removal of ARGs.

Photocatalysis, as a novel technique, has been widely used for antibiotic pollution remediation in wastewater. In the processes of degradation and removal of antibiotics, the impact of photocatalysts on microenvironment is very important but remains poorly understood. In the present study, the effect of typical photocatalyst g-C3N4 (Graphitic carbon nitride) on microbial community was investigated in two sediment types (riverbed sediment and pig-farm sediment) polluted by tetracycline (TC) in central southern China. The riverbed sediment and pig farm sediment samples were respectively exposed to g-C3N4 (25, 75, 125 mg⋅kg–1) and TC (60, 120, 180 mg⋅L–1) treatments alone or combination for 30 days, respectively. The bacterial community and antibiotic resistance genes (ARGs) of the treated sediments were analyzed by Illumina sequencing and metagenomic sequencing. Studies had shown that: TC, g-C3N4, and TC/g-C3N4 have significant effects on the changes of microbial communities and components in riverbed sediment, but they do not exist in pig farm sediment. The most alterations of microbial taxa were Acidobacteriota, Actinobacteriota, and Desulfobacterota in riverbed sediment, and Elusimicrobiota in the pig farm sediment under various treatments. Through network analysis, it was found that the distribution of microorganisms in the pig farm sediment is more complex and more stable. The addition of g-C3N4 reduced the absolute abundance of ARGs in the two examined sediments, but not significantly changed their relative abundance of ARGs. The g-C3N4 application was beneficial to the removal of TC residues and to the prevention of the generation and transmission of ARGs in sediments. Our results suggested that g-C3N4 was a suitable photocatalyst with excellent application prospect for the removal of TC residues and the control of ARGs in environment.

The pharmaceutical factories of oxytetracycline (OTC) massively produce OTC fermentation residues (OFRs). The high content of residual OTC and antibiotic resistance genes in OFRs must to be considered and controlled at an acceptable level. This study therefore investigated the applicability of Fenton oxidation in OTC degradation and resistant gene inactivation of OFRs. The results revealed that Fe 2+ as catalyzer could very rapidly activate H 2 O 2 to produce HO • , leading to instantaneous degradation of OTC. The optimum conditions for OTC removal were 60 mM H 2 O 2 and 140 mg/L Fe 2+ under pH 7. After Fenton oxidation treatment, the release of water-soluble polysaccharides , NO 3 –N, and PO 4 –P was enhanced, whereas for proteins and NH 3 –N were reduced. Three soluble fluorescence components (humic, tryptophan-like, and humic acid-like substances) were identified through fluorescence spectra with parallel factor analysis, and their reduction exceeded 50% after Fenton oxidation. There were twelve intermediates and three degradation pathways of OTC in OFRs during Fenton process. According to toxicity prediction, the comprehensive toxicity of OTC in OFRs was alleviated via Fenton oxidation treatment. In addition, Fenton oxidation showed the ability to reduce antibiotic resistance genes and mobile genetic elements, and even tetO , tetG , intI1 , and intI2 were eliminated completely. These results suggested that Fenton oxidation treatment could be an efficient strategy for removing OTC and resistance genes in OFRs.

With the rapid development of nanotechnology, nanomaterials have been prepared with diverse sizes and surface properties. However, the influences of these properties on the horizontal transfer of antibiotic resistance genes (ARGs) are still unclear. Here, two groups of Ag particles were prepared, one group with the same negative charge and different sizes (9, 72 and 1885 nm), the other group with similar sizes (72 and 67 nm) and different charges (−12.7 and +22.6 mV) and their effects on ARGs transformation were investigated. The results revealed that 9 nm Ag at 1000 μg/L induced a 14.56-fold increase in transformation, which was 2.05 and 3.91 times higher than that of 72 nm and 1885 nm Ag particles, respectively. When the size was similar, the positively charged Ag stimulated an 11.59-fold increase in transformation, which is 1.63-fold higher than that of the negatively charged Ag. Investigation of the underlying mechanisms suggested that Ag particles with a smaller size and/or positive charge induced more intercellular reactive oxygen generation and membrane damage than Ag particles with large size and/or negative charge. This study exposed the potential ecological risks of Ag particles with small size and positive charge in accelerating the dissemination of ARGs and highlighted concerns about the management of nanomaterials with varying structural properties.

Chlortetracycline (CTC), a widely used typical tetracycline antibiotic, has raised increasing concerns due to its potential health and environmental risks. Biodegradation is considered an effective method to reduce CTC in environment. In this study, a strain Aspergillus sp. LS-1, which can efficiently degrade CTC, was isolated from CTC-rich activated sludge . Under optimal conditions, the maximum removal efficiency of CTC could reach 95.41%. Temperature was the most significant factor affecting the degradation efficiency of LS-1. The 19 products were identified in the CTC degradation by strain LS-1, and three degradation pathways were proposed. All the degradation pathways for CTC exhibited ring-cleaving, which may accelerate the mineralization of CTC. To gain more comprehensive insights into this strain, we obtained the genome of LS-1, which had high GC content (50.1%) and completeness (99.3%). The gene annotation revealed that LS-1 contains some vital enzymes and resistance genes that may carry functional genes involved in the CTC degradation. In addition, other antibiotic resistance genes were found in the genome of LS-1, indicating that LS-1 has the potential to degrade other antibiotics. This study provides a more theoretical basis for the investigation of CTC degradation by fungi and new insights into the biodegradation of CTC.

Ciprofloxacin (CIP), as a quinolone antibiotic, has broad-spectrum antibacterial properties and can affect methanogenic performance in anaerobic digestion (AD). While previous studies focused on synthetic wastewater, the fate of CIP in real distillery wastewater (RDW) and its impact on microbial adaptation mechanisms remain unclear (such as biotransformation pathways, population dynamics, and the enzymes involved) in RDW is largely unclear. In this study, we investigated AD performance, metabolic pathways, and antibiotic resistance gene (ARG) dynamics using real wastewater spiked with CIP (0.3–2 mg/L). Results indicate that 0.5 mg/L CIP (631.83 mL CH 4 /g·VS) enhanced the methane yield by 6.67 % (592.34 mL CH 4 /g·VS in control), correlating with upregulated enzyme in glycosis, TCA cycle, and methanogenesis (F 420 increased). With full use of short-chain acids, transient volatile fatty acid (VFA) inhibition (≤ 50 mg/L on Day 3) was overcome by Day 10. Metagenomics revealed CIP promoted the production of stress proteins (e.g., cysteine synthase activity doubled). Furthermore, CIP (0.3–1 mg/L) suppressed mobile genetic elements (MGEs) encoding horizontal gene transfer, including isfinder (15.15 %) and integrases (6.25 %), while ARG and virulence factor abundances remained unchanged versus control. This study firstly shows that low-dose CIP in RDW increases methanogenesis via metabolic adaptation without exacerbating ARG risks. MGE suppression implies that CIP may lessen the possibility of ARG diffusion in AD systems. These results offer vital information for improving AD performance in the treatment of wastewater contaminated by antibiotics and developing methods to strike a balance between antibiotic removal and ARG control.
