Nanozymes are nanoparticles with enzymatic activity, which are widely used in environmental and antibacterial research. Herein, we designed and synthesized novel amorphous nanozyme Cu-Im NPs with multiple enzyme-mimicking activities. Cu-Im NPs have the same active sites as natural laccase. In addition, the active center is similar to that of carbonic hydrolase, replacing the zinc ions with copper ions. Meanwhile, Cu-Im NPs also possess peroxidase-like activity. We reveal the multi-enzyme catalytic mechanisms of Cu-Im NPs. Notably, phosphate inhibits the laccase-like and peroxidase-like activities of Cu-Im NPs while activating their hydrolase activity. Based on these findings, we have developed a sensitive and selective method for detecting phosphate anions.

Ten substituted 1,3-dihydroxyxanthones were synthesized in one step. The yields ranged from 40 to 76%. Compounds 8–10 were first reported. Next, the compounds’ in vitro anti-proliferative activities against nine human cancer cell lines, antityrosinase, and antioxidant activities were evaluated. Compounds 1, 4, 6–7, and 9–10 exhibited enhanced cytotoxicity against certain cancer cells. Compounds 2, 8, 9, and 10 inhibited tyrosinase activity to a certain extent. In addition, compound 4 exhibited the best antioxidant activity, which was consistent with theoretical calculations. These results demonstrated that compounds 1–2, 4, and 6–10 were promising leads for further investigation.

Ag NPs were generated by reducing silver ions with sodium borohydride and then loaded into a zirconium-based MOF, PCN-222, resulting in a deep purple powder, PCN-222@AgNPs, which exhibited photocatalytic activity. The material was characterized and found to have Ag NPs uniformly dispersed in the pores of PCN-222, with a content of 6.0306%. PCN-222@AgNPs had rod-shaped structures of micrometer sizes with irregular spherical Ag NPs on their surfaces, as revealed by scanning electron microscopy and transmission electron microscopy. In experiments on Ag NPs release and reactive oxygen species (ROS) detection, PCN-222@AgNPs effectively released Ag NPs into an aqueous solution and promote ROS generation. Finally, the antibacterial activities of PCN-222@AgNPs against Escherichia coli and Staphylococcus aureus were assessed by in vitro experiments. Results indicated that PCN-222@AgNPs had lower minimum bactericidal concentration (MBC) compared to PCN-222 in both light and dark environments. The MBC further decreased under light conditions due to the enhanced photocatalytic performance of the composite material and the promotion of Ag NPs release by ROS. These findings demonstrate that PCN-222@AgNPs have synergistic antibacterial effects under visible light irradiation.

Finger citron (Citrus medica L. var. sarcodactylis Swingle) is a traditional Chinese herb and considered as a healthy food. Flavonoids are the major bioactive substances in Finger citron. In this study, the major flavonoids of finger citron (FFC) were purified with AB-8 macroporous resins, and then three of them were identified as diosmetin-6-8-di-C-glucoside, hesperidin and diosmetin-6-C-glucoside, and other two were preliminarily inferred as limocitrol 3-alpha-l-arabinopyranosyl-(1->3)-galactoside and scutellarein 4′-methyl ether 7-glucoside by high-performance liquid chromatography and ultraperformance liquid chromatography to quadrupole time-of-flight mass spectrometry. Further, their antioxidation and antiaging activities were determined in vitro and in vivo. In vitro, chemical assays revealed that the purified FFC had strong antioxidative activity as demonstrated by its strong DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS [2,2-azinobis (3-ethyl-benzothiazoline-6-sulphonic acid) diammonium salt] radical scavenging activities and ORAC (oxygen radical absorbance capacity). In vivo, the purified FFC significantly increased the mean and maximum lifespan of Caenorhabditis elegans by 31.26 and 26.59%, respectively, and showed no side effects on their physiological functions. Under normal and oxidative stress conditions, purified FFC reduced the accumulation of reactive oxygen species (ROS) and malondialdehyde, while increased superoxide dismutase (SOD) and catalase (CAT) enzyme activities in C. elegans. Together, we successfully identified three major substances in purified FFC of finger citron and determined the excellent antiaging activity of FFC, which is attributed to its strong antioxidative activity and effect on homeostasis of ROS.

Background Paeonol is extracted and isolated as a rich and sustainable natural bioresource from the root bark of Paeonia suffruticosa , the derivatives of which exhibit numerous biological activities. It is well known that ester compounds play a very important role in pest control, such as organophosphorus, carbamate and pyrethroid pesticides. Results To discover biorational natural product-based pesticides, three series of (60) paeonol ester derivatives ( 7a–t , 8g , p , 9g , p , 10g–j , n–u , 11g , u , 12g , u , 13a–p , 14b , c , and 15b , c ) were prepared by structural modification of paeonol, and their structures were well characterized by proton nuclear magnetic resonance ( 1 H-NMR), carbon-13 nuclear magnetic resonance ( 13 C-NMR), high-resolution mass spectrometry (HRMS), and melting point. Furthermore, we assessed the compounds as insecticidal, nematicidal, and anti-oomycete agents against three serious agricultural pests, Mythimna separata , Heterodera glycines , and Phytophthora capsici . Among all tested compounds: (i) compound 8p showed more significant insecticidal activity than toosendanin, and the final mortality rates of 8p and toosendanin against M. separata (1 mg mL −1 ) were 70.4%, and 51.9%, respectively; (ii) compound 7a exhibited more promising nematicidal activity than paeonol, and the median lethal concentration (LC 50 ) values of 7a and 1 against H. glycines were 15.47 and 50.80 mg L −1 , respectively; (iii) compounds 7n and 13m displayed more significant anti-oomycete activity compared to zoxamide against Phytophthora capsici , and the median effective concentration (EC 50 ) values of 7n , 13m , and zoxamide were 23.72, 24.51, and 26.87 mg L −1 , respectively; and the protective effect of the compounds against Phytophthora capsici in vivo further confirmed the effectiveness of the agents. Conclusion This study suggested that the introduction of a nitro at the C5 or C3 position of paeonol could improve its bioactivity against M. separata , H. glycines , and Phytophthora capsici . © 2022 Society of Chemical Industry.

Similar to the enzymatic process, there might also be an active fragment in polysaccharides, how to obtain is important for investigating the bioactivity and pharmacological mechanism of polysaccharides. Presently, a Gynostemma pentaphyllum endophytic fungus Chaetomium globosum CGMCC 6882 polysaccharide [Genistein Combined Polysaccharide (GCP)] was degraded by ultrasonic treatment, two polysaccharide fragments of GCP-F1 and GCP-F2 were obtained. Physicochemical results showed that GCP-F1 and GCP-F2 had the same monosaccharide composition of arabinose, galactose, glucose, xylose, mannose, and glucuronic acid as compared to GCP with slightly different molar ratios. However, weight-average molecular weights of GCP-F1 and GCP-F2 decreased from 8.093 × 104 Da (GCP) to 3.158 × 104 Da and 1.027 × 104 Da, respectively. In vitro scavenging assays illustrated that GCP-F1 and GCP-F2 had higher antioxidant activity against 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical, superoxide anions, and hydroxyl radical than GCP, the order was GCP < GCP-F1 < GCP-F2. Meanwhile, antibacterial tests showed that ultrasonic degradation increased the antibacterial activity of GCP-F1 as compared to GCP, but GCP-F2 almost lost its antibacterial activity with further ultrasound treatment. Changes in the antioxidant and antibacterial activities of GCP-F1 and GCP-F2 might be related to the variation of their molecular weights.

The structural variances of adsorbents play a crucial role in determining the number of effective adsorption sites and pretreatment performance. However, there is still a gap in comprehending the impact of different carbon structural adsorbents on membrane fouling . Therefore, this study aimed to compare the efficacy of granular activated carbon (GAC), powdered activated carbon (PAC), and activated carbon fiber (ACF) in mitigating membrane fouling during municipal sewage reclamation using an aerobic granular sludge membrane bioreactor (AGMBR). The results demonstrated that the utilization of PAC significantly enhanced the normalized flux and reduced fouling resistance in comparison to GAC and ACF systems. PAC effectively adsorbed low and medium-molecular-weight pollutants present in raw sewage, resulting in an increase in average particle size and a decrease in foulant content on the membrane surface. The Hermia model indicated that adsorption pretreatment minimized standard blocking while promoting the formation of a sparse and porous cake layer. Moreover, according to the extended Derjaguin–Landau–Verwey–Overbeek theory, PAC has been demonstrated as the optimal antifouling system owing to its enhanced repulsion between membrane-foulant and foulant-foulant interactions. Correlation analysis revealed that the exceptional antifouling performance of the PAC system was due to its high removal rates of chemical oxygen demand (~78 %) and suspended solids (~97 %). This research offers valuable insights into the mitigation of membrane fouling through the utilization of adsorbents featuring diverse carbon structures .

Metal oxides with peroxidase (POD)-like and halogen peroxidase (HPO)-like catalytic activities effectively eliminate bacteria by catalyzing reactive oxygen species (ROS) production, including hydroxyl radicals (·OH) and hypobromous acid (HBrO)/hypochlorous acid (HClO). While metal phosphides generally exhibit higher catalytic activity than metal oxides, bimetallic compounds tend to show better catalytic activity than monometallic compounds. However, there is currently a lack of bimetallic phosphides nanozymes with enhanced POD- and HPO-like activities to selectively produce ·OH and HBrO/HClO for antibacterial applications. Here, FeCu–P/carbon dots (FeCu–P/CDs) nanozymes show multi-enzyme activity to catalyze ROS production for enhanced antibacterial activity. FeCu–P/CDs nanozymes demonstrate POD-like, HPO-like and superoxide dismutase (SOD)-like activities. POD-like activity of FeCu–P/CDs nanozymes with a specific activity of 37.9 U/mg is 8.2 times improved than Cu–P/CDs. FeCu–P/CDs nanozymes show enhanced antibacterial activity by catalyzing the production of ·OH and HBrO/HClO through dual pathways, respectively. Based on the POD-like catalytic pathway, FeCu–P/CDs nanozymes achieved antibacterial rates exceeding 99.9 % against E. coli and S. aureus at 25 μg/mL and 80 μg/mL, respectively, which are significantly higher than those of Cu–P/CDs and Fe–P/CDs. Based on the HPO-like catalytic pathway, FeCu–P/CDs nanozymes demonstrated antibacterial effects against E. coli and S. aureus , achieving an antibacterial rate of 98.7 % against E. coli at 400 μg/mL. This work will inspire the exploration of a variety of bimetallic species-phosphate/carbon nanomaterials with high catalytic activity to enhance antibacterial properties via multi-enzyme activity pathway.

In this work, a simple one-step thermal oxidation process was established to achieve a significant surface increase in {110} and {111} nanofacets on well-defined, pure and Pr-doped, ceria nanocubes. More importantly, without changing most of the bulk properties, this treatment leads to a remarkable boost of their enzymatic activities: from the oxidant (oxidase-like) to antioxidant (hydroxyl radical scavenging) as well as the paraoxon degradation (phosphatase-like) activities. Such performance improvement might be due to the thermally generated sawtoothlike {111} nanofacets and defects, which facilitate the oxygen mobility and the formation of oxygen vacancies on the surface. Finally, possible mechanisms of nanoceria as artificial enzymes have been proposed in this manuscript. Considering the potential application of ceria as artificial enzymes, this thermal treatment may enable the future design of highly efficient nanozymes without changing the bulk composition.

Monitoring the quality consistency of traditional Chinese medicines, or herbal medicines (HMs), is the basis of assuring the efficacy and safety of HMs during clinical applications. The purpose of this work was to characterize the difference in hydrophilic antioxidants and related bioactivities betweenFlos Chrysanthemum(JH) and its wild relatives (Chrysanthemum indicumL.; YJH) based on the establishment of fingerprint–efficacy relationship modeling. The concentrations of the total phenolics and flavonoids of JH samples were shown to be generally higher than those of YJH, but the concentration distribution ranges of YJH were significantly greater compared to JH samples, possibly related to environmental stress factors leading to the concentration fluctuations of phytochemicals during the growth and flowering ofChrysanthemumcultivars. Correspondingly, the total antioxidant capabilities of JH were greatly higher than those of YJH samples, as revealed by chemical assays, including DPPH and ABTS radical scavenging activities and FRAP assays. In addition, cellular-based antioxidant activities confirmed the results of chemical assays, suggesting that the differences in antioxidant activities among the different types ofChrysanthemumswere obvious. The extracts from YJH and JH samples showed significant α-glucosidase inhibitory activity and lipase-inhibitory activity, implying the modulatory effects on lipid and glucose metabolisms, which were also confirmed by an untargeted cell-based metabolomics approach. The selected common peaks by similarity analysis contributed to the discrimination of YJH and JH samples, and the modeling of the fingerprint–bioactivity relationship identified neochlorogenic acid, isochlorogenic acid A, and linarin as efficacy-associated chemical markers. These results have demonstrated that integrating HPLC fingerprints and the analysis of similarity indexes coupled with antioxidant activities and enzyme-inhibitory activities provides a rapid and effective approach to monitoring the quality consistency of YJH/JH samples. Keywords:Chrysanthemum;antioxidant;cellular antioxidant activity;α-glucosidase inhibitory activity;lipase-inhibitory activity;fingerprint–activity relationship

The magnetization strategy of isoquinoline alkaloids has been successfully used in the extraction and isolation, but the effect of the magnetization on biological activities of those alkaloids still deserves further investigation. Therefore, the antibacterial, lipid-lowering and antioxidant activities of five isoquinoline alkaloids (berberine, tetrahydroberberine, palmatine, tetrahydropalmatine and tetrahydropapavine) before and after magnetization were compared in this study, and the results showed that the relevant activities were enhanced after magnetization. Additionally, among the five magnetic derivatives studied, berberine magnetic derivative ([Ber·H] + [FeCl 4 ] − ) had the best antibacterial effect on S. aureus and E. coli with MIC of 200 and 800 µM, respectively; palmatine magnetic derivative ([Pal·H] + [FeCl 4 ] − ) showed the strongest lipid-lowering activity with IC50 of 429 mM, and the inhibitory effect and type on lipase was reversible and mixed inhibition of competition and non-competition type; tetrahydroberberine magnetic derivative ([THBer·H] + [FeCl 4 ] − ) had the strongest antioxidant activity. This study provides new ideas and references for the further application of magnetization strategy.

The present study was designated to evaluate the antioxidant, antibacterial and antischistosomal activities of Grateloupia livida (GL) extracts in vitro . A GL Ethanol extract (EE) was separated into petroleum ether (PE), ethyl acetate (EA), n-butyl alcohol (BuOH) and aqueous (AQ) fractions to fractionate the polar and non-polar compounds in the EE. Extracts antioxidant activities were evaluated in vitro by DPPH radical-scavenging, deoxyribose radical scavenging, and β-carotene bleaching assays, all using butylated hydroxytoluene (BHT) as the reference antioxidant compound. The most effective antioxidant properties were observed in the PE fraction in all three assays. Antimicrobial testing showed that the PE fraction exhibited broad-spectrum antimicrobial activity, with the PE fraction also exhibiting strong activity against the human pathogenic trematode S. japonicum adult worm. In order to investigate the relationships between bioactivity and chemical composition, the chemical composition of the PE fraction was analyzed by gas chromatography-mass spectrometry (GC-MS). In total, 25 components were identified in the PE fraction, most of which have known antioxidant and antimicrobial activities. However, none of the compounds have reported activity against Schistosoma, suggesting that the schistosomicidal activity of the PE fraction may be related to minor constituents present in the extract, or governed by more intricate synergistic or additive relationships. Finally, fractions with the greatest biological activity displayed neither cellular cytotoxicity, at concentrations up to 100 ug/ml, or acute oral toxicity in mice, at doses up to 2000 mg/kg. Based on antioxidant, antimicrobial, antischistosomal activities, and low toxicity, the PE fraction possesses properties useful for food preservation and overall improvement of human health.
