Giant panda feces: potential raw material in preparation of nitrocellulose for propellants

Duan Xikai,Li Zhaoqian,Shi Xianrui,Pei Chonghua

It is highly anticipated to explore low-priced raw materials for preparing nitrocellulose (NC). The use of low-priced raw materials reduces environmental pollution while utilizing waste. In this paper, it is fascinating to prepare NC for propellants based on giant panda feces. The synthesis of NC based on giant panda feces and bitter bamboo stems as raw materials were labeled as P-NC and B-NC, respectively. Compared with cotton-based NC (NC12.5%), P-NC has certain advantages in decomposition heat, burning flame and speed, and mechanical properties. However, this advantage is very slight. The gastrointestinal digestion of giant pandas shortens the time for extracting α-cellulose. P-NC and B-NC are very similar in structure and properties, except that P-NC maintains a higher degree of crystallinity. The results show that P-NC can replace cotton-based NC in terms of performance for propellants. It should be emphasized that this paper is based on the use of animal excrement, which not only protects the environment and saves resources, but also reduces the workload of the staff at the giant panda conservation base. In addition, this paper also provides us with a new idea about waste utilization.

Tongue Prick Bionic Angularly Adjustable Microneedles for Enhanced Scarless Wound Healing

Ni Gan,Xin Li,Meng Wei,Zhijie Li,Shu Zhou,Bingbing Gao

High-tension site wounds are frequently accompanied by challenges associated with hypertrophic scarring. The key to achieving scar-free healing is the creation of a mechanical environment conducive to scar-free skin regeneration. Herein, simple rolling punctures are utilized on angle transform molds to develop cat tongue prick bionic angle-adjustable microneedles (TPMNs) to maintain a firm grip on the periwound skin, thereby reducing tissue tension. The integration of TPMNs with triboelectric nanogenerators (TENGs) enables excellent conductive and triboelectric properties. The system can provide a stable spatial electric field around the wound to promote cell migration. As the microfluid reaches the TPMNs, the self-driving force is enhanced by a unique angle design to control the microfluid flow rate. Sufficient evidence has shown that TPMNs expedite wound contraction and skin tissue regeneration while concurrently reducing scar formation in mouse trauma model experiments. The innovative TPMNs-TENGs synergistically provides a highly functional platform for wound tension relief, which is suitable for scar treatment in this study and potentially extends to the construction and regulation of smart wearable devices.

Targeted and direct intracellular delivery of native DNAzymes enables highly specific gene silencing

Xia Li,Fang Yang,Wenjiao Zhou,Ruo Yuan,Yun Xiang

DNAzymes exhibit high potential as gene silencing agents for therapeutic applications. Such purposes, however, are significantly challenged by the targeted and successful delivery of unmodified DNAzymes into cells with minimal side effects. Here, we set out to formulate and demonstrate a new stimuli-responsive and constrained aptamer/DNAzyme (Apt/Dz) catenane nanostructure for highly specific gene silencing. The rational design of the Apt/Dz catenane nanostructure with the respective integration of the aptamer sequence and the completely closed catenane format enables both the targeted capability and significantly improved nuclease resistance, facilitating the stable and targeted delivery of unmodified Dz into cancer cells. Moreover, the Dz enzymatic activity in the constrained structure can only be conditionally regulated by the specific intracellular mRNA sequences to silence the target gene with highly reduced side effects. Results show that the Apt/Dz catenane nanostructure can effectively inhibit the expression of the target gene and the proliferation of cancer cells with high specificity.

Controlling cathepsin B activity in Jinhua ham through multifactorial analysis and modeling of temperature, pH and salt content

Shiqi Hu,Xinglian Xu,Wangang Zhang,Chunbao Li,Guanghong Zhou

The activity of cathepsin B in Jinhua ham was affected by various processing parameters, which were investigated and quantified using a multi-layer artificial neural network (ANN). The key factors that influence ham quality are processing temperature, pH, and sodium chloride content. The study discussed the univariate analysis and multivariate interactions between two or three factors. The established ANN model was used for further investigation, and the actual enzyme activity of cathepsin B was characterized. The results showed that high temperature (greater than 60 °C) and an alkaline environment or the addition of Na + cation significantly reduced the enzyme activity ( p  < 0.05). The optimal conditions for protease activity were found to be at 40 °C and pH 6.0. The enzyme was found to be unstable at a temperature of 20 °C or pH 6.5–7.0 in the presence of NaCl, indicating significant interactions between temperature, pH value and NaCl content ( p  < 0.01). Furthermore, the ANN model showed good match between true and predicted values. The correlation coefficient was 0.98468, and the model provided supplementary information about the effect of NaCl on cathepsin B activity within 0.4M–0.6M. The findings could be useful in Jinhua ham production and the study of the structure-activity relationship of cathepsin B.

Molybdenum Disulfide Nanoflakes Covered Carbonized Catkin Microtube Hybrids as Superior Catalysts for Electrochemical Hydrogen Evolution

Shanshan Tong,Shenghua Ma,Yingchun Su,Qingchuan Li,Xuejing Wang,Xiaojun Han

The synergistic effect of non-noble-metal, low-cost, and high-performance hybrid electrocatalysts was often used for hydrogen evolution reaction (HER). Herein, the molybdenum disulfide nanoflakes were in situ grown on carbon microtube derived from natural tubular willow catkins as templates to construct the three-dimensional catkins/MoS2 hybrids. Catkins/MoS2 hybrids exhibited excellent HER activity in acidic conditions with a remarkable small onset overpotential of 50 mV, a Tafel slope of 62 mV decade–1, a comparatively small overpotential of 136 mV at 10 mA cm–2, and a Faraday yield of ∼100%. The as-synthesized catalysts also presented impressive durability and long-term stability. The superior electrocatalytic activity of catkins/MoS2 hybrids resulted from the unique hybrid structure of the catkins microtube and MoS2 nanoflakes providing abundant catalytically active sites and the annealed-catkins providing the good conductive support. The hybrids may find potential in practical hydrogen generation.

Effects of spatial distribution of the nuclear localization sequence on gene transfection in catiomer–gene polyplexes

Xiaojun Cai,Haiqing Dong,Junping Ma,Haiyan Zhu,Wei Wu,Meng Chu,Yongyong Li,Donglu Shi

A spatial effect of targeted signal distribution is found in catiomer–gene polyplexes which strongly influences the gene transfection profiles. The nuclear localization sequence PKKKRKV (NLS) is chosen as the target ligand. For a given gene delivery system of constant composition, size, and charge, it is engineered such that the NLS is the only structural variable in polyplexes. This unique architecture of polyplexes relies on successive electrostatic interaction. The structural features of the PKKKRKV sequence in polyplexes are investigated by NMR. The gene transfection profiles of the polyplexes are found to be well correlated with the spatial distribution of the nuclear localization sequence. Upon modification of nuclear localization, the gene transfection efficiency is found to increase remarkably from 40% to 60%. The drastic improvement of gene transfection is explained by a NLS spatial distribution mechanism. High gene transfection based on spatial NLS distribution provides a new base for the design and development of non-viral gene delivery vectors.

Development and validation of an LC–MS/MS method for the simultaneous quantification of milbemycin oxime and praziquantel in plasma: application to a pharmacokinetic study in cats

Shiting Xie,Yixing Lu,Jun Wang,Changcheng Lin,Peiyu Ye,Xiaolin Liu,Wenguang Xiong,Zhenling Zeng,Dongping Zeng

Introduction Milbemycin oxime (MBO) and praziquantel (PZQ) have a broad spectrum of biological activity and are commonly used to treat the parasitic infection in the veterinary clinic. In this study, a fast and efficient LC-MS/MS method was established and validated for the simultaneous determination of MBO, PZQ, cis-4-hydroxylated-PZQ (C-4-OH-PZQ) and trans-4-hydroxylated-PZQ (T-4-OH-PZQ) and in cat plasma. Methods Extraction of analytes and internal standards from cat plasma by acetonitrile protein precipitation, allows rapid processing of large batches of samples. MBO, PZQ, C-4-OH-PZQ, T-4-OH-PZQ, and internal standard (IS) were eluted for 13.5 min on a C 18 column with a 0.1% formic acid water/acetonitrile mixture as the mobile phase. Results Results showed that the method had good precision, accuracy, recovery, and linearity. The linearity range was 2.5–250 ng/mL for MBO, and 10–1000 ng/mL for PZQ, C-4-OH-PZQ, and T-4-OH-PZQ. The intra-day and inter-day precision CV values of the tested components were within 15%. The extraction recoveries of the four components ranged from 98.09% to 107.46%. The analytes in plasma remained stable for 6 h at room temperature, 26 h in the autosampler (4 °C), after freeze–thaw (−20°C) cycles, and 60 days in a −20°C freezer. Method sensitivity sufficed for assessing pharmacokinetic parameters of MBO, PZQ, C-4-OH-PZQ, and T-4-OH-PZQ in plasma samples with LLOQ of 2.5 ng/mL for MBO and 10 ng/mL for PZQ, C-4-OH-PZQ, and T-4-OH-PZQ. Conclusion In this study, a selective and sensitive LC-MS/MS method for the simultaneous quantification of MBO, PZQ, C-4-OH-PZQ, and T-4-OH-PZQ in cat plasma was developed and validated. This method had been successfully applied to evaluate the pharmacokinetics of MBO, PZQ, C-4-OH-PZQ, and T-4-OH-PZQ after a single oral administration of 8 mg MBO and 20 mg PZQ in cats.

Targeting Catenibacterium mitsuokai with icariin modulates gut microbiota and improves hepatic lipid metabolism in intrauterine growth restriction

Wei Yusen,Mao Jiangdi,Tang Wenjie,Ma Yanfei,Li Jiachen,Su Songtao,Ni Zhixiang,Wu Jinhong,Liu Daren,Wang Haifeng

Male offspring with intrauterine growth restriction exhibit more pronounced hepatic lipid metabolism abnormalities than females, necessitating earlier intervention. Icariin has been shown to effectively modulate hepatic lipid metabolism in male piglets with intrauterine growth restriction. However, the role of gut microbiota in this process remains to be elucidated. This study aimed to explore the influence of gut microbiota on icariin-induced enhancement of hepatic lipid metabolism. By examining changes in microbiota composition and hepatic lipid metabolism following icariin intervention, the study demonstrated an association between microbial alterations and hepatic lipid regulation through fecal microbiota transplantation. The impact of Catenibacterium on gut microbiota structure and hepatic lipid metabolism was assessed in vivo, and the direct effect of icariin on Catenibacterium was explored in vitro. Results revealed that icariin intervention modified fecal, ileal, and colonic microbiota in male piglets with intrauterine growth restriction, enhanced gut morphology and barrier function, and normalized the expression of hepatic peroxisome proliferator-activated receptor (PPAR) signaling pathway-related genes. Fecal microbiota transplantation from piglets with intrauterine growth restriction impaired intestinal barrier function and led to hepatic lipid deposition, whereas transplantation from icariin-treated donors showed no pathological changes, an outcome associated with reduced abundance of Catenibacterium. Mechanistically, icariin inhibits adenosine triphosphate synthesis to suppress Catenibacterium, remodels gut microbiota, reduces lipopolysaccharide production and translocation, and activates the hepatic PPARα/CD36 axis. In conclusion, icariin intervention alleviates hepatic lipid metabolic disorders in male offspring with intrauterine growth restriction by suppressing Catenibacterium, restoring gut microbial balance, and enhancing intestinal barrier integrity to limit lipopolysaccharide translocation.

Development and Validation of an LC–MS/MS Method for Quantifying Gabapentin in Plasma: Application to a Pharmacokinetic Study in Cats

Feifei Zhao,Changcheng Lin,Yunying Wu,Xinyue Luo,Ning Han,Wenguang Xiong,Zhenling Zeng

Simple SummaryGabapentin is an analogue of the neurotransmitter γ-aminobutyric acid, which is commonly used to treat neuropathic pain and epilepsy. However, studies have shown that early use of gabapentin can significantly reduce cats’ stress and increase their compliance with veterinary examinations. There is currently little research on the pharmacokinetics of gabapentin tablets for oral administration in cats. Consequently, monitoring gabapentin plasma levels provides critical insights into therapeutic adherence and facilitates early identification of toxicity thresholds. In this study, a novel LC-MS/MS assay was established and comprehensively validated to achieve high-throughput quantification of gabapentin in feline plasma resolution within 7 min. Additionally, this study provides further clinical evaluation for the management of sedation and anti-stress in cats.Gabapentin (GBP), a γ-aminobutyric acid analogue used for neuropathic pain and epilepsy, can reduce cat stress and improve veterinary exam compliance. A sensitive LC-MS/MS assay was established and rigorously validated for quantifying GBP in feline plasma. The method employed acetonitrile-mediated protein precipitation to efficiently extract GBP and its internal standard, pregabalin (PGB). Chromatographic separation was achieved within 7 min using a C18 column (2.1 × 50 mm, 1.7 μm) with a gradient mobile phase comprising 0.1% (v/v) formic acid in water and acetonitrile. The pharmacokinetics of 25  mg/kg GBP was studied by single-dose oral and intravenous administration. The results demonstrated that the method exhibited satisfactory precision, accuracy and linearity. The pharmacokinetic results showed that the Tmax, Cmax, T1/2and AUC0–tof GBP in cats after oral administration of 25  mg/kg were (1.83 ± 0.75) h, (13.94 ± 3.75) μg/mL, (5.60 ± 1.79) h and (115.54 ± 27.56) (μg/mL) h, respectively. The results of the study indicated that after intravenous administration of 25 mg/kg GBP, the C0, T1/2and AUC0–twere (58.82 ± 15.34) μg/mL, (3.87 ± 0.64) h and (160.44 ± 32.65) (μg/mL) h, respectively. The oral bioavailability of GBP in cats was (78.71 ± 18.55)%. In this study, a selective and sensitive LC-MS/MS method for the quantification of GBP in cat plasma was developed and validated. This method was successfully employed to assess the pharmacokinetics following the oral and intravenous administration of GBP in cats.

Ultralight Poly(vinyl alcohol)/Cattail Fiber Cellulose Aerogels as Green and Recyclable Organic Liquid Captors

Zhi Li,Kai Shen,Miao Liu,Min Hu,Shengjie Yao,Qiong Liu,Chuangang Fan,Xiaoxu Wu

Oil–water separation has attracted widespread attention from researchers because of increasing worldwide water pollution. Therefore, it is urgent to design an environmentally friendly material with a high efficiency oil–water separation property. Herein, a facile strategy for fabricating ultralight, biodegradable, and high porosity methyltrimethoxysilane (MTMS) modified poly(vinyl alcohol) (PVA)/cellulose aerogels (PCAs) by chemical vapor deposition is reported. The obtained results show that the oil–water separation ability of PCA is enhanced with the increase of the PVA load. Besides, the hydrophobicity of the modified PCA is improved relative to that of pristine PCA. In particular, the water contact angle of MTMS-P0.4CA was 140.3°. Furthermore, PCA modified MTMS exhibits an excellent repeated adsorption capacity for organic liquids. The as-prepared aerogels with outstanding hydrophobicity and oil–water separation ability show great potential as recyclable organic liquid captors.

Cathepsin B and thermal dual-stimuli responsive linear-dendritic block copolymer micelles for anticancer drug delivery

Wenjie Song,Junwu Wei,Lindong Li,Yangyang Qian,Yujia Wang,Yunmei Bi

In this study, two linear poly( N -vinylcaprolactam)s (PNVCLs) with different molecular weights were first synthesized as polymeric supporters by reversible addition-fragmentation chain transfer polymerization and then were used to prepare cathepsin B and thermal dual-stimuli responsive amphiphilic linear–dendritic block copolymers (LDBCs) PNVCL n - b - d (Phe-Lys) 1–3 ( n = 66, 100) with linear PNVCL and dendritic phenylalanyl-lysine (Phe-Lys) dipeptides via stepwise peptide chemistry. The copolymers were characterized by 1 H NMR spectra and gel permeation chromatography and exhibited a controlled molecular weight and a narrow molecular weight distribution (polydispersity index ≤ 1.27). They self-assemble into stable micelles at relatively low critical micelle concentrations (0.007–0.019 mg mL −1 ) in aqueous solution. These micelles are capable of dissociating to release the encapsulated anticancer drug doxorubicin upon cathepsin B treatment. The doxorubicin release rates from PNVCL n - b - d (Phe-Lys) 1–3 micelles decreased with increasing generation of Phe-Lys dipeptide dendrons and decreasing molecular weight of PNVCL. In addition, the lower critical solution temperatures of the copolymers increased with an extension of the enzyme incubation time. The results of in vitro cytotoxicity and cell uptake experiments showed that LDBCs show no cytotoxicity even at a high LDBC concentration of 0.4 mg mL −1 and the drug-loaded micelles can be enriched in tumor cells. Such smart copolymers could be efficiently used in targeted therapy systems. © 2021 Society of Industrial Chemistry.

In vitro modeling of feline gut fermentation: a comprehensive analysis of fecal microbiota and metabolic activity

Qianle Ren,Yuling Li,Mingmei Duan,Jinjun Li,Fangshu Shi,Yun Zhou,Wanjing Hu,Junfu Mao,Xiaoqiong Li

The gut microbiota (GM) is a large and diverse microbial community that plays essential roles in host health. The in vitro fermentation model of the fecal GM serves as a valuable complement to food and health research in both humans and animals. Despite advancements in standardized protocols for culturing human GM, research concerning animals—particularly companion animals—remains limited. This study aims to identify the optimal in vitro fermentation method for cat gut microbiota by comprehensively analyzing fecal microbiota and fermentation characteristics. We evaluated seven culture media previously used to simulate the gut microenvironment in humans, dogs, and cats: anaerobic medium base (AMB), Minimum medium (MM), Pet medium (PM), VI medium (VI), VL medium (VL), Yeast culture medium (JM), and yeast casitone fatty acid agar medium (YCFA). Fresh fecal samples were fermented in these media for 48 h, followed by 16S rRNA sequencing to assess bacterial community composition and targeted metabolite monitoring during fermentation. The results revealed that the substrate composition in the medium differentially impacts bacterial community structure and fermentation characteristics. High levels of carbon and nitrogen sources can substantially increase gas production, particularly CO2, while also significantly enhancing the production of short-chain fatty acids (SCFAs). Additionally, substrates with a high carbon-to-nitrogen ratio promote the production of more SCFAs and biogenic amines, and enrich the Bacteroidaceae family, even when the total substrate amount is lower. Comprehensive analysis of gut microbiota and metabolites reveals that PM medium effectively simulates a nutrient-deficient microenvironment in the cat gut during in vitro fermentation. This simulation maintains bacterial community stability and results in lower metabolite levels. Therefore, using PM medium to culture cat gut microbiota for 48 h, without focusing on specific bacterial genera, represents the most suitable in vitro model. This finding contributes to understanding the optimal conditions for simulate cat gut microbiota and may provide a new approach for investigating the food pharmaceuticals on the cat gut microbiota and related health.
