Bioengineered Microrobot for Targeted and Autonomous Eradication of Helicobacter pylori

Shali Tan,Ruijue Dan,Junfeng Guo,Qinglin Wei,Kaige Xu,Qiang Luo,Yuchen Zhang,Chaoqiang Fan,Xiaoyan Zhao,Donghui Zhu,Quan Wang,Malcolm Xing,Shiming Yang,Yu Huang

Helicobacter pylori ( H. pylori ) infection remains a major challenge in gastric disease treatment, with conventional antibiotic therapies hindered by poor gastric retention, lack of specificity, and the growing threat of bacterial resistance. To address these limitations, we introduce a novel bioengineered robotic system inspired by icebreakers, integrating multi-functional nanomaterials for targeted, autonomous, and efficient H. pylori eradication. Our micro-robots combine a platinum nanozyme-based calcium carbonate complex with chitosan (Cs) modification, gastric epithelial cell membranes (CM) coating, and pronase (P) incorporation, enabling selective targeting and autonomous motility. Upon gastric acid exposure, this system undergoes controlled decomposition, generating carbon dioxide to propel movement, releasing pronase to penetrate and degrade mucus barriers, and deploying Pt nanozymes for deep bacterial eradication. This multi-modal mechanism significantly enhances gastric retention, precision targeting, and therapeutic effectiveness. Our in vitro and in vivo studies validate its superior antibacterial activity, biocompatibility, and its unprecedented ability to mitigate H. pylori -induced inflammation while preserving intestinal microbiota balance. By combining bioengineering, nanotechnology, and autonomous propulsion, this innovation represents a paradigm shift in H. pylori therapy, offering a precise, efficient, and resistance-mitigating alternative to conventional treatments.

Autonomous Liquid Metal Droplets Actuated by Ion Diffusion

Qianqiang Tang,Fuhong Wang,Mingchao Zhang,Wenfu Chen,Pengyu Chen,Jiqian Tan,Shiyang Shi,Xiaosa Li,Wenhui Shang,Ben Wang,Yaokang Zhang,Metin Sitti,Hongzhang Wang,Xuechang Zhou

The autonomous systems with embodied energy and intelligence show great potential to build future robots that are inspired by living organisms. This work proposes a liquid metal droplet-based machine that can be self-propelled in a controllable way through surrounding [CuCl 4 ] 2- ions interaction. Different autonomous behaviors of liquid metal droplets can be tuned by altering the distribution [CuCl 4 ] 2- of ions and the immersing ratio of CuCl 2 solution on liquid metal droplets. The underlying mechanisms behind the deformation and locomotion of the liquid metal droplet are attributed to the unbalanced surface tension and surface Marangoni flow induced by synergistic effect of galvanic replacement reaction and in situ displacement reaction. The autonomous liquid metal droplets based on microfluidics and sodium dodecyl benzene sulfonate (SDBS)-modified CuCl 2 solution are demonstrated, which induces the realization of shuttling a narrow channel, offset correction, climbing low-rise stairs, medium transportation, circuit repair, chemotactic transfer and escape from a maze independently. This work elucidates the autonomous liquid metal droplet (ALMD) driven by surrounding [CuCl 4 ] 2- ions, highlighting motion mode switching, interpretation of motion mechanisms, multifunctional manipulation and application, which offers a new path for further development of liquid robots with autonomous capability and embodied energy, and progressing toward physically intelligent robots.

Fenobam modulates distinct electrophysiological mechanisms for regulating excessive gamma oscillations in the striatum of dyskinetic rats

Pengfei Wang,Weina Dai,Hongbin Liu,Han Liu,Yuming Xu

Gamma oscillations have been frequently observed in levodopa-induced dyskinesia (LID), manifest as broadband (60-120 Hz) and narrowband (80-110 Hz) gamma activity in cortico-striatal projection. We investigated the electrophysiological mechanisms and correlation of gamma oscillations with dyskinesia severity, while assessing the administration of fenobam, a selective metabotropic glutamate receptor 5 (mGluR5) antagonist, in regulating dyskinesia-associated gamma activity. We conducted simultaneous electrophysiological recordings in Striatum (Str) and primary motor cortex (M1), together with Abnormal Involuntary Movement Scale scoring (AIMs). Phase-amplitude coupling (PAC), power, coherence, and Granger causality analyses were conducted for electrophysiological data. The findings demonstrated increased beta oscillations with directionality from M1 to Str in parkinsonian state. During on-state dyskinesia, elevated broadband gamma activity was modulated by the phase of theta activity in Str, while M1 → Str gamma causality mediated narrowband gamma oscillations in Str. Striatal gamma power (both periodic and aperiodic power), periodic power, peak frequency, and PAC at 80 min (corresponding to the peak dyskinesia) after repeated levodopa injections across recording days (day 30, 33, 36, 39, and 42) increased progressively, correlating with total AIMs. Additionally, a time-dependent parabolic trend of PAC, peak frequency and gamma power was observed after levodopa injection on day 42 from 20 to 120 min, which also correlated with corresponding AIMs. Fenobam effectively alleviates dyskinesia, suppresses enhanced gamma oscillations in the M1-Str directionality, and reduces PAC in Str. The temporal characteristics of gamma oscillations provide parameters for classifying LID severity. Antagonizing striatal mGluR5, a promising therapeutic target for dyskinesia, exerts its effects by modulating gamma activity.

Drug-Loaded Zwitterion-Based Nanomotors for the Treatment of Spinal Cord Injury

Tianyi Bao,Nan Li,Huan Chen,Zinan Zhao,Jin Fan,Yingfang Tao,Chenglong Chen,Mimi Wan,Guoyong Yin,Chun Mao

Spinal cord injury (SCI) treatment requires a nanosystem for drug delivery that can effectively penetrate the blood–spinal cord barrier (BSCB). Herein, we designed poly(2-methacryloyloxyethyl phosphorylgallylcholine) (PMPC)/l-arginine (PMPC/A)-based nanomotors that can release nitric oxide (NO). The nanomotors were loaded with the inducible NO synthase inhibitor 1400W and nerve growth factor (NGF). PMPC with a zwitterionic structure not only provided good biocompatibility for the nanomotors but also facilitated their passage through the BSCB owing to the assistance of a large number of choline transporters on the BSCB. Additionally, the l-arginine loaded on the nanomotors was able to react with reactive oxygen species in the microenvironment of the injured nerve to produce NO, thereby conferring the ability of autonomic movement to the nanomotors, which facilitated the uptake of drugs by cells in damaged areas and penetration in pathological tissues. Moreover, in vivo animal experiments indicated that the PMPC/A/1400W/NGF nanomotors could effectively pass through the BSCB and restore the motion function of a rat SCI model by regulating its internal environment as well as the release of therapeutic drugs. Thus, the drug delivery system based on nanomotor technology offers a promising strategy for treating central nervous system diseases.

Design of Near Infrared Light-Powered Copper Phyllosilicate Nanomotors for Cuproptosis-Based Synergistic Cancer Therapy

Yibo Song,Guowu Zhan,Shu-Feng Zhou

Cuproptosis, a newly discovered cell death pathway, has shown great potential in cancer treatment. Herein, near-infrared (NIR) light-driven nanomotors (CuSiO 3 @Au-Pd NMs) are designed for cuproptosis-assisted synergistic therapy with autonomous mobility and improved cellular uptake and tumor penetration. Specifically, the released Cu 2+ ions from CuSiO 3 @Au-Pd NMs can induce the Fenton-like reaction, leading to the generation of hydroxyl radicals (·OH), accompanied by the depletion of glutathione within the MCF-7 cells. Additionally, the designed CuSiO 3 @Au-Pd NMs also exhibit excellent photothermal effects, which can further promote the production of ·OH, resulting in intensified oxidative stress and cellular apoptosis. Moreover, the enhanced tumor permeation efficiency and cellular uptake of CuSiO 3 @Au-Pd NMs via autonomous movement under self-thermophoretic forces are proved using 2D cellular experiments and 3D multicellular tumor spheroids. The resultant intracellular accumulation of Cu 2+ can induce the oligomerization of lipoylated proteins, leading to cuproptosis, along with the mitochondrial dysfunction pathway. More importantly, both in vitro and in vivo experiments show that CuSiO 3 @Au-Pd NMs could penetrate deeply into tumors and exhibit great anticancer efficacy through multimodal therapeutic methods. These findings manifest promising potentials of NIR-powered Cu-based NMs with high maneuverability for future smart and synergistic cancer therapy.
