The widespread application of neonicotinoid insecticides (NNIs) has attracted widespread attention to their potential ecotoxicological effects. In this study, we systematically evaluated the toxic effects of thiamethoxam (TMX) and its metabolite clothianidin (CLO) on earthworms ( Eisenia fetida ). Specifically, the antioxidant system responses and endogenous metabolite metabolism responses in earthworms were analyzed in the temporal dimension after 7, 14, 21 and 28 days of exposure to TMX and CLO. The results found that TMX and CLO could inhibit the growth phenotype of earthworms and cause significant changes in antioxidant system related indicators. More importantly, we found that TMX and CLO could cause significant changes in the metabolic profiles of earthworms through NMR-based metabolomics. From the changes in endogenous metabolites, the toxicity effects of TMX on earthworms gradually increases with prolonged exposure time. Differently, the toxicity effects of CLO on earthworms is significantly higher than that of TMX in the early stages of exposure. Meanwhile, these impacts will not weaken with prolonged exposure time. Furthermore, the results of KEGG enrichment pathway analysis indicated that TMX and CLO could significantly interfere with energy homeostasis, redox homeostasis, osmotic regulation, amino acid metabolism and protein synthesis in earthworms. These findings further deepen our understanding of the ecotoxicological effects of NNIs on soil organism.

Arsenic and perfluorooctane sulfonate (PFOS) are widely distributed pollutants in the soil environment and could coexist as mixture, but little studies have been reported about their combined toxicity to terrestrial invertebrates. In the present work, we exposed earthworm Eisenia fetida to sub-lethal levels of arsenate (As(V)), PFOS, and their mixture for 30 days. Multiple biomarker responses and bioaccumulation of the two toxicants in the organisms were analyzed to assess the joint toxicity of As(V) and PFOS. Bioaccumulation analysis results demonstrated that the coexistence of As(V) and PFOS in soil could increase the bioavailability of As(V) while decreasing the bioavailability of PFOS. As the level of exposure increased, significant variations were observed in all of the biomarkers analyzed, indicating growth inhibition and oxidative damage. The multiple biomarker responses were summarized by using the Integrated Biomarker Response (IBR) index, and significant dose–response relationships were found in treatments of As(V), PFOS and As(V)/PFOS mixture. Then the toxic interaction between As(V) and PFOS was assessed by combining IBR and two mixture toxicity indices, Effect Addition Index (EAI) and Concentration Addition Index (CAI). Results showed that the joint toxicity of As(V) and PFOS depended on the exposure concentration of As(V). The combined pollution of PFOS with lower concentration of As(V) led to synergistic interaction at the whole range of effect levels, while that with higher concentration of As(V) resulted in the toxicity interaction changing from antagonism to synergism as the effect level increased. As(V) was the main toxicant that caused adverse effects on earthworm biomarkers and affected the joint toxicity of As(V)/PFOS mixture. This study, for the first time, provides valuable toxicological data for the risk evaluation of combined toxicity of As(V) and PFOS in the soil environment. Moreover, this work indicates that the integration of IBR index and EAI/CAI could be an effective and practical method for the characterization of toxicity interaction within binary mixture systems.

Ubiquitous contamination of microplastics and arsenic in soil ecosystems can induce many health issues to nontarget soil organisms, and will also cause many potential threats to the gut bacterial communities of soil fauna . However, the changes in the gut bacterial communities of soil fauna after exposure to both microplastics and arsenic remain unknown. In this study, the toxicity and effects on the gut microbiota of earthworm Metaphire californica caused by the combined exposure of microplastics and arsenic were examined by using arsenic species analysis and high throughput sequencing of gut microbiota. Results showed that total arsenic and arsenic species in the earthworm gut and body tissues after exposure to combination of microplastics with arsenate (As(V)) were significantly different from that treated with As(V) alone. Microplastics lessened the accumulation of total arsenic and the transformation rate of As(V) to arsenite (As(III)). Microplastics alleviated the effect of arsenic on the gut microbiota possibly via adsorbing/binding As(V) and lowering arsenic bioavailability , thus prevented the reduction of As(V) and accumulation of total arsenic in the gut which resulted in a lower toxicity on the earthworm. The study broadens our understanding of the ecotoxicity of microplastics with other pollutants on the soil animals and on their gut microbiota.

In this paper, several studies were conducted to elucidate the binding mechanism of earthworm fibrinolytic enzyme (EFE) with imidocloprid (IMI) by using theoretical calculation, fluorescence, UV–vis, circular dichroism spectroscopy and an enzymatic inhibition assay. The spectral data showed that the binding interactions existed between IMI and EFE. The binding constants, binding site, thermodynamic parameters and binding forces were analyzed in detail. The results indicate a single class of binding sites for IMI in EFE and that this binding interaction is a spontaneous process with the estimated enthalpy and entropy changes being 2.195 kJ mol −1 and 94.480 J mol −1 K −1 , respectively. A single class of binding site existed for IMI in EFE. The tertiary or secondary structure of EFE was partly destroyed by IMI. The visualized binding details were also exhibited by the theoretical calculation and the results indicated that the interaction between IMI and Phe (Tyr, or Trp) or EFE occurred. Combining the experimental data with the theoretical calculation data, we showed that the binding forces between IMI and EFE were mainly hydrophobic force accompanied by hydrogen binding, and π–π stacking. In addition, IMI did not obviously influence the activity of EFE. In a word, the above analysis offered insights into the binding mechanism of IMI with EFE and could provide some important information for the molecular toxicity of IMI for earthworms.

The use of plastics has increased significantly with consequent rise in the generation of wastes. Microplastics (MPs) with particle size <5 mm are produced in natural terrestrial habitats by weathering of the discarded plastic debris and therefore are likely to impact soil biota . Earthworms are the dominant soil fauna which play vital role in soil formation and decomposition of organics. Since these animals are soil feeders, MP particles contaminating soil are likely to enter in to the gut of these animals affecting their physiology. MPs have been shown to be potent adsorbents of various other pollutants such as heavy metals and agrochemicals . This study reports the effects of two MPs, polyvinyl chloride (PVC) and polypropylene (PP) alone and in combination with the pesticide monocrotophos in soil on tissue protein, lipid peroxidation (LPX), activities of lactate dehydrogenase (LDH) and catalase (CAT) of an epigeic earthworm Eudrillus eugeniae over an exposure period of 48h. Results from molecular docking and laboratory experiment confirmed that both the MPs are potent adsorbents of the pesticide and enhanced oxidative stress on the animal with significant reduction in protein, increased LPX level and enzyme activities . PP indicated significantly higher pesticide adsorption relative to PVC.

Microplastics (MPs) are currently receiving widespread attention worldwide, and their co-occurrence with antibiotics is unavoidable. However, our understanding of how protists respond to co-pollution and mediate antibiotic resistance genes (ARGs) profiles remains exceedingly limited, particularly within non-target animals’ guts. To bridge these gaps, we investigated the individual and combined effects of polyethylene and sulfamethoxazole (SMZ) on microbial communities and ARGs in soil and earthworm guts. We found that the MP–SMZ combination significantly elevated the abundance and richness of ARGs in the soil and earthworm. Protistan compositions (particularly consumers) responded more strongly to pollutants than did bacterial and fungal communities, especially under combined pollution. Interkingdom cooccurrence network analysis revealed that protists had stronger and more effective interactions with the resistome in the earthworm guts, suggesting that the impact of these protists on ARGs compositional changes was potentially modulated through the “top-down” regulation of bacteria and fungi. Meta-cooccurrence networks further confirmed that protist-related networks had more keystone pollution-sensitive ASVs ( ps ASVs) and these ps ASVs were mostly associated with protistan consumers. Our study highlights protists as promising agents for regulating and monitoring microbial functions, as well as the ecological risks of the antibiotic resistome associated with MPs and SMZ pollution in agricultural ecosystems.

N-ethyl perfluorooctane sulfonamide (N-EtFOSA) is commonly known as the active ingredient of sulfluramid. It can be degraded to perfluorooctane sulfonic acid (PFOS) in biota and environment. Earthworms ( Eisenia fetida ) were exposed with N-EtFOSA to examine the bioaccumulation, elimination and metabolism of N-EtFOSA by the earthworms after in vivo and in vitro exposure. N-EtFOSA could be biodegraded in quartz sands to perfluorooctane sulfonamide (FOSA) and PFOS. In the in vivo tests, in addition to parent N-EtFOSA, three metabolites, including perfluorooctane sulfonamide acetate (FOSAA), FOSA and PFOS also accumulated in earthworms as a result of N-EtFOSA biotransformation , with FOSA as the predominant metabolite. The bioaccumulation factor (BAF) and uptake rate coefficient ( k u ) of N-EtFOSA from sand were 20.4 and 2.41·d −1 , respectively. The elimination rate constants ( k e ) decreased in the order FOSAA (0.130·d −1 ) > N-EtFOSA (0.118·d −1 ) > FOSA (0.073·d −1 ) > PFOS (0.051·d −1 ). The biotransformation of N-EtFOSA in earthworm was further confirmed by the in vitro test involving incubation of earthworm homogenates with N-EtFOSA. This work provides evidence on the accumulation and transformation of N-EtFOSA in terrestrial invertebrates and will be helpful to explore the indirect sources of FOSA and PFOS in environmental biota.

Antibiotics in wastewater present significant ecological risks. However, their effects on nitrogen (N) removal and greenhouse gas (GHG) emissions in constructed wetlands (CWs) have been largely neglected in previous studies. This study investigated the effects of antibiotics on N removal and GHG emissions in earthworm-enhanced CWs constructed with three earthworm densities (0 g m -2 , 156 g m -2 , and 312 g m -2 ). The results showed that (1) antibiotics significantly increased effluent total inorganic nitrogen (TIN) concentrations by 49.17 % in systems with the high-density earthworm, whereas no significant effects were observed in the other treatments; (2) in systems without earthworms, antibiotics significantly reduced N₂O emissions by 139 %. In systems with low-density earthworms, CH₄ and CO₂ emissions were significantly reduced by 366 % and 70.3 %, respectively. In contrast, in high-density earthworm systems, N₂O emissions significantly increased by 289 %; (3) antibiotics led to a significant reduction in non-CO₂ global warming potential (GWP non-CO₂ , -143 %) in the systems without earthworms and a reduction in total global warming potential (GWP CH₄+N₂O+CO₂ , -57.3 %) in the systems with low-density earthworms. However, both indices significantly increased by 289 % and 51.2 % in systems with high-density earthworms; (4) antibiotics altered the microbial community composition and significantly increased nitrite reductase activity in the systems with low-density earthworms (+24.5 %) but significantly decreased it in systems with high-density earthworms (-33.0 %). In summary, antibiotic contamination markedly influenced both N removal performance and GHG emissions in earthworm-enhanced CWs, with effects varying depending on earthworm density.

In this study, we investigated the bioavailability and toxicity of metals (Cu and Ni) in the soil containing polyethylene microplastics (PE-MPs). The bioavailability of the metals determined by the five-step chemical sequential extraction method increased with the addition of MPs (0.1%, 1%, 10%) in the soil, which was confirmed by the adsorption-desorption characteristics. To further examine the bioavailability and toxicity of metals, earthworms ( Eisenia fetida ) were exposed to soil containing Cu 2+ (100 mg/kg) or Ni 2+ (40 mg/kg) with different amounts (0.01%, 0.05%, and 0.1%) of PE-MPs for 21 days. The highest concentrations of Cu 2+ and Ni 2+ in earthworms reached to 73.3 and 36.3 mg/kg, respectively. Moreover, metal concentrations in earthworms increased with MP contents in the soil, which was consistent with the bioavailability measured by the sequential extraction method. Furthermore, changes in biomarkers including peroxidase (POD), catalase (CAT) and superoxide dismutase (SOD) activity, malondialdehyde (MDA) contents, and related gene expression levels in earthworms suggested that the pollutants caused toxicity to earthworms. Overall, MPs increased the bioavailability of metals in the soil and the toxic effects to earthworms. These findings provide insights regarding the impacts of MPs on the bioavailability of metals and the combined toxic effects of these two kinds of pollutants on terrestrial animals.

Triazole contaminants in water and soil environments can form complexes with metal ions, and therefore affect the bioavailability and toxicity of some heavy metals. In present study, significant increase of copper (Cu) uptake by earthworm ( Eisenia fetida ) was observed when combined pollution of benzotriazole (BTR) presented in soil. For instance, Cu accumulation in earthworms increased 55% approximately when BTR presented at the BTR/Cu molar ratio of 1:2.5. While the single Cu exposure (at 32 mg kg −1 in soil) resulted in increased malondialdehyde (MDA) content in earthworms from 0.319 to 0.668 nmol mg protein −1 , joint exposure to BTR at BTR/Cu molar ratio of 1:10 significantly decreased the MDA content to 0.405 nmol mg protein −1 . This indicates a potential detoxification effect of BTR to Cu induced oxidative damage in earthworms. Varied Cu subcellular distribution can be observed in earthworms of the single and combined exposure treatments. With the combined exposure of BTR, the proportion of Cu associated with granular fraction, the toxically inert fraction in earthworms, increased from 25% to 39%. This phenomenon can be used to explain the protective effects of BTR against oxidative damage.

Microcystins (MCs) have a significant influence on aquatic ecosystems, but little is known about their terrestrial fate and impact. Here, we investigated the fate of two MCs (MC-LR and MC-RR) in the soil-earthworm system, with consideration of their congener-specific impact on earthworm health, soil bacteria, and soil metabolome. Although MCs had little acute lethal effect on earthworms, they caused obvious growth inhibition and setae rupture. Relative to MC-RR, MC-LR exhibited higher bioaccumulation and the resulting dermal lesions and deformation of longitudinal muscles. While the incorporation of both MCs into soils stimulated pathogenic bacteria and depressed oxidative stress tolerant bacteria, the response among soil nitrification and glutathione metabolism differed between the two congeners. The dissipation kinetics of MCs obeyed the first-order model. Earthworms stimulated soil N-cycling enzyme activities, increased the abundance of MC-degrading bacteria, and promoted bacterial metabolic functions related to glutathione metabolism, xenobiotics biodegradation, and metabolism of amino acids that comprise MCs, which accelerated the dissipation of MC-LR and MC-RR by 227% and 82%, respectively. These results provide evidence of significant congener differences in the terrestrial fate and impact of MCs, which will enable a better understanding of their role in mediating soil functions and ecosystem services.

PS, one of the most extensively used synthetic polymers, persists in terrestrial ecosystems owing to its high chemical stability and inherent resistance to degradation. Yet, the biological mechanisms for its degradation remain unclear. This study investigated the degradation of PS-MPs mediated by intestinal microorganisms from two earthworm species, Eisenia fetida and Perionyx guillelmi . An integrative approach combining SEM, GC-MS, and microbial diversity analysis was applied to characterize alterations, identify degradation intermediates, and assess gut microbial dynamics. The PS-MP content in soil decreased progressively, achieving degradation efficiencies of 2.33 % on day 7 and 56.67 % on day 28. Three bacterial strains , Bacillus cereus, Klebsiella variicola, and Citrobacter portucalensis , exhibited PS degradation efficiencies of 2.33 %, 2.48 %, and 2.60 %, respectively. Collectively, our findings provide compelling evidence that the earthworm intestinal microbiota actively mediate the biodegradation of PS-MPs. These results elucidate a microbial biodegradation pathway and highlight earthworm microorganisms as promising biocatalysts for sustainable plastic bioremediation.
