A 3D-printed integrated maneuverable device for sensitive colorimetry of Pseudomonas aeruginosa

Jizhou Li,Shiruoyu Zhou,Ruining Bai,Zhifeng Fu

Timely and sensitive monitoring of pathogens in clinical specimen is highly demanded for efficient control and precise treatment of infectious diseases. Herein, a 3D-printed maneuverable device integrating incubation, washing, and detection functions was manufactured for colorimetry of Pseudomonas aeruginosa ( P. aeruginosa ) using transparent resin. The device combined with magnetic beads (MBs) can achieve specific separation and efficient enrichment of P. aeruginosa . Furthermore, it can be directly fixed onto a 96-well plate holder for colorimetry. Specifically, a P. aeruginosa bacteriophage termed as JZ1 acquired from river water was applied as a recognition reagent to functionalize the separation vectors MBs. Then, a nanoconfinement MOFs material termed as PCN-222(Pt) with remarkable peroxidase-like activity was conjugated with polymyxin B to act as a signal tracer. With the formation of target bacterial complexes, the bound PCN-222(Pt) catalyzed the color reaction of 3,3′,5,5′-tetramethylbenzidine to enable quantitative colorimetry of P. aeruginosa by the maneuverable device. With this device, P. aeruginosa can be quantified within 40 min, with a dynamic range of 1.9 × 10 2 ∼ 1.9 × 10 6 cfu mL −1 . The results for colorimetry of P. aeruginosa in diverse sample matrixes demonstrated its satisfactory practicability. This work provides a facile, timely, and cost-effective technique for point-of-care testing of pathogens.

Beyond denitrification: Alginate production by mucoid Pseudomonas aeruginosa using wastewater nitrate as electron acceptor

Xinyi Tao,Kun Zou,Mengze Yuan,Yixuan Wen,Benben Liu,Taiping Qing,Bo Feng,Peng Zhang

Microbial synthesis is an effective method for carbon and nitrogen recovery in the biological nitrogen removal process. The recycling of alginate-like exopolymers has attracted the attention of many researchers; however, the composition of the polymers and alginate differs. In this study, the capacity and mechanism of a mucoid Pseudomonas aeruginosa to remove nitrate and synthesize alginate were investigated. The results showed that the P. aeruginosa achieved higher nitrate removal efficiency (95.3 %) and alginate yield (298 mg/L) after 36 hours of microaerobic cultivation compare to aerobic condition. The optimal conditions for nitrogen removal and alginate recovery are 35 °C, pH 7 and C/N 15. The transcriptional activities of denitrifying genes in P. aeruginosa were upregulated at the low dissolved oxygen level. Although O 2 may upregulate the expression of alginate synthesis key genes ( algD , alg8 and alg44 ) by activating AlgZ-AlgR two-component signal transduction system, microaerobic condition facilitates P. aeruginosa proliferation and results in a higher alginate yield. Moreover, the P. aeruginosa strain was further coupled with an anoxic/aerobic reactor for deeper nitrogen removal, and the nitrate removal efficiency and alginate yield were maintained above 91 % and 100 mg/L, respectively. The conversion efficiency of sodium acetate to alginate reached 15 %. This work provides an economically and environmentally sustainable strategy for wastewater treatment and the recovery of high-value bioresource.

In situ monitoring of quorum sensing signalling molecules using a SERS chip with a micro -chamber array

Lulu Liu,Wenrui Ma,Haolan Tan,Chuang Ge,Bei Zhang,Chunyan Wang,Yang Zhang,Yi Xu

The in situ monitoring of quorum sensing (QS) signal molecules in bacterial biofilms is crucial for the diagnosis and treatment of bacterial biofilms. In this work, a surface-enhanced Raman scattering (SERS) chip integrated with functional units of bacterial biofilms and in situ SERS detection is proposed for the monitoring of pyocyanine, an important signal molecule and biomarker secreted by Pseudomonas aeruginosa. The SERS chip was composed of a top self-assembled nano-silver SERS substrate layer, a PDMS layer containing a micro-chamber array and bottom quartz layer. The nano-silver SERS substrate showed good homogeneity and stability with an enhancement factor of up to 1.84 × 108 for pyocyanine, and the design of the SERS substrate located at the top layer of the chip could effectively attenuate interference from complex matrices. Using the SERS chip, a detection limit of as low as 1 nM for pyocyanine with a wide detection range of 1 nM–100 μM was observed. The proposed SERS chip could achieve the monitoring of pyocyanine during the growth of Pseudomonas aeruginosa biofilms with and without the treatment of different types of antibacterial drugs. Furthermore, pyocyanine secreted by Pseudomonas aeruginosa was confirmed using mass spectrometry (MS), and the concentration relationship obtained via MS/MS was consistent with that obtained via SERS detection. The proposed method based on the SERS chip for monitoring QS in bacterial biofilms offers the advantages of in situ detection, flexibility and efficiency.

Efficient Aflatoxin B1 degradation by a novel isolate, Pseudomonas aeruginosa M-4

Yanhua Xu,Huiyan Dong,Chenxi Liu,Haiwei Lou,Renyong Zhao

Aflatoxin B 1 (AFB 1 ) is the most common mycotoxin with the highest toxicity, posing a threat for food contamination. In this study, the bacterium Pseudomonas aeruginosa M-4 was isolated from rotten wood, and its culture supernatant showed a 56.79% AFB 1 degradation rate. Treatment with proteinase K, sodium dodecyl sulfate, and trichloroacetic acid decreased the AFB 1 degradation rate in the culture supernatant to 20.71%, 10.83%, and 19.62%, respectively. Moreover, heating the culture supernatant at 121 °C for 30 min increased in the AFB 1 degradation rate to 84.76%, indicating effective degradation by heat-resistant and inducible extracellular proteins or enzymes. P. aeruginosa M-4 reduced the content of AFB 1 in contaminated maize (90.57%, 14 d). Additionally, high-performance liquid chromatography and ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry analyses showed that AFB 1 was biotransformed into structurally different compounds, (C 17 H 16 O 6 ), (C 16 H 14 O 5 ), (C 17 H 14 O 5 ), and (C 16 H 10 O 6 ). According to the structure-activity relationship, these degradation products were produced by the destruction of partial toxic sites of AFB 1 , so the toxicity of these products may be lower than that of AFB 1 . Thus, P . aeruginosa M-4 has excellent potential for application in the detoxification of AFB 1 in food and feed.

Highly Specific Bacteriophage-Affinity Strategy for Rapid Separation and Sensitive Detection of Viable Pseudomonas aeruginosa

Yong He,Mengyao Wang,Enci Fan,Hui Ouyang,Huan Yue,Xiaoxiao Su,Guojian Liao,Lin Wang,Shuguang Lu,Zhifeng Fu

A virulent bacteriophage highly specific to Pseudomonas aeruginosa (P. aeruginosa) was isolated from hospital sewage using a lambda bacteriophage isolation protocol. The bacteriophage, named as PAP1, was used to functionalize tosyl-activated magnetic beads to establish a bacteriophage-affinity strategy for separation and detection of viable P. aeruginosa. Recognition of the target bacteria by tail fibers and baseplate of the bacteriophage led to capture of P. aeruginosa onto the magnetic beads. After a replication cycle of about 100 min, the progenies lysed the target bacteria and released the intracellular adenosine triphosphate. Subsequently, firefly luciferase-adenosine triphosphate bioluminescence system was used to quantitate the amount of P. aeruginosa. This bacteriophage-affinity strategy for viable P. aeruginosa detection showed a linear range of 6.0 × 102 to 3.0 × 105 CFU mL–1, with a detection limit of 2.0 × 102 CFU mL–1. The whole process for separation and detection could be completed after bacteria capture, bacteriophage replication, and bacteria lysis within 2 h. Since the isolated bacteriophage recognized the target bacteria with very high specificity, the proposed strategy did not show any signal response to all of the tested interfering bacteria. Furthermore, it excluded the interference from inactivated P. aeruginosa because the bacteriophage could replicate only in viable cells. The proposed strategy had been applied for detection of P. aeruginosa in glucose injection, human urine, and rat plasma. In the further work, this facile bacteriophage-affinity strategy could be extended for detection of other pathogens by utilizing virulent bacteriophage specific to other targets.

Improved polyhydroxybutyrate production by Cupriavidus necator and the photocatalyst graphitic carbon nitride from fructose under low light intensity

Junting Wang,Mengying Xu,Pier-Luc Tremblay,Tian Zhang

The photocatalyst graphitic carbon nitride (g-C 3 N 4 ) is known to photostimulate the production of the bioplastic polyhydroxybutyrate (PHB) by Cupriavidus necator . In previous studies, the combination of C. necator and g-C 3 N 4 increased PHB yield from either an organic or inorganic carbon substrate under a light intensity of 4200 lx. Here, different parameters including light intensity, pH, temperature, nitrogen and carbon concentrations, aeration, and inoculum size were explored to maximize PHB production by hybrid photosynthesis from fructose and visible light. A g-C 3 N 4 / C. necator culture grown with a lower light intensity of 2100 lx, an inoculum size of 128.30 × 10 6   CFU  ml −1 , and constant aeration produced 7.16 g l −1 d −1 PHB with a product yield from fructose of 60.94%. Furthermore, the ratio of incident photons harvested by g-C 3 N 4 converted into NADPH+H + by C. necator for PHB production was improved to 19.74% after the process optimization. In comparison, the PHB production rate of a non-optimized g-C 3 N 4 / C. necator system exposed to 4200 lx was only 2.94 g l −1 d −1 with a product yield from fructose of 33.29%. These results demonstrate that hybrid photosynthesis productivity can be significantly augmented by decreasing light intensity and adjusting other parameters, which is promising for future bioproduction applications.

Co-culture of benzalkonium chloride promotes the biofilm formation and decreases the antibiotic susceptibility of a Pseudomonas aeruginosa strain

Caihong Wang,Qiao Ma,Jiaxin Zhang,Nan Meng,Dan Xu

Benzalkonium chloride (BAC) is a disinfectant with broad-spectrum antibacterial properties, yet despite its widespread use and detection in the environments, the effects of BAC exposure on microorganisms remain poorly documented. Herein, the impacts of BAC on a Pseudomonas aeruginosa strain Jade-X were systematically investigated. The results demonstrated that the minimum inhibitory concentration (MIC) of BAC against strain Jade-X was 64 mg/L. Exposure to BAC concentrations of 8, 16, 32, and 64 mg/L significantly augmented biofilm formation by 2.03-, 2.43-, 2.96-, and 2.56-fold respectively. The swimming and twitching abilities, along with the pyocyanin production, were inhibited. Consistently, quantitative reverse transcription PCR assays revealed significant downregulation of genes related to flagellate- and pili-mediated motilities (flgD, flgE, pilB, pilQ, and motB), as well as phzA and phzB genes involved in pyocyanin production. The results of disk diffusion and MIC assays demonstrated that BAC decreased the antibiotic susceptibility of ciprofloxacin, levofloxacin, norfloxacin, and tetracycline. Conversely, an opposite trend was observed for polymyxin B and ceftriaxone. Genomic analysis revealed that strain Jade-X harbored eleven resistance-nodulation-cell division efflux pumps, with mexCD-oprJ exhibiting significant upregulation while mexEF-oprN and mexGHI-opmD were downregulated. In addition, the quorum sensing-related regulators LasR and RhlR were also suppressed, implying that BAC might modulate the physiological and biochemical behaviors of strain Jade-X by attenuating the quorum sensing system. This study enhances our understanding of interactions between BAC and P. aeruginosa, providing valuable insights to guide the regulation and rational use of BAC.

Antimicrobial blue light inactivation of Pseudomonas aeruginosa: Unraveling the multifaceted impact of wavelength, growth stage, and medium composition

Yucheng Wang,Xue Li,Hongtong Chen,Xinyi Yang,Lei Guo,Rui Ju,Tianhong Dai,Guoqing Li

Pseudomonas aeruginosa , a notable pathogen frequently associated with hospital-acquired infections, displays diverse intrinsic and acquired antibiotic resistance mechanisms, posing a significant challenge in infection management. Antimicrobial blue light (aBL) has been demonstrated as a potential alternative for treating P. aeruginosa infections. In this study, we investigated the impact of blue light wavelength, bacterial growth stage, and growth medium composition on the efficacy of aBL. First, we compared the efficacy of light wavelengths 405 nm, 415 nm, and 470 nm in killing three multidrug resistant clinical strains of P. aeruginosa . The findings indicated considerably higher antibacterial efficacy for 405 nm and 415 nm wavelength compared to 470 nm. We then evaluated the impact of the bacterial growth stage on the efficacy of 405 nm light in killing P. aeruginosa using a reference strain PAO1 in exponential, transitional, or stationary phase. We found that bacteria in the exponential phase were the most susceptible to aBL, followed by the transitional phase, while those in the stationary phase exhibited the highest tolerance. Additionally, we quantified the production of reactive oxygen species (ROS) in bacteria using the 2′,7′-dichlorofluorescein diacetate (DCFH-DA) probe and flow cytometry, and observed a positive correlation between aBL efficacy and ROS production. Finally, we determined the influence of growth medium on aBL efficacy. PAO1 was cultivated in brain heart infusion (BHI), Luria-Bertani (LB) broth or Casamino acids (CAA) medium, before being irradiated with aBL at 405 nm. The CAA-grown bacteria exhibited the highest sensitivity to aBL, followed by those grown in LB broth, and the BHI-grown bacteria demonstrated the lowest sensitivity. By incorporating FeCl 3 , MnCl 2 , ZnCl 2 , or the iron chelator 2,2′-bipyridine (BIP) into specific media, we discovered that aBL efficacy was affected by the iron levels in culture media.

Imidazole-Rich, Four-Armed Host-Defense Peptidomimetics as Promising Narrow-Spectrum Antibacterial Agents and Adjuvants against Pseudomonas Aeruginosa Infections

Yuwei Zheng,Jiaying Chi,Jiayu Ou,Ling Jiang,Liqing Wang,Rui Luo,Yilang Yan,Zejun Xu,Tingting Peng,Jianfeng Cai,Chuanbin Wu,Peng Teng,Guilan Quan,Chao Lu

The development of narrow-spectrum antimicrobial agents is paramount for swiftly eradicating pathogenic bacteria, mitigating the onset of drug resistance, and preserving the homeostasis of bacterial microbiota in tissues. Owing to the limited affinity between the hydrophobic lipid bilayer interior of bacterial cells and most hydrophilic, polar peptides, the construction of a distinctive class of four-armed host-defense peptides/peptidomimetics (HDPs) is proposed with enhanced specificity and membrane perturbation capability against Pseudomonas aeruginosa by incorporating imidazole groups. These groups demonstrate substantial affinity for unsaturated phospholipids, which are predominantly expressed in the cell membrane of P. aeruginosa , thereby enabling HDPs to exhibit narrow-spectrum activity against this bacterium. Computational simulations and experimental investigations have corroborated that the imidazole-rich, four-armed peptidomimetics exhibit notable selectivity toward bacteria over mammalian cells. Among them, 4H10, characterized by its abundant and densely distributed imidazole groups, exhibits impressive activity against various clinically isolated P. aeruginosa strains. Moreover, 4H10 has demonstrated potential as an antibiotic adjuvant, enhancing doxycycline accumulation and exerting effects on intracellular targets by efficiently disrupting bacterial cell membranes. Consequently, the hydrogel composed of 4H10 and doxycycline emerged as a promising topical agent, significantly diminishing the skin P. aeruginosa burden by 97.1% within 2 days while inducing minimal local and systemic toxicity.

Constructing nanostructured functional film on EH40 steel surface for anti-adhesion of Pseudomonas aeruginosa

Xin Cheng,Jiashun Shi,Weigang Wang,Huimin Liao,Shiqiang Chen,Guangzhou Liu,Jvna Chen

Biofilm formation is an important cause of microbiologically influenced corrosion (MIC), and inhibition of microbial adhesion is one of the most effective ways to control biofilm formation. In this work, nanostructured functional film of EH40 steel is prepared by one-step anodization in NaOH solution to inhibit adhesion of Pseudomonas aeruginosa ( P. aeruginosa ) without and with light. Results show that the functional films fabricated on EH40 steel surface in a series of NaOH solutions inhibit the adhesion of P. aeruginosa , and its anti-adhesion performance first increases and then decreases with varying concentrations of NaOH solution. The optimal anti-adhesion performance under dark condition, i.e., anti-adhesion rate of 92.7% compared with bare steel, is obtained for the nanostructured oxide film fabricated in 12 M NaOH solution under anodization potential of 1.0 V for 2 h. Under visible light irradiation, the best anti-adhesion rate increases to 99.9% due to hydroxyl radicals ( OH) produced by the nanostructured oxide film with photocatalytic activity .

Glycomimetic-Conjugated Photosensitizer for Specific Pseudomonas aeruginosa Recognition and Targeted Photodynamic Therapy

Yu Zhao,Zhentan Lu,Xiaomei Dai,Xiaosong Wei,Yunjian Yu,Xuelei Chen,Xinge Zhang,Chaoxing Li

Due to the rapid development of bacterial resistance, there is an urgent need to explore new antibacterial agents to substitute for traditional antibiotic therapy. Photodynamic therapy has been identified as a promising bactericidal method to conquer antibiotic-resistant pathogens. To solve the problem of photosensitizer damage to normal tissues in vivo, we developed a boron-dipyrrolemethene (BODIPY)-based glycosylated photosensitizer for ablating Pseudomonas aeruginosa (P. aeruginosa). This glycosylated photosensitizer exhibited good water solubility and generated 1O2 rapidly in an aqueous solution under light exposure. The photosensitizer did not cause detectable toxicity to human cells in the dark. Importantly, the photosensitizer was able to selectively attach to P. aeruginosa over normal cells, thus resulting in effective pathogen ablation by reactive oxygen species. Moreover, the photosensitizer inhibited over 90% of the biofilm formation produced by P. aeruginosa. The results indicate that the design of the macromolecular photosensitizer-induced bacterial death and inhibited biofilm formation provide a novel strategy for overcoming bacterial infection.

Selective and Sensitive Detection of Pseudomonas aeruginosa Based on Aminoguanidine-Functionalized Carbon Dots Synthesized by One-Step Hydrothermal Method

Liu Mei,Sun Zhaomeng,Guo Rong

Developing effective detection systems of pathogenic microbes is urgent to prevent human from bacterial infection. In this study, Pseudomonas aeruginosa ( P. aeruginosa ) as a model foodborne pathogen, we report a versatile strategy for construction of functionalized carbon dots-based sensors for detection of P. aeruginosa . The aminoguanidine-functionalized carbon dots (AG-CDs) with high biocompatibility and low toxicity were synthesized by a greener hydrothermal reaction between ascorbic acid and aminoguanidine. The AG-CDs exhibit good trapping ability toward P. aeruginosa due to the binding of the aminoguanidine residues to lipopolysaccharide (LPS) on P. aeruginosa . The fluorescence provided by AG-CDs/ P. aeruginosa conjugates is closely correlated with P. aeruginosa content. The linear detection range is 3–3 × 10 7  CFU/mL and the limit of detection is 1 CFU/mL. The fluorescence sensor exhibited successful practical applications of detecting P. aeruginosa in water and food. The finding provides the great potential of simple, inexpensive and efficient advantages in bacteria determination.
