A miscellaneous polysaccharide, PAP55–3-1, with a molecular weight of 23.03 kDa, was isolated from Periplaneta americana through extraction with dilute alkali solution, ethanol precipitation, and column chromatography purification. Structural analysis shows that PAP55–3-1 is mainly composed of five monosaccharides: galactosamine hydrochloride, glucosamine hydrochloride, galactose, glucose and mannose. Its main glycosidic bonds are: Manp-(1→, Galp-(1→, →3)-Galp-(1→, →3,6)-Manp-(1→, →2,6)-Manp-(1→, →6)-Manp-(1→, →4)-Galp-(1→, →6-Glcp-(1→, →6)-Galp-(1→, →2)-Manp-(1 →, →3,4)-Glcp-(1→, →3,6)-Galp-(1→. In vitro experiments demonstrated that PAP55–3-1 can effectively inhibit reactive oxygen species (ROS) and O 2 − production following H 2 O 2− induction. After H 2 O 2− induction, HIF-1α (hypoxia-inducible factor) was translocated in mitochondria PAP55–3-1 increased localization of HIF-1α was located on mitochondria to maintain the stability of mitochondrial function stability, thereby effectively inhibiting H 2 O 2 -induced mitochondrial oxidative damage. Additionally, PAP55–3-1 inhibited the M1 polarization of macrophages stimulated by H 2 O 2 and promoted the phenotype polarization of macrophages from M1 to M2, displaying anti-inflammatory and pro-repair properties. In vivo experimental results indicated that PAP55–3-1 promoted wound healing in mice. Immunohistochemical experiments revealed a reduction in CD68 expression and increase in CD206 expression in both positive and the high-dose polysaccharide group control group. This further demonstrated that PAP55–3-1 promotes the phenotype polarization of macrophages from M1 to M2, exerting anti-inflammatory and wound-healing activities.

Abstract Lam16A is a novel GH16 β-1,3–1,4-lichenase isolated from the genus Caldicellulosiruptor which can utilize untreated carbohydrate components of plant cell walls. Its catalytic module has been characterized that the six carbohydrate-binding modules (CBMs) were queued in the C-terminus, but their roles were still unclear. Here, full-length and CBM-truncated mutants of Lam16A were purified and characterized through heterologous expression in Escherichia coli . The profiles of these proteins, including the enzyme activity, degrading efficiency, substrate-binding affinity, and thermostability, were explored. Full-length Lam16A with six CBMs showed excellent thermostability and the highest activity against barley β-glucan and laminarin with optimum pH of 6.5. The CBMs stimulated degrading ability of the catalytic module, especially against β-1,3(4)-glucan-based polysaccharides. The released products from β-1,3–1,4-glucan by Lam16A or its truncated mutants revealed an endo-type glycoside hydrolase. Lam16As exhibited strong binding affinities to the insoluble polysaccharides, especially Lam16A-1CBM. The degradation of yeast cell walls by Lam16A enzyme solution relative to the control reduced the absorbance values at OD 800 by ~ 85% ± 1.2, enabling the release of up to ~ 0.057 ± 0.0039 µg/mL of the cytoplasmic protein into the supernatant, lowering the viability of the cells by ~ 70.3% ± 6.9, thus causing significant damage in the cell wall structure. Taken together, CBMs could influence the substrate specificity, thermal stability, and binding affinity of β-1,3–1,4-glucanase. These results demonstrate the great potential of these enzymes to promote the bioavailability of β-1,3-glucan oligosaccharides for health benefits.
Key points
Carbohydrate-binding modules strongly influenced the enzyme activity and binding affinity, and further impacted glycoside hydrolase activity.
Lam16A enzymes have sufficient ability to hydrolyze β-1,3–1,4-glucan-based polysaccharides.
Lam16As provide a powerful tool to promote the bioavailability of β-1,3-glucan oligosaccharides.

Lithium (Li) metal anode has attracted extensive research interest, while lithium dendrites and the resulting irreversible effects have restricted its development. In this study, a thin and dense solid electrolyte interface is constructed by introducing 3-(1-pyridinio)-1-propane sulfonate (PPS), a pyridine functional groups additive in DME/DOL based electrolyte to inhibit the lithium dendrite growth. The preferential decomposition of PPS has resulted in a reduction of gas generation. Subsequently, this process leads to forming of an N-rich solid electrolyte interface , which has been found to cause a decrease in polarization potential and change the behavior of lithium deposition from dendrites to a more cellular Li metal surface. As a result, our proposed electrolyte provides multiple functionalities, including remarkable rate capability, cyclic stability, and coulombic efficiency. Significantly, Li | Li symmetrical cells exhibited a stable cycle over 1200 h at a high current density and capacity of 5 mA cm −2 /15 mAh cm −2 . We are optimistic that our proposed method can be employed in future research to identify electrolytes appropriate for use in lithium metal anodes.

Polymer binders have been shown to efficiently conquer the notorious lithium polysulfide (LiPS) shuttle effects in lithium–sulfur (Li–S) batteries for years, but more study is needed. Herein, a water dispersible and molecular interaction regulated polymer binder (PNAVS) for Li–S batteries was elaborately designed by co-polymerizing N-acryloyl glycinamide and 3-(1-vinyl-3-imidazolio)propanesulfonate. We demonstrate that by modulating the multiple interactions between the functional groups through copolymerization the binder was able to coordinate the LiPSs with higher binding energy for shuttle effect alleviation and cycling performance improvement. In addition, the Li+ diffusion coefficient is also optimized in the PNAVS binder, which facilitates acceleration of the redox kinetics during cycling. Consequently, the PNAVS binder renders the Li–S battery with an ultrastable open circuit voltage for more than 3000 h. Even with a high sulfur loading of 11.7 mg cm–2, the battery can still exhibit excellent areal capacity of 12.21 mA h cm–2. As proof of concept, a pouch cell was also demonstrated with the stable cycling performance for 110 cycles. The binder engineering strategy in this work will propel the practical applications of high-performance batteries.
