Docetaxel-loaded redox-sensitive nanoparticles self-assembling from poly(caprolactone) conjugates with disulfide-linked poly(ethylene glycol)

Yongli Shi,Chunyan Li,Mingbo Yang,Xiaofei Pan,Jie Hu

In this study, novel redox-sensitive nanoparticles (NPs) were fabricated from the poly(caprolactone) conjugates with disulfide-linked poly(ethylene glycol) (DDMAT- mPEG-S-S-PCL, DPSP). The DPSP polymer was synthesized by ring-opening polymerization (ROP) and reversible addition-fragmentation chain transfer (RAFT) polymerization. The obtaining of the DPSP polymer was confirmed by the 1H nuclear magnetic resonance (1H NMR) and Fourier transform infrared spectroscopy (FTIR) spectra. The DPSP NPs were fabricated with the solvent-evaporation method. Docetaxel (DTX) was employed as a model drug and encapsulated into the DPSP NPs. The in vitro anti-tumor activity of the DTX-loaded DPSP NPs and free DTX against the breast cancer cells (4T1) were evaluated by MTT assay. The cargo-free DPSP NPs were in circular shapes with an average diameter of 107.8 ± 0.4 nm. These NPs displayed redox-responsive behavior in the presence of glutathione. Animal experiments indicated that the DPSP NPs showed excellent blood compatibility and good bio-security. Cell tests suggested that the DPSP NPs could be taken in by 4T1 cells, smoothly, which improved the anti-tumor activity of free DTX.

Binary Pt/Te Nanoheterostructures with High Photothermal Conversion Efficiency and Anti-inflammatory Action for Enhanced Photothermal Therapy of 4T1 Breast Tumors Guided by Photoacoustic Imaging

Chao-Qing Li,Xiao-Lin Hou,Dong-Xue Jiang,Bin Zhang,Meng-Wen Ma,Xiao-Ting Xie,Yuan-Di Zhao,Tian-Cai Liu,Bo Liu

Photothermal therapy is a powerful candidate for tumor treatment. However, photothermal therapy still faces some challenges, such as lacking photothermal agents with high photothermal conversion efficiency and undesirable inflammatory responses, which may result in tumor recurrence and therapeutic resistance. Here, the Pt/Te nanoheterostructures (PT) were synthesized by a simple hydrothermal reaction. The photothermal conversion efficiency was up to 51.84%. The outstanding photothermal conversion capacity of PT was attributed to the unique localized surface plasmon resonance frequency of metals and semiconductors and the increased circuit paths of electron transitions from nanoheterostructures. After coating with the murine mammary carcinoma (4T1) cell membrane, the camouflaged PT (mPT) exhibits excellent biocompatibility and effective homologous targeting capacity. Benefiting from antioxidative activity, mPT can efficiently scavenge inflammation-related reactive oxygen species and cytokines (such as tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6) caused by hyperthermia to alleviate inflammation in vitro and in vivo. The in vitro and in vivo therapeutic results showed that mPT could effectively inhibit 4T1 breast tumors. In addition, the in vivo therapy could be guided by photoacoustic imaging. These results demonstrated that these multifunctional mPT provide a paradigm for biomimetic metal and semiconductor nanoheterostructures for enhanced photothermal therapy and anti-inflammatory action on tumors.

Design, preparation and pharmacodynamics of ICG-Fe(Ⅲ) based HCPT nanocrystals against cancer

Qiongzhe Ren,Xuefeng Tang,Yi Lu,Qing Li,Zhiqian Liao,Shinan Jiang,Haoli Zhang,Zhigang Xu,Lei Luo

The use of nanocrystal technology to manufacture drug delivery systems intended to enhance therapeutic efficacy has attracted the attention of the pharmaceutical industry. However, the clinical application of nanocrystal drugs for injection is restricted by Ostwald ripening and the large-scale use of stabilizers such as polysorbate and lecithin, which have potential toxicity risks including hemolysis and allergies. Here, we designed an amorphous nanocrystal drug complex (IHNC), which is stabilizer-free and composed of indocyanine green (ICG) framework loading with a chemotherapeutic agent of 10-hydroxycamptothecin (HCPT). Considering the possibility of industrial manufacturing, IHNC was simply prepared with the assistance of ferric ion (III) via supramolecular assembly strategy. The theoretical result of Materials Studio simulation indicated that the prepared ICG-Fe(III) framework showed a stable spherical structure with the appropriate cavity for encapsulating the two drugs of HCPT and ICG with equal mass ratio. The IHNC was stable at physiological pH, with excellent PTT/PDT efficacy, and in vivo probing characteristics. The nanoscale size and reductive stimuli-responsiveness can be conducive to drug accumulation into the tumor site and rapid unloading of cargo. Moreover, such combination therapy showed synergistic photo/chemotherapy effect against 4T1 breast cancer and its tumor inhibition rate even up to 79.4%. These findings demonstrated that the nanocrystal drug delivery strategy could avoid the use of stabilizers and provide a new strategy for drug delivery for combination therapy.

Application of dual chemotherapeutic drug delivery system based on metal-organic framework platform in enhancing tumor regression for breast cancer research

Lin Zhan,Xuelian Yin,Li Qiu,Chenchen Li,Yanli Wang

The nanomedicine system based on dual drug delivery systems (DDDs) can significantly enhance the efficacy of tumor treatment. Herein, a metal-organic framework, Zeolite imidazole salt frames 8 (ZIF-8), was successfully utilized as a carrier to load the dual chemotherapeutic drugs doxorubicin (DOX) and camptothecin (CPT), named DOX/CPT@ZIF-8 (denoted as DCZ), and their inhibitory effects on 4T1 breast cancer cells were evaluated. The study experimentally demonstrated the synergistic effects of the dual chemotherapeutic drugs within the ZIF-8 carrier and showed that the ZIF-8 nano-carrier loaded with the dual drugs exhibited stronger cytotoxicity and inhibitory effects on 4T1 breast cancer cells compared to single-drug treatment. The use of a ZIF-8-based dual chemotherapeutic drug carrier system highlighted its potential advantages in suppressing 4T1 breast cancer cells.

Biological pH sensing based on the environmentally friendly Raman technique through a polyaniline probe

Li Songyang,Liu Zhiming,Su Chengkang,Chen Haolin,Fei Xixi,Guo Zhouyi

The biological pH plays an important role in various cellular processes. In this work, a novel strategy is reported for biological pH sensing by using Raman spectroscopy and polyaniline nanoparticles (PANI NPs) as the pH-sensitive Raman probe. It is found that the Raman spectrum of PANI NPs is strongly dependent on the pH value. The intensities of Raman spectral bands at 1225 and 1454 cm −1 increase obviously with pH value varying from 5.5 to 8.0, which covers the range of regular biological pH variation. The pH-dependent Raman performance of PANI NPs, as well as their robust Raman signals and sensitivities to pH, was well retained after the nanoparticles incorporated into living 4T1 breast adenocarcinoma cells. The data indicate that such PANI NPs can be used as an effective biological pH sensor. Most interestingly, the PANI spherical nanostructures can be acquired by a low-cost, metal-free, and one-pot oxidative polymerization, which gives them excellent biocompatibility for further biological applications.

Chitosan-based nanoparticles for survivin targeted siRNA delivery in breast tumor therapy and preventing its metastasis

Ping Sun,Wei Huang,Mingji Jin,Qiming Wang,Bo Fan,Lin Kang,Zhonggao Gao

Nanoparticle-mediated small interfering RNA (siRNA) delivery is a promising therapeutic strategy in various cancers. However, it is difficult to deliver degradative siRNA to tumor tissue, and thus a safe and efficient vector for siRNA delivery is essential for cancer therapy. In this study, poly(ethylene glycol)-modified chitosan (PEG-CS) was synthesized successfully for delivering nucleic acid drug. We deemed that PEGylated CS could improve its solubility by forming a stable siRNA loaded in nanoparticles, and enhancing transfection efficiency of siRNA-loaded CS nanoparticles in cancer cell line. The research results showed that siRNA loaded in PEGylated CS (PEG-CS/siRNA) nanoparticles with smaller particle size had superior structural stability in the physical environment compared to CS nanoparticles. The data of in vitro antitumor activity revealed that 4T1 tumor cell growth was significantly inhibited and cellular uptake of PEG-CS/siRNA nanoparticles in 4T1 cells was dramatically enhanced compared to naked siRNA groups. The results from flow cytometry and confocal laser scanning microscopy showed that PEG-CS/siRNA nanoparticles were more easily taken up than naked siRNA. Importantly, PEG-CS/siRNA nanoparticles significantly reduced the growth of xenograft tumors of 4T1 cells in vivo. It has been demonstrated that the PEG-CS is a safe and efficient vector for siRNA delivery, and it can effectively reduce tumor growth and prevent metastasis.

A novel hypoxia-activated polymeric Tirapazamine derivative for enhanced antitumor therapy

Yajun Xu,Jianlin Lv,Chaoying Kong,Yanran Li,Kun Wang,Na Shen,Zhaohui Tang

Tirapazamine (TPZ, 3-amino-1,2,4-benzotriazine 1,4-dioxide), a representative hypoxia-activated prodrugs (HAPs), has entered Phase II/III clinical trials. Despite a promising phase II clinical trial results, TPZ does not bring benefits to cancer therapy in most phase III clinical trials, which making the clinical trials failed. To improve its therapeutic effects, in this study, we synthesize a novel TPZ derivative, 3-(3-(5-hydroxypentyl)ureido)benzo[e][1,2,4]triazine 1,4-dioxide (TPZH), and covalently conjugated it to poly (L-glutamic acid) (PLG) to obtain TPZH nanoparticles (TPZH-NPs). The TPZH-NPs shows prolonged blood circulation, enhanced tumor accumulation, and increased maximum tolerated dose (MTD) than TPZH. In addition, TPZH-NPs exhibits significantly enhanced antitumor therapeutic efficiency than free TPZH against 4T1 breast tumors. Graphical

Diagnostic and Therapeutic Integration Prodrug: Dynamic Probe Engineering via Cys/GSH Depletion to Amplify Type I Photodynamic Therapy for Breast Cancer

Yongfei Huang,Junping Liang,Wenbo Lu,Chenzhong Li,Zhefeng Fan

Cancer is a serious threat to human health, and it is essential to find effective methods for detecting and treating cancer. Diagnostic integration is a new strategy for cancer diagnosis and treatment that enables localized detection and treatment of tumors in a single administration and effectively reduces the toxic side effects on normal tissues. It remains a challenge for small-molecule probes to achieve multifunctionality in sensing, targeting, imaging, and therapy. Photodynamic therapy (PDT), which uses photosensitizers to generate reactive oxygen species under specific wavelengths of light to ablate tumor tissue, has demonstrated unique advantages in tumor treatment. However, the high expression of Cys/GSH in the tumor region will consume the reactive oxygen species generated by photosensitizers during PDT and reduce the therapeutic effect of the tumor. In this study, the highly expressed Cys/GSH in the tumor microenvironment was used as the activation source, and benzopyrylium salt and coumarin dyes were selected as fluorophores, with the ether bond and aldehyde group as recognition sites, to construct a near-infrared-activated Cys/GSH fluorescent probe (Bpl-O-Cou). Based on the intramolecular rearrangement mechanism and intramolecular cyclization mechanism, Bpl-O-Cou could discriminate the detection of Cys/GSH accompanied by different fluorescence emissions. Under near-infrared laser irradiation, Bpl-O-Cou + Cys/GSH could produce •OH and O2–•, leading to 4T1 cell death. In vivo experiments further illustrated the interventional role of Bpl-O-Cou in the PDT process. This study provides a new idea for the design of multifunctional probes for cancer diagnostics and therapeutics.

A copper missile triggered power coalescence and death vortex within tumor cell mitochondria for synergistic cuproptosis/phototherapy/chemotherapy

Yicheng Jiang,Shuhan He,Niu Xiang,Linghui Duan,Yuxiang Lin,Wenyu Huang,Zhenghong Wu,Xiaole Qi

Definitely, copper homeostasis in mitochondria, and copper-triggered modality of mitochondrial cell death have been further confirmed. However, existing copper-based nanoplatforms were focused on synergistic therapies when the intracellular therapeutic targets were relatively scattered. Effective integrating of all targets within mitochondria to generate power coalescence remains a challenge. Herein, we develop a novel copper-based delivery system to trigger power coalescence and death vortex within tumor cell mitochondria. Specifically, a mitochondrial targeting "copper missile" loaded with curcumin (termed as Cur@CuS-TPP-HA, CCTH) was designed for cuproptosis/phototherapy/chemotherapy synergistic anti-tumor therapy. Once the CCTH NPs are shuttled to the mitochondria, near-infrared (NIR) irradiation initiates the release of copper ions and curcumin for in situ drug accumulation in cancer cell mitochondria. Excess of copper ions and curcumin can activate cuproptosis and mitochondrial apoptosis pathways respectively. When combined, they can cause an increase in reactive oxygen species (ROS), damage to mitochondrial DNA (mt-DNA), and a decrease in energy supply, thereby leading to a “vicious circle” of mitochondrial damage that further enhances tumor-killing efficacy. As a consequence, this "copper missile" exhibits advanced anti-tumor effects as verified through in vitro assessments and in vivo evaluations using the 4T1 breast tumor model, providing a promising approach for cuproptosis-based synergistic anti-tumor therapy.

Doxorubicin nanomedicine based on ginsenoside Rg1 with alleviated cardiotoxicity and enhanced antitumor activity

Chaoqi Li,Xiangbo Gou,Hui Gao

Aim: The authors aimed to develop Dox@Rg1 nanoparticles with decreased cardiotoxicity to expand their application in cancer. Materials & methods: Dox@Rg1 nanoparticles were developed by encapsulating doxorubicin (Dox) in a self-assembled Rg1. The antitumor effect of the nanoparticles was estimated using 4T1 tumor-bearing mice and the protective effect on the heart was investigated in vitro and in vivo. Results: Different from Dox, the Dox@Rg1 nanoparticles induced increased cytotoxicity to tumor cells, which was decreased in cardiomyocytes by the inhibition of apoptosis. The study in vivo revealed that the Dox@Rg1 nanoparticles presented a perfect tumor-targeting ability and improved antitumor effects. Conclusion: Dox@Rg1 nanoparticles could enhance the antitumor effects and decrease the cardiotoxicity of Dox.Lay abstractDoxorubicin (Dox) is a drug used to treat cancer; however, it can be toxic to the heart. In this study, researchers made nanoparticles containing Dox and a component of ginseng, a root similar to ginger. They tested the nanoparticles in mice with tumors. The nanoparticles appeared to gather at the tumor site in greater amounts than free Dox. In healthy mice, the nanoparticles gathered less in the heart than free Dox. This means that putting Dox into nanoparticles such as these could improve their anticancer effect and decrease harm to the heart.Graphical abstract

Anti-Tumor Efficacy of an Adjuvant Built-In Nanovaccine Based on Ubiquitinated Proteins from Tumor Cells

Fang Huang,Jinjin Zhao,Yiting Wei,Zhifa Wen,Yue Zhang,Xuru Wang,Yanfei Shen,Li-xin Wang,Ning Pan

Background and Aim We have previously identified ubiquitinated proteins (UPs) from tumor cell lysates as a promising vaccine for cancer immunotherapy in different mouse tumor models. In this study, we aimed at developing a highly efficient therapeutic adjuvant built-in nanovaccine (α-Al2O3-UPs) by a simple method, in which UPs from tumor cells could be efficiently and conveniently enriched by α-Al2O3 nanoparticles covalently coupled with Vx3 proteins (α-Al2O3-CONH-Vx3).Methods The α-Al2O3 nanoparticles were modified with 4-hydroxybenzoic acid followed by coupling with ubiquitin-binding protein Vx3. It was then used to enrich UPs from 4T1 cell lysate. The stability and the efficiency for the UPs enrichment of α-Al2O3-CONH-Vx3 were examined. The ability of α-Al2O3-UPs to activate DCs was examined in vitro subsequently. The splenocytes from the vaccinated mice were re-stimulated with inactivated tumor cells, and the IFN-γ secretion was detected by ELISA and flow cytometry. Moreover, the therapeutic efficacy of α-Al2O3-UPs, alone and in combination with chemotherapy, was examined in 4T1 tumor-bearing mice. Results Our results showed that α-Al2O3-UPs were successfully synthesized and abundant UPs from tumor cell lysate were enriched by the new method. In vitro study showed that compared to the physical mixture of α-Al2O3 nanoparticles and UPs (α-Al2O3+UPs), α-Al2O3-UPs stimulation resulted in higher upregulations of CD80, CD86, MHC class I, and MHC class II on DCs, indicating the higher ability of DC activation. Moreover, α-Al2O3-UPs elicited a more effective immune response in mice, demonstrated by higher IFN-γ secretion than α-Al2O3+UPs. Furthermore, α-Al2O3-UPs also exhibited a more potent effect on tumor growth inhibition and survival prolongation in 4T1 tumor-bearing mice. Notably, when in combination with low dose chemotherapy, the anti-tumor effect was further enhanced, rather than using α-Al2O3-UPs alone. Conclusion This study presents an adjuvant built-in nanovaccine generated by a new simple method that can be potentially applied to cancer immunotherapy and lays the experimental foundation for future clinical application.

Decorating Delivery Vehicles Using Hyaluronic Acid Oligosaccharides Enables Active Targeting Toward Cancer and Minimizes Adverse Effect of Chemotherapeutics

Weibin Jia,Runrun Li,Fengjuan Zou,Min Li,Hongjuan Weng,Qianqian Shen,Guozhen Qi,Ruipiao Zhou,Yikang Shi,Guofeng Gu,Fengshan Wang,Zonggang Chen

The major drawback of conventional chemotherapeutic treatment is the non-specificity or inability to ascertain and target cancerous cells directly. In this study, an active targeting strategy that is poised to carry the anticancer agents to the desired sites for therapeutic action while avoiding toxicity to normal organs is provided. The active targeting of delivery vehicles is achieved by ligand-receptor interactions, in particular the specific binding between hyaluronic acid oligosaccharides (oHAs) and CD44 receptors. This study first prepares oHAs by the size-exclusion chromatography and utilizes them to decorate chitosan (CTS) as basic materials (oHAs-CTS) for drug delivery, then fabricates oHAs-CTS into micro/nanoscale carriers to encapsulate agents for cancer chemotherapy. The oHAs-CTS micro/nanocarriers exhibit high drug encapsulation efficiency (58–87%), and the drug releases present a sustained behavior. Notably, oHAs-CTS delivery vehicles display an enhanced active targeting toward cancers and alleviate the cytotoxic effects on normal cells. Additionally, in vivo results show that drug-laden oHAs-CTS nanocarriers demonstrate a significant inhibitory effect on 4 T1 tumors without any toxicity to the major organs. Taken together, the findings highlight the potential of oHAs-CTS micro/nanospheres as delivery vehicles with enhanced active targeted capability toward cancers and minimized adverse effects of chemotherapeutic agents for cancer treatment.
