Wolfberry, a medicinal and edible plant rich in nicotinamide adenine dinucleotide (NADH), is noted for its antiaging and antioxidant properties. However, the trace levels and instability of NADH present significant detection challenges. To overcome the inherent limitations of traditional electrochemical linear peak fitting, three machine learning algorithms were introduced: random forest, support vector machine regression (SVR), and long short-term memory neural networks. This pioneering approach established a complex nonlinear relationship model between concentration and response current, effectively mitigating high-concentration linear distortion and interference constraints within the detection system. The Shapley additive prediction method (SHAP) was ultimately employed to quantify the contribution of each electrode potential to the prediction of response currents. This study developed an electrochemical biosensor based on a NbC@NC-ZIF-8/ADH heterojunction substrate, combined with a machine learning–based nonlinear modeling approach, achieving precise quantitative detection of NADH concentration in wolfberry. It exhibits a broad detection range from 1 μM to 1.5 mM, with a detection limit as low as 0.075 μM. The linear prediction model achieved R 2 = 0.976. After constructing a nonlinear model using the SVR machine learning algorithm, the NADH concentration detection accuracy improved to R 2 = 0.996, significantly enhancing detection precision. SHAP analysis indicated that the 0.264 V potential contributed most significantly to the model output. This study established a trace detection research framework encompassing “heterojunction material preparation–density functional theory analysis–nonlinear model construction–SHAP interpretability analysis,” providing a reliable solution for the rapid and accurate detection of NADH in wolfberry. It demonstrates broad application prospects in the field of bioanalysis.

Starch is an important raw material in the food industry, and the ratios of amylose to amylopectin within it is closely related to the quality of starch products. Therefore, the development of rapid detection methods for the amylose/amylopectin ratios in starch is a practical demand to meet industrial development. Consequently, the development of a rapid, sensitive, and portable detection method is of great significance. In this study, an electrochemical detection method for the amylose-to-amylopectin ratios was established by utilizing alcohol dehydrogenase (ADH) and pullulanase to fabricate an ADH@pullulanase-AuNPs biosensor. Specifically, protein-metal nanoflowers were synthesized by regulating the interactions of Cu 2+ and PO 4 3− with the enzymes to achieve the spatially localized immobilization of ADH and pullulanase; meanwhile, AuNPs were modified to enhance electrical conductivity. Additionally, the applicability of this method to rice products was investigated. The experimental results showed that the ADH@pullulanase cluster exhibited a porous flower-like structure, good enzymatic activity, and excellent electrochemical performance. Under optimal conditions, this method exhibited a wide linear range (1 × 10 −6 –1 × 10 −4 mol/L) for the amylose/amylopectin ratios, a low limit of detection (0.5 × 10 −6 mol/L), and a short detection time (< 10 min). When applied to the detection of rice products, the average relative deviation between it and the iodine colorimetric method was 6.9%. The successful development of this method provides a technical reference and theoretical basis for the on-site rapid and sensitive detection of the amylose/amylopectin ratios.

This work reports a novel signal amplification strategy based on three-dimensional ordered macroporous C 60 -poly(3,4-ethylenedioxythiophene)-1-butyl-3-methylimidazolium hexafluorophosphate (3DOM C 60 -PEDOT-[BMIm][BF 6 ]) for ultrasensitive detection of thrombin by cascade catalysis of [email protected] 2 microspheres and alcohol dehydrogenase (ADH). [email protected] 2 microspheres were constructed not only as nanocarriers to anchor the large amounts of secondary thrombin aptamers but also as nanocatalysts to catalyze the oxidation of ethanol efficiently. Significantly, the electrochemical signal was greatly enhanced based on cascade catalysis: First, ADH catalyzed the oxidation of ethanol to acetaldehyde with the concomitant generation of NADH in the presence of β-nicotinamide adenine dinucleotide hydrate (NAD + ). Then, gold nanoparticles (AuNPs) as nanocatalysts could effectively catalyze NADH to produce NAD + with the help of PEDOT as redox probe. Under the optimal conditions, the proposed aptasensor exhibits a linear range of 2 × 10 −13 to 2 × 10 −8 M with a low detection limit of 2 × 10 −14 M for thrombin detection and shows high sensitivity and good specificity.

Alcoholic liver disease (ALD) is a worldwide disease caused by excessive ethanol consumption. In current study, hepatoprotective peptides were isolated from the porcine liver while their activities and interactions with ethanol metabolizing enzymes (alcohol dehydrogenase: ADH/acetaldehyde dehydrogenase: ALDH) were analyzed. The alcalase digested porcine liver hydrolysate (PLH) was confirmed to possess ADH/ALDH activation ability in a dose-dependent manner. The peptide sequences of PLH were identified by mass spectrometry. Molecular docking and in silico analysis were federatively used for screening. NTLPHPTAP had the lowest binding energy with ADH (−0.88 kcal/mol) and low toxicity. Fluorescence quenching data showed a dynamic fluorescence quenching constant (Kq) for NTLPHPTAP interacting with ADH/ALDH (ADH: 1.57 × 10 −10 M −1 s −1 ; ALDH:1.961 × 10 −10 M −1 s −1 ). Circular dichroism (CD) spectra and infrared spectrometer (FTIR) revealed that the enzyme secondary structures became flexible and disordered after interaction with NTLPHPTAP. Meanwhile, the ADH/ALDH was speculated to bind NTLPHPTAP via amido and carbonyl groups, respectively. These results indicated that the formation of ADH/ALDH-NTLPHPTAP complexes expanded the overall structure of the original enzymes and further increased the possibility of enzyme-substrate binding. In conclusion, the study proposed a method for the extraction of porcine liver hepatoprotective peptides and suggested further explanation for the interaction between NTLPHPTAP and ADH/ALDH in vitro .

Neutral polysaccharides such as konjac glucomannan, starch and pullulan are abundant in nature and have unique property. Their nanofibers hold great potential for biomedicine, which however, are seldom applied in the field due to the lack of crosslinking method. In this work, we report a periodate oxidation - adipic acid dihydrazide (ADH) crosslinking strategy to prepare robust and biocompatible neutral polysaccharide nanofibers. Neutral polysaccharides with adjacent dihydroxyl groups are firstly partially oxidized with periodate to give dialdehyde polysaccharides, and their electrospun nanofibers are then crosslinked with ADH to form dihydrazone crosslinkers. The resulting crosslinked neutral polysaccharide nanofibers exhibit high water resistance and excellent mechanical properties because of the high reactivity of Schiff base crosslinking reaction. Moreover, the crosslinked neutral polysaccharide nanofibers show good biocompatibility due to the low toxicity of ADH. These robust and biocompatible neutral polysaccharide nanofibers are expected to seek extensive applications in a variety of biomedical fields.

Chiral amines are valuable intermediates widely applied in the pharmaceutical industry. As an alternative to classical synthetic methods for the production of chiral amines, biocatalytic cascade synthesis for these molecules from their alcohol precursors has been developed. However, currently available enzyme cascade systems remain unstable and unsustainable in the practice of chiral amine synthesis. Herein, the catalytic cascade system of chimeric amine dehydrogenase (AmDH) and alcohol dehydrogenase (ADH) fused with silica binding peptide (SiBP) was site-specifically co-immobilized on silica nanoparticles (SNPs). SiBP enabled the fusion enzymes to firmly bind to SNPs, and both the single immobilized enzymes showed higher catalytic efficiencies (kcat/Km) than their free counterparts to different degrees. Using 10 mM (S)-2-hexanol as the substrate, the co-immobilized enzyme system (SiBP–ADH&SiBP–AmDH@SNPs) showed 90% (R)-2-aminohexane yield after 48 h reaction, which was 1.85-fold higher than the free enzyme system due to the enhanced catalytic efficiency of the immobilized enzymes and the possible proximity effect of the SiBP–ADH&SiBP–AmDH@SNPs. The catalyst productivity of the SiBP–ADH&SiBP–AmDH@SNPs was 11 to 34 times higher than literature data, demonstrating the high economic benefits and catalytic efficiency of this system in producing chiral amines. SiBP–ADH&SiBP–AmDH@SNPs retained 87% of its original activity after eight cycles of reuse. These results highlighted the potential of the co-immobilization system for the effective and sustainable synthesis of chiral amines in industrial practice.

Injectable hydrogels, of which the cover area and volume can be flexibly adjusted according to the shape and depth of the wound, are considered to be an ideal material for wound dressing. Konjac glucomannan (KGM) is a natural polysaccharide with immunomodulatory capability, while γ-polyglutamic acid (γ-PGA) is a single chain polyamino acid with moisturizing, water-retention and antibacterial properties. This work intended to combine the advantages of the two materials to prepare an injectable hydrogel (P-OK) by mixing the adipic acid dihydrazide (ADH) modified γ-PGA with oxidized KGM. The chemical structures, the physical and chemical properties, and the biological properties of the P-OK hydrogel were evaluated. The optimal conditions to form the P-OK hydrogel were fixed, and the cytotoxicity, qPCR, antibacterial and animal experiments were performed. Results showed that the P-OK hydrogel had a fast gelation time, good water-retention rate, little cytotoxicity, good immunomodulating and antibacterial capabilities, and could shorten the healing period in the rat full-thickness defect model, which makes it a potential candidate for wound repair dressing.

Pectin-based drug delivery systems hold great potential for oral insulin delivery, since they possess excellent gelling property, good mucoadhesion and high stability in the gastrointestinal (GI) tract. However, lack of enterocyte targeting ability and premature drug release in the upper GI tract of the susceptible ionic-crosslinked pectin matrices are two major problems to be solved. To address these issues, we developed folic acid (FA)-modified pectin nanoparticles (INS/DFAN) as insulin delivery vehicles by a dual-crosslinking method using calcium ions and adipic dihydrazide (ADH) as crosslinkers. In vitro studies indicated insulin release behaviors of INS/DFAN depended on COOH/ADH molar ratio in the dual-crosslinking process. INS/DFAN effectively prevented premature insulin release in simulated GI fluids compared to ionic-crosslinked nanoparticles (INS/FAN). At an optimized COOH/ADH molar ratio, INS/DFAN with FA graft ratio of 18.2% exhibited a relatively small particle size, high encapsulation efficiency and excellent stability. Cellular uptake of INS/DFAN was FA graft ratio dependent when it was at/below 18.2%. Uptake mechanism and intestinal distribution studies demonstrated the enhanced insulin transepithelial transport by INS/DFAN via FA carrier-mediated transport pathway. In vivo studies revealed that orally-administered INS/DFAN produced a significant reduction in blood glucose levels and further improved insulin bioavailability in type I diabetic rats compared to INS/FAN. Taken together, the combination of dual crosslinking and FA modification is an effective strategy to develop pectin nano-vehicles for enhanced oral insulin delivery.
