A new method was developed for simultaneous determination of levels of AMP, ADP and ATP in blood by anion-exchange chromatography. The three adenosine phosphates were separated by Dionex IonPac AG18 (50 mm × 4 mm) guard column and IonPac AS18 (250 mm × 4 mm) analytical column using a gradient method and detected with a suppressed conductivity detector. The detection limits (S/N = 3) of AMP, ADP and ATP were 38, 47, 108 ng/L, respectively. The relative standard deviations of retention time, peak area and peak height were all less than 1.87% and a good linear relationship was obtained. This method was applied to analyze human blood samples.

ADP-P@Mn and ADP-P@Ni composites were prepared by the interfacial assembly of ammonium dihydrogen phosphate (ADP) with transition metal–phytic acid (PA) complexes. These composites were then mixed with cellulose fibers in an aqueous solution to produce flame-retardant paper. Morphological analysis revealed that the original ADP particles exhibited a smooth, cubic morphology; however, after functionalization with phytic acid, the particles became rougher. The presence of P–O–Mn and P–O–Ni bonds confirmed the bridging effect between phytic acid and ADP, which led to reduced crystallinity and promoted synergistic flame-retardant effects during combustion. Within all concentration ranges tested, both ADP-P@Mn and ADP-P@Ni exhibited significantly improved flame retardancy compared with pristine ADP and reduced the flame spread rate by up to 50%, showing remarkably enhanced flame-retardant efficiency. Notably, even at low concentrations, these two novel flame retardants endowed cellulose paper with self-extinguishing properties. Additionally, increased carbon and oxygen contents in residual char, along with cross-linking via P–C, P–O, and P═O bonds, indicated the formation of a dense carbonaceous layer. Consequently, ADP-P@Mn and ADP-P@Ni significantly enhanced the flame resistance of cellulose paper while maintaining its excellent writing performance, moisture resistance, and mechanical properties.

In this study, a series of dopant-free, low-cost hole-transporting materials (HTMs) based on triphenylamine-functionalized azadipyrromethene dyes 1–3 (TPA-ADPs 1–3) were designed and synthesized. The properties of these new HTMs were investigated by optical spectroscopy, cyclic voltammetry, thermogravimetric analysis, differential scanning calorimetric, atomic force microscopy, and X-ray diffraction, as well as theoretical calculations. The results indicated that the TPA-ADPs 1–3 presented well-matched energy levels with perovskite, higher hole mobility, as well as more effective defect passivation at the perovskite/HTM interface by the coordination interaction between the ADP moiety and the undercoordinated Pb2+. The n-i-p perovskite solar cells (PSCs) employing HTMs 1–3 as well as doped Spiro-OMeTAD were fabricated and characterized. The TPA-ADP 1-based PSCs exhibited the best performance with a champion power conversion efficiency (PCE) of 22.13% and an fill factor of 0.81, which was superior to that of the devices based on the doped Spiro-OMeTAD. Long-term device performance studies indicated that the TPA-ADP 1-based PSCs maintained 80% of the initial PCE after 1800 h of storage in the ambient condition of 40–60% RH, which was also higher than the stability of doped Spiro-OMeTAD-based devices under the same conditions.

Polyamide 6 (PA6) is inherently flammable and prone to severe melt-dripping, which severely restricts its application in aerospace, rail transit, and public safety sectors. To address this, we developed a complementary flame-retardant system by incorporating electrochemically exfoliated graphene (GR), melamine polyphosphate (MPP), and aluminum diethylphosphinate (ADP) into PA6 via in situ polymerization. The thermal stability and interactions among the three components were thoroughly investigated. The results indicate that the optimized formulations exhibit significant flame-retardant enhancement. Specifically, the GR/MPP/ADP/PA6-012 and GR/MPP/ADP/PA6-75 composites achieved limiting oxygen index (LOI) values of 35% and 29%, respectively, and both passed the vertical burning test and achieved V-0 rating, without any dripping. In cone calorimetry, the GR/MPP/ADP/PA6-57 composite demonstrated the most effective heat-release suppression, with the peak heat release rate (pHRR) and total heat release (THR) significantly reduced to 76.3 kW m −2 and 37.7 MJ m −2 , respectively. Mechanistic analysis reveals that GR forms a continuous, compact physical barrier during combustion, effectively hindering heat transfer. MPP promotes the formation of an expanded char layer, enhancing condensed-phase protection. Meanwhile, ADP releases PO · radicals in the gas phase to quench flame-active species and simultaneously promotes char formation. The three components work complementarily, establishing a multi-level “barrier–charring–quenching” flame-retardant network. This study proposes an efficient and environmentally friendly flame-retardant strategy for PA6. By optimizing the ratio of each flame retardant, the resulting composites exhibit superior flame-retardant performance, offering new insights into the flame-retardant modification of engineering plastics and the design of multi-component systems. Highlights GR/MPP/ADP flame-retardant system for PA6 was developed via electrochemistry and in situ polymerization. Electrochemically exfoliated GR forms a continuous two-dimensional physical barrier in PA6 with excellent dispersibility. The ternary system achieves an LOI of 35% and vertical burning test V-0 rating without any dripping. Optimized composite reduced pHRR by 85.3% and THR by 28.5% in cone calorimetry. Synergistic GR/MPP/ADP system builds a barrier–charring–quenching flame-retardant network.

Pathological cardiac hypertrophy and heart failure remain mechanistically incompletely defined despite their clinical significance in cardiovascular disease. While ADP-ribosyltransferase cholera toxin-like (ARTC) enzymes modulate membrane protein function and downstream signaling via post-translational modifications, their role in cardiac pathology remains unexplored. Herein, we investigated the regulatory involvement of ADP-ribosyltransferase 3 (ART3) in myocardial remodeling. Cardiomyocyte-specific ART3 knockdown and overexpression murine models were established using adeno-associated virus serotype 9 (AAV9), with RNA-sequencing employed to profile ART3-dependent transcriptional responses in pathological cardiac hypertrophy. Functional validation was performed in complementary in vitro and in vivo hypertrophy models, complemented by immunoprecipitation-coupled mass spectrometry to identify ART3 substrates. ART3 was found to be cardiomyocyte-enriched and transcriptionally downregulated during progressive myocardial remodeling and heart failure. Cardiomyocyte-specific ART3 knockdown 3 accelerated the decompensatory transition of cardiac hypertrophy, whereas ART3 overexpression exerted significant anti-hypertrophic and anti-remodeling effects. Mechanistically, ART3 preserved cardiomyocyte mono-ADP-ribosyltransferase activity, catalyzing the site-specific mono-ADP-ribosylation (mADPr) of Integrin Subunit Alpha 7 (ITGA7) at residue R129. This post-translational modification was requisite for integrin signaling pathway activation, which mediated the observed cardioprotective effects. Collectively, these findings establish ART3 as a novel regulator of pathological cardiac hypertrophy by modulating ITGA7 via mADPr, highlighting its therapeutic potential as a target for heart failure intervention.

Fresh shiitake ( Lentinus edodes ) is prone to brown, pileus-opening and flavor-loss during storage. Therefore, it is important to find an effective preservation method for fresh shiitake. Negative air ions (NAI) are negatively-charged molecules or atoms in the air, and can affect the physiological metabolism of live cells and be conveniently used with low cost. In this study, NAI treatment was performed at different times and the physico-chemical characteristics, microstructure, membrane potential and energy metabolism of shiitake were determined during storage. Results showed that NAI treatment for 40 min could reduce 29% of browning index and maintain the hardness of shiitake. NAI treatment groups had higher content of sweetness amino acids, umami amino acids, 5′-IMP, eight-carbon alcohols compounds and cyclic sulfides compounds than the control, and comprehensive quality of the group being treated for 40 min was the best. The mitochondria of shiitake swelled and the membrane potential decreased after being treated by NAI. However, NAI treatment for 40 min could improve the contents of ATP and ADP, maintain a relatively stable energy charge level, and promote energy utilization of shiitake during storage. The results demonstrated that NAI treatment had the potential to improve the quality shiitake during storage.

The recent use of PARP inhibitors (PARPi) in the maintenance treatment of ovarian tumor has significantly improved the survival rates of cancer patients. However, the current oral administration of PARP inhibitors fails to realize optimal therapeutic effects due to the low bioavailability in cancerous tissues, and often leads to a range of systemic adverse effects including hematologic toxicities, digestive system reactions, and neurotoxicities. Therefore, the demand for an advanced drug delivery system that can ensure effective drug administration while minimizing these unfavorable reactions is pressing. Injectable hydrogel emerges as a promising solution for local administration with the capability of sustainable drug release. In this study, we developed an injectable hydrogel made from aminated hyaluronic acid and aldehyde-functionalized pluronic127 via Schiff base reaction. This hydrogel exhibits excellent injectability with short gelation time and remarkable self-healing ability, and is applied to load niraparib. The drug-loaded hydrogel (HP@Nir hydrogel) releases drugs sustainably as tested in vitro as well as displays significant anti-proliferation and anti-migratory properties on human epithelial ovarian cancer cell line. Notably, HP@Nir hydrogel effectively suppresses the growth of ovarian cancer, without significant adverse reactions as demonstrated in animal studies. Additionally, the developed hydrogel is gradually degraded in vivo for around 20 d, while maintaining good biocompatibility. Overall, the injectable hydrogel loaded with niraparib provides a secure and efficient strategy for the treatment and management of ovarian cancer.

ADP-ribosylation factor 1 (HcARF1) is one of theHaemonchus contortus(H. contortus) excretory/secretory proteins involved in modulating the immune response of goat peripheral blood mononuclear cells (PBMC). Here, we evaluated the immunogenic potential of recombinant HcARF1 (rHcARF1) againstH. contortusinfection in Institute of Cancer Research (ICR) mice. Briefly, rHcARF1 was entrapped in poly (D, L-lactide-co-glycolide) (PLGA) and chitosan (CS) nanoparticles (NP) and injected into mice as a vaccine. Fifty-six ICR mice were assigned randomly into seven groups, with eight animals in each group, and they were vaccinated subcutaneously. At the end of the experiment (14th day), the blood and the spleen were collected from euthanized mice to detect lymphocyte proliferation, cytokine analysis, and the production of antigen-specific antibodies. Scanning electron microscope was used to determine the size, morphology, and zeta potential of nanoparticles. Flow cytometry was performed, which presented the increase percentages of CD4+T cells (CD3e+CD4+), CD8+T cells (CD3e+CD8+) and dendritic cells (CD11c+CD83+, CD11c+CD86+) in mice vaccinated with rHcARF1+PLGA NP. Immunoassay analysis show raised humoral (Immunoglobulin (Ig)G1, IgG2a, IgM) and cell-mediated immune response (Interleukin (IL)-4, IL-12, and IL-17, and Interferon (IFN)-γ) induced by rHcARF1+PLGA NP. Experimental groups that were treated with the antigen-loaded NP yield higher lymphocyte proliferation than the control groups. Based on these results, we could propose that the rHcARF1 encapsulated in NP could stimulate a strong immune response in mice rather than administering alone against the infection ofH. contortus. Keywords:Haemonchus contortus;rHcARF1;PLGA;CS;nanoparticle;ICR mice

Adenosine-5′-triphosphate (ATP) is the energy equivalent of the living system. Polyphosphate (polyP) is the ancient energy storage equivalent of organisms. Polyphosphate kinases (PPKs) catalyze the polyP formation or ATP formation, to store energy or to regenerate ATP, respectively. However, most PPKs are active only in the presence of long polyPs, which are more difficult and more expensive to generate than the short polyPs. We investigated the PPK preference towards polyPs by site-directed mutagenesis and computational simulation, to understand the mechanism and further design enzymes for effective ATP regeneration using short polyPs for in vitro cascade reactions, which are highly desired for research and applications. The results suggest that the short polyPs inhibit PPK by blocking the ADP-binding pocket. Structural comparison between PPK ( Corynebacterium glutamicum ) and PPK ( Sinorhizobium meliloti ) indicates that three amino acid residues, i.e., lysine, glutamate, and threonine, are involved in the activity towards short polyP by fixing the adenosine group of ADP in between the subunits of the dimer, while the terminal phosphate group of ADP still offers an active site, which presents a binding pocket for ADP. A proposed triple mutant PPK (SMc02148-KET) demonstrates significant activity towards short polyP to form ATP from ADP. The obtained high glutathione titer (38.79 mM) and glucose-6-phosphate titer (87.35 mM) in cascade reactions with ATP regeneration using the triple mutant PPK (SMc02148-KET) reveal that the tailored PPK establishes the effective ATP regeneration system for ATP-dependent reactions.

Background Catalpol (CAT), a naturally occurring iridoid glycoside sourced from the root of Rehmannia glutinosa , affects mitochondrial metabolic functions. However, the mechanism of action of CAT against pyrexia and its plausible targets remain to be fully elucidated. Purpose This study aimed to identify the specific targets of CAT for blocking mitochondrial thermogenesis and to unveil the unique biological mechanism of action of the orthogonal binding mode between the hemiacetal group and lysine residue on the target protein in vivo . Methods Lipopolysaccharide (LPS)/ carbonyl cyanide 3-chlorophenylhydrazone (CCCP)-induced fever models were established to evaluate the potential antipyretic effects of CAT. An alkenyl-modified CAT probe was designed to identify and capture potential targets. Binding capacity was tested using in-gel imaging and a cellular thermal shift assay. The underlying antipyretic mechanisms were explored using biochemical and molecular biological methods. Catalpolaglycone (CA) was coupled with protein profile identification and molecular docking analysis to evaluate and identify its binding mode to UCP2. Results After deglycation of CAT in vivo , the hemiacetal group in CA covalently binds to Lys239 of UCP2 in the mitochondria of the liver via an ɛ-amine nucleophilic addition. This irreversible binding affects proton leakage and improves mitochondrial membrane potential and ADP/ATP transformation efficiency, leading to an antipyretic effect. Conclusion Our findings highlight the potential role of CA in modulating UCP2 activity or function within the mitochondria and open new avenues for investigating the therapeutic effects of CA on mitochondrial homeostasis.

The quest for ever-growing energy density of lithium-ion batteries dictates urgent concerns about their thermal safety. Phase change materials (PCMs) are considered promising for battery thermal management, yet their extensive application is restricted by inherent limitations such as leakage risk, low thermal conductivity, and flammability. Here, we fabricate a multilayered PCL@PEG/Al₂O₃-ADP (CEAD) composite film through strategy of coaxial electrospinning and dip-coating. Benefiting from the effective PEG encapsulation, thermally conductive Al₂O₃ fillers, and an ADP coating for flame retardancy, the composite film demonstrates excellent shape stability with no leakage even after 12 h of continuous exposure at 50 °C, high phase change enthalpy, improved thermal conductivity, and self-extinguishing behavior. Its notable foldability also ensures facile installation. When wrapped around a lithium-ion battery, the film maintains the battery temperature below 45 °C under a 5C discharge rate, which is 13 °C lower compared to the bare one. Thus, the composite film provides simultaneous efficient thermal management and flame retardancy, meeting critical safety requirements for high energy density lithium-ion batteries. This work offers a viable pathway for developing multifunctional thermal management materials suitable for advanced battery systems and other electronics.

Thrombosis is one of the biggest obstacles in the clinical application of small-diameter tissue-engineered blood vessels (TEBVs). The implantation of an unmodified TEBV will lead to platelet aggregation and further activation of the coagulation cascade, in which the high concentration of adenosine diphosphate (ADP) that is released by platelets plays an important role. Inspired by the phenomenon that endothelial cells continuously generate endogenous antiplatelet substances via enzymatic reactions, we designed a reduced graphene oxide (RGO) based dual-enzyme biomimetic cascade to successively convert ADP into adenosine monophosphate (AMP) and AMP into adenosine. We used RGO as a support and bound apyrase and 5′-nucleotidase (5′-NT) on the surface of RGO through covalent bonds, and then, we modified the surface of the collagen-coated decellularized vascular matrix with the RGO-enzyme complexes, in which RGO functions as a platform with a large open surface area and minimal diffusion barriers for substrates/products to integrate two catalytic systems for cascading reactions. The experimental results demonstrate that the two enzymes can synergistically catalyze procoagulant ADP into anticoagulant AMP and adenosine successively under physiological conditions, thus reducing the concentration of ADP. AMP and adenosine can weaken or even reverse the platelet aggregation induced by ADP, thereby inhibiting thrombosis. Adenosine can also accelerate the endothelialization of TEBVs by regulating cellular energy metabolism and optimizing the microenvironment, thus ensuring the antithrombotic function and patency of TEBVs even after the RGO-enzyme complex loses its activity.
