Musa spp. (banana) is consumed globally as a healthy fruit and improves the immune system. Banana blossoms are a by-product of banana harvesting rich in active substances such as polysaccharides and phenolic compounds; however, these blossoms are typically discarded as waste. In this report, a polysaccharide, MSBP11, was extracted, purified and identified from banana blossoms. MSBP11 is a neutral homogeneous polysaccharide with a molecular mass of ∼214.43 kDa and composed of arabinose and galactose at a ratio of 0.303:0.697. MSBP11 exhibited potent antioxidant and anti-glycation activities in a dose-dependent manner and can be used as a potential natural antioxidant and inhibitor of advanced glycosylation end products (AGEs). In addition, banana blossoms have been shown to decrease the levels of AGEs in chocolate brownies, which might possibly be developed as functional foods for diabetic patients. This study provides a scientific basis to further research the potential application of banana blossoms in functional foods.

Advanced glycation end-products (AGEs) play a crucial role in the pathogenesis of diabetic osteoarthritis (DOA), contributing to cartilage degradation and impaired joint lubrication, which complicate clinical management. However, no therapeutics specifically targeting AGEs are developed for DOA treatment. Here, a composite hydrogel (PTC-MP) incorporated with polydopamine-coated tannic cerium (PTC) nanozymes and magnesium ions (Mg 2+ ), implementing a strategy of “restrain-restore-reinforce (3R)” for AGEs-directed DOA therapy is reported. PTC-MP hydrogel effectively restrains the formation of AGEs by scavenging free radicals, chelating ferrous ions, and competitive hydrophobic site binding. By leveraging mild photothermal therapy (mPTT), PTC-MP hydrogel restores cartilage homeostasis by reducing AGEs-induced reactive oxygen species (ROS) overproduction and mitochondrial dysfunction. Furthermore, Mg 2+ in PTC-MP hydrogel reinforces joint repair by stimulating endogenous hyaluronic acid (HA) secretion to improve lubrication. In a rat DOA model, PTC-MP hydrogel with mPTT significantly attenuated DOA progression, reduced osteophytes formation and synovial inflammation, and improved motor function. Therefore, the “3R” strategy targeting AGEs provides a promising therapeutic approach for recalcitrant DOA.

Long-term hyperglycemia is a typical symptom of diabetes mellitus (DM) which can cause a high level of protein glycation and lead to the formation of advanced glycation end products (AGEs). The accumulation of AGEs in turn deteriorates DM and its complications. Insulin, the only hormone that directly decreases blood sugar in vivo, is vulnerable to glycation which causes the loss of its biological activity. In this study, we used a porcine insulin (PI)–methylglyoxal (MGO) model to investigate the inhibitory effect of leonurine (LN), a natural alkaloid extracted from Herba leonuri, on AGE formation. Assays including AGE-specific fluorescence, and fructosamine level and carbonyl group content determination showed that LN can dose-dependently suppress PI glycation. A significantly decreased cross-linking level on the glycated PI was also proven by SDS-PAGE electrophoresis. A further liquid chromatography-mass spectrometry study suggested that LN may inhibit PI glycation through trapping MGO and keeping it from reacting with PI. Our results thus indicate that LN is a promising anti-glycation agent for the prevention of diabetes and its complications via inhibiting AGE formation.

Advanced glycosylation end products (AGEs) are becoming a prominent issue in the biomedical and food safety industries because of their strong relationship to human health. Here, a MIL@PANI@LDH photocatalyst was prepared by in-situ growth method, realizing thorough elimination of AGEs for the first time. The synthesized photocatalysts used MIL-101 holding remarkably specific surface area as the core and PANI with a large number of amino and imine groups as the linker to ensure excellent adsorption capacity. The lamellar LDH structure grown in the outermost layer, on the other hand, introduces a narrower band gap and stronger light absorption ability, capable of activating sodium persulfate (PS) and generating free radicals with powerfully oxidizing ability. They work in concert to achieve the effective degradation. Through the optimization of the reaction conditions, the best photochemical performance could be achieved with a Zn/Al molar ratio of 3:1, and the degradation rates of AGEs were greater than 99.8 % (within 150 min). Additionally, multiple radical synergistic and non-radical action pathways involved in the system were revealed. The precision-engineered catalysts also exhibited high stability, reusability, and desirable biosafety, offering an effective and sustainable solution to the problem of purifying hazardous substances in real application.

Background Cardiovascular metabolic diseases (CMDs) are a major contributor to global mortality and disability, yet their pathogenesis remains incompletely understood, partly because existing in vitro models fail to capture disease complexity. Conventional engineered heart tissues (EHT), which typically contain only a limited set of cell types and lack neural components, cannot replicate the intricate neuro-cardiac interactions involved in CMDs. Objective This study aimed to develop a neuron-like-Integrated Engineered Heart Tissue for investigating neuro-cardiac interactions under both physiological and pathological conditions, offering a new tool for CMD research. Methods We constructed a Sympathetic-like-Integrated Engineered Heart Tissue (SIEHT) by incorporating sympathetic-like neuronal cells into EHT. The structural and functional properties of SIEHT were systematically compared with conventional EHT using morphological analysis, immunofluorescence staining, contractility measurements, qPCR, and RNA sequencing. The model was then exposed to advanced glycation end products (AGEs) to assess pathological remodeling through multiple parameters, including cell viability, oxidative stress, structural and functional integrity, and transcriptomic profiles. Results SIEHT exhibited greater structural and functional maturation than EHT, as indicated by improved cardiomyocyte alignment, increased contraction amplitude, and upregulated expression of connexin 43. Transcriptomic analysis revealed enriched pathways associated with multi-system development. Under AGEs-induced pathological conditions, SIEHT demonstrated a more pronounced reduction in cell viability, elevated reactive oxygen species levels, more severe contractile dysfunction, a higher frequency of abnormal spontaneous beating, and greater neural injury relative to controls. Transcriptome profiling further identified significant enrichment of the AGE-RAGE signaling pathway in diabetic complications. Conclusions We successfully established a novel SIEHT model that recapitulates physiological neuro-cardiac interactions and AGEs-induced adverse remodeling across multiple dimensions, providing a powerful and innovative tool for elucidating the pathophysiological mechanisms of neuro-cardiac dysregulation in CMDs.

The dietary advanced glycation end products (AGEs) contribute to the development of major chronic diseases. Maillard reactions are the main mechanism for AGEs formation but their formation involving ascorbic acid (AA) is far from being fully understood. This study investigated the effect of pH (6–10) and temperature (65, 100 and 120 ℃) on AGEs formation in three model systems: glucose (Glu) + lysine (Lys), AA + Lys and Glu + Lys + AA. In addition, the formation pathway of AGEs in Glu + Lys + AA model system was proposed by carbon module labeling (CAMOLA) technique. The results suggested alkaline environment can promote the production of N -carboxymethyllysine (CML) and N -carboxyethyllysine (CEL), but inhibit the production of pyrraline (Pyr). Meanwhile the high temperature favored AGEs formation. In the U- 13 C-Glu + Lys + AA model, AA produced glyoxal (GO), methylglyoxal (MGO), CML and CEL, which was significantly higher than with Glu alone. This study provides a theoretical basis for the formation mechanism of AGEs in the Maillard reaction involving AA.

Background and objectives: On account of the long-term inflammatory microenvironment, diabetic wounds are challenging to heal in which advanced glycation end products are considered important factors hindering the healing of diabetic wounds. Gum Arabic has demonstrated significant potential in the treatment of various diseases owing to its anti-inflammatory and antioxidant properties. Nonetheless, there is still insufficient research on the role of Arabic gum in facilitating diabetic wounds healing and its mechanisms. This study aims to investigate the pharmacological targets and therapeutic mechanisms of Arabic Gum on diabetic wound healing by adopting network pharmacology, molecular docking, and experimental validation. Methods: Key active components of Arabic Gum and disease targets were identified through network pharmacology and bioinformatics. GO/KEGG enrichment was performed to identify critical pathways. Cytoscape and AutoDock were used for targets prediction and molecular docking validation. In vitro, Transwell assay and tube formation assay were performed to evaluate the effect of Arabic Gum on human fibroblasts migration and human umbilical vein endothelial cells angiogenesis. Western blotting analyzed Pro-caspase-1, ASC, NLRP3 and NF-κB pathway-related proteins. In vivo, a full-thickness diabetic wound model was established. Histological changes were assessed via H&E and Masson’s staining, oxidative stress levels through DHE staining, inflammation levels with IL-1β, CD68 and CD206 staining, angiogenesis and cell proliferation levels were assessed by CD31 and Ki67 staining. The levels of pathway-related proteins were analyzed by NLRP3 and Phospho-NF-κB P65 staining. Results: Network pharmacology analysis identified key targets, encompassing HSP90AA1, STAT3, and PRKCB, involved in the AGEs-NF-κB-NLRP3 signaling axis. Molecular docking demonstrated strong binding affinity between AG components and these targets. In vitro, AG lessened AGEs-induced activation of the NLRP3 inflammasome via modulation of the NF-κB pathway and reinforced cell migration and angiogenesis. In vivo, AG-treated diabetic wounds exhibited accelerated healing, with augmented collagen deposition, lowered oxidative stress and inflammation, and strengthened cell migration and angiogenesis. AG promotes diabetic wound healing by modulating the AGEs-NF-κB-NLRP3 axis, exerting anti-inflammatory, antioxidant, pro-angiogenic, and cell-proliferative effects. Conclusion: This study provides new insights into diabetic wound repair and suggests that AG is a promising therapeutic agent for improving diabetic wound healing.

L-theanine, a non-protein amino acid naturally occurring in tea leaves, is recognized for its antioxidant, anti-inflammatory, and neuroprotective properties. Despite its known benefits, the mechanisms by which L-theanine influences lifespan extension remain poorly understood. This study investigated the effects of L-theanine on the lifespan ofCaenorhabditis elegansand explored the underlying mechanisms. Our findings indicate that L-theanine significantly diminishes the accumulation of advanced glycation end products (AGEs), which are biomarkers closely linked to aging and age-related diseases. Through an AGE-level analysis, we observed that L-theanine, when administered during early adulthood, notably extended the lifespan ofCaenorhabditis elegansunder both normal and high-glucose-induced stress conditions. L-theanine enhanced the lifespan under typical conditions and provided protective effects against high-glucose-induced stress. A further analysis demonstrated that L-theanine extends the lifespan ofCaenorhabditis elegansby modulating the DAF-2/DAF-16 insulin-like signaling pathway and reducing the accumulation of advanced glycation end products (AGEs). In summary, this study identified L-theanine as a potential anti-aging intervention that extends the lifespan by reducing AGE accumulation and regulating insulin-like signaling pathways. These findings provide new insights for developing anti-aging strategies and lay the groundwork for further research on the potential benefits of L-theanine in mammals. Future studies could explore the molecular mechanisms, test L-theanine in mammalian models, and assess the long-term side effects.

Background Punicalagin (PUN), the main bioactive in pomegranate peel polyphenols, plays a key therapeutic effect against diverse ailments, especially inflammatory diseases. Advanced Glycation End Products (AGEs) can disrupt lipid metabolism, trigger inflammation, and are closely linked to metabolic inflammatory diseases. Based on PUN's known protective effects, this study uses lipidomic analysis to examine its influence on AGEs-induced metabolic inflammation, providing a basis for anti-inflammatory drug research. Purpose To develop the medicinal value of PUN by studying its effects on AGEs-induced inflammation and lipid metabolism disorders. Methods RAW264.7 macrophages were first stimulated with AGEs (200 μg/mL), followed by treatment with PUN (25–100 μg/mL). Next, TNF-α and IL-1β levels were measured by ELISA. Western blotting was then performed to assess the expression of PPARγ and key proteins in the NF-κB and JNK signaling pathways, nuclear translocation of NF-κB p65 was visualized via immunofluorescence. By using the antagonist GW9662, the upstream and downstream relationship between PPARγ and NF-κB, as well as JNK, was demonstrated. Finally, non-targeted lipidomics using UPLC-Q/TOF-MS, combined with PCA and OPLS-DA multivariate analysis, identified differential lipid metabolites. Results PUN significantly suppressed AGEs-induced secretion of TNF-α and IL-1β in a concentration-dependent manner. Mechanistically, PUN upregulated PPARγ expression, inhibited the phosphorylation of IKKα/β, IκB-α, and NF-κB, reduced p-JNK, c-Jun, and c-Fos levels, and blocked nuclear translocation of p65. The use of the PPARγ antagonist GW9662 led to a re-upregulation of the phosphorylation levels of the NF-κB and JNK pathways downstream of PPARγ. The original inhibitory effect of PUN was blocked. This confirmed that the anti-inflammatory mechanism of PUN depends on the activation of PPARγ. Lipidomics analysis revealed that PUN reversed 31 dysregulated lipid metabolites induced by AGEs, with glycerophospholipid metabolism identified as the most significantly enriched pathway. Conclusion This study demonstrates that PUN alleviates AGEs-induced inflammation by restoring glycerophospholipid metabolism homeostasis, upregulating PPARγ expression, and subsequently suppressing NF-κB and JNK signaling pathways.

A novel and promising reheating method has been explored to address the issue of meat consumption and safety quality degradation after reheating. This study compared the effects of boiling, steaming, microwaving, stir-frying, and microwave synergistic steam (MW + ST) reheating on the flavor and advanced glycation end products (AGEs) of braised pork. The results demonstrated that MW + ST was effective in reducing AGEs formation with an Nε-carboxymethyllysine (CML) content of 4.21 mg/kg and a fluorescent AGEs level of 208.3 AU, which only increased 17.13% and 1.98%, respectively, compared with the content before reheating. This can be attributed to the high moisture content (49%) and immobilized water proportion (92.65%) maintained by MW + ST, resulting in the reduced accumulation and production of AGEs precursors (Schiff base and glyoxal) by suppressing lipid oxidation, protein oxidation, and the Maillard reaction. Additionally, the electronic nose and electronic tongue results showed that MW + ST was closest to the original odor of braised pork and exhibited excellent taste characteristics. Overall, the high sensory scores and safety qualities of the MW+ST suggested that it promises to be a new way of reheating for consumers and food distribution companies compared to conventional reheating methods.

Heavy-metal pollution has been established to affect ginseng quality. However, this effect is still unknown in ginseng of different ages, emphasizing the need to investigate the effects of heavy metals in soils on ginseng growth. Herein, we determined the content of heavy metals (Cu, Cd, Pb, Hg, and As) in ginseng of different ages (2 to 6-year-old) and the corresponding soil samples. Then, the total ginsenosides content of ginseng and rate-limiting enzyme (HMGR, SQE, CYP450) activity in the synthesis of ginsenosides were assessed. Results from 200 differently-aged Chinese ginseng showed that increased ginsenoside content in 3 to 5-year-old ginseng was paralleled by increased heavy metal element content in ginseng and its soil. The activity of rate-limiting enzymes increased in the first four years of ginseng growth and then exhibited a steady or downward trend. Further analysis suggested that heavy metal elements in soils could directly affect ginsenoside content. Moreover, we found that Cu significantly affected the rate-limiting enzyme CYP450 activity. Further principal component analysis and correlation analysis found that heavy metals could obviously inhibit ginseng growth during the 5th and 6th years. Heavy metal content in soils has huge prospects for predicting ginsenoside, Cu and As content in ginseng. This study provided support for ginseng cultivation, quality research and quality assessment.

Methylglyoxal (MGO) is a reactive carbonyl compound that forms advanced glycation end products (AGEs), which are associated with diseases such as diabetes, cancer, and Alzheimer's disease. Turmeric, which contains bioactive diarylheptanoids, has compounds like curcumin that can trap MGO and inhibit the formation of AGEs. However, diarylheptanoids suffer from poor stability and solubility, complicating their use in standard methods. Natural deep eutectic solvents (NADES), particularly the BG12–10 % system (betaine and glycerol mixed in a 1:2 ratio with 10 % water added), can improve these properties by enhancing solubility and stability, thus enabling more accurate reaction kinetics. The NADES extract of turmeric can also directly react with MGO, simplifying the experimental process. Liquid chromatography-mass spectrometry has identified 21 diarylheptanoids in turmeric, 10 of which can trap MGO, curcumin being the only one previously reported in the literature.
