Dramatic enhancement of mineralization capability for ozone/peroxymonosulfate process by single atom Co/CMK-3 catalyst

Gaozu Liao,Peng Xu,Ping Lu,Xiaojiao Qing,Zhanjiang Yu,Yu Chang,Weirui Chen,Yiming Tang,Jing Wang,Xi Wang,Xukai Li,Laisheng Li,Dehua Xia

Single Co atoms dispersed on CMK-3 was synthesized and employed as efficient catalyst to degrade APAP in O 3 /PMS process. The results of morphology and structure characterization indicated that the as prepared catalyst possessed uniform array of mesoporous channel structure with a long-range. Co atoms were single atomic dispersed in CMK-3 skeleton with the form of CoN 4 coordination. The BET surface area and average pore diameter of 3.6% SA Co/CMK-3 were measured as 956 m 2 /g and 3.6 nm, respectively. For APAP degradation by 3.6% SA Co/CMK-3 + PMS/O 3 process, TOC removal efficiency reached to 60% and 78.9% in initial 1 min and 15 min, respectively. The influence of initial pH was investigated and demonstrated that APAP could be efficiently mineralized in a wide pH range. In addition, high mineralization rates were also achieved for other refractory organics (nitrobenzene and sulfamethoxazole). Radical quenching experiments and ESR indicated that SO 4 •− , •OH and 1 O 2 were all responsible for APAP degradation. DFT simulations elucidated that single Co atoms and N-heteroatoms on 3.6% SA Co/CMK-3 served as dual reaction sites for PMS and O 3 decomposition, which enhanced the ROS generation and APAP mineralization.

The Self-Assembly Soluplus Nanomicelles of Nobiletin in Aqueous Medium Based on Solid Dispersion and Their Increased Hepatoprotective Effect on APAP-Induced Acute Liver Injury

Jinrong Ning,Guodong Zheng,Yi Cai,Yunguang Hu,Yiqi Liu,Enping Lai,Baizhong Chen,Yujie Liu,Ziqi Liang,Jijun Fu,Minyan Wei

Purpose APAP-induced liver injury (AILI) is a common cause of acute liver failure (ALF). Nobiletin (NOB) is a potential hepatoprotective agent for the treatment of APAP-induced liver injury. However, the poor solubility and low bioavailability of NOB hinders its application. In this study, a novel self-assembly nano-drug delivery system of nobiletin (solid dispersion of NOB, termed as NOB/SD) was developed based on solid dispersion technology to improve the bioavailability and hepatoprotective ability of NOB for APAP-induced liver injury therapy. Methods The optimized NOB/SD system was constructed using the amphiphilic copolymers of Soluplus and PVP/VA 64 via hot melt extrusion technology (HME). NOB/SD was characterized by solubility, physical interaction, drug release behavior, and stability. The bioavailability and hepatoprotective effects of NOB/SD were evaluated in vitro and in vivo. Results NOB/SD maintained NOB in matrix carriers in a stable amorphous state, and self-assembled NOB-loaded nanomicelles in water. Nanostructures based on solid dispersion technology exhibited enhanced solubility, improved release behavior, and promoted cellular uptake and anti-apoptosis in vitro. NOB/SD displayed significantly improved bioavailability in healthy Sprague Dawley (SD) rats in vivo. Furthermore, NOB/SD alleviated the APAP-induced liver injury by improving anti-oxidative stress with reactive oxygen species (ROS) scavenging and nuclear factor erythroid 2-related factor 2 (Nrf2) activation. Conclusion These results suggested that NOB/SD could be considered as a promising hepatoprotective nano-drug delivery system for attenuating APAP-induced acute liver injury with superior bioavailability and efficient hepatoprotection, which might provide an effective strategy for APAP-induced acute liver injury prevention and treatment.

A Biomimetic Nanoparticle Exerting Protection against Acute Liver Failure by Suppressing CYP2E1 Activity and Scavenging Excessive ROS

Qing Yao,Yingying Tang,Sheng Dai,Lihui Huang,Zewei Jiang,Shiming Zheng,Meng Sun,Yitianhe Xu,Ruijie Lu,Tuyue Sun,Huirong Huang,Xinyu Jiang,Xiaomin Yao,Guangyong Lin,Longfa Kou,Ruijie Chen

Acute liver failure (ALF) is a severe liver disease caused by many reasons. One of them is the overdosed acetaminophen (APAP), which is metabolized into N -acetyl-p-benzoquinone imine (NAPQI), an excessive toxic metabolite, by CYP2E1, resulting in excessive reactive oxygen species (ROS), exhausted glutathione (GSH), and thereafter hepatocyte necrosis. N -acetylcysteine is the Food and Drug Administration-approved drug for detoxification of APAP, but it has limited clinical application due to the short therapeutic time window and concentration-related adverse effects. In this study, a carrier-free and bilirubin dotted nanoparticle (B/BG@N) is developed, which is formed using bilirubin and 18 β -Glycyrrhetinic acid, and bovine serum albumin (BSA) is then adsorbed to mimic the in vivo behavior of the conjugated bilirubin for hitchhiking. The results demonstrate that B/BG@N can effectively reduce the production of NAPQI as well as exhibit antioxidant effects against intracellular oxidative stress via regulating the nuclear factor erythroid 2-related factor 2/heme oxygenase-1 signal axis and reducing the production of inflammatory factors. In vivo study shows that B/BG@N can effectively improve the clinical symptom of the mice model. This study suggests that B/BG@N own increases circulation half-life, improves accumulation in the liver, and dual detoxification, providing a promising strategy for clinical ALF treatment.
