Aptamer-based DNAzyme walker electrochemical biosensing strategy for Acinetobacter baumannii detection

Linhong Cao,Tianyu Wang,Jingling Xie,Yihua Wang,Yaxin Huang,Sijian Luo,Xiaoting Zhan,Hui Jiang,Liuxin Ran,Xing Jin,Jinbo Liu,Baolin Li

In this study, an innovative electrochemical biosensor was developed for the rapid, specific, and sensitive detection of Acinetobacter baumannii without the need for sample pretreatment. The biosensor utilized an aptamer as a specific capture probe for A. baumannii and employed a self-powered DNAzyme walker cleavage cycle reaction to achieve signal amplification. Upon introduction of the target bacteria, the aptamer captured the bacteria and released the Trigger, activating the DNAzyme to cleave the substrate chain containing methylene blue (MB). This led to the release of MB-labeled DNA fragments from the electrode surface, resulting in a significant decrease in the square wave voltammetry (SWV) signal of MB on the sensing platform. The limit of detection (LOD) for A. baumannii was determined to be 30 CFU/mL, enabling discrimination of the target bacteria from other common clinical isolates. Furthermore, the biosensor’s potential for real sample analysis was demonstrated in cerebrospinal fluid (CSF), showcasing its efficacy and versatility as a biosensing tool with wide-ranging applications in disease diagnosis and bioanalysis.

Biodegradation of typical pharmaceutical compounds by a novel strain Acinetobacter sp.

Shizong Wang,Yuming Hu,Jianlong Wang

A novel sulfamethoxazole (SMX)-degrading strain, Acinetobacter sp., was used to degrade other pharmaceutical compounds, including sulfadiazine (SD), sulfamethazine (SMT), trimethoprim (THM), triclosan (TCS), diclofenac (DFC) and carbamazepine (CBZ). The experimental results showed that Acinetobacter sp. can completely degrade SMX, SD and SMT, but with different mineralization efficiency. Acinetobacter sp. can mineralize 98.8% of SMX, while only 17.5% and 20.5% for SD and SMT, respectively. The intermediate products of SMX, SD and SMT degradation were tentatively identified. Based on the intermediates, it is inferred that the initial step for degrading sulfonamides by Acinetobacter sp. was the amidation of the amino groups in the benzene ring . The presence of methyl in the heterocyclic ring could induce the formation of methylase. By comparing the intermediates of SMX, SD and SMT degradation, it is concluded that Acinetobacter sp. preferred attacking the oxazole ring. However, Acinetobacter sp. cannot degrade THM, TCS, DFC and CBZ, while Acinetobacter sp. can still degrade SMX in the respective presence of THM, DFC and CBZ, although the degradation rate decreased. Moreover, the presence of TCS could completely inhibit the degradation of SMX by Acinetobacter sp.

Hybrid Cu2O/TiO2 Nanocomposites with Enhanced Photocatalytic Antibacterial Activity toward Acinetobacter Baumannii

Huifan Li,Jiahui Zhong,Huijuan Zhu,Yuping Yang,Mengna Ding,Liulin Luo,Yuning Huo,Hexing Li

The hybrid Cu2O-TiO2 photocatalytic composite with a p-n heterojunction was synthesized by the supercritical solvothermal route. The uniformly distributed Cu2O was stably combined with TiO2 to benefit the increase of the specific surface area, the harvesting of visible light, and the separation of photogenerated holes and electrons. As a result, photocatalytic inactivation of Acinetobacter baumannii under visible-light irradiation was facilitated. Based on the photogenerated holes acting as the main active species, the Cu2O-TiO2 photocatalytic heterojunction could induce the leakage of K+ ions and serious damage of the cell structure, leading to the final cell death. During the photoantibacterial process on Cu2O-TiO2, the RecA gene played a significant role in enhancing the survival rate of Acinetobacter baumannii. Furthermore, the great bactericidal effect of Cu2O-TiO2 nanocomposites to different bacteria indicated the potential application for the environment-friendly disinfection.

Photoelectrocatalytic bacterial inactivation of Acinetobacter baumannii on Cu2O/TiO2@Cu mesh photoanodes

Mengna Ding,Fengyan Shi,Huijuan Zhu,Yuping Yang,Jiahui Zhong,Liulin Luo,Yuning Huo,Hexing Li

A hybrid Cu2O/TiO2 composite loaded on a Cu mesh (Cu2O/TiO2@Cu mesh) was developed via a supercritical solvothermal route. A uniformly distributed Cu2O/TiO2 p–n hetero-junction was stably coated on a Cu mesh, benefiting the visible-light harvesting. In comparison to electrochemistry and visible-light photocatalysis, the photoelectrochemistry process on the photoanode of the Cu2O/TiO2@Cu mesh presented improved bacterial inactivation activity for Acinetobacter baumannii. By means of the main active species of ˙O2− radicals, the photoelectrocatalytic process on the Cu2O/TiO2@Cu mesh also realized complete leakage of K+ ions, much stronger mineralization of the cell membrane and more efficient elimination of the self-recovery function, finally leading to cell death. In comparison, the electrochemistry process only led to the leakage of K+ ions without inactivation of bacteria, and the visible-light photocatalysis process induced partial leakage of K+ ions and cell mineralization after the death of most bacteria.

A Novel Nitrite Reductase from Acinetobacter haemolyticus for Efficient Degradation of Nitrite

Xiao-Yan Yin,Emmanuel Mintah Bonku,Jian-Feng Yuan,Zhong-Hua Yang

Nitrite reductases play a crucial role in the nitrogen cycle, demonstrating significant potential for applications in the food industry and environmental remediation, particularly for nitrite degradation and detection. In this study, we identified a novel nitrite reductase ( Ah NiR) from a newly isolated denitrifying bacterium, Acinetobacter haemolyticus YD01. We constructed a heterologous expression system using E. coli BL21/pET28a- Ah Nir, which exhibited remarkable nitrite reductase enzyme activity of 29 U/mL in the culture broth, substantially higher than that reported for other strains. Structural analysis of Ah NiR revealed the presence of [Fe-S] clusters, with molecular docking studies identifying Tyr-282 and Ala-289 as key catalytic sites. The enzymatic properties of Ah NiR demonstrated an optimal pH of 7.5 and an optimal catalytic temperature of 30 °C. Its kinetic parameters, K m and v max, were 1.53 mmol/L and 10.18 mmol/min, respectively, fitting with the Michaelis–Menten equation. This study represents the first report of a nitrite reductase from a denitrifying bacterium, providing a new enzyme source for nitrite degradation applications in the food industry and environmental remediation, as well as for biosensing technologies aimed at nitrite detection.

Strategically reducing carbapenem-resistant Acinetobacter baumannii through PDCA cycle-driven antibiotic management

Xuekun Nie,Minhua Lin,Shuanglin Xu,Lingling Zhang,Xiaohui Lin,Weiying Huang

Purpose With the escalating global challenge of antibiotic resistance , particularly the resistance rate of Acinetobacter baumannii , the need to rationalize carbapenem antibiotic use in clinical settings has become paramount. Our study tapped into a fishbone diagram to uncover the irrationalities in applying these antibiotics and highlight potential influencing factors. Methods Based on these analyses, we initiated targeted intervention strategies. A PDCA cycle-based scientific management approach was implemented through the combined efforts of our antimicrobial stewardship team and relevant departments. Results Our study showed a significant post-intervention increase in the rational use of carbapenem antibiotics (P < 0.01) and a concurrent decrease in the detection of carbapenem-resistant Acinetobacter baumannii . Conclusion Our findings underscore that carbapenem usage can be effectively minimized with the continuous refinements offered by the PDCA cycle, leading to a reduction in multidrug-resistant bacteria, thus fostering rational drug use in healthcare.

Biodegradation and metabolic pathway of sulfamethoxazole by a novel strain Acinetobacter sp.

Wang Shizong,Wang Jianlong

In this study, a novel strain capable of degrading sulfamethoxazole (SMX) was isolated and identified as Acinetobacter sp. The effect of influencing factors, such as initial SMX concentration (5–240 mg/L), temperature (15–35 °C), and pH (5–7), on SMX degradation was investigated. The results showed that when the initial SMX concentration was in the range of 5–240, the removal efficiency was 100%. The optimal condition for SMX biodegradation and microbial growth was determined to be 25 °C and pH = 7.0 in terms of the removal efficiencies of SMX and total organic carbon (TOC). Four metabolite compounds were detected during the process of SMX biodegradation, and the degradation pathways were tentatively proposed. In summary, Acinetobacter sp. was highly efficient in mineralizing SMX, which has the potential to be used for degrading SMX in water and wastewater.

Phenol biodegradation by Acinetobacter radioresistens APH1 and its application in soil bioremediation

Liu Yifan,Wang Weiwei,Shah Syed Bilal,Zanaroli Giulio,Xu Ping,Tang Hongzhi

Phenol accounts for a large proportion of the contamination in industrial wastewater discharged from chemical plants due to its wide use as a raw chemical. Residual phenol waste in water and soil significantly endangers human health and the natural environment. In this study, an Acinetobacter radioresistens strain, APH1, was isolated and identified for its efficient capability of utilizing phenol as sole carbon source for growth. A draft genome sequence containing 3,290,330 bases with 45 contigs was obtained, and 22 genes were found to be involved in phenol metabolism and 51 putative drug-resistance genes were annotated by genomic analysis. The optimal conditions for cell culture and phenol removal were determined to be 30 °C, pH 6.0, and a phenol concentration of 500 mg/L; the upper limit of phenol tolerance was 950 mg/L. Based on GC-MS analysis, the key metabolites including cis,cis -muconic acid, catechol, and succinic acid were detected. During bioremediation experiment using 450 mg/kg (dry weight) of phenol-contaminated soil, the strain APH1 removed 99% of the phenol within 3 days. According to microbial diversity analysis, the microbial abundance of Chungangia , Bacillus , Nitrospira , Lysinibacillus , and Planomicrobium increased after the addition of phenol. Furthermore, at day 23, the abundance of strain APH1 was greatly reduced, and the microbial diversity and structure of the whole microbial community were gradually recovered, indicating that strain APH1 would not affect this microbial ecosystem. These findings provide insights into the bioremediation of soil contaminated with phenol.

Supercharged precision killers: Genetically engineered biomimetic drugs of screened metalloantibiotics against Acinetobacter baumanni

Xianyuan Wei,Bin Xue,Shuangchen Ruan,Jintong Guo,Yujing Huang,Xiaorui Geng,Dan Wang,Cangtao Zhou,Jun Zheng,Zhen Yuan

To eliminate multidrug-resistant bacteria of Acinetobacter baumannii, we screened 1100 Food and Drug Administration–approved small molecule drugs and accessed the broxyquinoline (Bq) efficacy in combination with various metal ions. Antibacterial tests demonstrated that the prepared Zn(Bq)2 complex showed ultralow minimum inhibitory concentration of ~0.21 micrograms per milliliter with no resistance after 30 passages. We then constructed the nano zeolitic imidazolate framework-8 (ZIF-8) as a drug carrier of Zn(Bq)2 and also incorporated the photosensitizer chlorin e6 (Ce6) to trace and boost the antibacterial effect. To further ensure the stable and targeted delivery, we genetically engineered outer membrane vesicles (OMVs) with the ability to selectively target A. baumannii. By coating the ZnBq/Ce6@ZIF-8 core with these OMV, the resulted drug (ZnBq/Ce6@ZIF-8@OMV) exhibited exceptional killing efficacy (>99.9999999%) of A. baumannii. In addition, in vitro and in vivo tests were also respectively carried out to inspect the remarkable efficacy of this previously unknown nanodrug in eradicating A. baumannii infections, including biofilms and meningitis.

Analysis of the Metabolites of Indole Degraded by an Isolated Acinetobacter pittii L1

Zuoyi Yang,Junhui Zhou,Yanbin Xu,Yaping Zhang,Haien Luo,KenLin Chang,Yujie Wang

Indole and its derivatives are typical nitrogen heterocyclic compounds and have been of immense concern since they are known for the risk of their toxic, recalcitrant, and carcinogenic properties for human and ecological environment. In this study, a Gram-negative bacterial strain of eliminating indole was isolated from a coking wastewater. The strain was confirmed as Acinetobacter pittii L1 based on the physiological and biochemical characterization and 16S ribosomal DNA (rDNA) gene sequence homology. 400 mg/L indole could be completely removed within 48 h by the strain on the optimum condition of 37°C, pH 7.4, and 150 rpm. The organic nitrogen was converted to NH 3 -N and then to and the organic carbon was partially transferred to CO 2 during the indole biodegradation. The metabolic pathways were proposed to explain the indole degradation based on the liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis of indigo, 4-(3-Hydroxy-1H-pyrrol-2-yl)-2-oxo-but-3-enoic acid, and isatin. The toxicity of the biodegradation products was evaluated using the Microtox test, which revealed that the metabolites were more toxic than indole. Our research holds promise for the potential application of Acinetobacter pittii L1 for NHCs degradation, production of indigoids, and soil remediation as well as treatment of indole containing wastewater.

Characterization of phage HZY2308 against Acinetobacter baumannii and identification of phage-resistant bacteria

Wang Ruilin,You Xiaojuan,Liu Xinwei,Fei Bing,Li Yifan,Wang Dan,Zhu Rui,Li Yongwei

Background Acinetobacter baumannii (AB) is a notable cause of hospital-acquired infections, with carbapenem-resistant Acinetobacter baumannii (CRAB) classified as a high-priority critical pathogen. Bacteriophage therapy is emerging as a promising alternative to combat drug-resistant bacterial infections. In this study, a lytic phage, HZY2308, was isolated from hospital sewage, and the biological properties, biosafety and anti-biofilm properties of phage HZY2308 were characterized and identified. Moreover, the antibacterial effect of phage HZY2308 in combination with antibiotics was investigated, and the apparent characteristics of phage-resistant strain AB48-R were demonstrated, which provided data support for further studies to elucidate the mechanism of generating phage resistance. Methods Phage HZY2308 was isolated by double agar plate method using clinical strain AB48 as the host bacterium. The morphology of phage HZY2308 was identified by transmission electron microscopy (TEM), and biological characteristics of phage HZY2308 were identified by host range, the efficiency of plating (EOP), sensitivity to temperature, pH, and chloroform, one-step growth curve, the optimal multiplicity of infection (MOI), and detection of endotoxin and cytotoxicity. Besides, the complete genome map of HZY2308 was constructed using CGview, and the phylogenetic tree of HZY2308 was constructed with MEGA. Additionally, the full genomic sequence of phage HZY2308 and the selected phage were compared using Easyfig. Checkerboard test of phage HZY2308 in combination with tigecycline (TGC) was performed to investigate their synergistic effect and bactericidal kinetics. The effect of HZY2308 on biofilm was investigated by semi-quantitative staining of biofilm with crystal violet, determination of bacterial activity in biofilm by 2,3-Bis (2-methoxy-4-nitro-5-sulfophenyl) -2 H-tetrazolium-5-carboxanilide (XTT) assay and observation of biofilm structure by fluorescence microscopy. Finally, Phage-resistant bacteria AB48-R were characterized by colony-forming capacity, morphology, growth curves, adsorption efficiency, and antibiotic susceptibility assays. Results A lytic phage, HZY2308, was isolated from hospital sewage, which exhibited advantageous traits such as a brief incubation period, large burst size, and robust stability. Safety assessments conducted at both genetic and cellular levels also have yielded positive outcomes. Besides, phage HZY2308 effectively inhibited AB biofilm formation and disrupted established biofilm structures. Furthermore, a synergistic antibacterial effect was noted when phage HZY2308 was combined with tigecycline. Interestingly, the phage-resistant strain, AB48-R was screened through natural selection. Compared to the wild strain AB48, the adsorption efficiency of the phage to AB48-R diminished. However, AB48-R’s sensitivity to antibiotics such as cefepime, gentamicin, amikacin, and tobramycin increased, indicating an evolutionary trade-off. Conclusions Phage HZY2308 shows strong antimicrobial potential, especially in combination with tigecycline, and the phage-resistant strain exhibits increased antibiotic sensitivity.

CRISPR-Cas in Acinetobacter baumannii Contributes to Antibiotic Susceptibility by Targeting Endogenous AbaI

Yuhang Wang,Jie Yang,Xiaoli Sun,Mengying Li,Pengyu Zhang,Zhongtian Zhu,Hongmei Jiao,Tingting Guo,Guocai Li

Acinetobacter baumannii is a well-known human opportunistic pathogen in nosocomial infections, and the emergence of multidrug-resistant Acinetobacter baumannii has become a complex problem for clinical anti-infective treatments. The ways this organism obtains multidrug resistance phenotype include horizontal gene transfer and other mechanisms, such as altered targets, decreased permeability, increased enzyme production, overexpression of efflux pumps, metabolic changes, and biofilm formation. A CRISPR-Cas system generally consists of a CRISPR array and one or more operons of cas genes, which can restrict horizontal gene transfer in bacteria. Nevertheless, it is unclear how CRISPR-Cas systems regulate antibiotic resistance in Acinetobacter baumannii. Thus, we sought to assess how CRISPR-Cas affects biofilm formation, membrane permeability, efflux pump, reactive oxygen species, and quorum sensing to clarify further the mechanism of CRISPR-Cas regulation of Acinetobacter baumannii antibiotic resistance. In the clinical isolate AB43, which has a complete I-Fb CRISPR-Cas system, we discovered that the Cas3 nuclease of this type I-F CRISPR-Cas system regulates Acinetobacter baumannii quorum sensing and has a unique function in changing drug resistance. As a result of quorum sensing, synthase abaI is reduced, allowing efflux pumps to decrease, biofilm formation to become weaker, reactive oxygen species to generate, and drug resistance to decrease in response to CRISPR-Cas activity. These observations suggest that the CRISPR-Cas system targeting endogenous abaI may boost bacterial antibiotic sensitivity.IMPORTANCE CRISPR-Cas systems are vital for genome editing, bacterial virulence, and antibiotic resistance. How CRISPR-Cas systems regulate antibiotic resistance in Acinetobacter baumannii is almost wholly unknown. In this study, we reveal that the quorum sensing regulator abaI mRNA was a primary target of the I-Fb CRISPR-Cas system and the cleavage activity of Cas3 was the most critical factor in regulating abaI mRNA degradation. These results advance our understanding of how CRISPR-Cas systems inhibit drug resistance. However, the mechanism of endogenous targeting of abaI by CRISPR-Cas needs to be further explored.
