Introduction: Photothermal responsive, antimicrobial hydrogels are very attractive and have great potential in the field of tissue engineering. The defective wound environment and metabolic abnormalities in diabetic skin would lead to bacterial infections. Therefore, multifunctional composites with antimicrobial properties are urgently needed to improve the current therapeutic outcomes of diabetic wounds. We prepared an injectable hydrogel loaded with silver nanofibers for efficient and sustained bactericidal activity. Methods: To construct this hydrogel with good antimicrobial activity, homogeneous silver nanofibers were first prepared by solvothermal method and then dispersed in PVA-lg solution. After homogeneous mixing and gelation, injectable hydrogels (Ag@H) wrapped with silver nanofibers were obtained. Results: By virtue of Ag nanofibers, Ag@H exhibited good photothermal conversion efficiency and good antibacterial activity against drug-resistant bacteria, while the in vivo antibacterial also showed excellent performance. The results of antibacterial experiments showed that Ag@H had significant bactericidal effects on MRSA and E. coli with 88.4% and 90.3% inhibition rates, respectively.Discussion: The above results indicate that Ag@H with photothermal reactivity and antibacterial activity is very promising for biomedical applications, such as wound healing and tissue engineering.

Organic rectorite (OREC) was used to prepare the intercalated nanocomposites with N-(2-hydroxyl) propyl-3-trimethyl ammonium chitosan chloride (HTCC), and then the immobilization of the positively charged HTCC-OREC nanocomposites and the negatively charged sodium alginate (ALG)
on cellulose nanofibrous mats was performed through layer-by-layer (LBL) technique. Fiber diameter distribution results from Field Emission Scanning Electron Microscopy (FE-SEM) images showed that the average fiber diameter of (HTCC-OREC/ALG) n films coating obviously increased from 433 to
608 nm. Moreover, X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD) results further confirmed the interaction between HTCC and OREC and their successful immobilization on cellulose template. MTT assay indicated that the prepared nanofibrous mats exhibited strong inhibitory
activity against human hepatocellular carcinoma cells (SMMC-7721) but a little cytotoxic effect on human Chang liver (CCL-13) cells. Furthermore, the experimental results from FE-SEM and Inverted Fluorescence Microscope of SMMC-7721 cells cultured on LBL structured nanofibrous mats demonstrated
the significant antitumor activity of prepared samples. The developed approach to immobilize nanocomposites onto polymer nanofibers with controllable thickness may also be utilized to tumor therapy.

Objective Islet allotransplantation has demonstrated improved clinical outcomes using the hepatic portal vein as the standard infusion method. However, the current implantation site is not ideal due to the short-term thrombotic and long-term immune destruction. Meanwhile, the shortage of human organ donors further limits its application. To find a new strategy, we tested a new polymer combination for islet encapsulation and transplantation. Meanwhile, we explored a new site for xenogeneic islet transplantation in mice. Method We synthesized a hydrogel combining alginate plus poly-ethylene-imine (Alg/PEI) for the encapsulation of rat, neonatal porcine, and human islets. Transplantation was performed into the retroperitoneal retro-colic space of diabetic mice. Control mice received free islets under the kidney capsule or encapsulated islets into the peritoneum. The biochemical indexes were measured, and the transplanted islets were harvested for immunohistochemical staining of insulin and glucagon. Results Mice receiving encapsulated rat, porcine and human islets transplanted into the retroperitoneal space maintained normoglycemia for a median of 275, 145.5, and 146 days, respectively. In contrast, encapsulated xenogeneic islets transplanted into the peritoneum, maintained function for a median of 61, 95.5, and 82 days, respectively. Meanwhile, xenogeneic islets transplanted free into the kidney capsule lost their function within 3 days after transplantation. Immunohistochemical staining of encapsulated rat, porcine and human islets, retrieved from the retroperitoneal space, allowed to distinguish morphological normal insulin expressing β- and glucagon expressing α-cells at 70, 60, and 100 days post-transplant, respectively. Conclusion Transplantation of Alg/PEI encapsulated xenogeneic islets into the retroperitoneal space provides a valuable new implantation strategy for the treatment of type 1 diabetes.

Entrapment of enzyme in a cage structure of alginate/silica composite gel can maintain the chemical or configurational structures unchanged in the enzyme, providing a promising means of enzyme immobilization. The alginate/silica composite gel can also provide more adsorption sites for the substrate, which is biocompatible and will carry out degradation and adsorption of pollutants within a cage-like structure reactor simultaneously. In this study, we used a magnetic host-guest cage structured MNPs@ALG@SiO 2 composite with a specific enzyme embedded as its guest in the host matrix, to enhance its performance. Additionally, its swelling, leakage rate and immobilization conditions were optimized. The magnetic nanoparticles (MNPs) were used to compensate for the ease of collapse of the internal structure of alginic acid, while SiO 2 was used to enhance the swelling resistance of the carrier and prevent the degradation of polysaccharides in the natural environment, thereby maintaining the enzyme biological activity and immobilization rate. Finally, the effects on the degradation of tetrachloroguaiacol (TeCG) by the immobilized enzyme embedded in the MNPs@ALG@SiO 2 composite were investigated. When the concentration of ALG was 2% and the TEOS concentration was 40 %, the immobilized MNPs@ALG@SiO 2 was found to perform the best. The optimum conditions of immobilizing laccase are pH = 5, enzyme concentration 0.4 g/L, adsorption time 12 h and temperature 20 ℃. The degradation conditions of TeCG are pH = 6, degradation temperature 40 ℃, degradation time 10 h and the degradation products of TeCG were acetic acid and ethanol according to GC–MS.

Biomedical dressings are highly susceptible to bacterial infections, and most suffer from poor mechanical resistance, leading to structural damage during application and reduced effectiveness. Therefore, developing novel dressings with high mechanical strength and effective antibacterial ability is essential. In this study, a multi-component composite membrane is designed to address these challenges by leveraging the reinforcing effect of hydroxyapatite nanofibers (HANFs) and the photothermal antibacterial abilities of copper sulfide (CuS). This composite membrane exhibits excellent mechanical properties, with a maximum ultimate tensile strength and Young's modulus of 61 ± 1.25 MPa and 937.36 ± 16.15 MPa, respectively, in the dry state. Under near infrared (NIR) irradiation, the generated heat and released Cu 2+ produce a synergistic antibacterial effect, with the inhibition rate against Staphylococcus aureus ( S. aureus ) exceeding 90 % when the HANFs@CuS content reaches 40 wt%. Cellular experiments confirm the good biocompatibility of the composite membranes across different HANFs@CuS contents, with cell viability maintaining over 80 % at 40 wt%. This study seamlessly integrates the mechanical advantage of HANFs with the photothermal antibacterial ability of CuS nanoparticles , providing a promising strategy for designing advanced biomedical dressings.

Poly (lactic-co-glycolic acid) (PLGA) nanoparticles are an ideal paclitaxel (PTX)-carrying system due to its biocompatibility and biodegradability. But it possessed disadvantage of drug burst release. In this research, a layer-by-layer deposition of chitosan (CS) and sodium alginate (ALG) was applied to modify the PLGA nanoparticles. The surface charges and morphology of the PLGA, PLGA/CS and PLGA/CS/ALG particles was measured by capillary electrophoresis and SEM and TEM, respectively. The drug encapsulation and loading efficiency were confirmed by ultraviolet spectrophotometer. The nanoparticles were stable and exhibited controlled drug release performance, with good cytotoxicity to human lung carcinoma cells (HepG 2). Cumulatively, our research suggests that this kind of three-layer nanoparticle with LbL-coated shield has great properties to act as a novel drug-loaded system.

The contamination of chronic wound with bacteria especially methicillin-resistant Staphylococcus aureus (MRSA) is considered as the major factor interferencing normal wound healing. There still remain great challenges in developing safe and effective wound dressings with wide-spectrum antibacterial functions. Alginate hydrogel is a common dressing for wound treatment. Copper is one of the trace elements in human body with inherent antibacterial activity. Traditional methods for preparing a structure-controlled copper-alginate antibacterial matrix are difficult however, due to the fast and uncontrolled gelation between alginate and metal ions. In this work, we report an electrodeposition method for rapid fabrication of copper cross-linked alginate antibacterial films (Cu 2+ -Alg) with controlled structure and copper content, which is relied on an electrical signal controlled release of copper ions from the reaction of insoluble salt Cu 2 (OH) 2 CO 3 and the generated protons via water electrolysis on anode. The results prove that the physical structure and chemical composition of the electrodeposited Cu 2+ -Alg films can be continuously modulated by the imposed charges during electrodeposition. In vitro tests demonstrate the film has Cu 2+ content-dependent bactericidal activities. Film’s cytocompatibility is well controlled by the imposed charges for Cu 2+ -Alg fabrication. The MRSA infected wound model in vivo also indicates that Cu 2+ -Alg film can effectively eliminate bacterial infection and suppress host inflammatory responses. We believe this study demonstrates a convenient and controllable strategy to fabricate alginate antibacterial dressings with potential applications for infected wound treatment. More broadly, our work reveals electrodeposition is a general and simple platform to design alginate films with versatile functions.

Efficient bone regeneration requires scaffolds that combine mechanical strength with controlled osteoinductive factor delivery. Here, we present a porous Ti6Al4V scaffold (PXTC4) functionalized with polydopamine and integrated with dual-level BMP2–PLA–Alg core–shell microspheres (BMP2-HPLA-Alg) for sustained BMP2 release. The PXTC4 scaffold exhibited high porosity (∼75%) and an elastic modulus (∼6.5 GPa) compatible with cortical bone, while the microspheres ensured heparin-mediated BMP2 adsorption, tunable degradation, and reduced initial burst release by 53.5% compared to the single-level BMP2-Alg microspheres. In vitro studies demonstrated enhanced MC3T3-E1 adhesion, proliferation, cytoskeletal organization, early osteogenic differentiation, and late-stage matrix mineralization. These results indicate that BMP2 synergizes with scaffold topography to create a favorable osteogenic microenvironment. This BMP2-HPLA-Alg microsphere-functionalized porous Ti6Al4V scaffold provides a robust platform with mechanical stability and sustained pro-osteogenic activity, offering a promising strategy for bone regeneration that warrants further in vivo investigation.

A stable food-grade amphiphilic Lys/Alg nanoparticle emulsifier was prepared by double crosslinking. • An Edible, all-biomass-derived multifunctional Pickering emulsion was successfully developed via simple emulsification. • The Lys/Alg served as effective Pickering stabilizer enhanced the stability and bioavailability of curcumin. • The developed emulsion exhibited high antioxidant properties (99.02%) and broad-spectrum antibacterial activity. • The shelf life of various fruits was effectively extended by the emulsion coating. The integration of nano-encapsulation with edible coatings holds great potential for active fruit preservation. However, conventional emulsions often rely on chemical surfactants, posing safety risks and limited functionality. Here, we developed an edible, all-biomass-derived Pickering emulsion (CLAPE) coating stabilized by amphiphilic lysozyme/alginate composite nanoparticles (Lys/Alg). The Lys/Alg were fabricated through electrostatic self-assembly followed by Ca 2+ cross-linking, forming a robust interfacial layer that effectively encapsulates curcumin (Cur). Molecular docking simulations revealed a strong binding affinity between Lys and Cur (binding energy: −87.2 kcal/mol), which significantly enhances the aqueous dispersibility and stability of Cur. The resulting Pickering emulsion enables sustained release of curcumin over 60 h, with a peak cumulative release of 94.0%, approximately twice that of the unencapsulated control. This release profile follows Fickian diffusion, extending the bioactive duration and increasing the total released amount, thereby enhancing curcumin's bioavailability. Moreover, CLAPE exhibits excellent thermal stability, maintaining high stability indices under varied accelerated storage conditions. It also exhibited high antioxidant (99.02%) and broad-spectrum antibacterial activity. Additionally, the application of CLAPE on strawberries, bananas and grapes has led to a doubling of their shelf life while maintaining the freshness and nutrient content of the fruits. The edible active preservative material, based entirely on biomass, not only forms a multifunctional protective coating that prevents spoilage but can also be easily washed off. This study presents an innovative strategy for the designs of active preservation materials utilizing all-natural plant-derived components. Lacking self-repair abilities, injuries to articular cartilage can lead to cartilage degeneration and ultimately result in osteoarthritis. Tissue engineering based on functional bioactive scaffolds are emerging as promising approaches for articular cartilage regeneration and repair. Although the use of cell-laden scaffolds prior to implantation can regenerate and repair cartilage lesions to some extent, these approaches are still restricted by limited cell sources, excessive costs, risks of disease transmission and complex manufacturing practices. Acellular approaches through the recruitment of endogenous cells offer great promise for in situ articular cartilage regeneration. In this study, we propose an endogenous stem cell recruitment strategy for cartilage repair. Based on an injectable, adhesive and self-healable o-alg-THAM/gel hydrogel system as scaffolds and a biophysio-enhanced bioactive microspheres engineered based on hBMSCs secretion during chondrogenic differentiation as bioactive supplement, the as proposed functional material effectively and specifically recruit endogenous stem cells for cartilage repair, providing new insights into in situ articular cartilage regeneration.

Intervertebral disc degeneration (IVDD) can be caused by aging, injury, and genetic factors. The pathological changes associated with IVDD include the excessive accumulation of reactive oxygen species (ROS), cellular pyroptosis, and extracellular matrix (ECM) degradation. There are currently no approved specific molecular therapies for IVDD. In this study, we developed a multifunctional and microenvironment-responsive metal-phenolic network release platform, termed TMP@Alg-PBA/PVA, which could treat (IL-1β)-induced IVDD. The metal-phenolic network (TA-Mn-PVP, TMP) released from this platform targeted mitochondria to efficiently scavenge ROS and reduce ECM degradation. Pyroptosis was suppressed through the inhibition of the IL-17/ERK signaling pathway. These findings demonstrate the versatility of the platform. And in a rat model of IVDD, TMP@Alg-PBA/PVA exhibited excellent therapeutic effects by reducing the progression of the disease. TMP@Alg-PBA/PVA, therefore, presents clinical potential for the treatment of IVDD.
