A reaction-guided strategy enabled rapid screening of acrolein scavengers. • Eight acrolein-reactive compounds were identified from Anoectochilus roxburghii . • Narcissoside inhibited AGEs formation and protein glycoxidation. • NAR and ISO alleviated ACR-induced carbonyl stress and inflammation. • Potential application in mitigating carbonyl stress in food systems. Acrolein (ACR), a highly reactive α,β-unsaturated carbonyl compound generated during thermal food processing and lipid peroxidation, contributes to carbonyl stress and oxidative protein modification. In this study, a reaction-guided UHPLC-DAD-HRMS/MS strategy was developed to screen natural ACR scavengers from Anoectochilus roxburghii ( A. roxburghii ). Eight major ACR-reactive compounds were identified, and narcissoside (NAR) was selected for further validation due to its relatively high abundance and confirmed ACR reactivity. NAR directly trapped ACR in vitro through mono-, di-, and tri-adduct formation, while in vivo fecal analysis suggested that its metabolite isorhamnetin (ISO) contributed to ACR trapping. NAR also attenuated protein modification in ACR–BSA, MGO–BSA, Glu–BSA, and Fru–BSA models. Furthermore, low-dose subacute ACR exposure showed that NAR and ISO improved serum carbonyl stress, oxidative stress, and inflammatory markers. These findings identify NAR as a representative ACR-trapping constituent of A. roxburghii . Download: Download high-res image (232KB) Download: Download full-size image

Background: Plant growth-promoting bacteria (PGPB) represents a common biological fertilizer with remarkable effect in improving crop production and environmental friendliness. Methods: In the present work, we presented a detailed characterization of plant morphology and physiology, metabolism, and pharmacological activity of A. roxburghii between Burkholderia ambifaria LK-P4 inoculation and un-inoculation (CK) treatment by routine analytical techniques (include microscopy and enzymatic activity assays and so on) coupled with metabolomics approaches. Results: Morphological and physiological results showedthat the P4 bacteria could significantly increase plant stomatal density, freshweight, survival rate, and the content of total flavonoids in leaves but reducethe amount of free amino acid. Furthermore, metabolite data showed that fatty acids (linoleic acid, linolenic acid, stearic acid) and active substance (kyotorphin and piceatannol) were specifically up-regulated in P4 inoculation. It was also demonstrated that the differential metabolites were involved in citrate cycle, glyoxylate and dicarboxylate metabolism, and biosynthesis of unsaturated fatty acids pathway. In addition, pharmacological efficacy found that A. roxburghii under P4 inoculation can significantly decrease (p < 0.05) blood glucose levels and protect the organs of mice with similar effect of Glibenclamide tablets.Conlusion: Overall, it can be seen that the exogenous P4 bacteria can promote the growth and increase content of special metabolites in A. roxburghii. This study provided theoretical basis and supported for the high-yield and high-quality bionic cultivation of A. roxburghii.

Integrated screening and HSCCC identified narcissoside from A. roxburghii . • Narcissoside reversibly inhibited ADH through a mixed-type mechanism. • Narcissoside bound ADH and protected HepG2 cells from ethanol injury. Alcohol dehydrogenase (ADH)-mediated ethanol oxidation and oxidative stress are closely associated with alcohol-induced hepatocellular injury. In this study, an integrated bioactivity-guided strategy combining microfractionation, affinity ultrafiltration, and HPLC was developed to screen hepatoprotective constituents from Anoectochilus roxburghii . Narcissoside was identified as the leading dual-active candidate and was successfully isolated by one-step high-speed countercurrent chromatography (HSCCC) with a purity of 98.32%. Narcissoside showed strong ABTS radical-scavenging activity and apparent ADH-binding activity. Enzyme assays demonstrated that narcissoside inhibited ADH activity in a concentration-dependent manner, with an IC 50 of 39.73 μM. Kinetic analysis indicated that narcissoside acted as a reversible mixed-type inhibitor, with K i lower than K is , indicating a higher apparent affinity for free ADH than for the ADH–substrate complex. Fluorescence spectroscopy indicated changes in the local microenvironment of ADH, whereas far-UV circular dichroism indicated changes in its secondary structure. Molecular docking and 200-ns molecular dynamics simulations further provided a plausible structural model involving hydrogen-bonding and hydrophobic interactions between narcissoside and ADH. In an ethanol-injured HepG2 cell model, narcissoside improved cell viability and reduced ALT and AST activities in the culture supernatant, indicating protective effects against alcohol-induced cellular injury. These findings identify narcissoside as a bioactive flavonoid glycoside from A. roxburghii with antioxidant, ADH-inhibitory, and hepatocellular protective activities in vitro, although whether the cellular protection is mechanistically related to ADH inhibition remains to be determined. Anoectochilus roxburghii (AR) has been reported to exhibit immunomodulatory activity. However, the underlying mechanism has not yet been fully elucidated. The present study employed a gas chromatography–mass spectrometry–based metabolomics approach, combined with histopathological and biochemical analyses, to investigate the metabolomic changes and therapeutic effects of AR in cyclophosphamide-induced immunosuppressed rats. The rats were divided into four groups: control group, model group, positive group, and AR-treated groups. The immunosuppressed model was established by intraperitoneal injection of cyclophosphamide (CTX, 150 mg/kg/day). The model group showed significant reductions in spleen and thymus indices, along with elevated levels of tumor necrosis factor-α (TNF-α) and interleukin-2 (IL-2) but decreased interleukin-10 (IL-10). AR treatment restored spleen and thymus indices and normalized TNF-α, IL-2, and IL-10 levels compared to the model group. Pattern recognition was employed to identify potential biomarkers. Pattern recognition analysis identified potential biomarkers: 26, 25, and 28 differential endogenous metabolites in the model, positive control, and AR-treated groups, respectively. Among these, 15 endogenous metabolites were validated using standard compounds, which demonstrated excellent linearity ( R 2 > 0.9991) and precision (intraday RSD < 4.82%, interday RSD < 4.39%). Recovery rates for the standards ranged from 95.40% to 104.91%, with RSD values below 5.97%. Pearson correlation analysis revealed that lactic acid, threonine, 5-oxoproline, palmitic acid, and arachidonic acid are positively correlated with spleen and thymus indices, whereas valine, urea, octaethylene glycol, and 2-palmitoyl glycerol are negatively correlated with these indices. Collectively, these findings demonstrate that AR extract enhances immunity in immunosuppressed rats by modulating lipid and amino acid metabolism.

A novel, simple, and efficacious analytical method for determining of nucleotides in Anoectochilus roxburghii (Wall.) Lindl ( A. roxburghii ) was developed. Magnetic dispersive solid-phase micro-extraction (MDSPME) combined with high-performance liquid chromatography was applied for extraction and determination of three nucleotides, such as adenosine-5ʹ-monophosphate (AMP), uridine-5ʹ-monophosphate (UMP) and guanosine-5ʹ-monophosphate (GMP) in A. roxburghii from different sources. The structure and morphology of magnetic nanoparticles, Fe 3 O 4 @GO, were illustrated by Fourier-transform infrared (FTIR), X-ray diffraction (XRD), transmission electron microscopy (TEM), and Thermagravimetric analysis (TGA) techniques. The effects of different extraction conditions on extraction efficiency were investigated and optimized. The optimum extraction conditions were performed as follows: 40.0 mg Fe 3 O 4 @GO were dispersed in 30 mL adsorption solution (pH 3.50, 2 μg/mL), 50 mM NaOH was employed for elution with 12 min of ultra-sonication at 40 °C. Under the aforementioned extraction conditions, the Fe 3 O 4 @GO nano-adsorbent obtained an excellent adsorption property. The corresponding linearity range for all three analytes exhibited a good linearity (r 2 ≥ 0.9982) and notable added recoveries ranging from 88.4% to 109.8%, whereas the limit of quantitation was between 0.8–8 ng/mL. The enrichment factors (EF) were between 174 and 255. The proposed method showed the advantages of full purification, high EF, simplicity, and good recovery. The method was also successfully applied to nucleotides extraction and determination in A. roxburghii, showing superior reproducibility and high sensitivity. Based on this, the method could be expected to provide a novel experimental means and developmental direction for improving pretreatment and purification of nucleotides, reducing matrix effects as much as possible, in traditional Chinese medicinal materials or biological specimens.

Ethnopharmacological relevance Anoectochilus roxburghii ( A. roxburghii ) is a popular folk medicine in many Asian countries, which has been used traditionally for treatment of some diseases such as diabetes, tumors, hyperlipemia , and hepatitis. The ethanol extract from A. roxburghii was recently shown to exert better ability to scavenge free radicals in vitro and possess antioxidant on natural aging mice in vivo . Aim of the study This study is to characterize the chemical composition, and investigate the protective effect of the A. roxburghii flavonoids extract (ARF) against hydrogen peroxide (H 2 O 2 )-induced oxidative stress in LO2 cells in vitro and D-galactose (D-gal)-induced aging mice model in vivo , and explore the underlying mechanisms. Materials and methods The chemical components of the flavonoids extract fro m A. roxburghii were detected by ultraperformance lipid chromatography coupled with quadrupole-time-of-flight mass spectrometry (UPLC-QTOF-MS/MS). H 2 O 2 was used to establish an oxidative stress model in LO2 cells. Cytotoxic and protective effects of ARF on the LO2 cells were determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method. Moreover, the levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), and malondialdehyde (MDA) in cell supernatants were measured by commercial reagent kits. Kun-Ming mice were induced to aging with D-gal (400 mg/kg, BW) by subcutaneous injection for 58 days. From the 28th day to the 58th day of D-gal treatment, ARF (122.5, 245 and 490 mg/kg, BW) and vitamin E (100 mg/kg, BW) were orally administrated to aging mice once a day for consecutive 30 days. After 25 days of the treatment with ARF, learning and memory were assessed using Morris Water Maze (MWM). At the end of the test period, the animals were euthanized by cervical dislocation , and the levels of SOD, GSH-PX, and MDA in serum, liver homogenates and brain homogenates were measured. The levels of monoamine oxidase (MAO) and acetylcholinesterase (AchE) were determined in brain homogenates. Skin and liver histopathological morphology were observed by H&E staining. Furthermore, antioxidant-related gene expression levels in the liver were carried out by quantitative real-time polymerase chain reaction (qRT-PCR). Results Nine flavonoids were identified in the extracts of A. roxburghii . In vitro assay, a high concentration of ARF (>612.5 μg/ml) reduced the survival rate and had toxic effects on LO2 cells. In addition, ARF (245 μg/ml, 490 μg/ml) and Vitamin C (200 μg/ml) markedly inhibited generations of MDA and increased activities of SOD, GSH-PX in H 2 O 2 -induced LO2 cells supernatants. In vivo assay, ARF (122.5 mg/kg, 245 mg/kg and 490 mg/kg) and Vitamin E (100 mg/kg) not only ameliorated learning and memory ability but also improved skin and liver pathological alterations. Strikingly, ARF significantly decreased MDA and MAO levels, markedly enhanced antioxidant enzyme (SOD and GSH-PX) activities. Further, compared to the D-gal group, ARF could obviously up-regulate glutathione peroxidase-1 (GPx-1) and glutathione peroxidase-4 (GPx-4) mRNA levels. Conclusions These findings suggested that ARF protects LO2 cells against H 2 O 2 -induced oxidative stress and exerts the potent anti-aging effects in D-gal aging mice model, which may be related to the inhibition of oxidative stress. Flavonoid compounds may contribute to the anti-oxidative capability and modulating aging.

Luteolin is one of the bioactive components from the compound Anoectochilus roxburghii (Wall.) Lindl. oral liquid (CAROL), which was reported to have excellent hepatoprotective and anti-inflammatory activities. However, the enrichment and quantitation of luteolin from CAROL is challenging due to the low content and complex aqueous matrix. In this study, a bifunctional monomer surface molecularly imprinted polymer (MIP) with metal–organic frameworks (MOFs) as cores was prepared for the selective adsorption of luteolin from the aqueous system CAROL. Compared with conventional MIPs, this unique nanocomposite adsorbent (MOF@MIPs) has the advantages of short kinetic equilibrium time, good selectivity, and high adsorption capacity in aqueous solution. The theoretical maximum adsorption capacity of MOF@MIPs for luteolin was 36.99 mg/g. After adsorption enrichment of luteolin from CAROL using MOF@MIPs, liquid chromatography–tandem mass spectrometry was applied to analyze the target. The corresponding linearity range for analyte was 10–6000 ng/mL with good linearity (R 2 =0.9992), and the added recoveries varied from 85.70% to 99.25%. The present method has been successfully employed for the analysis of luteolin in five different batches of CAROL. Notably, we found no significant difference in the content of luteolin between these batches, which proved that the composition was stable between batches. The novel structure MIPs are suitable for the specific recognition of template molecules in aqueous solution. Therefore, this study provides a technical reference for the special identification and determination of trace components in complex samples, while the novel MOF@MIP nanocomposite can also provide valuable references for the extraction and purification methods of specific substances in traditional Chinese medicine and expand the application environment of MIPs material.

The analysis of plant growth regulators presents a challenge due to their trace quantities and complex matrices. A novel, simple, and effective analytical method for the determination of three trace acidic plant growth regulators in Anoectochilus roxburghii (Wall.) Lindl was developed to address this issue. Three-phase hollow fiber liquid-phase microextraction combined with high-performance liquid chromatography was applied for the enrichment, purification, and determination of three acidic plant growth regulators, namely, indole-3-acetic-acid, indole-3-butyric-acid, and (+)-abscisic acid. The factors affecting extraction performance, including extractant species, pH of donor and acceptor phases, salt addition dosage, extraction time, temperature, and stirring rate, were investigated and optimized. Under optimum conditions, the proposed method provided good linearity ( R 2 , 0.9994–0.9999), low limit of detection (0.038–0.12 ng/mL), and acceptable relative recoveries (56.7–117.6%). The enrichment factors were between 153 and 328. The developed method was successfully applied to the enrichment and determination of plant growth regulators in Anoectochilus roxburghii (Wall.) Lindl and exhibited increased purification capacity, higher sensitivity, and decreased organic solvent consumption compared with conventional sample preparation methods. This method may provide a testing platform for the monitoring of plant growth regulator residues, ensuring the safe and effective use of traditional Chinese medicine.

Plants with partial or complete loss of chlorophylls and other pigments are frequently occurring in nature but not commonly found. In the present study, we characterize a leaf color mutant ‘ arly01 ’ with an albino stripe in the middle of the leaf, which is an uncommon ornamental trait in Anoectochilus roxburghii . The albino “mutant” middle portion and green “normal” leaf parts were observed by transmission electron microscopy (TEM), and their pigment contents were determined. The mutant portion exhibited underdevelopment of plastids and had reduced chlorophyll and other pigment (carotenoid, anthocyanin, and flavonoid) content compared to the normal portion. Meanwhile, comparative transcript analysis and metabolic pathways mapping showed that a total of 599 differentially expressed genes were mapped to 78 KEGG pathways, most of which were down-regulated in the mutant portion. The five most affected metabolic pathways were determined to be oxidative phosphorylation, photosynthesis system, carbon fixation & starch and sucrose metabolism, porphyrin and chlorophyll metabolism, and flavonoid biosynthesis. Our findings suggested that the mutant ‘ arly01 ’ was a partial albinism of A. roxburghii , characterized by the underdevelopment of chloroplasts, low contents of photosynthetic and other color pigments, and a number of down-regulated genes and metabolites. With the emergence of ornamental A. roxburghii in southern China, ‘ arly01 ’ could become a popular cultivar due to its unique aesthetics.
