Insulin receptors (InR) are an integral component of the insulin/insulin-like growth factor signaling pathway, which plays a vital role in insect development, lifespan, reproduction, and olfactory sensitivity. However, whether InR participate in the peripheral olfactory system of insects remains unclear. Recently, we found that 2-heptanone (2-HT) affects AcerInR expression, the gene for an InR protein, in Apis cerana cerana . We then examined the spatiotemporal expression profile of the gene in A. cerana cerana . The mRNA of AcerInR was primarily expressed in the antennae, wings, and legs of forager bees, which are probable chemosensory tissues. The results of fluorescence competitive binding assays, combined with site-directed mutagenesis, demonstrated that AcerOBP6 and AcerOBP14 exhibit strong binding affinities to 2-HT. Furthermore, after foragers were fed with double-stranded AcerInR , the expression levels of AcerOBP6 and AcerOBP14 decreased significantly, as did the electroantennogram responsiveness to 2-HT and some other odorants. In conclusion, our findings provide a foundation for understanding the involvement of AcerInR in the odor perception of A. cerana cerana . Moreover, they offer novel insights into the olfactory recognition mechanism in insects.

Honey is a complex food product, and has been linked to a number of substantial health advantages. The aim of this study was to assess the phenolic compounds, organic acids, microbial community, antioxidant activity, and antimicrobial activity of multifloral honeys produced by Apis cerana cerana , A. dorsata , and Lepidotrigona flavibasis in Hainan province, China. Our results illustrated that chlorogenic acid was the major phenolic component in all honeys. L. flavibasis honey showed the greatest antioxidant activity (DPPH IC 50 value, 7.66 mg/mL; FRAP value, 5217.06 µmol TE/kg), as well as the highest levels of proline (548.64 mg/kg), total flavonoid (11.67 mg QE/100 g), total phenolic (132.73 mg GAE/100 g), and Lactobacillus (96.28 %). Besides, all honeys were found to have a broad spectrum of antibacterial activity. Overall, our data imply that Hainan honeys, particularly stingless bee honeys, have been beneficial components of human diets.

To comprehensively evaluate the physicochemical characteristics and bioactivities of three distinctiveApis cerana(A. cerana) honeys—Bauhinia championiihoney (BCH),Polygonum perfoliatumhoney (PPH), andRhus chinensishoney (RCH)—systematic analyses were performed on total phenolic content (TPC), in vitro antioxidant capacity (via DPPH, ABTS, and FRAP assays), antibacterial efficacy againstStaphylococcus aureus(S. aureus) andEscherichia coli, and volatile composition analysis. Among the varieties, BCH presented the most substantial TPC (636.21 ± 17.05 mg GAE/kg), a value statistically higher than that of PPH and RCH (p< 0.05), and correspondingly exhibited superior antioxidant performance across all assays. All honeys displayed a clear concentration-dependent antioxidant effect. Antibacterial assays revealed that both undiluted and diluted (0.5 g/mL) samples exerted significant inhibitory effects on the tested pathogens, furthermore, BCH and PPH showing significantly higher inhibition againstS. aureusthan the positive control (penicillin,p< 0.05). Volatile profiling by HS-SPME-GC-QQQ identified 42, 35, and 38 volatile compounds in BCH, PPH, and RCH, respectively, including esters, terpenes, aldehydes, and phenolics. Cedrol (in PPH), phenylethyl acetate (in BCH), and benzaldehyde (in RCH) were pinpointed as potential botanical markers that define their characteristic aroma profiles and may partially contribute to their bioactivity. Collectively, these honeys possess remarkable antioxidant and antibacterial properties, with BCH showing the greatest potential for functional food applications owing to its high phenolic concentration and robust overall bioactivity. These findings elucidate the intrinsic link between the botanical chemical signatures ofA. ceranahoney and its biological functions, offering a scientific foundation for their utilization in functional food innovation, geographical indication protection, and quality authentication.

O -Carboxymethyl chitosan nanoparticles ( O -CMC-NPs), which are organic pesticide carriers, have excellent application potential. Exploring the effects of O -CMC-NPs on non-target organisms, such as Apis cerana cerana , is critical for their effective application; however, such studies are limited. This study investigated the stress response of A. cerana Fabricius after O -CMC-NPs ingestion. The administration of high O -CMC-NP concentrations enhanced the activities of antioxidant and detoxifying enzymes in A. cerana , with the activity of glutathione- S -transferase increasing by 54.43 %–64.33 % after one day. The transit of O -CMC-NPs into the A. cerana midgut resulted in their deposition and adherence to the intestinal wall, as they cluster and precipitate in acidic conditions. The population of Gillianella bacteria in the middle intestine was remarkably reduced after 6 d of administration of high O -CMC-NP concentrations. Contrastingly, the abundance of Bifidobacteria and Lactobacillus in the rectum significantly increased. These results indicate that the intake of high concentrations of O -CMC-NPs causes a stress response in A. cerana and affects the relative abundance of crucial intestinal flora, which may pose a potential risk to the colony. This implies that even nanomaterials with favorable biocompatibility should be applied reasonably within a specific range to avoid adverse effects on the environment and non-target organisms in the context of large-scale research and promotion of nanomaterials.

Chlorantraniliprole (CAP), a diamide insecticide, is extensively applied to combat pests in various crops. However, the widespread use of insecticides has raised concerns about their potential impact on pollinators. In the present study, we explored the toxic effects of CAP in two important honey bee species, Apis mellifera and Apis cerana . The 48 h LC 50 values of CAP for A. mellifera and A. cerana was 256.052 mg/L and 109.709 mg/L, implying that A. cerana is more sensitive to CAP. Prolonged exposure to 40 mg/L CAP significantly impaired sucrose responsiveness and climbing activity in both bee species. Both species showed a decrease in GR activity and GSH content with increasing CAP concentration. By contrast, the activities of GST, CAT, P450 and NAD-MDH were increased in both A. mellifera and A. cerana , but the differences between the 10 mg/L and 40 mg/L treatments were less pronounced in A. mellifera . Moreover, the immune related genes exhibited differential responses to CAP when comparing the two species. Low CAP concentrations led to down-regulation in expression of toll but up-regulation in expression of apideacin and hymeopatecin in A. mellifera , whereas A. cerana exhibited minimal changes in these genes. Additionally, CAP significantly inhibited the expression of ER stress response genes gp - 93 and P58 in A. mellifera , while 10 mg/L of CAP promoted P58 expression in A. cerana . Our results highlight species-specific effects with the possible, distinct detoxification mechanisms and immune responses between A. mellifera and A. cerana . These findings serve as a foundation for further evaluating the safety of CAP for honey bee species and offer insights into the scientific use of pesticides.

The cyclin-dependent kinase (CDK) protein family plays an important role in regulating life functions, such as the cell cycle and metabolism. This study reports the first cloning and functional analysis of A. cerana cerana CDK1 (AccCDK1) . The distribution profile of AccCDK1 in different developmental periods and different tissues was determined. The experimental results showed that the distribution of AccCDK1 was tissue-specific. AccCDK1 distribution at the transcriptional and translational levels was affected by stress conditions induced by H 2 O 2 , UV, HgCl 2 , CdCl 2 , extreme temperatures (4 °C, 44 °C) and pesticides (avermectin, lambda-cyhalothrin, haloxyfop- R -methyl, and glyphosate), which resulted in changes in the expression levels. These results suggest that AccCDK1 may have an important part to play in honey bee resistance to stress. The expression of a recombinant AccCDK1 protein in vitro enhanced the antistress capacities of E. coli and yeast, which suggests that AccCDK1 is related to the stress response. When AccCDK1 was silenced, the expression of some antioxidant genes was downregulated, and the enzymatic potencies of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) were reduced, which suggests that AccCDK1 takes part in the body's resistance to oxidative stress upon external stimulation by influencing relevant antioxidants. Notably, the survival rate of A. cerana cerana under high-temperature-induced stress decreased after AccCDK1 silencing, which verifies our results. In conclusion, we found that AccCDK1 played an indispensable function in resisting oxidative stress and maintaining normal cellular functions.

Glyphosate (GLY) stands as one of the most extensively used pesticides globally. Although several studies have investigated the effects of sublethal doses of GLY on the gut microbiota of bees, limited information is available on how residual concentrations of GLY in bee food affect Apis cerana (Asian honey bees) and Apis mellifera (European honey bees) at different time points. To explore these effects, we employed 16S rRNA Illumina sequencing technology to evaluate the consequences of chronic GLY exposure (0.7 mg/kg, GLY residual in beebread) on the gut microbiome of A. cerana and A. mellifera on days 3 and 12 of the exposure period. Even though our findings revealed no significant difference in the microbiota abundance between control bees and GLY-treated bees on the 3rd day, GLY-treated bees exhibited 55.56 % abundance of the genus Snodgrassella (a core gut bacterium) and 4.54 % of the genus Commensalibacter (a non-core gut bacterium) compared to control bees in A. cerana and A. mellifera , respectively. In addition, our results showed that a noticeable trend indicated that the gut of A. mellifera was more susceptible to GLY than that of A. cerana . Considering the vital role of gut microbiota in the health of honey bee colonies, our results suggest that 0.7 mg/kg of GLY are likely to have adverse effects on the health of honey bees.

Simple SummaryThe keen olfactory capacity ofApis cerana ceranaprovides substantial plasticity in pollination and resource selection, with odorant receptors (ORs) playing indispensable roles in detecting and transducing environmental chemical signals. In this study, we systematically characterized the structural properties, evolutionary conservation, and spatiotemporal expression patterns ofAcerOR120, and evaluated alterations in olfactory sensitivity following gene knockdown. The protein exhibits a classical seven-transmembrane architecture, shares high sequence conservation with orthologs across the genusApis, and peaks in expression during the early foraging stage (15- and 20-day-old workers), predominantly localized in antennae, heads, thoraces, and legs. SilencingAcerOR120via RNA interference significantly weakened antennal sensitivity to key odorants, including geraniol, isoamyl acetate, decanal, (E)-2-octenal, ocimene, linalool, and (+)-dihydrocarvone. These results demonstrate thatAcerOR120is involved in foraging behavior by modulating the perception of floral scents and colonial pheromones, offering a functional molecular target for deciphering and manipulating olfactory recognition inA. c. cerana. This study aims to characterize the structural features and spatiotemporal expression patterns of the protein encoded by odorant receptor geneAcerOR120, and to explore its biological functions in olfactory recognition inApis cerana cerana. Bioinformatic software was used to predict the transmembrane topology, secondary/tertiary structures and conserved domains of the AcerOR120 protein. A neighbor-joining phylogenetic tree was constructed based on homologous sequences within the genus Apis. qRT-PCR was performed to detect the gene expression profiles in antennae of worker bees at 1, 5, 10, 15, 20 and 25 days old, as well as different tissues of foragers. EAG recording was applied to compare the electrophysiological responses of worker antennae to 11 volatile odorants before and after gene silencing via RNA interference. The open reading frame ofAcerOR120is 1203 bp, encoding a 401-amino-acid protein containing seven transmembrane domains with an intracellular N-terminus. Its secondary structure is dominated by α-helices accounting for 82.79%, which matches the typical structural signature of insect odorant receptors. Phylogenetic analysis revealed that AcerOR120 shares high conservation acrossApisspecies. Spatiotemporal expression assays showed that transcript abundance in antennae increased first and then decreased with worker age, reaching the minimum at day 1 and the maximum at day 20. Among tissues of foragers, the highest expression was detected in antennae and thoraxes, followed by heads and legs, while abdomens and wings possessed low expression levels. RNAi silencing significantly reduced overall EAG responses of antennae to all 11 tested odorants, with the most dramatic decreases observed for Geraniol, Decanal, Ocimene and (+)-Dihydrocarvone.AcerOR120encodes a canonical insect odorant receptor and is highly expressed in forager antennae. It mediates the recognition of multiple floral volatiles and pheromones and is speculated to participate in olfactory-related physiological processes, including nectar source localization and social behavior regulation inA. cerana cerana.
