Polybrominated diphenyl ethers (PBDEs) are widely used as flame retardants and challenges for water treatment due to their persistence and toxicity. In this study, the reduction of 2′2′4′4′-tetrabromodiphenyl ether (BDE-47) was investigated in a nano-bio-integrated system. Results showed that the introducing of P. putida could markedly accelerate the demineralization of BDE-47 in nZVI/Pd- P.p system; the continuous generation of acidic metaboliates by P. putida could decrease pH, which could alleviate the surface passivation to some extent, resulting in the releasing of Fe 2+ and high generation of H 2 O 2 , the shift in reactive oxygen species from Fe(IV) to •OH. The BDE-47 was firstly debrominated to the DE by the highly reductive [Pd·2H] generated by nZVI/Pd, then oxidized to bromophenol and phenol, catechol as well as hydroquinone via the P. putida strain and the Fenton-like system. The toxicity assays confirmed the combined system could avert generation of nocuous intermediates, and could be an alternative strategy for complete remediation of recalcitrant POPs.

The degradation of 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) by Pycnoporus sanguineus was investigated in order to explore the impact of the heavy metal Cu 2+ on BDE-47 decomposition and the subsequent formation of metabolites, as well as to further elucidate the degradation mechanism of BDE-47. An increase in degradation rate from 18.63% to 49.76% in the first four days and its stabilization at (51.26 ± 0.08)% in the following days of BDE-47 incubation were observed. The presence of Cu 2+ at 1 and 2 mg/L was found to promote the degradation rate to 56.41% and 60.79%, respectively, whereas higher level of Cu 2+ (≥ 5 mg/L) inhibited the removal of BDE-47. The similar concentration effects of Cu 2+ was also found on contents of fungal protein and amounts of metabolites. Both intracellular and extracellular enzymes played certain roles in BDE-47 transportation with the best degradation rate at 27.90% and 27.67% on the fourth and third day, individually. During the degradation of BDE-47, four types of hydroxylated polybrominated diphenyl ethers (OH-PBDEs), i.e., 6′-OH-BDE-47, 5′-OH-BDE-47, 4′-OH-BDE-17, 2′-OH-BDE-28, and two bromophenols, i.e., 2,4-DBP and 4-BP were detected and considered as degradation products . These metabolites were further removed by P. sanguineus at rates of 22.42%, 23.01%, 27.04%, 27.96%, 64.21%, and 40.62%, respectively.

The progressive deterioration of human male reproductive capacity has paralleled the mounting global dispersal of persistent organic pollutants (POPs) and plastic-associated contaminants. Nevertheless, the collective consequences of simultaneous exposure to these ubiquitous environmental stressors on spermatozoal function remain inadequately characterized. This study undertook a comprehensive evaluation of the interactive reproductive harm posed by 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) and 25 nm polystyrene nanoplastics (PS-NPs) on human spermatozoa. Whereas isolated challenges with ecologically pertinent doses of either BDE-47 or PS-NPs yielded negligible alterations in sperm viability and motility parameters, concurrent administration provoked a marked superadditive compromise of multiple functional attributes indispensable for successful fertilization. Co-treatment substantially attenuated both total and progressive motility, diminished the penetrative capacity through a cervical mucus surrogate, and abrogated the induction of capacitation and progesterone-induced acrosome reaction. Mechanistic elucidation uncovered that dual exposure synergistically intensified intracellular reactive oxygen species accumulation and profoundly destabilized mitochondrial membrane potential, culminating in a robust augmentation of apoptotic rates. Furthermore, convergent network pharmacology assessments pinpointed pivotal signaling hubs governing apoptosis, including Caspase-3 and constituents of the BCL-2 family members, as principal mediators of the combined toxic insult. Collectively, these findings provide direct evidence that PS-NPs function as potent vectors capable of amplifying the spermatotoxicity of BDE-47 via a cascade of oxidative imbalance, mitochondrial dysfunction and programmed cell death initiation. This work highlights the imperative for adopting mixture centric frameworks in environmental contaminant hazard evaluation and yields novel mechanistic perspectives regarding the plausible etiological role of pollutant co-exposure to idiopathic male infertility.

Polybrominated diphenyl ethers (PBDEs) as a typical brominated flame retardant (BFR) have attracted worldwide attention due to the high environmental risk and resistance to conventional remediation processes. In this study, thermally activated persulfate (TAP) process was applied to degrade 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47), which is the most toxic and representative PBDEs in e-waste dismantling sites. Impact factors such as PDS dosage, heating temperature, and initial pH were evaluated. Results showed that BDE-47 can be 100% degraded within 180 min under the condition of PDS:BDE-47 = 1000:1, 60 °C, and pH = 7. Quenching experiments combined with EPR analysis further proved the important role of SO 4 ·− in oxidating BDE-47. According to high-resolution mass spectrometry (HRMS) analysis, only one oxidation product of low toxicity was detected during the oxidation process. Theoretical calculations further revealed that the oxidation process mainly involved radical attack at C–Br bond, cleavage of C–Br bond, and fission of ether bond, and HSO 4 · may also play an important role in BDE-47 degradation in TAP system. In addition, TAP system exhibited universality as all selected PBDE congeners can be degraded, and the degradation rate of PBDEs was greatly affected by the number of substituted Br atoms in a negative trend. Overall, these findings indicate that TAP can be applied as an effective method for removal of PBDEs, and we provide a new insight for the practical application of TAP technology in BDE-47 degradation from experimental and theoretical aspects.

This study aims to use ultraviolet (UV) irradiation to decompose polybrominated diphenyl ethers (PBDEs) in the elutes and then reuse the surfactants. The results indicate that UV can remove 2,2′,4,4′-tetrabrominated diphenyl ether (BDE-47) from surfactant eluents and Triton X series surfactants also can remove BDE-47 from the soil. Triton X-100 (TX-100) is the most promising surfactant during the washing and photodegradation processes. Quench experiments suggest that both 1 O 2 and OH• were involved in the TX-100 decomposition but only 1 O 2 is responsible for the degradation of BDE-47. In analysis of the photoproducts of BDE-47 by Gas Chromatography Mass Spectrum (GC-MS) and Liquid Chromatography High Resolution Mass Spectrometry (LC-HRMS), BDE-47 was mainly debrominated to the lower-brominated BDEs and then oxidized to ring-opening products. The little loss of TX-100 can mainly be attributed to the breakage of polyethylene oxide (PEO) chain. Nevertheless, the washing wastes treated by UV light can exhibit higher solubility for BDE-47 than before, indicating they can be reused for BDE-47 removal from soil. The toxicity assessment experiments were performed using Escherichia coli ( E.coli ) as an indicator. The results indicate that the removal of BDE-47 by UV irradiation can reduce the toxicity of eluent.

To test whether manure amendment in soil reduces plant uptake of persistent organic pollutants, carrot ( Daucus carota L. ) was used as a model plant and 2,2′,4,4′-tetrabrominated diphenyl ether (BDE-47) was selected as a model persistent organic pollutant to conduct a pot experiment with contaminated soil amended by composted pig manure. The results showed that the concentration and bioconcentration factors (BCFs) of BDE-47 in the edible part of carrot significantly decreased from 229.7 ± 28.2 to 43.4 ± 20.4 ng g −1 and from 1.86 ± 0.5 to 0.15 ± 0.03, respectively, with increasing composted pig manure dose from 0 to 4%. Organic matter (OM) derived from composted pig manure played a dominant role in reducing persistent organic pollutant bioavailability in soil. Composted pig manure amendment and carrot cultivation jointly altered the bacterial community composition in soil, especially the rhizosphere. Rhizodegradation of BDE-47 was enhanced from 8.6 to 28.5% with increasing composted pig manure dose from 0 to 4%, corresponding to increased soil microbe diversity and polybrominated diphenyl ether-degrading bacteria ( Sphingomonas , etc.) abundance in the rhizosphere. This study is the first, to the best of our knowledge, to provide an effective agronomic strategy of manure amendment to reduce plant uptake and simultaneously enhance rhizodegradation of persistent organic pollutants in soil, and thus potentially reduce human health risks through dietary intake.

Polybrominated diphenyl ethers (PBDEs), a type of brominated flame retardant, are of global concern due to their environmental persistence, bioaccumulation, toxicity, and resistance to conventional remediation methods. In this study, the electrochemical reduction of 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) with Pd/Metal foam electrodes (Ni, Cu, and Ag) was investigated. The effect of Pd loadings was explored, and the results show that Pd loading enhances the debromination performance, with 15.16%Pd/Ni foam exhibiting the best efficiency, followed by 9.37%Pd/Cu and 10.26%Pd/Ag. The degradation mechanisms for Pd/Ni and Pd/Ag are primarily hydrogen atom transfer, while for Pd/Cu, electron transfer dominates. Among the reduction products, Pd/Ni foam shows the highest debromination capability. The impact of electrolytes, current intensity, and bromination degrees of PBDEs was evaluated for 15.16%Pd/Ni. The results reveal that the presence of electrolytes inhibits BDE-47 degradation; the degradation rate of BDE-47 increases with current density, peaks at 4 mA, and decreases as current rises; and 15.16%Pd/Ni foam can effectively degrade PBDEs with varying bromination levels. Additionally, cycling tests show a decrease in efficiency from 94.3% (first cycle) to 56.58% (fourth cycle), attributed to Pd loss and structural damage. The findings offer valuable insights for developing efficient, sustainable catalytic materials for the electrochemical degradation of PBDEs and other persistent organic pollutants.

The rising prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD)-associated hepatocellular carcinoma (HCC) is a growing global concern, with environmental pollutants implicated in its progression. 2,2′,4,4′-tetrabromodiphenyl ether (BDE 47) exhibits hepatotoxic potential, but its role in MASLD-to-HCC progression remains unclear. Here, we investigated the association between BDE 47 and MASLD using data from the National Health and Nutrition Examination Survey and explored the potential mechanisms through network toxicology and multiomics analyses. Individuals with higher serum BDE 47 levels showed positive associations with MASLD and advanced fibrosis. Network toxicology predicted that BDE 47 perturbed lipid metabolism, inflammation, and oxidative stress, thereby contributing to malignant progression. In vitro, integrated lipidomic and transcriptomic analyses revealed that BDE 47 disrupted lysosome, sphingolipid, and cholesterol metabolism, leading to lipid accumulation. Additionally, BDE 47 exposure enhanced inflammatory response, oxidative stress, and the development of immunosuppressive environment, which may facilitate MASLD-to-HCC progression. Pharmacological experiments further demonstrated that increased sphingolipid synthesis and impaired cholesterol efflux played important roles in BDE 47-induced lipid accumulation. Notably, only the parent BDE 47 compound was detected in exposed cells. Collectively, these findings provide mechanistic evidence of the carcinogenic potential of BDE 47 and its role in liver disease progression.

Although microbial degradation has been extensively studied for removing 2,2′,4,4′-tetrabrominated diphenyl ether (BDE-47), knowledge regarding the comparative efficiency and safety of degradation by facultative anaerobes under varying oxygen conditions remain limited. This study investigated the degradation of BDE-47 by Pseudomonas sp. strain QS-1 under both aerobic and anaerobic conditions. Our findings revealed that 47.5% and 12.4% of the initially added BDE-47 (0.5 mg/L) were transformed within 7 days under aerobic and anaerobic conditions, respectively. The aerobic transformation involved a complex pathway consisting of debromination, hydroxylation, ether bond cleavage, and ring opening, whereas the anaerobic transformation was limited to debromination. Toxicity assessments using an Escherichia coli model revealed that aerobic degradation products reduced cytotoxicity, whereas anaerobic degradation products induced higher rates of cell apoptosis than the parent BDE-47. Metabolomics analysis attributed this enhanced toxicity of anaerobic products to the specific inhibition of purine metabolism. In soil microcosm experiments, strain QS-1 exhibited stronger colonization and degradation performance under aerobic conditions, shifting the soil microbial community assembly towards more deterministic processes and strengthening network interactions with indigenous taxa. These findings provide insights into the biological effects of BDE-47 degradation products, and inform the assessment of efficacy and safety for bioremediation strategy. Application of crop residues is a conventional practice that contributes to crop production through nutrient returns and other benefits to soil health: driving soil physicochemical and biological functions. However, little is known about the impacts of straw residue incorporation on the bioavailability of organic pollutants and associated changes in microbial community structure in contaminated soils. In this study, maize straw was added to a soil contaminated with a model polybrominated diphenyl ether (BDE-47). A pot experiment was conducted and planted with carrot ( Daucus carota L.). We found that straw addition greatly reduced the bioavailability of BDE-47, changed the bacterial community structure and affected a range of soil physiochemical properties. Moreover, the amount of BDE-47 that had accumulated in carrot roots and aboveground tissues was significantly reduced. This study may therefore describe an effective agronomic strategy to reduce the bioavailability of polybrominated diphenyl ethers (PBDEs) in a soil used to grow high value vegetable crops. This strategy draws on traditional wisdom and shows promise as a practical method to support horticultural production systems, remediate soils, and help to ensure food safety.

Emerging pollutants such as polystyrene-nanoplastics (PS-NPs) and polybrominated diphenyl ethers (PBDEs) are a threat to human health and ecological safety. However, NP-mediated PBDE exposure and toxicity remain poorly understood. Herein, male frogs ( Pelophylax nigromaculatus ) were exposed to 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) and 6-hydroxy-2,2′,4,4′-tetrabromodiphenyl ether (6-OH-BDE-47) alone or in combination with PS-NPs for 30 days to explore the carrier toxicity effect and mechanism of co-exposure. Transcriptome data verified that both BDE-47 and 6-OH-BDE-47 could mediate the steroid hormone synthesis signaling pathway to induce reproductive toxicity. The presence of PS-NPs increased the bioconcentration of BDE-47 and 6-OH-BDE-47 by 54.36 % and 51.49 %, respectively, and aggravate the damage of the testes. To investigate the mechanism underlying the disrupted steroidogenesis, changes in hormones of the hypothalamic-pituitary-gonadal (HPG) axis and gene expression involved in synthesis and transformation of sex hormones were assessed. AR, StAR, CYP11A1 and HSD3B1 were all downregulated, while mRNA expression levels of CYP19A1 and ESR1 were upregulated, accompanied by a decrease in testosterone and an increase in oestradiol. The results demonstrated that PS-NPs can exacerbate the estrogen-like effects of BDE-47 and 6-OH-BDE-47 in male frogs, highlighting the potential risks posed by these pollutants in aquatic ecosystems.

Pyrolysis of agricultural waste into biochar for soil remediation is a useful solid waste management strategy. However, it is still unclear how different agricultural feedstocks affect the properties of biochars and their effectiveness in remediation of PBDE-contaminated soil. In this study, we systematically investigated dynamic alterations of soil properties, microbial communities , and PBDE dissipation and bioavailability induced by the application of biochars from manure (MBC) and straw (SBC) to PBDE-contaminated soil. The results showed that soil properties, microbial community structure, and diversity changed differently with the incorporation of the two biochars. MBC had a larger surface area (17.4 m 2 /g) and a higher nutrient content (45.1% ash content), making it more suitable for use as a soil additive to improve soil quality and nutrient conditions, as well as to stimulate microbial growth. SBC showed higher adsorption capacity for 2,2′,4,4′-Tetrabromodiphenyl Ether (BDE-47) (26.73 ± 0.65 mg/g), thus lowering the bioavailability and ecological risk of BDE-47 in soil. BDE-47 was stepwise debrominated into lower brominated PBDE by PBDE-degrading bacteria. MBC accelerated the debromination of BDE-47 (10.1%) by promoting PBDE-degrading microorganisms , while this was inhibited by SBC (3.5%) due to strong adsorption of BDE-47. In addition, we found that both types of biochar favored Nitrospirae bacteria and promoted N cycling. Overall, biochars from manure and straw can positively shape soil microbial communities differently by altering soil properties, soil fertility and nutrient availability, and the fate and the effects of contaminants, which ultimately led to a difference in the potential of biochars for their use in soil remediation.
