Nowadays, rapid and facile diagnosis of cancer using user friendly processes has attracted much attention. In this regard, an electrochemical (EC) biosensor with high sensitivity was fabricated by merging MIL156 MOF@COF nanocomposite with Au nanoparticles for the detection of CA15-3. Herein, metal clusters of MIL-156 as a Metal organic frameworks (MOF) were coated by a crystalline covalent organic frameworks (COF) through covalent bonding and created core-shell-like structures. The active part of the working electrode was modified in two consecutive steps. First, MIL-156 MOF@COF and then Au nanoparticles were electrodeposited on the glassy carbon electrode (GCE). The porosity of nanocomposite has significantly increased the surface area and improved the conductivity. Au nanoparticles also form an acceptable substrate for bonding antibodies due to their high affinity with amino groups. In addition, Au nanoparticles amplify the EC signal of the biosensor with their undeniable conductivity. Nanocomposite was characterized with XRD, SEM, and EDAX techniques. To investigate the proposed biosensor, differential pulse voltammetry (DPV) was used as an analytical technique. The CA15-3 calibration provided a linear range between concentrations of 30 and 100 nU/mL, thus expressing the powerful diagnostic potential of the designed biosensor. Furthermore, the suggested biosensor has been used in serum samples and discriminate breast cancer sufferers from healthy individuals with high confidence levels.

CA15–3 is a vital biomarker for monitoring and evaluating treatment efficacy in breast cancer patients, especially those with advanced-stage disease. Conventional immunoassays face limitations, including high cost, limited sensitivity, and dependence on potentially error-prone single-fluorescence signal outputs. To address these challenges, we designed a dual-signal-emitting sensor based on CdTe quantum dots@ZIF-8 nanomaterials, incorporating both the CRISPR/Cas12a system and catalytic hairpin assembly (CHA) technology. The binding of CA15–3 to its specific aptamer inhibits the CHA cascade reaction, thereby preventing the generation of double-stranded DNA activators necessary to initiate Cas12a-mediated trans-cleavage activity. As a result, FAM-ssDNA is extensively adsorbed onto the CdTe QDs@ZIF-8 surface, where efficient electron transfer effectively quenches its fluorescence. The incorporation of the aptamer facilitates dual-signal amplification through both the CHA and CRISPR/Cas12a systems, achieving a detection limit of 0.131 U/mL. This dual-signal output mechanism provides the system with an internal reference function, thereby significantly improving the reliability of the results. Overall, this work establishes a novel tumor biomarker detection platform that combines high sensitivity with excellent robustness.

ICP-MS-based multiplex immunoassays have the advantages of low sample consumption and minimized repetitions of tedious procedures. However, this technique is seldom used in clinical tests because of the complicated matrix effect in clinical samples and the discrimination effect to the target proteins. In this work, we developed an ICP-MS-based magnetic immunoassay by using magnetic beads as highly efficient affinity probes and optimizing the magnetic immunoassay parameters. We applied this method to the simultaneous determination of three crucial gynecological tumor biomarkers that are vital in the prognosis and monitoring of the recurrence of breast cancer and ovarian cancer. The measurable ranges of HER-2, HE4 and CA15-3 were 12–1000 ng mL−1, 8–2000 pmol L−1 and 5–200 U mL−1, with detection limits (3σ) of 3.94 ng mL−1, 2.59 pmol L−1 and 1.62 U mL−1, respectively. The relative standard deviations (RSDs) were 2.61% for HER-2 at 15 ng mL−1, 4.90% for HE4 at 25 pmol L−1 and 6.68% for CA15-3 at 10 U mL−1, indicating good accuracy of the method. The spiking test indicated that the present method had a low matrix effect. Under optimized conditions, the simultaneous detection of HER-2, HE4, and CA15-3 in clinical serum samples was performed and the values correlated well with those obtained by the time resolved fluorescence immunoassay (r2 = 0.9568 for HER-2, 0.9485 for HE4, 0.9133 for CA15-3). The present work shows that the ICP-MS-based magnetic immunoassay meets the requirements for the quantification of three crucial gynecological tumor biomarkers in real clinical tests.

The screening of glycoprotein markers has become an integral part of the in vitro diagnosis of malignant tumors. Herein, an electrochemical method based on alkaline phosphatase (ALP)-mediated enzymatic silver deposition is reported for the highly sensitive detection of glycoprotein tumor markers, in which ALP enzymes are decorated to the glycan moieties of targets via the lectin-carbohydrate interactions. As glycoproteins are conjugated with multiple glycan chains, lectin-mediated labeling can result in the decoration of each target with multiple ALP enzymes. Moreover, the enzymatic hydrolysis of ascorbic acid 2-phosphate into ascorbic acid can result in the deposition of a high density of silver particles, which can then be sensitively assayed via the robust Ag/AgCl solid-state voltammetric process. As a result, the enzymatic silver deposition-based electrochemical method exhibits high sensitivity. Using the aptamer-based electrochemical detection of the breast cancer-associated glycoprotein CA15–3 as a proof of concept, a detection limit of 0.32 mU/mL has been demonstrated. Results show that the synergism of the aptamer-based capture and the glycoform-specific discrimination capability of the lectin-carbohydrate interactions can endow this method with high selectivity, and its practical use in the assay of CA15–3 levels in serum samples has been illustrated. With benefits from the high efficiency, mild reaction conditions, and user-friendly operation, the lectin-mediated labeling of ALP enzymes for enzymatic silver deposition is highly applicable to the electrochemical detection of low-abundance glycoprotein tumor markers.

Herein we reported the construction of dual-mode electrochemiluminescence (ECL) and electrochemical (EC) sensing platform using the bipolar silica nanochannel array (bp-SNA) confined with the bifunctional probe (tris(2,2′-bipyridyl) ruthenium , Ru(bpy) 3 2+ ) inside the nanochannels (designated as Ru@bp-SNA). With the asymmetric surface charges, bp-SNA on the indium tin oxide electrode can effectively function as electrostatic nanocage for the stable confinement of Ru(bpy) 3 2+ via dual electrostatic force , namely electrostatic attraction of inner negative surface and electrostatic repulsion of outer positive surface. Ru(bpy) 3 2+ immobilized into the nanospace acts as both highly efficient ECL luminophore and well-defined EC redox molecule to generate strong signal. Since the modification of bio-recognition units on the top of Ru@bp-SNA is able to confer excellent recognitive capacity and obtain the formation of complex in the presence of specific analyte, the decrease of ECL and EC signals enables the quantitative detection of this analyte. As proof-of-concept experiments, two tumor related biomarkers, CEA and CA15–3, have been successfully determined in aqueous solution and human serum by ECL and EC modes, respectively.

Timely, convenient, and efficient detection of carbohydrate antigen 15-3 (CA15-3) levels in serum holds significant importance in early screening, diagnostic assistance and prognosis prediction of breast cancer. The development of efficient and convenient electrochemical aptasensors with immobilized redox probes for label-free detection of CA15-3 is highly desirable. In this work, a bipolar silica nanochannel array film (bp-SNA) with two distinct functional domains including nanochannels and an outer surface was employed for the immobilization of recognition ligands and electrochemical redox probes, enabling the construction of a probe-integrated aptasensor for reagentless electrochemical detection of CA15-3. Cost-effective and readily available indium tin oxide (ITO) was used as the supporting electrode for sequential growth of a negatively charged inner layer (n-SNA) followed by a positively charged outer layer (p-SNA). The preparation process of bp-SNA is convenient. Functionalization of amino groups on the outer surface of bp-SNA was modified by aldehyde groups for covalent immobilization of recognition aptamers, further establishing the recognition interface. Within the nanochannels of bp-SNA, the electrochemical redox probe, tri (2,2′-dipyridyl) cobalt (II) (Co(bpy) 3 2+ ) was immobilized, which experienced a dual effect of electrostatic attraction from n-SNA and electrostatic repulsion from p-SNA, resulting in high stability of the immobilized probes. The constructed aptasensor allowed for reagentless electrochemical detection of CA15-3 ranged from 0.001 U/mL to 500 U/mL with a low detection limit (DL), 0.13 mU/mL). The application of the constructed aptasensor for CA15-3 detection in fetal bovine serum was also validated. This sensor offers advantages of a simple and readily obtainable supporting electrode, easy bp-SNA fabrication, high probe stability and good stability.

In this study, a fascinating photocatalytic property of EDTA under light irradiation was observed, much the same as that of natural horseradish peroxidase. The small EDTA molecules enable it easily attaches to amine terminal on antibody surface, an ELISA-like assay may be fabricated through combining with gold nanoparticles (AuNPs) in suspensions can exhibit various colors. The labeled EDTA on the proteins which may catalyze decomposition of hydrogen peroxide and in situ mediate the growth of hydrogen peroxide-induced formation of AuNPs, displaying various color nanoparticle dispersion as a read-out means. A straightforward plasmonic sensor was accordingly designed to enable naked-eye observation and quantitative determination of ultra-trace target analytes based on peroxidase-like EDTA. The highly sensitive assay was utilized to determine cancer antigen 15-3 (CA15-3, a breast cancer biomarker) and methamphetamine, and their limit of detection CA15-3 and methamphetamine are 7.5 × 10 −15 U/mL and 2.8 × 10 −20 mg/mL by naked eyes, respectively. Furthermore, practical applications for detection of CA15-3 and MA in human blood samples were also carried out. The results of detecting CA15-3 of breast cancer serum samples are close agreement with those given by hospital, implying this sensor with high accuracy. This assay has a great potential to be developed into a platform to quantitatively determine analytes as long as the specific antibodies against them were available. Its versatile features will broaden the applicability toward the ultrasensitive detection of target molecules in clinical diagnosis, environmental monitoring, and food quality control only using naked eyes.

A new method for simultaneously determining cytokeratin fragment antigen 21–1 (CYFRA21-1), carcinoembryonic antigen (CEA), and carbohydrate antigen (CA15–3) was developed using single-particle, ICP-MS-based, magnetic immunoassay . Gold nanoparticles (AuNPs), ZnSe quantum dots (QDs), and Silver nanoparticles (AgNPs) were employed to tag antibodies. A highly efficient capture of CYFRA21-1, CEA, and CA15–3 was achieved using biotinylated, antibody-coated magnetic beads. AuNPs/ZnSe QDs/AgNPs-tagged second antibodies helped realize sensitive quantification of the target antigen using single particle ICP-MS. Under optimum conditions, CYFRA21-1, CEA, and CA15–3 linear concentration ranges were 0.05–50 ng mL −1 , 0.02–100 ng mL −1 , and 0.6–250 mU mL −1 . The correlation coefficients were 0.9999, 0.9987, and 0.9970, respectively. CYFRA21-1, CEA, and CA15–3 detection limits were 0.02 ng mL −1 , 0.006 ng mL −1 , and 0.25 mU mL −1 , respectively. Relative standard deviations (RSD) for CYFRA21-1, CEA, and CA15–3 were 3.2%, 4.4%, and 4.2%, respectively. The method simultaneously detected CYFRA21-1, CEA, and CA15–3 in human lung cancer serum samples with recoveries of between 89.8% and 101%.
