Here, straight upward Au nanowires (NWs) were successfully grown onto Fe 3 O 4 @TiO 2 matrix through a seed-mediated strategy to intensively improve its photocatalysis and SERS performances, facilitating a peculiar recyclable surface-enhanced Raman spectroscopy (SERS)-based immunoassay of CA19–9 in liquid form based on visible light irradiation. Such immunoassay was also supported by a smart heterobifunctional cross-linking agent-mediated probe of anti-CA19–9/4-MBA without metal nanoparticles . A low limit of detection of 5.65 × 10 −4 IU mL −1 and a wide linear range from 1000 to 0.001 IU mL −1 were demonstrated through repeated constructing the sandwich immunostructure with only one batch of nanocomposites . Moreover, the actual levels of CA19–9 for colorectal cancer patients were readily measured by the recyclable immunoassay, the results of which are principally consistent with the conventional CLIA detection. Thus, such a green strategy of visible light-induced recyclable immunoassay could be expected to have a potential practicability in the clinical diagnoses of cancer.

Convenient and sensitive detection of tumor biomarkers is crucial for the early diagnosis and treatment of cancer. Herein, we present a probe-integrated and label-free electrochemical immunosensor based on binary nanocarbon composites and surface-immobilized methylene blue (MB) redox probes for detection of carbohydrate antigen 199 (CA19-9), which is closely associated with gastric malignancies. Nanocarbon composites consisting of electrochemically reduced graphene oxides and carbon nanotubes (ErGO-CNT) are electrodeposited onto an indium tin oxide (ITO) electrode surface to form a 3D nanocomposite film, which could provide high surface area to immobilize abundant MB probes, facilitate the electron transfer of MB, and therefore, improve sensitivity. Polydopamine (PDA) served as a bifunctional linker is able to immobilize anti-CA19-9 antibodies and stabilize the inner probe, conferring the sensing interface with specific recognition capacity. Electrochemical detection of CA19-9 is achieved based on the decrease of the redox signal of MB after specific binding of CA19-9 with a wide linear range of 0.1 mU/mL to 100 U/mL and a limit of detection (LOD) of 0.54 nU/mL (S/N = 3). The constructed electrochemical immunosensor has good selectivity, repeatability, reproducibility, and stability. Furthermore, determination of CA19-9 in human serum samples is also realized. Keywords:electrochemically reduced graphene oxide;carbon nanotubes;methylene blue;CA19-9;immunosensor

Molecularly imprinted nanozymes (MINs) offer a compelling alternative to antibody-based assays by integrating specific molecular recognition with enzyme-like catalytic activity. Nevertheless, conventional molecular imprinting of glycoproteins remains technically challenging. In this work, employing CA19-9 as a model glycoprotein biomarker, we develop a surface-directed glycan imprinting strategy by integrating molecular imprinting technology with iron-based metal-organic framework (MIL-88B-NH 2 ) nanozymes. The characteristic glycan of CA19-9 (sialyl Lewis A, SLe a ) is pre-adsorbed onto the amino-rich MIL-88B-NH 2 surface via hydrogen bonding and boronic acid affinity, followed by sol-gel polymerization to grow an imprinted shell. After template removal, the recognition cavities are automatically positioned within nanometers of the intrinsic Fe catalytic sites. Upon specific binding of CA19-9, steric hindrance exerted by the protein backbone impedes diffusion of the chromogenic substrate 3,3′,5,5′-tetramethylbenzidine (TMB) to the surface of MOF, resulting in quantifiable suppression of color development. The developed sensor demonstrates sensitive detection of CA19-9 across a wide linear range from 0.001 to 50 U mL −1 , with a detection limit of 1.01 × 10 −4 U mL −1 (3σ/slope), and shows excellent selectivity in serum samples. This work provides a stable, cost-effective, and visual biosensing platform with significant potential for point-of-care biomarker testing. Graphical

Carbohydrate antigen 19-9 (CA19-9) is a key biomarker for early diagnosis of various malignancies, including lung cancer. Herein, a ZnPc/Ti 3 C 2 heterojunction photosensitive material was synthesized via a novel one-pot strategy and utilized for the first time to construct a photoelectrochemical sandwich immunosensor. Taking advantage of the reducing property of Ti 3 C 2 , gold nanoparticles (AuNPs) were in-situ generated on ZnPc/Ti 3 C 2 to form ZnPc/Ti 3 C 2 /AuNPs nanocomposite, which was then immobilized onto a fluorine-doped tin oxide (FTO) electrode. The AuNPs provided a large surface area and biocompatible interface for efficient loading of capture antibody (Ab1). Upon target CA19-9 recognition, the polydopamine-labeled detection antibody (PDA-Ab2) was introduced, triggering a dual photocurrent quenching mechanism through which PDA acted as an electron trap, capturing photoelectrons from ZnPc/Ti 3 C 2 /AuNPs, while the bulky immunocomplex competed with the electrode for light absorption, reducing irradiation of the photosensitive material. This synergistic quenching enabled highly sensitive CA19-9 detection with a linear range of 0.001–200 U/mL and a detection limit of 0.53 mU/mL. The proposed sensor was successfully applied to CA19-9 determination in serum samples with a relative error within 10% compared with hospital reference values, demonstrating its potential for clinical diagnostics.

This work took advantage of 3D biomimetic hydrangea-like bismuth oxychloride (BiOCl) and PtNi nanocubes to fabricate a facile electrochemical immunosensor for the detection of CA19–9. Herein, APTES functionalized BiOCl (APTES-BiOCl) with large specific surface areas can provide eco-friendly microenvironment for biomolecules. The PtNi nanocubes prepared by the classic hydrothermal method have good conductivity, which can facilitate electron transfer highly efficient. Meanwhile, PtNi nanocubes can immobilize the anti-CA19–9 via covalent Pt-N bonds to anchor a great number of biomolecules more firmly. In addition, reduced graphene oxide (rGO) with excellent conductivity was implemented as a matrix, which greatly promoted electron transfer at the sensing interface. Under the optimal conditions, the proposed immunosensor can detect CA19–9 sensitively, from which a linear range of 0.01–150 U·ml–1 was derived accompanied with a low detection limit of 0.0034 U·ml–1. The results illustrated that the designed immunosensor exhibited excellent specificity, good reproducibility, and satisfactory stability. Moreover, it can be applied in the detection of real samples, which indicated that the immunosensor has great potential for early screening and diagnosis of diseases.

Porous carbon sphere materials have a large variety of applications in several fields due to the large surface area, adaptable porosity, and good conductivity they possess. Obtaining a steady carbon sphere using the green synthesis method remains a significant challenge. In this experiment, covalent organic frameworks (COFs) were used as a precursor and Fe 3 O 4 NPs were integrated into the precursor in order to synthesize a porous carbon sphere material using the one-step pyrolysis method. COFs have an ordered porous structure, perpetual porosity, large surface area, and low density and display good environmental tolerance. These properties make them an excellent precursor for synthesizing porous carbon sphere, which maintains good morphology at high temperatures, and it is not involved in the removal of dangerous reagent and small size restrictions during the synthesis process. In addition to the formation of a porous carbon sphere, transition metal carbon material that contains N element can be an active catalyst. The composites exhibit better activity when Fe is doped into carbon materials containing N element than that of other doped transition metals including Mn and Co. In this situation, the integration of Fe 3 O 4 NPs and N element in the COF precursor exposed the active sites of the composites and the two substances synergistically improved the electrocatalytic properties, and the composites were named Fe 3 O 4 @NPCS. The constructed Fe 3 O 4 @NPCS/GCE immunosensor was applied as a means of detecting CA19-9 antigen and presented a wide linear range from 0.00001 to 10 U/mL with a low detection limit of 2.429 μU/mL (S/N = 3). In addition, the prepared immunosensor was utilized for detecting CA19-9 antigen in the real human serum, and the recovery rates were in the range from 95.24% to 106.38%. Therefore, a porous carbon sphere prepared by COFs as a precursor can be applied for the detection of CA19-9 antigen in real samples, which could be an excellent strategy for CA19-9 antigen detection and could potentially promote the development of COF materials in various electrochemical fields.

The detection of carbohydrate antigen 19 − 9 (CA19-9), a critical serum marker for gastrointestinal malignancies, is particularly challenging because of its typically low concentration and substantial matrix interference, creating a critical demand for improved detection technologies. Herein, we rationally designed and synthesized a nanozyme with dual-ligand cerium-organic framework (Ce-MOF-BN) nanozyme for glycosyl-imprinted colorimetric biosensing via a one-pot method. The crystalline porous framework is densely functionalized on its surface with spatially accessible amino and boronic acid groups, enabling the recognition and oriented immobilization of imprinting templates. A well-defined core–shell molecularly imprinted composite was successfully fabricated through the systematic optimization of the ligand ratio. The composite exhibits intrinsic oxidase-like activity without requiring H 2 O 2 , and the molecularly imprinted polymer (MIP) shell supplies specific cavities for the highly selective recognition of CA19-9. Under optimized assay conditions, the sensor achieves the quantitative detection of CA19-9 across a broad linear range of 0.0001–100 U/mL, with a limit of detection as low as 2.7 × 10 − 5 U/mL. This work provides valuable insights for designing next-generation high performance biomimetic sensing materials and advancing their clinical diagnostic applications. In this work, label-free electrochemical immunosensing of the cancer biomarker carbohydrate antigen 19-9 (CA19-9) is reported using [Fe(CN)6]3−/4− as a signal probe and a copper organic framework (Cu-BTC) nanocomposite for the amplification of the signal. The immunosensor was fabricated by the following process. First, the Cu-BTC nanomaterial with a larger surface area and good biocompatibility was synthesized to improve the dispersion of gold nanoparticles (Au NPs). Then, nitrogen-doped graphene (N-GR) was combined with Cu-BTC to form the nanocomposite. The synthesized Cu-BTC@N-GR@AuNPs@CS nanocomposite was employed to modify the surface of the immunosensor to accelerate the electron transfer rate and improve the immobilization amount of CA19-9 antibodies (Ab). Various techniques, including TEM, SEM and XPS were used to characterize Cu-BTC and nanocomposites. Differential pulse voltammetry (DPV) was used to measure the electrochemical response of the immunosensor in [Fe(CN)6]3−/4−. The signal intensity of the immunosensor was linearly changed upon increasing the concentration of CA19-9 antigen from 10 μU mL−1 to 100 U mL−1, and a detection limit of 4.2 μU mL−1 was achieved. Furthermore, the immunosensor showed good stability, reproducibility and specificity, indicating its potential application in clinical analysis.
