Metabolomics analysis of MnO2 nanosheets CDT for breast cancer cells and mechanism of cytotoxic action

Jian Liu,Changchun Wen,Miaomiao Hu,Juan Long,Jing Zhang,Minzhe Li,Xiang-Cheng Lin

Chemodynamic therapy (CDT) has received more and more attention as an emerging therapeutic strategy, especially transition metals with Fenton or Fenton-like activity have good effects in CDT research, manganese dioxide nanosheets (MnO2 NSs) and their complexes have become one of the most favored nanomaterials in CDT of tumors. CDT is mainly based on the role of reactive oxygen species (ROS) in tumor treatment, which have clear chemical properties and produce clear chemical reactions. However, their mechanism of interaction with cells has not been fully elucidated. Here, we performed CDT on mouse breast cancer cells (4T1) based on MnO2 NSs, extracted the metabolites from the 4T1 cells during the treatment, and analyzed the differences in metabolites by using high-resolution liquid chromatography-mass spectrometry (LC-MS). Untargeted metabolomics studies were conducted using the relevant data. This study mainly explored the changes in MnO2 NSs on the metabolite profile of 4T1 cells and their potential impact on tumor therapy, in order to determine the mechanism of action of MnO2 NSs in the treatment of breast cancer. The results of the study showed the presence of 11 different metabolites in MnO2 NSs CDT for 4T1 tumor cells, including phosphoserine, sphingine, phosphocholine, and stearoylcarnitine. These findings provide a deeper understanding of breast cancer treatment, and are beneficial for the further research and clinical application of CDT.

WO3-x@Ferrocene-Folic Acid Composites Induce Cancer Cell Death and Activate Immunity via PTT/CDT

Qi Wu,Mingyang Liu,Hui Zhang,Guanghao Li,Ziqing Yang,Xiaodan Wu,Guanghui Tan,Chenfeng Ji,Yingxue Jin

At present, tumor immune escape is a major problem in the treatment of tumors. The complex network of tumor microenvironments significantly impairs the efficacy of immunotherapy. This paper reports the preparation and immunoantitumor activity of a novel multifunctional defect tungsten trioxide@ferrocene-folic acid composite (WO 3-x @Fe-FA) with a high Fenton reaction rate. Ferrocene is modified on the surface of defective trioxide by the covalent coupling method for the first time, and the reaction rate of Fenton is increased by 10 times. WO 3-x @Fe-FA induces immunogenic cell death (ICD) through the powerful synergistic anti-tumor effect of PTT/CDT and decomposes H 2 O 2 to produce oxygen through the Fenton reaction, thus down-regulating the expression of immune checkpoint PD-L1 induced by tumor hypoxia. In vitro and in vivo experiments proved that WO 3-x @Fe-FA reverses the immunosuppressive tumor microenvironment, transforms the immunosuppressive “cold tumor” into the immune “hot tumor”, and activates the immune activity of the system. In vitro and in vivo experiments show that WO 3-x @Fe-FA has excellent immunoantitumor activity, and it is expected to be a candidate drug for immunoantitumor therapy.

GSH responsive AuNRs@TFF nanotheranostic for NIR-II photoacoustic imaging-guided CDT/PTT synergistic cancer therapy

Nan Leng,Yuzhu Zhou,Changchun Wen,Qi Fang,Xiaohuan Guo,Bingbing Cai,Ke-Bin Huang,Hong Liang

Gold nanorods (AuNRs) are important photothermal therapeutic agents; however, a single therapy does not achieve satisfactory outcomes, and the synthesis process often leads to the adsorption of cetyltrimethylammonium bromide on the surface of AuNRs, which reduces its biocompatibility. Natural polyphenols are abundant in natural plants and have good biocompatibility. The metal-polyphenol network is formed by the coordination of metal ions and polyphenols, which has good drug loading, surface adhesion, and biocompatibility. In this study, the metal-polyphenol network structure formed by a transition metal (iron) and natural polyphenol tannic acid was used to modify the surface of gold nanorods (AuNRs@TF). Additionally, the surfaces of AuNRs were modified using the targeted functional molecule mercapto folic acid (AuNRs@TFF). The constructed composite nanomaterials AuNRs@TFF has good biocompatibility and tumor targeting ability. Tannic acid‑iron degrades in the tumor microenvironment and releases iron ions that catalyze the Fenton reaction, thereby facilitating chemodynamic therapy. The good photo-thermal ability of AuNRs generate good photoacoustic signals to facilitate photoacoustic imaging mediation and enhances photothermal and chemodynamic therapy performance. This study expands on the application of AuNRs in the field of nanomedicine. The simple and effective design of AuNRs@TFF provides a strategy for the development of synergistic therapeutic agents for photothermal therapy and chemodynamic therapy.

Tumor microenvironment responsive mesoporous silica nanoparticles for dual delivery of doxorubicin and chemodynamic therapy (CDT) agent

Yuan Zhang,Omer Eltayeb,Yating Meng,Guomei Zhang,Yan Zhang,Shaomin Shuang,Chuan Dong

Chemodynamic therapy is a new antitumor strategy triggered by transition metal as chemodynamic agent via the in situ catalytic Fenton reaction. Herein, we engineer a smart nanocarrier activated by tumor microenvironment-specific weak acidic pH and high glutathione conditions for chemo/chemodynamic processes. This unique architecture orderly consists of mesosilica-encapsulated doxorubicin, self-polymerization of polydopamine Fe3+ clathrate, further bonding the targeting polymer MnO2 disguised albumin–folic acid. Polydopamine and albumin as intelligent nanovalves shield doxorubicin from premature leaking. Folic acid improves the targeting capability for folate receptor (FR) overexpressing cancer cells. After intake of the nanocarrier by tumor cells, the nanocarrier nanovalves are first biodegraded by native glutathione and pH, enabling on-demand anticancer drug escape, and also generate and release Fe3+ and Mn2+ (reduced by MnO2via glutathione depletion). Subsequently, the ions interact with H2O2, generating highly toxic hydroxyl radicals (˙OH) via a Fenton-like reaction to enhance the anticancer cell efficacy. This result proves that the designed nanocarrier more likely devours into FR-positive cancer cells and illustrates a high intracellular ˙OH generation, leading to the desired specific killing efficacy. We believe this innovative strategy will exhibit a new avenue for the potential of accurate chemo/chemodynamic co-therapy in cancer cells.

NIR-II-responsive AuNRs@SiO2–RB@MnO2 nanotheranostic for multimodal imaging-guided CDT/PTT synergistic cancer therapy

Changchun Wen,Xiaolu Guo,Cunji Gao,Zhongkai Zhu,Nianqi Meng,Xing-Can Shen,Hong Liang

Specific tumor-responsive capabilities and efficient synergistic therapeutic performance are the keys to effective tumor treatment. Herein, AuNRs@SiO2–RB@MnO2 was developed as a new type of tumor-responsive nanotheranostic for multimodal imaging and synergistic chemodynamic/photothermal therapy. In AuNRs@SiO2–RB@MnO2, the SiO2 layer wraps the AuNRs, providing light absorption in the second near-infrared (NIR-II) region. The SiO2 layer also adsorbs the MnO2 nanosheets, which have Fenton-like activity, resulting in a fluorescent sensing platform based on the fluorescence quenching properties of MnO2 for rhodamine B dye. The fluorescence can be recovered by the consumption of MnO2 by glutathione, which simultaneously produces Mn2+ in the tumor region. The recovery of fluorescence reflects the consumption of glutathione and the increase in Mn2+, which produces hydroxyl radicals via Fenton-like reaction in the tumor microenvironment to realize chemodynamic therapy. Meanwhile, the AuNRs are a good photothermal reagent that can effectively absorb NIR-II light and convert it into heat energy to kill tumor cells via photothermal therapy. The NIR-II absorption performance of the AuNRs provides good photoacoustic imaging and deep photothermal performance, which is favorable for efficient NIR-II photoacoustic imaging-guided photothermal therapy. As a result, the AuNRs@SiO2–RB@MnO2 nanotheranostic exhibits outstanding imaging and synergistic chemodynamic/photothermal therapeutic performance for tumor imaging and treatment.

The PTT, SDT, and CDT synergistic antitumor effect of Fe3+-coordinated PDA@PLGA-Cur nanoparticles in vitro

Fanfan Zhang,Xuemei Hu,Yue Zhang,Baiyuan Sha,Fengshi Shang,Wenjie Wang,Rui Liu,Hao Wang

Hepatocellular carcinoma (HCC) poses a severe threat to public health, with clinical therapy limited by narrow surgical indications, chemotherapeutic side effects, and targeted drug resistance. Herein, we developed Fe 3+ coordination Polydopamine@Poly (lactic-co-glycolic acid)-Curcumin (Fe 3+ -PDA@PLGA-Cur, FePPC) nanoparticles integrating photothermal therapy (PTT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), and curcumin (Cur)'s intrinsic anticancer activity for synergistic HCC treatment. FePPC was fabricated via emulsion solvent evaporation, in-situ dopamine polymerization, and Fe 3+ coordination chelation, with its structure and physicochemical properties characterized by FT-IR, UV-vis, SEM, XPS, and particle size analysis. FePPC exhibited uniform spherical morphology (198.58 nm), favorable stability, 73.60 ± 0.51% encapsulation efficiency, and NIR-triggered temperature-dependent Cur release. Under 808 nm NIR light (2 W/cm 2 , 4 cm), FePPC achieved a 24.4 °C temperature elevation with robust photothermal stability. Catalytic assays confirmed FePPC generated ·OH via Fe 3+ -mediated Fenton-like reactions, 1 O 2 under NIR irradiation, and ·O₂⁻ upon ultrasound stimulation, realizing synergistic reactive oxygen species (ROS) production. In vitro experiments on HepG2 cells showed time-dependent FePPC internalization; the FePPC + PTT + CDT + SDT group reduced cell viability to 53.45%, induced 47.55% apoptosis, elevated intracellular ROS, disrupted mitochondrial membrane potential, downregulated LDH, modulated Bax/Bcl-2 expression, and inhibited cell migration. This study highlights FePPC's potential as a multifunctional nanoplatform for HCC therapy via coordinated chemical-biological interactions.

Cell membrane camouflaged Cu-doped mesoporous polydopamine for combined CT/PTT/CDT synergistic treatment of breast cancer

Di Meng,Shuoye Yang,Lin Ju,Jinpeng Wang,Yanan Yang,Lu Zhang,Lan Cui

Currently, traditional monotherapy for cancer often results in indiscriminate attacks on the body, leading to the emergence of new health problems. To confront these challenges, multimodal combination therapy has become necessary. However, how to develop new smart nanomaterials through green synthesis methods, delivering drugs while simultaneously synergizing multimodal combination therapies for tumor treatment, remains a topic of great significance. In this study, a biomimetic composite nanomaterial (RM-Cu/P) composed of mesoporous polydopamine (MPDA) as the core and red blood cell membranes (RBCMs) as the shell was synthesized as a drug carrier to deliver doxorubicin (DOX) while achieving synergistic chemotherapy, photothermal and chemodynamic therapy (CT/PTT/CDT). Herein, the nanoparticles were extensively characterized to examine their morphological characteristics, elemental composition, and drug-carrying capacity. Notably, the coating of RBCM reduced the toxicity of the RM-Cu/P@DOX nanoparticles, improved their targeting ability and prolonged their circulation time in vivo . The Cu-doped nanoparticles were capable of initiating a Fenton-like reaction to generate reactive oxygen species (ROS) for CDT, while the photothermal conversion efficiency (η) reached 45.20 % under NIR laser irradiation. Subsequently, the particles were examined by in vivo and in vitro experimental studies in cytotoxicity, cellular uptake, ROS levels, lysosomal escape, and mouse tumor model to evaluate their potential application in antitumor. Compared with monotherapy, the RM-Cu/P@DOX nanoparticles had multiple-stimulation response properties under redox, pH, and NIR, which exhibited the advantage of combined trimodal therapy, resulting in remarkable synergistic antitumor efficacy. In conclusion, this innovative platform exhibited promising applications in smart drug delivery and synergistic treatment of cancer.

NIR-activatable proenzyme DSF/CuS@M for synergistic in situ chemotherapy/CDT/PTT/cuproptosis-based antitumor therapy

Lushuang Xu,Qinyue Chen,Yangwei Qian,Jin Wang,Chenxi Wang,Rui Fan,Jia-Wei Shen,Qiying Shen

Nanozymes provide therapeutic benefits against a diverse range of malignant tumors. However, their antitumor potencies are seriously impaired by their low catalytic efficiencies and the off-target toxicities caused by their “always-on” catalytic activity. Herein, we developed multifunctional nanoparticles co-loaded with ultra-small copper sulfide nanoparticles (CuS NPs) and disulfiram (DSF) (DSF/CuS@M). These nanoparticles had peroxidase-like activity and served as (near-infrared) NIR-activatable proenzymes to achieve synergistic in situ chemotherapy, chemodynamic therapy (CDT), photothermal therapy (PTT), and cuproptosis-based antitumor therapy. DSF/CuS@M not only provided PTT as NIR-activatable proenzymes but also enabled complete DSF and Cu 2+ release. Non-toxic DSF was converted into diethyldithiocarbamic acid (DDC) through a glutathione (GSH)-mediated metabolic reaction, leading to the generation of cytotoxic Cu(DDC) 2 in situ , which enhanced cell apoptosis. Cu 2+ was converted into Cu + by GSH, and this Cu + catalyzed the conversion of H 2 O 2 into hydroxyl radicals (·OH) to induce CDT. Moreover, Cu + could also bind to dihydrolipoyl transacetylase (DLAT) in the tricarboxylic acid (TCA) cycle, thereby inducing cuproptosis. These results demonstrate that the NIR-activatable proenzyme developed in this study is effective and safe for synergistic therapy against malignant tumors.

Ruthenium-gallic acid nanozymes encapsulated in 3D bioprintable photosensitive hydrogels for synergistic PTT/CDT therapy of glioblastoma

Jianyu Nie,Zhongyong Wang,Haotian Tian,Yang Qiao,Menghui Liu,Xianwen Wang,Liang Xu,Xingliang Dai

Confronted with the clinical challenges of glioma treatment, including incomplete resection and therapeutic resistance, we developed an integrative bioprinting strategy. At the core of our approach is a 3D-bioprintable, photosensitive hydrogel loaded with ruthenium-gallic acid (Ru-GA) nanoenzymes. This system is designed to seamlessly combine chemodynamic therapy (CDT) with photothermal therapy (PTT) to achieve precise and synergistic treatment of glioblastoma multiforme (GBM). Ru-GA, synthesized via Ru³⁺-gallic acid coordination, has potent peroxidase-like activity to catalyze reactive oxygen species generation from H₂O₂ in the tumor microenvironment. It also efficiently depletes glutathione to disrupt tumor redox homeostasis, and exhibits excellent near-infrared absorption and photothermal conversion efficiency. Based on GelMA and HAMA, the hydrogel is liquid at 37 °C and has the ability to be injected into tumor tissue, ensuring localized retention and sustained action of Ru-GA at tumor sites. In vitro tests verified Ru-GA kills GL261 glioma cells via CDT, and combining it with PTT further inhibits tumor cell proliferation, migration, and clonogenicity while boosting apoptosis. In 3D bioprinted tumor models, the hydrogel suppresses tumor cell aggregation and viability effectively. In vivo experiments showed intratumoral hydrogel injection plus near-infrared irradiation raises tumor temperature to 55 °C, exerting synergistic PTT/CDT effects to significantly inhibit tumor growth with excellent biosafety. Mechanistically, this therapy activates inflammatory pathways and inhibits metastasis-related signaling, offering a safe, efficient GBM treatment and laying a foundation for clinical translation of nanocatalyst-based photosensitive hydrogels.

Photothermal and Acid-Responsive Fucoidan-CuS Bubble Pump Microneedles for Combined CDT/PTT/CT Treatment of Melanoma

Jiaojiao Tao,Bingjie Wang,Yu Dong,XiangYan Chen,Shuang Li,Tianze Jiang,Xia Zhao

Transdermal cancer therapy faces great challenges in clinical practice due to the low drug transdermal efficiency and the unsatisfactory effect of monotherapy. Herein, we develop a novel bubble pump microneedle system (BPMN-CuS/DOX) by embedding sodium bicarbonate (NaHCO3) into hyaluronic acid microneedles (MNs) loaded with fucoidan-based copper sulfide nanoparticles (Fuc-CuS NPs) and doxorubicin (DOX). BPMN-CuS/DOX can generate CO2 bubbles triggered by an acidic tumor microenvironment for deep and rapid intradermal drug delivery. Fuc-CuS NPs exhibit excellent photothermal effect and Fenton-like catalytic activity, producing more reactive oxygen species (ROS) by photothermal therapy (PTT) and chemodynamic therapy (CDT), which enhances the antitumor efficacy of DOX and reduces the dosage of its chemotherapy (CT). Simultaneously, DOX increases intracellular hydrogen peroxide (H2O2) supplementation and promotes the sustained production of ROS. BPMN-CuS/DOX significantly inhibits melanoma both in vitro and in vivo by the combination of CDT, PTT, and CT. In short, our study significantly enhances the effectiveness of transdermal drug delivery by constructing BPMNs and provides a promising novel strategy for transdermal cancer treatment with multiple therapies.

Thermosensitive Multifunctional Hydrogel for Boosting Drug-Resistant Infected Diabetic Wound Healing by Integrating Gas Therapy and Cascaded CDT

Liyu Shi,Xian Li,Yunfei Mo,Xiying Chen,Zhanhui Tao,Yaqing Liu

The treatment of diabetic wounds remains unsatisfactory due to bacterial infection risks and impaired angiogenesis during the healing. Herein, a multifunctional hydrogel was constructed for boosting multidrug-resistant bacteria-infected diabetic wound healing by integrating gas therapy and cascaded chemical dynamic therapy (CDT). The hydrogel was based on Pluronic F127 and hyaluronic acid (HA), which was loaded with an iron-based metal organic framework (MIL-100(Fe)) coupled with S-nitrosoglutathione (GSNO) and glucose oxidase (GOX) (MIL-100(Fe)@GOX@GSNO@F127-HA, shortly named as MGG@FH). In the hyperglycemic diabetic microenvironment, GOX is activated to reduce local glucose levels and pH while generating hydrogen peroxide (H2O2). MIL-100(Fe) with peroxidase-like activity catalyzes H2O2 to produce reactive oxygen species (ROS), exerting antibacterial effects. GSNO spontaneously releases nitric oxide (NO) to modulate inflammation and promote angiogenesis. The MGG@FH hydrogel downregulates pro-inflammatory factors such as tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) while upregulating the anti-inflammatory factor IL-10. It also enhances angiogenesis and collagen deposition in vivo, significantly accelerating the healing of infected diabetic wounds. Meanwhile, the temperature-sensitive sol–gel transition property of the hydrogel makes it possible for in situ forming gels on the wound site, allowing it to conform to irregular shapes and well fit the depth of the wound. The strategy combines “endogenous improvement” (reducing glucose and pH) with “exogenous resistance” (antibacterial/anti-inflammatory effects), offering an effective approach for comprehensive infected diabetic wound management.

Cu-S-Mo interfacial sites in CuO2@MoS2/PLLA scaffolds for robust synergistic antibacterial efficacy via CDT and PTT

Cijun Shuai,Gao Pan,Qi Zhong,Shuping Peng

Bacterial infections considerably limit the success of scaffold transplantation. CuO 2 may generate hydroxyl radicals through a self-cascade catalytic process, which is effective for sterilization. However, reducing CuO 2 -derived Cu + ions to Cu° can lead to a decline in catalytic activity. To overcome these limitations, we anchored CuO 2 onto MoS 2 to establish stable Cu-S-Mo interfacial sites. The CuO 2 /MoS 2 nanozyme was fabricated using a low-temperature co-precipitation technique and introduced into poly- l -lactic acid scaffolds using selective laser sintering. The Cu-S-Mo interfacial sites showed a triad of functions that synergistically enhanced catalytic performance. These sites stabilized the Cu atoms, augmented catalytic activity by facilitating efficient electron transfer through the Mo 4+ /Mo 6+ redox couple, and improved the conduction of heat derived from the photothermal response of MoS 2 to the Cu catalyst. The synergistic action of chemodynamic and photothermal therapy, mediated by the Cu-S-Mo sites, resulted in remarkable antibacterial efficacy, with 92.3 % and 93.5 % inhibition rates against Staphylococcus aureus and Escherichia coli , respectively. Experimental evidence and simulations demonstrated that Cu-S-Mo structure stabilized the Cu atoms and improved electron transfer via interatomic charge transfer. These results highlight the potential of the CuO 2 @MoS 2 /PLLA scaffold in eliminating bacterial infections.
