Spatiotemporal Coding Terahertz Plasmonic Vortex on-Chip Interferometer

Yiming Wang,Huijun Zhao,Chengwei Song,Yunyun Ji,Jierong Cheng,Xianghui Wang,Fei Fan,Shengjiang Chang

Plasmonic vortices (PV) stimulated from metallic micro-nano structures hold great promise for improving on-chip data capacity. However, the manipulation freedom degree and reconfigurability of current PV devices still need further development in the terahertz regime, especially the simultaneous excitation of two or more PV modes and their coherent synthesis. In this letter, a terahertz PV interferometer with spatiotemporal coding based on liquid crystal integration is demonstrated, which can dynamically manipulate the transient evolution of spin-orbital angular momentum in the time domain, and the superposition field distribution in the frequency domain. By introducing sufficient surface wave path difference, different modes can be modulated independently within the separated time window based on the liquid crystal encodings. This space-time correlation coding enables to dynamically control the mode weightings, and finally obtains 16 superposition distributions. This reconfigurable on-chip interferometer and its dynamic spatiotemporal regulation mechanism pioneer an avenue for intelligent perception and photon computing.

Plasmon-exciton coupling for nanophotonic sensing on chip

Jun Dong,Yi Cao,Qingyan Han,Yongkai Wang,Minghan Qi,Wenwen Zhang,Lin Qiao,Jianxia Qi,Wei Gao

The monolayer graphene-noble metallic nanostructure hybrid system with excellent optical characteristic, which is deserved pay attentions in the study of surface-enhanced Raman scattering spectroscopy. In this work, a hybrid sandwich structure is designed to transfer single-layer graphene to the surface of discs substrate covered by silver film and assembly of the dense Au nanoparticles (AuNPs). Blu-ray disc has a cycle density of approximately 5.7 times that of DVD-R due to the different storage capacities of these optical discs. In the research, enhancement effects have been explored for two different periodic grating structures. Compared to spectra of Si/G structure, Graphene Raman spectra from Blu-grating/AuNPs/G structure and Blu-grating/G/AuNPs enhancement multiples at the 2D peak position possesses different Raman responses of 1.09 and 2.51 times, respectively. The sandwich hybrid structure of Ag grating/graphene/AuNPs obtains a Raman enhancement factor (EF) of 6.2×108 for Rhodamine 6G and surface-enhanced Raman Scattering(SERS) detection limit of 0.1 nM. These findings can be attributed to the electric field enhancement of the hybrid structure and the chemical enhancement of graphene. This study provides a new approach for SERS detection and offers a new technique for designing SERS sensors with grapheme-plasmon hybrid structures.

Study of Monolithic Integrated Micro Gas Chromatography Chip

Bo-Wen TIAN,Fei FENG,Bin ZHAO,Fan LUO,Xue-Lei YANG,Hai-Mei ZHOU,Xin-Xin LI

A monolithic micro gas chromatography (μGC) chip which integrated the micro separation column (μSC) and the micro thermal conductivity detector (μTCD) based on MEMS (Micro-electro-mechanical systems) technique was fabricated. Compared to the state of the art, the μSC with high depth-to-width ratio channels that was coated with mesoporous silica nanoparticles as stationary phase could effectively improve the column capacity and separation performance. Besides, the stable suspending μTCD, which was designed and fabricated in two ports of the μGC chip, could availably enhance the thermal isolation and reliability of the device. The mixture of light hydrocarbons (methane, ethane, propane and butane) could be separated from each other and detected by this monolithic integrated μGC chip, in which the overall analysis and detection time was only 33 seconds, the separation resolution of ethane and propane was 8.34, and the number of theoretical plate was as high as 11420. The monolithic integrated μGC chip has many advantages such as good separation resolution, high column efficiency and short analysis time, and is suitable for portable gas chromatographic field and onsite detection.

Screen-printed microfluidic dielectrophoresis chip for cell separation

Hongwu Zhu,Xiaoguang Lin,Yong Su,Hua Dong,Jianhua Wu

Dielectrophoresis (DEP), the induced motion of polarizable particles in a non-uniform electric field, has been proven as a perfect candidate to transport, accumulate, separate and characterize micro-/nano-scale bioparticles in microfluidic systems. However, conventional fabrication technologies are complex, time-consuming and relatively expensive, leading to low throughput of the DEP-based systems. In this paper, we report a novel microfluidic alternating current DEP (AC-DEP) chip fabricated via inexpensive screen printing method. The innovation of our work consists in the extreme simplicity of the fabrication procedure, i.e., the main components, including electrodes and channels, were constructed by layer-by-layer screen printing process , which is especially suitable for high-throughput mass production. Carbon paste, instead of metals, was used to print interdigitated electrodes with semi-3D structure which not only reduces dramatically the chip cost but also increases particle trapping efficiency. To test the chip performance, yeast cells, as model cells, were trapped and separated from a mixed suspension with PS microspheres . Our results show that high capture rate and separation efficiency can be achieved under optimized conditions.

On-Chip Sonoporation-Based Flow Cytometric Magnetic Labeling

Qianwei Zhu,Weiping Ding,Shibo Li,Fenfen Li,Yi Hu,Shengnan Ya,Tianzhi Luo,Dayong Gao,Bensheng Qiu

Tracing magnetically labeled cells with magnetic resonance imaging (MRI) is an emerging and promising approach to uncover in vivo behaviors of cells in cell therapy. Today, existing methods for the magnetic labeling of cells are cumbersome and time-consuming, which has greatly limited the progress of such studies on cell therapy. Thus, in this study, using the flow cytometric loading technology, we develop a sonoporation-based microfluidic chip (i.e., a microfluidic chip integrated with ultrasound; MCU), to achieve the safe, instant, convenient, and continuous magnetic labeling of cells. For the MCU we designed, a suitable group of operating conditions for safely and efficiently loading superparamagnetic iron oxide (SPIO) nanoparticles into DC2.4 cells was identified experimentally. Under the identified operating conditions, the DC2.4 cells could be labeled in approximately 2 min with high viability (94%) and a high labeling quantity of SPIO nanoparticles (19 pg of iron per cell). In addition, the proliferative functions of the cells were also well maintained after labeling. Furthermore, the in vivo imaging ability of the DC2.4 cells labeled using the MCU was verified by injecting the labeled cells into the leg muscle of the C57BL/6 mice. The results show that the excellent imaging outcome can be continuously achieved for 7 days at a density of 106 cells/mL. This work can provide insight for the design of magnetic cell labeling devices and promote the MRI-based study of cell therapies.

A hypoxic microfluidic organoid-on-a-chip system for studying the efficacy of metronidazole-modified nanomaterials against cholangiocarcinoma established within the chip

ao xie,Zipeng Yao,Qijun Du,Mengjiao Xia,Qinrui Lu,Jiashu Wang,Wenqi Hu,Lin Wu,Chenwei Sun,Youlong Yang,Di Wu,Haijie Hu,Guohua Wu,Shuqi Wang

Cholangiocarcinoma (CCA) is a highly aggressive biliary malignancy characterized by a dismal prognosis. Tumor progression relies heavily on the hypoxic tumor microenvironment (TME), a key factor that promotes drug resistance and reduces therapeutic efficacy. A major barrier to clinical translation is that standard in vitro cultures fail to maintain this stable low-oxygen state. Addressing this limitation, we designed a microfluidic platform incorporating patient-derived CCA organoids (CCOs) to act as a high fidelity tumor model. Moreover, we synthesized a hypoxia-activatable nanodrug, BM-MN@PDA (BMMNP). This agent consists of a Metronidazole (MN)-loaded Bismuth-TCPP framework shielded by a biocompatible polydopamine (PDA) coating. In hypoxic environment, the drug generates cytotoxic radicals. Validation studies confirmed that our microfluidic platform successfully established a highly biomimetic hypoxic TME (O2 < 2.5%), evidenced by the robust upregulation of HIF-1α and HIF-2α. Furthermore, BMMNP NPs demonstrated superior cellular uptake and potent cytotoxicity within the hypoxic CCOs. Notably, these nanoparticles effectively reversed hypoxia-induced resistance to gemcitabine, acting as a powerful chemo-sensitizer. Collectively, this platform not only establishes a physiologically relevant "hypoxia-on-a-chip" model for preclinical drug evaluation but also accelerates the optimization of nanoparticle-based strategies for hypoxia-targeted therapy.

TFAP2C-mediated transcriptional activation of STEAP3 promotes lung squamous cell carcinoma progression by regulating the β-catenin pathway

Sun Tong,Yang Zhiguang

Six-transmembrane epithelial antigen of prostate 3 (STEAP3) is associated with the progression of several human malignancies. However, its role in lung squamous cell carcinoma (LUSC) remains unclear. We measured STEAP3 expression in LUSC cell lines and tissues. LUSC cells with stable STEAP3 overexpression and knockdown were obtained through G418 selection. Multiple assays were used to evaluate the malignant phenotypes of LUSC cells and the activation of the β-catenin signaling. The potential transcriptional regulatory factors of STEAP3 were predicted using the JASPAR database, and the correlation between transcription factor AP-2 gamma (TFAP2C) and STEAP3 was analyzed through the GEPIA database. The study evaluated the regulatory relationship between a potential transcription factor and STEAP3 through ChIP and luciferase reporter assays. Additionally, rescue assays were utilized to ascertain whether TFAP2C serves as the upstream regulatory factor of STEAP3, contributing to LUSC progression. Finally, tumor growth and metastasis were evaluated in vivo. STEAP3 expression was notably higher in LUSC, and its overexpression was linked to a poor prognosis. Moreover, STEAP3 overexpression activated the β-catenin pathway, thereby accelerating cell proliferation and metastasis. Conversely, STEAP3 knockdown had an anti-tumor effect in LUSC. Additionally, TFAP2C bound directly to the STEAP3 promoter and positively regulate its expression in LUSC. The anti-tumor effects of TFAP2C knockdown were partially reversed by STEAP3 overexpression. The study indicates that the TFAP2C/STEAP3 axis may be a therapeutic target for LUSC treatment. This enhances our understanding of lung carcinogenesis.

Electronic Chip-Mimetic Medical Implants for Controllable Sonothermal Treatments

Zhengdong Zhang,Pan Liu,Jie Lei,Shuchang Peng,Qiu Chen,Wei Guan,Yizhou Zhu,Gang Liu,Xiaobo Feng,Cao Yang,Lei Tan

The sterility and controllable biological functionalities of implantable medical devices determine their lifespan, safety, and therapeutic efficacy. Inspired by the thermal effect of working electronic chips, which consist of various heterostructures, we have developed an electronic chip-mimetic sonothermal platform through constructing metal-semiconductor heterogeneous interface. Through magnetron sputtering on the surface of the sandblasted/acid-etched pretreated titanium (pTi), we find that semiconductor coatings (TiO 2 , Si, ZnO, and Te) endow pTi with different in situ sonothermal effects (ΔT > 18°C, 15 min) under ultrasound (US) irradiation, whereas conductor coatings do not. The sonothermal mechanism of pTi-semiconductor is associated with US-activated electron and phonon transport within the heterogeneous interface of implants, which is determined by the electrical and phonon characteristics of pTi-semiconductor, including their matching degree, thermal conductivity, defected structure, and type of semiconductor. Clinical titanium screws were introduced with defected structure in oxygen layer (TiO 2-x ) and bone-derived whitlockite, which shows great sonothermal/sonodynamic effects for efficient elimination of biofilm infection and improved osseointegration. In addition, the prepared NiTi-TiO 2-x guidewire enables rapid thrombolysis (30 min) in the deep vein thrombosis of beagles after 10 min of sonothermal/urokinase treatment. The electronic chip-mimetic sonothermal platform provides a promising and widespread clinical application prospect.

Photonic integrated chip for wearable physiological and biochemical sensing

Xiaolin Li,Hongqiang Li,Mingjun Ding,Ming Han,Xin Liu,Liyuan Yang,Cheng Zhang,Lizhen Zhang,Francisco Hernández-ramírez,Lu Cao,Enbang Li

With the increasing need to monitor physiological and biochemical signals, flexible wearable optical sensors have attracted considerable attention because of their ability to attach to human skin for monitoring purposes. However, existing optical sensors face limitations in terms of integration, sensitivity, and versatility, which restrict their practical application. In this study, a fully integrated polydimethylsiloxane/poly (methyl methacrylate) (PDMS/PMMA)-based photonic integrated chip is developed for human physiological and biochemical measurements. It integrates microring resonators (MRRs), Mach–Zehnder modulator (MZM), Mach–Zehnder interferometers (MZIs), and a microfluidic chip and can measure skin temperature, pulse, blood pressure, ECG, blood glucose, and sweat glucose. The good skin fit makes this photonic integrated chip an innovative solution for continuous physiological and biochemical measurements, which holds substantial promise in redefining standard clinical practices for early disease detection and management.

On-Chip Fabrication of Carbon Nanoparticle–Chitosan Composite Membrane

Weiping Ding,Cheng Liang,Sijie Sun,Liqun He,Dayong Gao

The on-chip fabrication of a carbon nanoparticle–chitosan composite membrane (i.e. a sorbent membrane or a mixed matrix membrane) using laminar flow-based interfacial deprotonation technology was presented in this paper. In addition, the effects of carbon nanoparticles and reactant flow rates on membrane formation were investigated. Finally, the permeability and adsorption capacities of the membrane were discussed. During fabrication, an acidic chitosan solution and a basic buffer solution that contained carbon nanoparticles were introduced into a microchannel. At the flow interface, a freestanding composite membrane with embedded carbon nanoparticles was formed due to the deprotonation of the chitosan molecules. The membrane growth gradually stopped with time from upstream to downstream and the thickness of the membrane increased rapidly and then slowly along the reactant flow direction. The formation of the membrane was divided into two stages. The average growth rate in the first stage was significantly larger than the average growth rate in the second stage. Carbon nanoparticles in the basic solution acted as nucleating agents and made the membrane formation much easier. As the flow rate of the chitosan solution increased, the averaged membrane thickness and the membrane hydraulic permeability initially increased and then decreased. Because of the addition of carbon nanoparticles, the formed membrane had adsorption abilities. The carbon nanoparticle–chitosan composite membrane that was fabricated in this study could be employed for simultaneous adsorption and dialysis in microdevices in the future.

Fluorescence Super-Resolution Imaging Chip for Gene Silencing Exosomes

Gaoqiang Yin,Tongsheng Qi,Jinxiu Wei,Tingyu Wang,Zhuyuan Wang,Yiping Cui,Shenfei Zong

Tumor cell-derived extracellular vesicles and their cargo of bioactive substances have gradually been recognized as novel biomarkers for cancer diagnosis. Meanwhile, the PD-L1 (Programmed Death-Ligand 1) protein, as an immune checkpoint molecule, is highly expressed on certain tumor cells and holds significant potential in immune therapy. In comparison to PD-L1 monoclonal antibodies, the inhibitory effect of PD-L1 siRNA (small interfering RNA) is more advantageous. In this article, we introduced a microfluidic chip integrating cell cultivation and exosome detection modules, which were intended for the investigation of the gene silencing effect of PD-L1 siRNA. Basically, cells were first cultured with PD-L1 siRNA in the chip. Then, the secreted exosomes were detected via super-resolution imaging, to validate the inhibitory effect of siRNA on PD-L1 expression. To be specific, a “sandwich” immunological structure was employed to detect exosomes secreted from HeLa cells. Immunofluorescence staining and DNA-PAINT (DNA Point Accumulation for Imaging in Nanoscale Topography) techniques were utilized to quantitatively analyze the PD-L1 proteins on HeLa exosomes, which enabled precise structural and content analysis of the exosomes. Compared with other existing PD-L1 detection methods, the advantages of our work include, first, the integration of microfluidic chips greatly simplifying the cell culture, gene silencing, and PD-L1 detection procedures. Second, the utilization of DNA-PAINT can provide an ultra-high spatial resolution, which is beneficial for exosomes due to their small sizes. Third, qPAINT could allow quantitative detection of PD-L1 with better precision. Hence, the combination of the microfluidic chip with DNA-PAINT could provide a more powerful integrated platform for the study of PD-L1-related tumor immunotherapy.Keywords:exosomes;immunotherapy;PD-L1;microfluidic chip;DNA-PAINT;integrated platform
