Traditional wound dressings frequently exhibit issues such as inadequate adhesion to irregular wound surfaces, a tendency to induce secondary tissue damage, and limited multifunctionality, posing significant challenges in clinical practice. This study presents a novel injectable hydrogel dressing (CMCS/ODEX/TA/HNTs) that integrates multiple mechanisms, thereby imparting superior hemostatic, antioxidant and antibacterial properties. CMCS and ODEX form a reversible three-dimensional network via a rapid Schiff base reaction. Upon injection, these materials can rapidly gelatinize in situ, adaptively filling the cavities of complex wounds. TA serves as a multifunctional crosslinking agent, significantly enhancing the hydrogel’s mechanical strength, self-healing properties, antioxidant capacity, and broad-spectrum antibacterial activity through robust hydrogen bonds and covalent interactions. A crucial aspect of this enhancement is the incorporation of halloysite nanotubes (HNTs). By leveraging their excellent dispersibility in both the aqueous phase and polymer matrix, along with their unique tubular structure and high specific surface area, HNTs not only improve tissue adhesion and mechanical integrity but also promote platelet aggregation, facilitate the sustained release of bioactive factors, and markedly enhance the hemostatic effect. In vivo experiments on rat tail cut off and liver injury models have revealed that the hemostatic efficacy of CMCS/ODEX/TA/HNTs hydrogel surpasses that of commercial hemostatic sponges, ensuring swift hemostasis. In vitro studies have showcased its substantial antibacterial capabilities, while animal trials have further validated its superior hemostatic performance compared to conventional sponges. The synergistic combination of its multifunctional attributes, encompassing antibacterial, antioxidant, and highly effective hemostatic properties, presents a promising approach for managing wound infections. Particularly suited for challenging emergency scenarios, this hydrogel holds significant promise for clinical translation.

The management of bone defects poses a significant challenge for clinicians, necessitating effective strategies to regulate immune inflammation, angiogenesis, and osteogenic differentiation for successful bone regeneration. In this study, we developed a novel hydrogel composed of carboxymethyl chitosan (CMCS) and gelatin methacryloyl (GelMA) designed for the sustained release of the bioactive component calcitonin gene-related peptide (CGRP) and zeolitic imidazolate framework-8 (ZIF-8). CGRP was initially encapsulated within ZIF-8 and subsequently integrated into the CMCS/GelMA hydrogel matrix. In vitro evaluations revealed that the hydrogel exhibited exceptional biocompatibility and antimicrobial properties, effectively promoting osteogenesis and angiogenesis while modulating M2 macrophage polarization. Furthermore, in vivo assessments indicated that the CGRP@MOF/CG hydrogel significantly regulated the local immune microenvironment and accelerated the healing of cranial defects in rat models. This study provides valuable references for the design and fabrication of multifunctional materials for enhancing bone regeneration.

Developing multifunctional pesticide delivery systems represents a crucial strategy to address challenges in conventional pesticide applications, including low active ingredient utilization, resistance development, and environmental risk accumulation. This study developed a pH/enzyme dual-responsive nano-delivery system based on carboxymethyl chitosan (LCT@CMCS-g-PCL) for smart targeted delivery and controlled release of lambda-cyhalothrin (LCT). The system's environmental stability, foliar affinity, and stimuli-responsive release behavior were systematically characterized. LCT@CMCS-g-PCL nanoparticles exhibited an average size of 312.5 nm with 23.69 % drug loading and excellent storage stability. Notably, the system showed over 2-fold enhanced resistance to enzymatic degradation and extended photodegradation half-life of LCT from 37.5 h to 187.9 h. Compared with commercial microemulsion (LCT ME), the system showed 17.06 % improvement in foliar deposition efficiency. In vitro release studies confirmed environmental responsiveness, with stable deposition under acidic conditions, triggered release in the alkaline microenvironment, coupled with enzyme activity modulation to delay resistance. Bioassays demonstrated superior pest control efficacy against Plutella xylostella larvae compared to conventional formulations, with the LC 50 value reduced by 39.7 %. Biosafety evaluations confirmed the relative safety for human cells and adult zebrafish, with minimal disturbance to aquatic microbial communities. This study provides an effective strategy for precision pest control, resistance management and environmental risk management in agriculture.

In Catharanthus roseus , the inherently low yield of bioactive alkaloids such as vindoline, catharanthine, ajmalicine, and vinblastine remains a major limitation to their broader therapeutic application, particularly in anticancer and antihypertensive treatments. To address this challenge, we developed a novel metabolic flux regulation strategy by combining the enzyme inhibitors lovastatin and naproxen with the abiotic elicitor copper(II)-chitosan (Cu-CS) complex. Compared to controls, treatment with 2 mg/L lovastatin significantly enhanced catharanthine, vindoline, ajmalicine, and vinblastine accumulation in C. roseus CMCs to 2.38, 0.63, 35.3, and 2.33 mg/L, respectively — increases of 48%, 121%, 71%, and 12%. Application of 8 mg/L naproxen further enhanced catharanthine, vindoline, and vinblastine levels to 3.78, 0.67, and 2.14 mg/L, representing increases of 151%, 135%, and 30%, respectively. The Cu-CS complex at 10 mg/L elevated catharanthine, vindoline, ajmalicine, and vinblastine to 2.13, 0.49, 32.11, and 2.46 mg/L, with corresponding increases of 49%, 63%, 55%, and 36%. Notably, the combined treatment of all three agents synergistically boosted catharanthine, vindoline, and vinblastine production to 5.94, 1.52, and 3.65 mg/L — representing 310%, 390%, and 98% increases over control levels. Quantitative RT-PCR analysis revealed that Cu-CS treatment upregulated the transcription of key genes and transcription factors involved in terpenoid indole alkaloid (TIA) biosynthesis. Overall, this integrated strategy effectively redirects metabolic flux toward enhanced TIA production in C. roseus CMCs, providing a practical and efficient approach for improving the yield of pharmaceutically important alkaloids in plant cell culture systems.

The medicinal value of vanillin and its isomers has not been well developed, so it is necessary to prepare crystals of vanillin and its isomers as well as to investigate their crystallization rules in detail using advanced crystallization techniques in polymer gel. Based on molecular simulation, the maximum number of hydrogen bonds between CMCS with Van, IsoVan and oVan were reached at molar ratios of 1:9 and 1:4 and 1:5, respectively. The gel hardness and apparent viscosity of CMCS/Van isomers were proportional to the mole ratio, while elongation at break and tensile strength decreased with an increase in molar concentration depending on the position of the side chain group of the Van isomer, exposure of the benzene ring, steric resistance and the number of hydrogen bonds formed. The crystallization of Van, IsoVan and oVan in CMCS gel unexceptionally follow classical supersaturation theory in the case that nVan mainly exhibits a unique growth pattern from needle to strip, IsoVan’s growth style changes from plate to bulk and oVan adapts growth pattern from needle to branch bifurcating. It was also found that the Van crystal changed from II-type to I-type under long-term heating. Studies have further confirmed that the discrepancy of physicochemical characteristics of CMCS/Van blend gel can be attributed to differences in the number of hydrogen bonds compared to CMCS with given group positions of Van isomers. This study provides powerful technical support for the gel crystallization of van isomers.

In this study, CMCS/OPC/LZM hydrogels with varying lysozyme (LZM) concentrations were constructed via simple mixing of carboxymethyl chitosan (CMCS), oligomeric proanthocyanidin (OPC), and LZM through non-covalent self-assembly, followed by evaluation of their physicochemical properties and biological functions. Characterization results showed all hydrogels featured a 3D porous network, shape adaptability, superhydrophilicity, self-healing ability, rapid gelation, and good injectability, with self-assembly mainly mediated by intermolecular hydrogen bonding and electrostatic interactions. Among them, CMCS/OPC 4 /LZM 1 hydrogel exhibited the highest cell proliferation activity, significantly promoting the migration of fibroblasts and endothelial cells, as well as endothelial angiogenesis. Additionally, the CMCS/OPC 4 /LZM hydrogel series achieved an antibacterial rate of nearly 100% against Escherichia coli and Staphylococcus aureus , possessed potent antioxidant activity, had a hemolysis rate of less than 5%, and showed superior hemostatic performance to traditional gauze. Collectively, these findings highlight the great application potential of CMCS/OPC 4 /LZM hydrogels in cutaneous wound repair. Graphical Schematic illustration depicting the fabrication of injectable, multifunctional carboxymethyl chitosan (CMCS)/oligomeric proanthocyanidin (OPC)/lysozyme (LZM) hydrogels via non-covalent self-assembly through hydrogen bonding and electrostatic interactions. The hydrogel exhibits rapid gelation, superhydrophilicity, self-healing, adhesion, and shape adaptability. In vitro, it promotes cell proliferation, migration, and angiogenesis, while demonstrating potent antibacterial activity and antioxidant capacity.

Surgical intervention is a primary treatment for osteosarcoma, often resulting in a tumorous bone defect with an irregular shape. Postoperative management is essential to minimize tumor recurrence risks and promote bone regeneration. To address these issues, we developed a multifunctional injectable, rapidly photo-curable hydrogel composed of gelatin methacryloyl/carboxymethyl chitosan/hyaluronic acid (GelMA/CMCS/HA), modified with vanadium-doped mesoporous bioactive glass (VMBG). The exceptional injectability enables seamless adaptation to irregular bone defects, offering a significant advantage over preformed implants, while the rapid photocurability of the hydrogel ensures stable fixation within minutes, thereby reducing potential risks during surgery. Furthermore, this platform exhibits dual therapeutic efficacy, characterized by antitumor activity and osteogenic induction. In vitro assessments demonstrated that V(V)/V(IV) valence cycling-driven ROS generation mediated its potent antitumor efficacy. Additionally, concurrent enhancement of alkaline phosphatase activity and osteogenic marker expression validated its osteogenic potential. The CMCS incorporation promoted healing at the defect site, while the HA addition created binding sites for cell adhesion and growth, thereby improving scaffold bioactivity. Collectively, this study presents the development and validation of a multifunctional GelMA/CMCS/HA hydrogel, highlighting its dual capability for bone regeneration and tumor suppression within tumor-associated bone microenvironments.

Infected traumatic wounds remain a formidable clinical challenge because existing bioadhesives provide only tissue adhesion and lack concurrent antimicrobial and regenerative capabilities. Here, we developed an injectable bioadhesive (CγR) that integrates an “antibacterial–adhesive–regenerative” (AAR) triad by precisely tuning the ratio of carboxymethyl chitosan (CMCS), autologous platelet-rich plasma (PRP), and γ-polyglutamic acid (γ-PGA). CγR exhibits > 99.9% bactericidal efficacy against Escherichia coli and Staphylococcus aureus , delivers instantaneous wet-tissue adhesion (> 3 kPa), and sustains a gradient release of PDGF, TGF-β, and VEGF. In a rat model of infected full-thickness wounds, a single application achieved sterile sealing and complete epithelialization within 6 days, increased collagen deposition by 1.5-fold, and markedly drove macrophage polarization toward the prohealing M2 phenotype. By condensing the conventional “suture–antibiotic–dressing” regimen into a one-step procedure, CγR offers a biosafe and readily scalable platform for managing contaminated complex wounds and holds promise for clinical translation. Graphical The injectable bioadhesive CγR integrates carboxymethyl chitosan (CMCS), platelet-rich plasma (PRP), and γ-polyglutamic acid (γ-PGA) to provide antimicrobial, adhesive, and regenerative effects. CγR bioadhesive exhibits sustained release of growth factors, promotes rapid wound closure, and accelerates tissue regeneration in infected wound models.

Sclerotinia sclerotiorum threatens crop production worldwide. Conventional prochloraz (Pro) suffers from photolysis, low utilization, and pollution. We developed a pH-responsive nanopesticide, Pro@ZnS@CMCS (PZC), using mesoporous ZnS to load Pro and coating it with carboxymethyl chitosan (CMCS). In the acidic microenvironment (pH ≈ 3) formed during fungal infection, the CMCS shell protonates and decomposes, enabling precise Pro release. Concurrently, the decomposition process consumes H + to modulate the pH of the infection microenvironment and disrupts the homeostasis of fungal oxalate metabolism. The exposed ZnS and residual CMCS synergistically enhance antibacterial activity, resulting in a triple mechanism of drug release, pH regulation, and multi-component action. PZC released 3.7-fold more Pro at pH 3 than pH 7. It improved leaf adhesion and rainfastness, with higher retention than free Pro. PZC exhibited an inhibition rate of 98.16% against Sclerotinia sclerotiorum , which was significantly higher than that of free Pro (30.23%). Transcriptomic analysis revealed that PZC treatment inhibits Sclerotinia sclerotiorum by inducing a “synergistic multiple stress” mechanism. This mechanism involves the simultaneous activation of stress responses, including catabolism, and the systemic inhibition of fundamental growth functions such as ribosome biogenesis, ultimately resulting in the collapse of its metabolic homeostasis. PZC also exhibited excellent biocompatibility with non-target organisms. This work offers a strategy for intelligent nanopesticides enabling targeted delivery, reduced dosage, higher efficacy, and environmental compatibility.

Ready-to-eat cooked meat products contain high levels of moisture and nutrients, making them highly vulnerable to microbial contamination and subsequent quality degradation during storage. Therefore, the development of green packaging materials that integrate active preservation functions with freshness indication is of great significance. In this work, a carboxymethyl chitosan (CMCS)/tannic acid (TA)-Fe 3+ coordination hydrogel (CMCS/TA-Fe) film with favorable biocompatibility and degradability was fabricated through a facile coordination-driven self-assembly strategy, leveraging the multiple interactions among CMCS, TA, and ferric ions (Fe 3+ ). The introduction of the TA-Fe 3+ metal-polyphenol markedly enhanced the mechanical performance of the film, increasing its storage modulus (G′) to approximately 2.4 × 10 4 Pa, and endowed it with efficient photothermal conversion capability. Under near-infrared irradiation at 808 nm, the film surface temperature rapidly increased to 103 °C within 15 min, enabling complete inactivation of Escherichia coli and Staphylococcus aureus . In addition, the dynamic coordination structure of TA-Fe 3+ exhibited pH-responsive chromatic behavior in a model system, resulting in distinct and reversible color changes under different pH conditions. These color variations suggest the potential of the CMCS/TA-Fe film as a visual indicator for freshness-related pH changes. When applied to the refrigerated preservation of cooked chicken breast, the CMCS/TA-Fe film combined with near-infrared irradiation effectively inhibited microbial proliferation, delayed pH increase, reduced moisture loss, and preserved textural and sensory quality. Overall, this study demonstrates a multifunctional and environmentally friendly hydrogel film that integrates photothermal antibacterial activity with a pH-responsive color indication function, offering a promising strategy for intelligent food packaging applications.

Osteoarthritis (OA) is an age-related joint disorder and one of the leading causes of physical disability. In this study, we designed and synthesized a new polysaccharide complex, carboxymethyl chitosan strontium (CMCS-Sr), which is believed to have positive effects on relieving OA. The synthesized CMCS-Sr was structurally verified by SEM, EDS, FTIR, etc. The therapeutic effects of CMCS-Sr were evaluated using various biological experiments. The cell viability and apoptosis results reveal that CMCS-Sr can significantly promote the proliferation and suppress OA chondrocytes apoptosis in vitro . The immunofluorescence staining results suggest that CMCS-Sr facilitates the promotion of the secretion of Type II collagen (Col-II). The transcriptomic results support the observed positive effects of CMCS-Sr on inhibiting chondrocytes apoptosis and alleviating inflammatory reactions. Moreover, animal study demonstrates that CMCS-Sr effectively reduced articular cartilage damage and subchondral bone degradation. Therefore, we propose the use of CMCS-Sr as a promising candidate for relieving OA.

Colorectal cancer chemotherapy often faces challenges such as rapid drug clearance, non-specific biodistribution, and burst release, leading to systemic toxicity and reduced therapeutic efficacy. To overcome these limitations, this study developed a colon-targeted sustained-release hydrogel bead system based on β-cyclodextrin (β-CD) and biocompatible polymers (CMCS/CMCNa/SA). The results demonstrated that the incorporation of β-CD significantly enhanced the drug loading capacity through host–guest interactions. Under the optimal mass ratio of drug complex to wall material (15 : 35), the encapsulation efficiency reached as high as 81.23%. Drug release kinetics showed high correlation coefficients (R2 > 0.9) in zero-order, first-order, Higuchi, and Korsmeyer–Peppas models, indicating a diffusion-controlled release mechanism. The β-CD-modified hydrogel matrix effectively sustained the release of 5-fluorouracil (5-Fu) within the therapeutic window while minimizing the initial burst release. In conclusion, this intelligent delivery system takes advantage of the host–guest inclusion of β-CD and the pH-responsive properties of the polymers, showing promise for localized treatment of colorectal cancer.
