High foliar adhesion carvacrol-chitosan nanoparticles assist cotton against Fusarium oxysporum infection

Jing Zhou,Rourou Wang,Ming Niu,Zhenghua Zhang,Minrui Zhu,Lumei Pu,Weibing Xu,Runtian Ma

The infection caused by Fusarium oxysporum severely threatens cotton production. Herein, carvacrol-chitosan nanoparticles (CS@CAR) are facilely fabricated using tripolyphosphate as a crosslinker. When the initial mass ratio of chitosan to carvacrol is 1:0.4, the nanoparticles show the best stability and drug loading. Morphological analysis reveals that the CS@CAR nanoparticles possesses a uniform spherical structure with an average diameter of approximately 150 nm. About 51.23 % of total CAR is released within the 48 h in the simulated fungal infection environment. The contact angle of drug-loaded particles decreases by 12.2 and 6.6 ° compares with that of pure water and unloaded particles. The fluorescence intensity of leaves before and after rain erosion is 1.3 times and 1.2 times that of the pure water control group by fluorescence imaging analysis. The bacteriostatic activity of drug-loaded nanoparticles is 6–7 times that of non-drug-loaded nanoparticles. The contents of nucleic acid, protein, and malondialdehyde leaked by spores after treatment with drug-loaded particles are 1.4, 1.5 and 2.3 times those in the non-drug-loaded particle treatment group, respectively. The CS@CAR entirely prevents Fusarium oxysporum from infecting cotton leaves in spore inoculation experiments. The CS@CAR nanoparticles are expected to be used to prevent and control the infection of Fusarium oxysporum on cotton.

Synergistic interactions of cinnamaldehyde in combination with carvacrol against food-borne bacteria

Haiqing Ye,Suxia Shen,Jingyue Xu,Songyi Lin,Yuan Yuan,Gregory S. Jones

In order to select effective and safe natural antibacterial ingredients, more than 30 kinds of plant extracts were selected for their suitability as antibacterial agents. A standard broth microdilution method was used to evaluate their antimicrobial activity, a cytotoxicity test was used to detect their safety, and a synergy assay was used to determine which combinations have a synergistic effect. Then time-kill curves were used to further verify their bactericidal capacity. As a result of these tests, cinnamaldehyde and carvacrol were identified first, then subsequently verified to be the most effective and safe natural active substances. It was found that all of the tested bacteria strains were sensitive to cinnamaldehyde and carvacrol. The combination of cinnamaldehyde with carvacrol also showed good synergistic antibacterial effect against 7 of the 11 tested bacterial strains. The time-kill assay verified synergism for the cinnamaldehyde/carvacrol combination toward  Escherichia coli and Staphylococcus aureus . These results indicated that the combination of cinnamaldehyde and carvacrol may serve as a promising naturally sourced food preservative with excellent bactericidal activity against common food spoilage microorganisms.

Synthesis of sulfonate derivatives of carvacrol and thymol as anti-oomycetes agents

Gen-Qiang Chen,Di Sun,Jin-Ming Yang,Song Zhang,Yue-E Tian,Zhi-Ping Che,Sheng-Ming Liu,Jia Jiang,Xiao-Min Lin

Two series of sulfonate derivatives of carvacrol and thymol were synthesized and screened in vitro for their anti-oomycete activity against Phytophthora capsici, respectively. Among all of 32 derivatives, five compounds 3a, 4a, 4k, 3n, and 4n exhibited more potent anti-oomycete activity against P. capsici with EC50 values of 66.66, 62.94, 68.65, 61.24, and 52.91 mg/L, respectively. This suggested that introduction of different substitutions at the hydroxyl position of 1/2 could have remarkable effect on anti-oomycete activity. Overall, when R1 = isopropyl and R2 = methyl, the anti-oomycete activities of the compounds were higher than that of the corresponding compounds of R1 = methyl and R2 = isopropyl.

Synthesis and antifungal activity of carvacrol and thymol esters with heteroaromatic carboxylic acids

Kaibo Wang,Shanshan Jiang,Yunhai Yang,Liming Fan,Fawu Su,Min Ye

Aiming to obtain the more effective pathogen inhibitive ingredients and explore the influence of introducing different heterocyclic units to carvacrol and thymol esters, twenty ester derivatives with different heterocyclic units were synthesized. And the in vitro antifungal activity of title compounds against five plant pathogenic fungi was evaluated by mycelium growth rate method. The results showed that some carvacrol and thymol esters showed good to excellent antifungal activity, and compound 9d (4-bromo-5-isopropyl-2-methylphenyl picolinate) exhibited a broad antifungal spectrum. Preliminary study indicated that the introduction of furan, thiophene and pyridine unit could enhance the antifungal activity of carvacrol and thymol esters against Botrytis cinerea and a bromine atom on the para position of benzene moiety could enhance their antifungal activity.

Mathematical modeling of carvacrol release from HD/PVA/starch-based active intelligent films

Mengge Li,Yingjun Cui,Meng Cheng,Xiangyou Wang,Shuaiyu Guo,Xinyu Yang,Rongfei Zhang

This study aimed to develop a hydrogel (HD)-based active intelligent packaging film for the controlled release of carvacrol (CA). HD was incorporated as a CA carrier within the matrix of acetylated distarch phosphate (ADSP) and polyvinyl alcohol (PVA). The structure and properties of HD and its effect on film properties were studied. Among them, the film containing 6 g HD (PRP-6 film) had the best overall performance. Scanning electron microscopy, Fourier-transform infrared spectroscopy, and X-ray diffraction confirmed the good compatibility between HD and the film base. Thermogravimetric analysis revealed enhanced thermal stability of PRP-6 compared with that of the films with directly added CA (PCO film). The release kinetics of CA in PRP-6 films and PCO films was investigated under varying temperatures, humidity, and acid–base conditions. The PRP-6 film exhibited controlled CA release in response to environmental changes, extending the release period to 12 d compared with that of the PCO film. The first-order kinetic model provided the best fit for describing CA release behavior, and the release process of CA followed Fick's diffusion law. The PRP-6 film showed sustained antibacterial activity over a 12-day storage period. In addition, PRP-6 films retarded the increase in fish pH and L values and inhibited the decrease in fish TVB-N values. This study established a theoretical foundation for developing advanced active packaging systems in aquatic product preservation.

Development and Characterization of Carvacrol Nanoemulsions Stabilized by Bovine Serum Albumin-Chitooligosaccharide Complexes

Yan Wang,Jun Mei,Jing Xie

The intrinsic hydrophobicity, high volatility, and instability of carvacrol (CAR) limit its preservation and practical applications. This study developed a bovine serum albumin (BSA) -chitooligosaccharide (COS) nanoemulsion-based carrier system to improve CAR encapsulation efficiency. The pH-dependent formation of BSA/COS complexes was systematically analyzed, demonstrating significantly enhanced electrostatic interactions between BSA and COS at pH 6.0, which promoted the formation of insoluble complexes with minimal hydrodynamic diameter. Both microscopic morphology and hydrodynamic diameter confirmed that BSA/COS complexes effectively stabilized the CAR-loaded nanoemulsion system. At a 1:1 BSA/COS mass ratio, the emulsion exhibited the most minor hydrodynamic diameter and optimal uniformity (< 200 nm). Furthermore, compared with other groups, the B1C1-4 M group showed better environmental tolerance, such as acid, heat, and salinity, as well as the highest encapsulation efficiency. For example, compared to the B2C1-4 M group and the B1C2-4 M group, the B1C1-4 M group had the lowest hydrodynamic diameter increase rate of 12.98% after being stored at 4°C for 30 days, demonstrating the strongest stability. The mass ratio of BSA/COS and CAR concentration significantly affected the emulsion's antibacterial and antioxidant properties. The B1C1-8 M group achieved the highest antibacterial activity and free radical scavenging rates (88.2% for ABTS and 89.3% for DPPH, respectively). This study presents an innovative approach for enhancing the stability and functionality of CAR in food-grade applications.

Efficient formation of carvacrol inclusion complexes during β-cyclodextrin glycosyltransferase-catalyzed cyclodextrin synthesis

Tonghui Liu,Chuqi Feng,Zhaofeng Li,Zhengbiao Gu,Xiaofeng Ban,Yan Hong,Li Cheng,Caiming Li

Carvacrol has strong antibacterial and antiinflammatory effects and has been increasingly employed in the food and pharmaceutical industries. However, carvacrol is volatile, easily oxidized, and difficult to dissolve in water. In this study, carvacrol was encapsulated with mixed cyclodextrins during enzymatic cyclodextrin synthesis and microencapsulated carvacrol was obtained by spray drying. The encapsulation efficiency and encapsulation yield of microencapsulated carvacrol were 92.16% and 72.30%, respectively. The encapsulation yield for the pure β-cyclodextrin/carvacrol inclusion complex (38.96%) was lower than that of microencapsulated carvacrol (50.44%) when the guest/host ratio was 1:4. No significant difference ( P  > 0.05) in the amount of encapsulated carvacrol was found when the guest/host ratio was less than 1:6. Microencapsulated carvacrol appeared in scanning electron micrographs as spherical particles with obvious invaginations. Determination of the inclusion formation constant, NMR analysis, and molecular modeling revealed that cavity size affected the ability of cyclodextrins to include carvacrol. Finally, the rate constants ( k ) of carvacrol release from microencapsulated carvacrol (1.09–8.53 × 10 −4 ) at various temperatures and relative humidities were systematically lower than those of β-cyclodextrin/carvacrol inclusion complexes (5.20–199 × 10 −4 ). This study transcended traditional essential oil-cyclodextrin inclusion formation by developing a mixed-cyclodextrin inclusion compound that achieved better encapsulation and superior release.

Effect of Sonication on the Properties of Flaxseed Gum Films Incorporated with Carvacrol

Shiyuan Fang,Weiqiang Qiu,Jun Mei,Jing Xie

Carvacrol is a natural compound known to be a highly effective antibacterial; however, it is a hydrophobic molecule, which is a limitation to its use within food packaging. Flaxseed gum (FG) films containing different contents of carvacrol (C) were produced by a film-casting method with sonication. The effects of sonication power and time on the properties of the FG-C films were investigated by measuring the film thickness, mechanical properties, contact angle, opacity, water vapor permeability (WVP), water sorption isotherm, Fourier transform infrared spectroscopy(FTIR), differential scanning calorimetry (DSC), antibacterial and antioxidant activities, and microstructure. The results showed that sonication power and time had significant effects on mechanical and barrier properties, film opacity, and degradability (p< 0.05). The tensile strength (TS) and elongation at break (EB) values exhibited an obvious improvement after sonication, and FG-0.5C-6030 had the lowest TS (33.40 MPa) and EB (4.46%) values. FG-C films formed a denser structure and the contact angle was improved as a result of sonication, which improved the integration of carvacrol into the FG matrix. In terms of microstructure, sonication resulted in a homogeneous and continuous crosssection of FG-C films, and regular surface and cross-sectional images were obtained through the highest acoustic intensity and longest time treatment. The FG films incorporated with carvacrol displayed antibacterial properties againstStaphylococcus aureus,Vibrio parahaemolyticus,Shewanella putrefaciens, andPseudomonas fluorescens, as well as increased antioxidant properties, and sonication was proven to enhance both of them. Keywords:sonication;active films;flaxseed gum;antibacterial agent;carvacrol

The effects of carvacrol on development and gene expression profiles in Spodoptera frugiperda

Jiafu Liu,Yanzheng Lin,Yuting Huang,Luyang Liu,Xueming Cai,Jintian Lin,Benshui Shu

The fall armyworm , Spodoptera frugiperda , is a highly polyphagous agricultural pest that is widely distributed around the world and causes severe crop yield loss. Carvacrol showed adverse effects on many pests, such as larval death and growth inhibition. While the effects of carvacrol on S. frugiperda larvae are not yet known. In this study, the effects of carvacrol on S. frugiperda , including larval growth inhibition and mortality induction, were observed. The detoxification and digestive enzyme activities of larvae with 1.0 and 2.0 g/kg carvacrol treatments were analyzed. Carvacrol boosted the enzyme activities of carboxylesterase (CarE) and glutathione S-transferase (GST) while decreasing the activities of α-amylase (AMS), lipase (LIP), and trypsin. A total of 3422 differentially expressed genes were identified in the larvae treated with 2.0 g/kg carvacrol, of which the DEGs involved in xenobiotic detoxification, food digestion, and insecticidal targets were further examined. These results suggest that carvacrol could regulate growth and development by affecting the process of food digestion, and exert its toxicity on the larvae through interaction with a variety of insecticidal targets. While the altered expressions of detoxification enzymes might be related to the detoxification and metabolism of carvacrol. Our findings offer a theoretical foundation for the use of carvacrol for S. frugiperda control in the field.

Exploring inhibition of Listeria monocytogenes biofilm by carvacrol based on action to quorum sensing

Xiaona Gao,Xinke Gao,Zichun Hua,Majid Alhomrani,Chao Shi,Lin Lin,Yulin Zhu

The contamination of L. monocytogenes on different foods mainly achieves by the formation of biofilm, and L. monocytogenes biofilm poses great risks to food industry. Hence, exploring the safe and nontoxic substances to interfere and destroy L. monocytogenes biofilm is of great concern. In this research, carvacrol was selected as the antibiofilm substance to investigate its effect on the three-dimensional structure, extracellular polymer matrix, aggregation, adhesion and Agr quorum sensing system of L. monocytogenes biofilm. The results revealed that L. monocytogenes showed the greatest biofilm formation ability in 5 days cultivation on stainless steel sheet. The minimum biofilm inhibition concentration and minimum biofilm elimination concentration of carvacrol against L. monocytogenes biofilm were 0.4 and 0.8 mg/mL. SEM and fluorescent pictures demonstrated that carvacrol could destroy and eliminate polymer matrix of L. monocytogenes biofilm. Furthermore, carvacrol could impact the swimming motility and aggregation ability of L. monocytogenes to inhibit the adhesion and initial formation of biofilm. Finally, carvacrol also could effectively inhibit the secretion and production of exopolysaccharide and proteins, downregulating the gene expression and binding with signaling proteins in Agr quorum sensing system to interfere the formation of biofilm. In conclusion, carvacrol could be applied as the ideal antibiofilm substance to inhibit and destroy L. monocytogenes biofilm in food industry.

Near-infrared light-responsive mesoporous polydopamine/carvacrol films for synergistic preservation of refrigerated shrimp

Yunxiao Xia,Weizhuo Xu,Jianzhuo Du,Huijing Chao,Tai Gao,Xu Zhao

The photothermal effect holds considerable promise for applications in food sterilization and preservation. Mesoporous polydopamine (MPDA), a green photothermal material with high drug-loading and sustained-release properties, was loaded with carvacrol (Car) to prepare MPDA-Car nanoparticles, which were incorporated into a PVA/Gel matrix to fabricate a multifunctional film with synergistic photothermal and Car's broad-spectrum antibacterial activity. Near-infrared (NIR) irradiation-induced photothermal effect directly damaged bacterial membranes and triggered controlled Car release from MPDA mesopores, enhancing antibacterial efficacy. Characterization confirmed that uniform MPDA-Car dispersion significantly improved the film's mechanical strength (tensile strength: 22.14 MPa, elongation at break: 218.36%) and reduced water vapor permeability (WVP) by 32.7% compared with the pure PVA/Gel film. The film also exhibited excellent antioxidant capacity (DPPH: 74.4%, ABTS: 77.9%) and potent inhibition against Staphylococcus aureus and Escherichia coli . In Chinese shrimp preservation, the 1.00% PG-MC film mitigated weight loss, retarded TVB-N and TBA accumulation, inhibited pH elevation, and extended shelf life by over 2 days. Notably, MPDA-mediated mild photothermal heating caused no significant degradation of shrimp soluble protein or LOX activity, retaining nutritional quality. This film provides a promising strategy for developing eco-friendly food packaging materials.

Multifunctional Mg-MOF-74@Carvacrol/Chitin nanofiber-based nanocomposite film for enhanced food packaging

Muhammad Junaid Khan,Muhammad Rehan Islam,Farooq Hafeez,Chunyan He,Jun Zhang,Yunlei Xianyu

The proliferation of pathogenic microorganisms is a significant cause of food spoilage, resulting in substantial economic losses and public health risks. To ensure food safety and extend shelf life, packaging must not only maintain product integrity but also prevent contamination by multidrug-resistant (MDR) pathogens such as Escherichia coli ( E. coli ), Listeria monocytogenes ( Listeria ), and Candida albicans ( C. albicans ). In this study, we developed a multifunctional nanocomposite (Mg-MOF-74@CA/CNF) by integrating magnesium-based metal–organic framework (Mg-MOF-74), carvacrol (CA), and chitin nanofibers (CNF). This nanocomposite was subsequently incorporated into a polyvinyl alcohol (PVA) matrix to fabricate the final film (Mg-MOF-74@CA/CNF/PVA). The resulting film demonstrated excellent antimicrobial activity, achieving up to 98 % inhibition of MDR pathogens through reactive oxygen species (ROS) generation and disruption of microbial membranes. In addition, the film exhibited improved thermal stability, enhanced tensile strength, and superior barrier properties against oxygen and water vapor. Application tests on cherry, cauliflower, and chicken confirmed significant reductions in microbial growth and weight loss, alongside prolonged shelf life while maintaining product quality. These findings highlight Mg-MOF-74@CA/CNF/PVA as a sustainable and effective alternative to conventional packaging materials, offering both food preservation and enhanced safety.
