Rational design of highly dispersed CeO 2 nanozymes with abundant oxygen vacancies is critical for achieving efficient and sensitive H 2 O 2 detection. Herein, we report a deep eutectic solvent (DES)-guided engineering strategy to construct an N-doped carbon (NC) framework integrated with mussel-inspired, uniformly dispersed CeO 2 nanoparticles. The DES acts both as a green C/N source and a self-templating medium, regulating heteroatom distribution and graphitization degree to yield a conductive carbon scaffold. Dopamine undergoes oxidative self-polymerization under mild alkaline conditions, and its catechol–amine functionalities strongly chelate Ce 3+ ions, directing homogeneous nucleation and suppressing nanoparticle aggregation. This molecularly engineered architecture produces an oxygen-vacancy-rich interface enriched with redox-active Ce 3+ /Ce 4+ sites, enabling rapid electron transfer and accelerated H 2 O 2 redox kinetics. The optimized CeO 2 /NC electrode delivers a diffusion-controlled amperometric response with dual linear ranges (0.05–60 and 60–300 μM), a low detection limit of 0.036 μM, and sensitivities of 94.8 and 49.3 mA M -1 cm -2 , respectively, surpassing most reported CeO 2 -based sensors. Furthermore, the sensor exhibits excellent selectivity and retains 89 % of its initial response after prolonged operation. This work provides a scalable, eco-friendly pathway for constructing bioinspired, oxygen-vacancy-rich nanozymes, offering broad applicability in advanced electrocatalytic sensing platforms.

Redox-active nanozymes offer low-cost controlled synthesis, high stability, and tunable catalytic properties over natural enzymes, which have attracted wide attention in the field of disease diagnosis and treatment. However, the improvement of catalytic activity remains an important challenge for nanozymes. Herein, the Au/CeO 2 nanozymes is developed to achieve enhanced multiple enzyme-mimetic activity. The Au/CeO 2 nanozymes at 5% doping possess best peroxidase-like activity with threefold higher catalytic rate than CeO 2 . For catalase-mimic catalysis, the Au/CeO 2 nanozymes at 5% doping also exhibited a 1.5-fold enhanced reaction rate higher than pure CeO 2 . The superoxide dismutase (SOD)-like capacity of Au/CeO 2 nanozymes is proportional to Au content. The Au/CeO 2 nanozymes at 10% doping show optimal SOD-like capacity of 60.2 U mg −1 . In vitro experiments validate the regulation ability of intracellular oxidative stress and inflammation. Au/CeO 2 nanozymes can reduce lipopolysaccharide- or H 2 O 2 -induced oxidative damage by scavenging excess ROS in nerve cell. Therefore, Au/CeO 2 can be used as a promising antioxidant in disease treatment, and the study offers general guidelines for achieving enhanced biocatalytic property through atomic doping.

The effective intracellular accumulation of doxorubicin (DOX) is crucial for improving antitumor efficacy, which is severely impeded by limited drug penetration, uncontrollable drug release, and drug resistance. In this study, a thermal-deformative polymer embedding ultrasmall ceria (CeO 2 ) was rationally designed for deep tumor drug shuttling and hypoxia reversal to improve chemotherapy. Structurally, the CeO 2 nanozyme was covalently grafted with a polymer of p(NIPAM-co-AM) that could sharply shrink for DOX loading, which was consolidated with polydopamine (PDA) film encapsulation. Thereafter, a tumor penetration guide of apolipoprotein A-I (apoA-I) conjugated iRGD peptide (apoA-I-iRGD) was further decorated onto the PDA shell via Michael addition for preparing CeO 2 P/DOX@iAPDA. With the aid of apoA-I-iRGD, CeO 2 P/DOX@iAPDA penetrated both the tumor spheroids (∼78 μm) and the tumors of the mouse model deeply. After internalization by tumor cells and triggering by low pH in lysosomes, rapid DOX release was achieved by peeling off the PDA shell and thermosensitive deformation of p(NIPAM-co-AM). CeO 2 P/DOX@iAPDA provided 66.4% tumor suppression in 4T1-derived tumor spheroids and 63.2% in 4T1-tumor-bearing mice, respectively. Preliminary mechanistic research involving western blotting and immunohistochemistry revealed that CeO 2 P/DOX@iAPDA reversed resistance through the through HIF-1α-P-gp/lipid axis. Collectively, this study intelligently integrated CeO 2 nanozymes, temperature-sensitive polymers, and imitated biochemical modifications to improve chemotherapy for breast cancer.

Artificial enzymes show prospects in biomedical applications due to their stable enzymatic catalytic activity and ease of preparation. CeO2 nanozymes represent a versatile platform showing multiple enzyme-mimicking activities, although their biocatalytic activities and selectivity are relatively poor for biomedical use. Herein, we developed Mn- and Co-doped CeO2 nanozymes (M/CeO2, M = Mn or Co) via atomic engineering to achieve a significant increase in enzyme-like activity. The M/CeO2 nanozymes exhibited outstanding peroxidase-like activity with a reaction rate about 8–10 times higher than that of CeO2. Importantly, the Co/CeO2 nanozyme preferred for catalase-like activity with a 4–6-fold higher catalytic rate than CeO2, while the Mn/CeO2 nanozyme had a predilection for improving the superoxide dismutase-like capacity. This indicated the selective modulation of enzyme-mimicking activities via atomic doping engineering. Cellular level experiments revealed the in vitro therapeutic effects of the nanozymes. Mn/CeO2 and Co/CeO2 selectively modulated the intracellular redox imbalance in lipopolysaccharide (LPS)- or H2O2-stimulated nerve cells and improved cell survival. This work provides a feasible strategy for the design of catalytically selective artificial enzymes and facilitates the widespread application of CeO2 nanozymes in redox-related diseases.

Sepsis, a life-threatening condition caused by a dysregulated immune response to infection, leads to systemic inflammation, immune dysfunction, and multiorgan damage. Various oxidoreductases play a very important role in balancing oxidative stress and modulating the immune response, but they are stored inconveniently, environmentally unstable, and expensive. Herein, we develop multifunctional artificial enzymes, CeO2 and Au/CeO2 nanozymes, exhibiting five distinct enzyme-like activities, namely, superoxide dismutase, catalase, glutathione peroxidase, peroxidase, and oxidase. These artificial enzymes have been used for the biocatalytic treatment of sepsis via inhibiting inflammation and modulating immune responses. These nanozymes significantly reduce reactive oxygen species and proinflammatory cytokines, achieving multiorgan protection. Notably, CeO2 and Au/CeO2 nanozymes with enzyme-mimicking activities can be particularly effective in restoring immunosuppression and maintaining homeostasis. The redox nanozyme offers a promising dual-protective strategy against sepsis-induced inflammation and organ dysfunction, paving the way for biocatalytic-based immunotherapies for sepsis and related inflammatory diseases.

CeO2 nanozymes have demonstrated the potential to enhance biological scaffolds with chemodynamic therapy. However, their catalytic efficacy is limited by the slow conversion of Ce4+ to Ce3+ and the lack of substrates like H2O2 and H+. To address these challenges, we adopted a dual-pronged strategy that utilized the plasmonic resonance of Au nanoparticles and their glucose-oxidase mimicry to boost electron and hydrogen transfer. Specifically, we integrated Au/CeO2 nanozymes into poly-l-lactic acid scaffolds via selective laser sintering. This conversion of Ce3+ to Ce4+ in the scaffolds enhanced the reduction of H2O2 to a hydroxyl radical, inducing oxidative stress in tumor cells. The Au nanoparticles played a crucial role in boosting the Ce3+/Ce4+ catalytic cycle by providing both the energy and the catalytic substrates. They recycled Ce4+ back to Ce3+ by exploiting plasmonic-induced hot electrons and catalyzed glucose oxidation to supply H2O2 and H+. Our nanoscale and atomic-scale simulations confirmed that the Au/CeO2 hybrid structure utilized near-field coupling to amplify the plasmonic resonance and the Au–O–Ce bridge reduced the electron transfer barrier. Consequently, the Au/CeO2 scaffold decreased the activation energy from 22.57 to 9.92 kJ/mol. These findings highlight the significant promise of the Au/CeO2 nanozyme scaffold for NIR-enhanced chemodynamic therapy.
