Scalable succinate biomanufacturing via in-situ Citrobacter freundii-CdS biohybrids: Light-driven redox balancing enhances reductive succinate biosynthesis

Shuyan Chi,Hanbang Ding,Miao Cai,Yuze Xu,Wenjie Xia,Xuejiao Zhang,Liwei Liu

In situ biomineralized C. freundii -CdS biohybrid enabled light-driven fermentation. • Photoelectrons increased intracellular NADH/NAD + by 59%. • Succinate production increased by 26.2% in a 5 L bioreactor. • rTCA and electron transport pathways were coordinately activated. • Light-driven redox balancing enhanced reductive succinate biosynthesis. Microbial fermentation provides a sustainable route to succinic acid production, yet the reductive tricarboxylic acid (rTCA) pathway demands more NADH than anaerobic glucose catabolism can supply, thereby limiting the yield. Here, we constructed a Citrobacter freundii -CdS biohybrid in which CdS nanoparticles, biomineralized on the cell surface, harvest visible light and are proposed to supply photoelectrons to the host cell, augmenting intracellular NADH availability. Under optimized conditions, the illuminated biohybrid elevated the intracellular NADH/NAD + ratio by 59% relative to controls. In a 5 L bioreactor, the illuminated biohybrid achieved a succinate titer of 42.8 g L −1 (yield: 0.49 g g −1 glucose), a 26.2% increase over the dark control, with concurrent reductions in formate, acetate, and lactate. Transcriptomic analysis identified coordinated upregulation of electron transport, NADH synthesis, and rTCA pathway genes alongside downregulation of competing branch-point enzymes. These findings support a model in which light-driven electron supply can serve as a non-genetic approach for redirecting intracellular reducing power and carbon partitioning toward succinate biosynthesis in facultative anaerobes, although the precise electron-transfer route remains to be established. Biogenic palladium nanoparticles reclaimed by a novel strain Citrobacter braakii Z1 accelerating RDX biodegradation in the munition effluent

Shichong Zhao,Bin Hou,Kunchan Zhang,Yating Jia,Jing Lu

The microbial degradation of high-energy explosives has emerged as a cost-effective environmental remediation strategy, but is hindered by low degradation efficiency due to the slow electron transfer. Biogenic Pd 0 nanoparticles (bio-Pd 0 ) demonstrate dual functionality, maintaining catalytic activity for N-NO 2 bond reduction while substantially enhancing electron transfer efficiency. This study introduces an innovative single-cell system that synergistically combines chemical catalysis with biological enhancement through in situ synthesis of nanoparticles. Comprehensive characterization through electron microscopy, spectroscopic analysis, and electrochemical measurements revealed that bio-Pd 0 synthesized by Citrobacter braakii Z1 ( C. braakii Z1) were preferentially localized in the periplasmic space and extracellular matrix, forming a stable hybrid named bio-Pd 0 @ C. braakii Z1. This process relied on the bio-reduction mediated by biohydrogen and extracellular electron transfer processes via c-type cytochromes (c-Cyts) and flavins. Notably, RDX (hexahydro-1,3,5-trinitro-1,3,5-triazine) degradation efficiency increased from 11 % with microbes alone to nearly 100 % with bio-Pd 0 . Although the observed RDX degradation in inactivated bio-Pd 0 @ C. braakii Z1 and the detected hydrogenolysis intermediates indicated the participation of chemical catalysis, the negative correlation between RDX degradation rate with the Pd:biomass ratios suggested the biology process dominated RDX degradation. Meanwhile, the higher output current in bio-Pd 0 @ C. braakii Z1 and the extracellular electron consumption by RDX indicated the bio-Pd 0 -mediated extracellular biodegradation was the primary driver for improved RDX removal efficiency. Toxicity evaluation confirmed that the in-situ combination of Pd-catalyzed chemical destruction and biological enhancement in a single cell is a feasible and environmentally friendly strategy for RDX removal with a reduced ecological risk.

FDH/Hases-S-chain mediated electron redistributing in Citrobacter freundii JH@FeS during degradation of sulfamethoxazole and nitrate

Huimin Liu,Dejin Liu,Yuancai Chen

Considering the negligent degradation of sulfamethoxazole (SMX) by Citrobacter freundii JH , the incorporation of bio-FeS could initiate the SMX biodegradation to 0.0444 (S-FeS), and further to 0.0564 mg L −1 mg −1 protein d −1 (SN-FeS) when coexisted with nitrate. Electrochemical (LSV, I-t , DPV, EIS and EDC) and respiratory inhibition experiments clarified that the bio-FeS could greatly switch/redistribute electron transmembrane-transfer from intracellular to extracellular mainly via FDH/Hases-S-chain, as revealed by the significant increase of ipa- FDH/Hases / ipa- FC-Cyts and ipc- FDH/Hases / ipc- FC-Cyts (from 1.09 and 1.07 (SN-native) to 1.50 and 3.58 (SN-FeS)), while nitrate (linear fitting with NADH ( R 2 = 0.9903)) mainly intensified CoQ-L-chain related INET from Complex I to CoQ to compensate for the electronic competition with SMX. SN-FeS system detoxified the SMX on microbial metabolism (such as membrane rupture and oxidative stress induction) with high SOD activity (737.93 U g FW −1 ). Structural equation modeling indicated that bio-FeS up-regulated PMF-mediated ATP synthesis ( P PMF-ATPs from 0.12 (SN-native) to 0.74 (SN-FeS)) and PMF-mediated NADH ( P PMF-NADH from -0.72 (SN-native) to 0.63 (SN-FeS)), and the nitrate addition intensified this positive feedback. Overall, this study provides a new perspective for bionanoparticles via electron transfer/redistribution to detoxify and launch the antibiotics biodegradation in ecological environment.

Resveratrol attenuates Citrobacter rodentium-infected colitis by remodeling the gut microbiota structure and inhibiting CD4+ cell activation in mice

Junzhu Li,Xin Feng,Yizhi Jing,Haidong Qian,Yun Ji,Yanling Hao,Zhengyuan Zhai

Infectious enteritis is a chronic inflammatory condition of the intestine, mainly characterized by immune cell infiltration and gut microbiota dysbiosis. Resveratrol, a natural polyphenol compound from grapes, berries and peanuts, is known for its antioxidant and anti-inflammatory properties. This study investigated the effects of resveratrol in a murine model of Citrobacter rodentium-induced colitis. Resveratrol ameliorated disease symptoms, body weight loss, colon shortening, and increased intestinal permeability. Histological and immunofluorescence analyses revealed that resveratrol improved intestinal barrier integrity by increasing goblet cells, upregulating MUC-2 expression, reducing crypt hyperplasia, and suppressing epithelial cell apoptosis. Resveratrol significantly decreased the mRNA levels of pro-inflammatory cytokines, namely Tumor Necrosis Factor-α (TNF-α), Interleukin-6 (IL-6) and Interleukin-22 (IL-22), while restoring the percentage of CD4+ cells to normal levels. It also attenuated oxidative stress by enhancing the activities of key antioxidant enzymes, including catalase (CAT), total superoxide dismutase (T-SOD), and glutathione peroxidase (GSH-Px). Furthermore, resveratrol reshaped the gut microbiota by increasing the abundance of obligate anaerobes Clostridium and decreasing the abundance of facultative anaerobes Enterobacteriaceae. The abundance of Lactobacillus and Dubosiella was also significantly increased by resveratrol. In summary, these findings demonstrate that resveratrol supplementation may serve as a promising nutritional strategy to ameliorate C. rodentium colitis by inhibiting CD4+ cell activation and remodeling the gut microbiota.

The effect of triclosan on intergeneric horizontal transmission of plasmid-mediated tigecycline resistance gene tet(X4) from Citrobacter freundii isolated from grass carp gut

Xinxin Jiang,Jingfei Long,Yanzhen Song,Xiaoyu Qi,Ping Li,Kuiquan Pan,Chenyang Yan,Hongzhou Xu,Haixia Liu

The transmission of antibiotic resistance genes (ARGs) in pathogenic bacteria affects culture animal health, endangers food safety, and thus gravely threatens public health. However, information about the effect of disinfectants - triclosan (TCS) on ARGs dissemination of bacterial pathogens in aquatic animals is still limited. One Citrobacter freundii ( C. freundii ) strain harboring tet (X4)-resistant plasmid was isolated from farmed grass carp guts, and subsequently conjugative transfer frequency from C. freundii to Escherichia coli C600 ( E. coli C600) was analyzed under different mating time, temperature, and ratio. The effect of different concentrations of TCS (0.02, 0.2, 2, 20, 200 and 2000 μg/L) on the conjugative transfer was detected. The optimum conditions for conjugative transfer were at 37 °C for 8h with mating ratio of 2:1 or 1:1 ( C. freundii : E. coli C600). The conjugative transfer frequency was significantly promoted under TCS treatment and reached the maximum value under 2.00 μg/L TCS with 18.39 times that of the control group. Reactive oxygen species (ROS), superoxide dismutase (SOD) and catalase (CAT) activities, cell membrane permeability of C. freundii and E. coli C600 were obviously increased under TCS stress. Scanning electron microscope showed that the cell membrane surface of the conjugative strains was wrinkled and pitted, even broken at 2.00 μg/L TCS, while lysed or even ruptured at 200.00 μg/L TCS. In addition, TCS up-regulated expression levels of oxidative stress genes ( kat E, hem F, bcp , hem A, kat G, ahp F, and ahp C) and cell membrane-related genes ( fim C, bam E and omp A) of donor and recipient bacteria. Gene Ontology (GO) enrichment demonstrated significant changes in categories relevant to pilus, porin activity, transmembrane transporter activity, transferase activity, hydrolase activity, material transport and metabolism. Taken together, a tet (X4)-resistant plasmid could horizontal transmission among different pathogens, while TCS can promote the propagation of the resistant plasmid.

Proteomic signatures of synergistic interactions in antimicrobials

Gang Zhou,Ying-si Wang,Hong Peng,Su-juan Li,Ting-li Sun,Qing-shan Shi,Jordi Garcia-Ojalvo,Xiao-bao Xie

Mounting evidence has shown that antimicrobial agents can interfere synergistically with bacterial viability and proliferation when acting together at both the planktonic and biofilm levels without clear underlying molecular mechanisms. Here, multiplexed proteomics by iTRAQ was used to study the interplay between two biocides, the isothiazolone 1,2-benzisothiazolin-3-one (BIT) and the chelating agent disodium ethylenediaminetetraacetic acid (EDTA-2Na), employing the Citrobacter werkmanii as a model system. We first confirmed that these two biocides act synergistically on this bacterial species and then extracted the proteomic profiles of C. werkmanii cells in the presence of BIT, EDTA-2Na, and their combinations. In particular, we identified 43 core proteins that are differentially expressed in all three conditions simultaneously. Meanwhile, we found that these core proteins are consistently up-regulated when these two biocides are present, but not for single biocides, where we found a balanced mix of up- and down-regulation. Meanwhile, most of the deletion mutants of the core DEPs exhibited biofilm growth inhibition under joint biocide action, while their response was very heterogenous, with respect to the wild-type strain. Together, our results show that while BIT and EDTA-2Na act on multiple protein targets, they interact synergistically at the protein level in a very consistent manner. Significance Our preliminary experiments have demonstrated that a combination of 1,2-benzisothiazolin-3-one (BIT) and EDTA-2Na shows higher inhibitory effects on planktonic growth and biofilm formation in both C. werkmanii and Staphylococcus aureus than when these two biocides act alone. However, the mechanistic basis of such synergistic interaction is still unknown. Therefore, the key proteins involved in the above-mentioned enhanced antimicrobial synergy were elucidated using multiplexed proteomics analysis by isobaric tags for relative and absolute quantification (iTRAQ). Our results reveal that the joint action of BIT and EDTA-2Na induces consistent protein expression alteration in a set of core proteins of C. werkmanii , which underlies a strong synergistic antimicrobial effect, which increase our understanding of the action modes of BIT and EDTA-2Na as well as their combinations.

One bead, triple feats: Alginate-encapsulated Fe/Ni-Citrobacter microsphere (AFC-Syn) unlocks concurrent contaminant elimination, nutrient recovery and self-regeneration in municipal wastewater treatment

Tianyu Li,Siying Li,Yuzhe He,Xingxing Peng

Brominated flame retardants (BFRs) resist degradation in conventional wastewater treatments, posing ecological and health risks. Herein, we innovatively developed an alginate-encapsulated Fe/Ni- Citrobacter synergistic system (AFC-Syn) for synergistic BFRs degradation in a continuous stirred-tank reactor (CSTR) treating real municipal wastewater. Remarkably, the system achieved sustained removal efficiencies of >90% for Tetrabromobisphenol A (TBBPA), Decabromodiphenyl ether (BDE-209), and Hexabromocyclododecane (HBCD), over 30 days, with complete degradation within 48 h, while simultaneously controlling COD and nutrient levels (>85% removal). In the system, in addition to direct electron transfer between Fe/Ni and membrane-binding proteins, the carboxyl groups in alginate are proposed to potentially serve as electron-hopping sites to enable long-range electron transport. Meanwhile, the alginate-encapsulated Fe/Ni continuously release Fe 2+ /Ni 2+ , which act as soluble electron carriers to accelerate electron transfer—either to BFRs for debromination or to microorganisms to stimulate their growth. Ultimately, the electrons are captured by dehalogenase, cleaving C Br bonds and driving the stepwise debromination. Metagenomic and metatranscriptomic analyses revealed persistent enrichment of keystone functional genera, collectively accounting for over 30% of the microbial community. These microorganisms activated interconnected gene networks, thereby establishing an integrated functional axis that synergistically links pollutant degradation, electron transfer, and nutrient cycling. This integration prevented toxic intermediate accumulation while achieving complete decontamination.
