Quenched solid-state electrochemiluminescence of gold nanoclusters and the application in the ultrasensitive detection of concanavalin A

Shihong Chen,Yu Fan,Cong Zhang,Yanyan He,Shaping Wei

This work investigated the quenching effect of ferrocenecarboxylic acid (Fc-COOH) on the bovine serum albumin-stabilized Au nanoclusters-triethylamine (Au 25 -Et 3 N) electrochemiluminescence (ECL) system. Based on the annihilation between the oxidation product of Fc (Fc + ) and Et 3 N radical, a quenching-based sandwich-configuration ECL biosensor was established for sensitive and specific detection of concanavalin A (ConA). Au 25 with strong initial ECL signal were modified onto the glassy carbon electrode (GCE) for immobilizing glucose oxidase (GOD). With the further immobilization of target ConA, the quenching probe of GOD@Fc-COOH-PtPd nanocubes were incubated onto the electrode through a specific carbohydrate-ConA interaction, thus achieving a sandwich structure. The resulting ECL biosensor showed a wide linear range from 0.004   ng/mL to 90   ng/mL and a low detection limit of 0.001   ng/mL (S/N   =   3) for ConA detection. The integration of Fc-COOH with Au 25 -Et 3 N system would hold a great promise in bioanalysis.

Glucose-sensitive hydrogels from covalently modified carboxylated pullulan and concanavalin A for smart controlled release of insulin

Kunhua Lin,Juzhen Yi,Xuhong Mao,Hao Wu,Li-Ming Zhang,Liqun Yang

Pullulan , which is a linear glucan with non-toxicity, good biocompatibility and biodegradability , was carboxylated with succinic anhydride to yield a pullulan derivative bearing COOH groups (CPUL). The CPUL structure was determined by Fourier-transform infrared (FTIR) and 1 H nuclear magnetic resonance (NMR) spectroscopies. 1 H NMR integration showed that the degree of substitution of COOH residues was 0.2. A covalently modified concanavalin A (ConA)-CPUL hydrogel with glucose sensitivity was prepared by an amidization reaction between the COOH group of CPUL and the NH 2 group of ConA via a safe and mild 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/ N -hydroxysuccinimide activation method. Insulin-loaded CPUL-ConA (insulin/CPUL-ConA) hydrogels were prepared by absorption of insulin in situ . Analysis of the release profiles in vitro confirmed that smart controlled release of insulin from the insulin/CPUL-ConA hydrogels could be achieved because of specific ConA-glucose binding. The insulin/CPUL-ConA hydrogels showed the characteristic viscoelasticity and shear thinning properties of covalently modified hydrogels. Scanning electron microscopy showed that the hydrogels had crosslinked network structures containing uniform pores and insulin particles. This work provides a pullulan-based glucose-sensitive hydrogel that enables smart controlled release of insulin in response to changes in glucose concentration. Such hydrogels could be used in closed-loop systems for diabetes therapy.

Smart controlled release of acarbose from glucose-sensitive hydrogels comprising covalently modified carboxylated pullulan and concanavalin A

Ju-Zhen Yi,Kunhua Lin,Hao Wu,Xuhong Mao,Li-Ming Zhang,Liqun Yang

A hydrogel comprising covalently modified carboxylated pullulan and concanavalin A (CPUL-ConA) was prepared with glucose-sensitivity in a simulated small intestinal fluid containing glucose. The acarbose-encapsulated CPUL-ConA (Acar/CPUL-ConA) hydrogel, with an acarbose encapsulation ratio of 0.63, was prepared by immersing the dried CPUL-ConA hydrogel in an acarbose/PBS (pH 7.2) solution. In the simulated gastric fluid, less than 20% acarbose was released from the Acar/CPUL-ConA hydrogel, which could be beneficial to protecting the stomach. In contrast, more than 80% acarbose was smartly controlled released from the Acar/CPUL-ConA hydrogel in the simulated small intestine fluid containing glucose. Scanning electron microscopy provided further evidence of the smart release of acarbose from the Acar/CPUL-ConA hydrogel in simulated gastrointestinal fluids, which is advantageous to inhibiting α-glucosidase activity and subsequently reducing the postprandial blood glucose levels, as well as helpful for reducing the gastrointestinal dysfunction of acarbose. Rheological analysis indicated that the Acar/CPUL-ConA hydrogel showed characteristics of chemical crosslinked hydrogels. There was no obvious shear thinning behavior, suggesting that the Acar/CPUL-ConA hydrogel could be stable under gastric and intestinal peristalsis. Therefore, the Acar/CPUL-ConA hydrogel is anticipated to improve the therapeutic efficacy of postprandial hyperglycemia and type 2 diabetes.

Formononetin protects against concanavalin-A-induced autoimmune hepatitis in mice through its anti-apoptotic and anti-inflammatory properties

Guangwei Liu,Wenxia Zhao,Jiameng Bai,Jianjiao Cui,Haowei Liang,Baoping Lu

Autoimmune hepatitis (AIH) is a chronic inflammatory liver disease that seriously threatens the health of humans globally. Formononetin (FMN) is a natural herb extract with multiple biological functions. In this study, an experimental model of AIH was established in mice through the use of concanavalin A (ConA). To investigate the effects of FMN on ConA-induced hepatitis, the mice were pretreated with 50 or 100 mg/kg body mass of FMN. The results show that FMN alleviated ConA-induced liver injury of mice in a dose-dependent manner. Moreover, pretreatment with FMN inhibited the apoptosis of hepatocytes in the ConA-treated mice through downregulating the expression of pro-apoptotic proteins (Bax, cleaved caspase 9, and cleaved caspase 3) and upregulating the expression of anti-apoptotic protein (Bcl-2). It was also found that the levels of proinflammatory cytokines were greatly reduced in the serum and liver tissues of mice pretreated with FMN. Further studies showed that FMN reduced the level of phosphorylated nuclear factor kappa B (p-NF-κB) p65 and enhanced the level of IκBα (inhibitor of NF-κB), suggesting that FMN inhibits the activation of the NF-κB signaling pathway. In addition, FMN inhibited activation of the NOD-like receptor protein 3 (NLRP3) inflammasome. Therefore, FMN could be a promising agent for the treatment of AIH.

Regulated basal and bolus insulin release from glucose-responsive core-shell microspheres based on concanavalin A-sugar affinity

Meirong Bai,Jing He,Liangfa Kang,Jun Nie,Ruixue Yin

Individual insulin therapy considering the heterogeneity of insulin resistance between patients may bring more benefits than conventional therapy. Therefore, in glucose-responsive insulin delivery systems, more attention should be paid on further regulation of insulin release to meet individual requirements. Our study shows the feasibility of using a photo-crosslinkable shell layer to regulate basal and bolus insulin release from glucose-responsive Con A-polysaccharides network. Core-shell microspheres were fabricated through a two-step high-speed shear-emulsification method. The morphology was observed by SEM and TEM, and the core-shell structure was confirmed by the differences in chemical composition between core-shell and single-layer microspheres obtained from XPS and IR analysis. In vitro insulin release test revealed that the core-shell microspheres with or without light-irradiation could maintain corresponding bolus and basal insulin release in response to different glucose concentration but enable much lower burst release compared with single-layer microspheres without shell. Meanwhile, insulin release rate and amount could be further decreased upon light-irradiation owing to the photo-induced cycloaddition of cinnamate pendant groups of the shell material. The released insulin was proved to remain active according to fluorescence and circular dichroism analysis. The HDF cell viability assessment suggested that the core-shell microspheres possessed no in vitro cytotoxicity.

An electrochemiluminescence biosensor based on boron nitride quantum dots as novel coreactant for quantitative determination of concanavalin A

Wang Caixia,Li Mengsi,Wang Peijin,Liu Defang

An electrochemiluminescence (ECL) analytical platform is constructed based on boron nitride quantum dots (BNQDs) as a novel coreactant of luminol for quantitative assay of concanavalin A (Con A). Different from previous research that mainly focuses on its superior optical properties, BNQDs are used for the first time as a coreactant for boosting ECL intensity of luminol, which has a 10-fold enhancement compared with individual poly(luminol/aniline) nanorods loaded on reduced graphene oxide (PLA-rGO) using GCE. On the basis that BNQDs contain an abundance of active amino, a possible mechanism of amino oxidation facilitating ECL emission is proposed. Firstly, luminol as light spices are oxidized to luminol •− and BNQDs generate an abundance of BNQDs-NH +• via electrochemical oxidization, producing reductive intermediates BNQDs-N • in alkaline conditions. Finally, BNQDs-N • react with luminol •− to obtain the excited species AP 2−* , returning to ground state and emitting light. Due to the hindrance effect of Con A, the ECL intensity decreases gradually as various concentrations of Con A are modifying the electrode surface. Therefore, a sensitive ECL biosensor for detecting Con A is constructed exhibiting a wide linear range of 1.0 pg·mL −1 to 1.0 μg·mL −1 and a low detection limit of 0.15 pg·mL −1 . Schematic representation of an electrochemiluminescence (ECL) biosensor based on boron nitride quantum dots (BNQDs) as an efficient coreactant of reduced graphene oxide functionalized poly(luminol/aniline) (PLA-rGO) for quantitative assay of concanavalin A (Con A).

A novel “on-off” electrochemiluminescence sensor for the detection of concanavalin A based on Ag-doped g-C3N4

Yu Fan,Xingrong Tan,Xin Ou,Qiyi Lu,Shihong Chen,Shaping Wei

In this work, a novel “on-off” electrochemiluminescence (ECL) biosensor was successfully constructed for ultrasensitive detection of concanavalin A (ConA). The “signal-on” state with strong initial ECL signal was obtained by Ag-doped graphitic carbon nitride nanosheet (Ag-g-C 3 N 4 ), which was modified onto the glassy carbon electrode (GCE) for immobilizing phenoxy dextran (DexP) through π-π interaction. Then, the target ConA was anchored onto the DexP/Ag-g-C 3 N 4 modified film via the specific carbohydrate-ConA interaction. Successively, polyaniline-3,4,9,10-perylenetetracarboxylic acid-DexP conjugate (denoted as PANI-PTCA-DexP), as the ECL signal quenching probe, was incubated onto the electrode through the carbohydrate-ConA interaction to achieve the “signal-off” state. Here, PTCA was used as a matrix for the high loading of PANI and DexP, PANI served as the quencher towards Ag-g-C 3 N 4 system, and DexP as the recognition element for bounding ConA. With the formation of the sandwiched structure of DexP, ConA and PANI-PTCA-DexP, a desirable quenching ECL signal was measured with S 2 O 8 2− as the co-reactant of Ag-g-C 3 N 4 . The quenching effect of PANI towards Ag-g-C 3 N 4 is positively correlated with the concentration of ConA. With such an “on-off” system, the detection of ConA was achieved with a wide linear range from 0.001   ng/mL to 50   ng/mL as well as a low detection limit down to 0.0003   ng/mL.

Proximity Ligation Assay with Three-Way Junction-Induced Rolling Circle Amplification for Ultrasensitive Electronic Monitoring of Concanavalin A

Bingqian Liu,Bing Zhang,Guonan Chen,Huanghao Yang,Dianping Tang

A new signal amplification strategy based on target-induced proximity ligation assay accompanying three-way junction-based rolling chain amplification was designed for ultrasensitive detection of concanavalin A (Con A) by coupling with a sequential injection mode. To construct such a proximity ligation assay system, two types of magnetic sensing probes including glucosamine/DNA1-conjugated magnetic bead (GA-MB-DNA1) and glucosamine/DNA2-labeled magnetic bead (GA-MB-DNA2) were first synthesized and prepared through a typical carbodiimide coupling. In the presence of target Con A, GA-MB-DNA1 and GA-MB-DNA2 were ligated together based on the interaction between Con A and the conjugated glucosamine on the MB, thereby resulting in the formation of a three-way DNA junction because of partial base pairing on the DNA1/DNA2. With the aid of ligase and polymerase, the formed three-way DNA junction could be used as the primer to produce numerous repeated oligonucleotide sequences through rolling circle amplification (RCA) reaction. The formed long oligonucleotide strand could cause the intercalation of numerous positively charged methylene blue molecules with a negatively charged DNA backbone. During the electrochemical measurement, each of the intercalated indicators could produce an electrochemical signal within the applied potentials, resulting in the amplification of detectable electronic signal. By monitoring the change in the signal, we could indirectly determine the concentration of target Con A in the sample. Under the optimal conditions, the developed sensing platform exhibited high sensitivity for detection of Con A with a wide dynamic range of 1.96 pM to 98 nM and a low limit of detection (LOD) of 1.5 pM at the 3sB level. Intra-assay and interassay coefficients of variation were less than 8.9% and 9.7%, respectively. In addition, the methodology was validated by assaying Con A spiked samples including newborn cattle serum and peanuts, and the recovery in all cases was 88.8–134.7%.

Selective extraction of bioactive glycoprotein in neutral environment through Concanavalin A mediated template immobilization and dopamine surface imprinting

Xue Qu,Feifei Wang,Yi Sun,Yu Tian,Rui Chen,Xiaoyu Ma,Changsheng Liu

Glycoproteins play important roles in various biological events. Extraction of specific bioactive glycoproteins from physiological fluids is highly required for clinical diagnosis and treatment. Concanavalin A (Con A) is a tetramer lectin protein that can specifically bind glycospecies containing sugar units, and this binding occurs in physiological environments. Inspired by this, we propose a sugar–lectin recognition based glycoprotein surface imprint, which is anticipated to selectively extract bioactive glycoproteins in physiological environments. The glycosylated biomarker, transferrin, was used as a model template. Transferrin was first immobilized on a Con A modified Fe3O4 surface via the sugar–lectin interaction at pH 7.4. Dopamine was then polymerized onto this surface for glycoprotein imprinting at the same pH, and continuous oxygen bubbling into the dopamine solution improved the polymerization speed. After removal of the template by boronate buffer, transferrin imprinted Fe3O4 was obtained. The subsequent binding experiment indicated that the transferrin imprint has pH dependent binding affinity towards this protein. The optimal transferrin binding capacity, as well as the maximum imprinting factor was observed at neutral pH. The isotherm adsorption study reveals that imprinted and non-imprinted material are both fitted with the Langmuir adsorption model. Single selective adsorption and competitive adsorption experiments show that the obtained surface imprint has selectivity towards template glycoprotein, and circular dichroism (CD) spectral analysis indicates that boronate buffer washing has no negative influence on the natural structure of transferrin and Con A. These results demonstrate that this novel glycoprotein surface imprint can work under physiological conditions, and the glycoprotein extraction method is mild enough to keep the bioactivity of these targets for further bio-application.

Efficient Preparation of High-Purity Sulforaphene: Selective Immobilization of Myrosinase by Designing a Concanavalin A-Loaded Metal–Organic Framework

Mintong Tian,Yuman Guo,Meng Zhou,Purui Yu,Mingming Li,Zehua Li,Qipeng Yuan

Isothiocyanates (ITCs) in cruciferous plants are natural active ingredients with cancer-preventive, anti-inflammatory, and antibacterial functions. ITCs are generated through the hydrolysis of glucosinolates by myrosinase (MYR). However, the presence of specifier proteins, especially epithionitrile-specifier proteins (ESPs), significantly impacts the yield, purification, and application of ITCs. Herein, we designed a metal–organic framework to selectively immobilize MYR by constructing a concanavalin A (ConA)-loaded ZIF-8 core. The 1.55-fold higher sulforaphane (SFE)-generation activity of ConA-MYR@ZIF-8 compared to ConA-free groups demonstrated the selectivity of ConA@ZIF-8 toward MYR. Impurities, including nitrile generated by ESP, were effectively excluded. ConA-MYR@ZIF-8 maintained over 93.9% activity after 1 week of storage at 4 °C and retained 77.9% activity after 20 cycles. SFE with 97.8% purity was prepared in a yield of 14.9 mg/(g·h) through enzymatic reactions coupled with real-time dichloromethane column extraction, achieving a recovery rate of 90.1%. This study provided new insights for the efficient preparation of SFE and other ITCs.

Concanavalin A inhibits human liver cancer cell migration by regulating F‑actin redistribution and assembly via MAPK signaling pathway

Haoran Jiang,Xianxin Wen,Xue Zhang,Bingyu Zhang

Concanavalin A (Con A), the first and most typical representative of the legume lectin family, has a potent anti‑liver cancer effect by inducing cell apoptosis and autophagy. However, its function in human liver cancer cell migration remains unclear. The present study investigated the effect of Con A on the viability and migration of human liver cancer cells with different metastatic abilities. It was found that Con A could reduce the viability of human liver cancer cells (HCCLM3, MHCC97L and HepG2) and human hepatocytes (MIHA). In addition, Con A could inhibit human liver cancer cell migration specifically without affecting hepatocytes. Sugar inhibition assay showed that glucose‑related sugar binding sites played an important role in the inhibition of Con A on human liver cancer cell migration. In addition, Con A could affect HCCLM3 migration by activating ERK1/2, JNK1/2/3 and p38 signaling in the MAPK pathway. Moreover, Con A could regulate fibrous actin (F‑actin) redistribution and assembly via the MAPK signaling pathway. However, Con A had no significant effect on the activation of matrix metalloproteinase (MMP)‑2 and MMP‑9 in HCCLM3 cells. In conclusion, Con A may bind to glucose‑related receptor protein, activating ERK1/2, JNK1/2/3 and p38 signaling in the MAPK pathway, further contracting cells and reducing the content of F‑actin of single cells to inhibit HCCLM3 cell migration. These results would deepen the links between human liver cancer cell migration and related glycoproteins or signaling molecules, and may provide a new perspective for Con A as a potential anticancer agent.

Joint concanavalin A-aptamer enabled dual recognition for anti-interference visual detection of Salmonella typhimurium in complex food matrices

Fengxia Zhao,Hangli Yan,Yi Zheng,Yu Zu,Shengyuan Yang,Hongmei Hu,Shengyuan Shi,Hao Liang,Xiangheng Niu

Given that food poisoning and infectious diseases caused by Salmonella typhimurium ( S. typhimurium ) draw intensive public health concerns, developing rapid, accurate, and cost-effective approaches to detect the pathogen is of crucial importance. Herein, we proposed a concanavalin A (Con A)-aptamer joint strategy to realize dual recognition for the strongly specific, visual, and highly sensitive determination of S. typhimurium . Compared with currently used single identification strategies, Con A and aptamer could recognize different sites of S. typhimurium to enhance the utilization rate of these sites for better sensing. The developed assay offered specific detection of S. typhimurium against other bacteria in a remarkably wide concentration range of 7.0 × 10 1  ∼ 7.0 × 10 9 CFU/mL, along with a detection limit as low as 23 CFU/mL. Real sample analyses of milk and pork demonstrated the excellent reliability and practicability of our assay, providing great potential for food safety analysis.
