Multifunctional nanomaterials have become a promising nanoplatform for anticancer treatment with tumor specific activation. Here, a cancer cell-activated nanocomposite Cu/ZIF-8@MPN@DOX@F127 (CZMDF) was constructed with a metal phenolic network (MPN)-modified Cu/ZIF-8 and loaded with doxorubicin (DOX). CZMDF was degraded to release tannic acid (TA), Fe3+, Cu2+, and DOX in cancer cells. Benefiting from the reducibility of TA, Fe3+ was converted to Fe2+, which could react with endogenous H2O2 to produce a highly toxic hydroxyl radical (•OH) by the Fenton reaction. The Fenton reaction-mediated chemodynamic therapy (CDT) was promoted by the accelerated Fe3+/Fe2+ conversion, which was also enhanced with a Cu2+-based Fenton-like reaction. In addition, endogenous glutathione (GSH) could reduce Fe3+ and Cu2+ to Fe2+ and Cu+, which led to further enhancement of CDT performance. DOX acted as a traditional drug to realize chemotherapy. Furthermore, the heat from MPN after near-infrared irradiation (NIR) could be applied to realize photothermal therapy (PTT). The multifunctional nanocomposite was designed to achieve synergistic tumor treatment with CDT/chemotherapy/PTT. In vitro experiments showed that the CZMDF caused about 70% of cell apoptosis, and the tumor growth in mice was also inhibited in vivo study. This multifunctional nanomaterial enriches the nanocatalyst types for synergistic tumor therapy, providing a nanoplatform for potential clinical transitions.

Immune checkpoint blockade therapy has made great strides in cancer treatment, but conventional immune checkpoint blockade antibodies often lead to overactivation of the immune system and severe immune-related adverse effects. Herein, we report the design of a nanomedicine (denoted as CPD@M−TDN) based on multifunctional tetrahedral DNA nanostructures (M−TDN). In this system, a bimetallic metal–organic framework (Cu/ZIF-8) is synthesized as the core; doxorubicin-loaded polydopamine acts as a shell to reduce the immunogenicity of the nanomaterials in peripheral blood. Finally, the CpG oligodeoxynucleotides and PD-L1 aptamer-based M−TDN are coupled to the core–shell structure nanomedicine through the biotin-avidin-system. Under the acidic tumor microenvironment, the decomposition of Cu/ZIF-8 and the release of copper ions and doxorubicin can lead to immunogenic death of tumor cells through chemodynamic/chemotherapy. Furthermore, the M−TDN has the dual effects of weakening the immunosuppressive microenvironment with PD-L1 aptamer and promoting dendritic cell activation with the immunostimulant CpG. Overall, the developed CPD@M−TDN can effectively enhance the response rate to immunotherapy and has excellent potential in the field of integrated tumor treatment.

Therapeutic nanosystems triggered by a specific tumor microenvironment (TME) offer excellent safety and selectivity in the treatment of cancer by in situ conversion of a less toxic substance into effective anticarcinogens. However, the inherent antioxidant systems, hypoxic environment, and insufficient hydrogen peroxide (H2O2) in tumor cells severely limit their efficacy. Herein, a new strategy has been developed by loading the chemotherapy prodrug disulfiram (DSF) and coating glucose oxidase (GOD) on the surface of Cu/ZIF-8 nanospheres and finally encapsulating manganese dioxide (MnO2) nanoshells to achieve efficient DSF-based cancer chemotherapy and dual-enhanced chemodynamic therapy (CDT). In an acidic TME, the nanocatalyst can biodegrade rapidly and accelerate the release of internal active substances. The outer layer of MnO2 depletes glutathione (GSH) to destroy the reactive oxygen defensive mechanisms and achieves continuous oxygen generation, thus enhancing the catalytic efficiency of GOD to burst H2O2. Benefiting from the chelation reaction between the released Cu2+ and DSF, a large amount of cytotoxic CuET products is generated, and the Cu+ are concurrently released, thereby achieving efficient chemotherapy and satisfactory CDT efficacy. Furthermore, the release of Mn2+ can initiate magnetic resonance imaging signals for the tracking of the nanocatalyst.
