3D-Printed Scaffold Achieves Synergistic Chemo-Sonodynamic Therapy for Tumorous Bone Defect

Cijun Shuai,Zihao Zhang,Min Chen,Bingxin Sun,Xingming Long,Guoyong Wang,Shuping Peng

Various smart scaffolds have recently been developed to address the regeneration of tumor bone defect. However, the recurrence of residual tumor cells poses a serious challenge to postoperative management, highlighting the need for effective therapeutic interventions. In this study, a multifunctional antitumor nanoplatform (Ti3C2/CuO2) for synergistic chemo-sonodynamic tumor therapy was developed and then rationally integrated into a poly(l-lactic acid) (PLLA) scaffold via selective laser sintering. CuO2 not only releases Cu2+ ions to facilitate chemodynamic antitumor therapy through the Fenton reaction but also generates H2O2, which further oxidizes Ti3C2 to produce TiO2 sonosensitizers. More importantly, the carbon-based substrates after oxidation of Ti3C2 have created favorable conditions for carrier transmission in the sonodynamic process, thereby amplifying the sonodynamic therapy. Additionally, moderate local hyperthermia form periodic sonodynamic therapy produces moderate localized heat therapy to further stimulate bone tissue regeneration. Meanwhile, the sustained release of bioactive ions (such as Cu and Ti ions) from the scaffold also fosters vascularization, further accelerating bone regeneration. This work presents a viable approach to developing multifunctional scaffolds for repairing tumorous bone defects.

Glucose-triggered self-perpetuating catalytic cascade in GOx@CuO2 nanoreactors for treating bacteria-infected diabetic wounds

Jun Wang,Yi Huang,Yan Cao,Wenjiao Zhu,Yuhui Chen,Yan Chen,Yuanyuan Zhang,Jianfeng Cui,Yuanxiang Wang,Kai Lan,Yanxia Lin

The hyperglycemic microenvironment of diabetic wounds is highly susceptible to bacterial infection, persistent inflammation, and impaired angiogenesis, severely delaying the healing process. To overcome the limitations of current therapeutic strategies, this study innovatively developed an intelligent cascade nanoreactor, GOx@CuO 2 , which utilizes excess glucose (Glu) as a driving force. The key innovation of this system is its exploitation of the excess glucose at the lesion site. Catalyzed by glucose oxidase (GOx), the Glu is converted into gluconic acid and hydrogen peroxide (H 2 O 2 ) which simultaneously accomplishes the tripartite microenvironmental modulation of Glu depletion, acid production, and H 2 O 2 supply. Under acidic conditions, GOx@CuO 2 exhibits efficient peroxidase-like (POD-like) activity, catalyzing the generation of hydroxyl radicals (·OH) to achieve potent antibacterial effects. As glucose is gradually consumed and the pH increases, GOx@CuO 2 intelligently switches to a catalase-like (CAT) mode, decomposing residual H 2 O 2 into oxygen (O 2 ). This alleviates oxidative stress and promote angiogenesis, forming an adaptive synergistic therapeutic cycle of “antibacterial-promoting healing”. In vivo experiments confirmed that the nanoreactor significantly improved the wound microenvironment, achieving a wound closure rate of 93.4 % within 14 days. This study not only proposes a novel non-antibiotic therapeutic strategy responsive to the lesion microenvironment but also provides new insights for designing intelligent enzyme-catalyzed systems for tissue repair related to metabolic diseases.

Jiajie Chen,Yi Cao,Shiyang Lin,Huicong Niu,Hongjian Zhang,Lei Guan,Chaoqin Shu,Aijun Wu,Yuhai Bian,Yufang Zhu

Traditional therapies for melanoma suffer from many restrictions, that calls for the development of new treatments with high effectiveness and safety. Recently, employing transdermal microneedle (MN) systems for delivering therapeutic agents has obtained increasing interest as the efficient, minimally invasive, painless, and safe natures. Designing an “all-in-one” multifunctional MN system without complicated integration that can realize enhanced tumor therapy has presented huge promising for clinical application but is still a challenging strategy. Herein, a simple responsive MN system is developed for efficient anti-melanoma by combining self-enhanced chemodynamic therapy (CDT) and photothermal therapy (PTT). This MN system, constructed by biocompatible poly(vinylpyrrolidone) (PVP) as substrate and PVP-stabilized CuO 2 nanoparticles (CuO 2 NPs) as therapeutic agent, shows good skin-insertion capability and dissolving property that benefit in penetrating skin efficiently to deliver the therapeutic CuO 2 NPs into melanoma site. The released CuO 2 NPs from MN system can not only produce copper ion (Cu 2+ ) and hydrogen peroxide (H 2 O 2 ) in response to the acidic environment for catalytic hydroxyl radical ( OH) generation and thus achieve the H 2 O 2 self-supplying CDT, but also can be utilized as a glutathione (GSH)-scavenging agent for further enhancing CDT. Moreover, owing to the loading of CuO 2 NPs, such MN system exhibits excellent near-infrared (NIR) photothermal property that can induce local hyperthermia at the melanoma area for PTT. In result, the designed MN system realizes the efficient tumor inhibition with minimal side effects in vivo by the combination therapy , offering a safe and effective melanoma treatment option for patients.

Multifunctional SGCG/CuO2 composite hydrogel for integrated wastewater remediation: Copper ions removal, phenolic pollutant degradation, and bacterial inactivation

Xifeng Ma,Yujing Liu,Xiaokang Zhang,Yanhui Wei,Hongchao Ma

Hydrogel-based materials have emerged as promising candidates for wastewater remediation due to their remarkable adsorption capacity, recyclability, and environmental compatibility. In this study, we developed a multifunctional sodium alginate-glutaraldehyde-chitosan-gelatin (SGCG) composite hydrogel with enhanced mechanical properties and abundant functional groups (-COOH, -NH 2 ) for efficient Cu 2+ sequestration. Investigation of adsorption parameters revealed optimal conditions and underlying mechanisms for heavy metal removal. Subsequent in situ reaction of the Cu 2+ -loaded hydrogel (SGCG/Cu 2+ ) with H 2 O 2 yielded a nanozyme composite (SGCG/CuO 2 ) with dual enzyme-mimetic activities: laccase-like activity for phenolic pollutant degradation and peroxidase-like activity for antibacterial applications. The SGCG/CuO 2 hydrogel exhibited exceptional performance in removing phenolic contaminants and inactivating pathogenic bacteria ( Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) ), owing to its robust catalytic properties. This work presents an innovative multifunctional hydrogel system that integrates heavy metal adsorption, catalytic organic degradation, and antimicrobial activity into a single platform, offering a sustainable and efficient solution for complex wastewater remediation.

H2O2 self-supplying Mo/Fe@CuO2 nanozyme with NIR light enhanced catalytic activity and photothermal synergistic antibacterial application

Feijian Xiao,Dezhi Yang,Chun Xun,Haiyan Li,Qiulan Li,Zitao Zhong,Daqiao Wei,Yaling Yang

Nanozymes have drawn increasing attention due to unique enzyme-mimic catalytic activities for the treatment of bacterial infections. Herein, a novel tri-metal nanozyme for synergistic eradication of Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) was generated by Mo/Fe doped carbon dots (Mo/Fe-CDs) wrapping copper peroxide (CuO 2 ) nanozyme (Mo/Fe@CuO 2 ). Mo/Fe@CuO 2 showed near-infrared (NIR) light-enhanced peroxidase-like catalytic activity, and H 2 O 2 self-supply to promote the generation of reactive oxygen species (ROS) for highly efficient antibacterial photothermal therapy. The CuO 2 could generate H 2 O 2 in the acidic microenvironment, and Mo/Fe-CDs consequently could convert H 2 O 2 to hydroxyl radicals (·OH), resulting in the improvement of sterilization efficiency. Interestingly, Mo/Fe-CDs could activate the transformation of Cu 2+ to Cu + through the co-catalytic reaction based on the Mo 4+ /Mo 6+ and Fe 3+ /Fe 2+ redox couples, and provided photonic hyperthermia (PTT) to enhance the peroxidase-like activity. Meanwhile, Mo/Fe@CuO 2 showed glutathione (GSH) oxidase-like activity, which can achieve the depletion of glutathione (GSH) in bacteria, thus significantly improving the bactericidal effect. The collaborative eradication of bacteria, through innovative metal carbon-based nanozyme technology, provides a powerful and feasible approach to treating bacterial infections that threaten human health.

Dual-mode colorimetric-electrochemical biosensor for Vibrio parahaemolyticus detection based on CuO2 nanodot-encapsulated metal-organic framework nanozymes

Wenhai Wang,Shu Xiao,Mengli Zeng,Hongzhen Xie,Ning Gan

Rapid and accurate detection of Vibrio parahaemolyticus (VP) in a variety of testing environments is critical for preventing the spread of foodborne illnesses. Therefore, a dual-mode colorimetric-electrochemical biosensor was prepared for the rapid and sensitive detection of VP . Phenylboronic acid-decorated CuO 2 nanodot-mediated organic metal-organic framework (CP@MOF) nanozymes were constructed to produce colorimetric and electrochemical signals. Furthermore, an electrode surface was fabricated using tetrahedral DNA nanostructure-functionalized poly(amidoamine) dendrimer-Au nanoparticles to specifically bind VP . Under acidic environments, the CP@MOF nanozymes disintegrated, enabling the simultaneous release of Cu 2+ and H 2 O 2 . This in turn causes a Fenton-type reaction, generating a hydroxyl radical , which then oxidizes 3,3′,5,5′-tetramethylbenzidine, thereby enabling visual detection of VP . Moreover, the produced copper ions can be used for the electrochemical detection of VP . This dual-mode biosensor achieved an accuracy of 88.7 %, which was higher than that of single-mode assays (81.1 %). Hence, this dual-mode assay may facilitate the effective on-site detection of pathogens in shrimp samples.

NIR-Responsive Nanoplatform Based on CuO2 with Self-Supplied H2O2/O2 for Synergistic Photodynamic-Chemodynamic-Photothermal Therapy of Skin Abscesses

Yingnan Zhu,Jiahang Si,Tiantian Xu,Lan Zheng,Xinyu Xiao,Xiaohan Sun,He Ding,Yuze Dong

Chemodynamic therapy (CDT) and photodynamic therapy (PDT) have emerged as promising strategies for treating skin abscesses. However, their therapeutic efficacy is fundamentally constrained by the hypoxic and hydrogen peroxide-deficient microenvironment characteristic of abscesses. Herein, we developed a near-infrared (NIR) light-responsive nanoplatform (ICG/CuO2@PCM) by encapsulating copper peroxide (CuO2) and indocyanine green (ICG) within a phase-change material (PCM), which enables control over self-supplied H2O2/O2 release via an NIR-triggered cascade. Upon irradiation, ICG-mediated photothermal conversion elevates the temperature beyond the PCM melting threshold, triggering a burst release of CuO2 and ICG. CuO2 generates hydroxyl radicals (•OH) through a Fenton-like reaction in the acidic wound microenvironment. Additionally, CuO2 can thermally decompose under photothermal conditions to produce a sustained oxygen supply, which ICG immediately converts into singlet oxygen (1O2), thereby enhancing PDT efficiency. Furthermore, photothermal therapy not only enhances the efficiency of CDT but also provides additional antibacterial support. Remarkably, ICG/CuO2@PCM (NPs), coupled with irradiation, accelerate abscess wound recovery by dispersing biofilm, killing bacteria, and promoting macrophage polarization toward an anti-inflammatory phenotype. Overall, this NIR-responsive nanoplatform highlights the promise in controlled modulation of microenvironment-adaptive antimicrobial strategies to overcome current limitations of hypoxic infections.

In situ synthesized H2O2 self-supplying composite peroxidase mimic CuO2@S/Cu-NC for portable detection of histamine and seafood freshness monitoring

Chenyu Zhou,Jianxin Qiang,Wanyi Zhang,Yuntai Lv,Xingguang Su

The application of peroxidase-like nanozymes in on-site detection was often restricted due to the cumbersome operation caused by necessary step of adding H 2 O 2 . Herein, a novel composite peroxidase mimic CuO 2 @S/Cu-NC with self-supplying H 2 O 2 function was prepared by in situ synthesis of the copper peroxide nanodots (CuO 2 ) on the S/N co-doping carbon-based copper nanozyme (S/Cu-NC). The CuO 2 could produce H 2 O 2 in acidic environment, and the in situ formed H 2 O 2 could immediately bind to the active sites of S/Cu-NC, resulting in the greatly accelerated catalytic reaction efficiency. Because of the property of H + -induced H 2 O 2 production of CuO 2 @S/Cu-NC, the changes of pH value could affect its peroxidase-like activity. Therefore, a sensitive and convenient method for histamine sensing was established. Furthermore, a convenient test swab was designed for the portable histamine detection and visual monitoring the of seafood freshness, which showed great potential of multifunctional nanozyme in the field of food monitoring.

An Intelligent Cupreous Nanoplatform with Self-Supplied H2O2 and Cu2+/Cu+ Conversion to Boost Cuproptosis and Chemodynamic Combined Therapy

Xiangjie Tian,Hui Xu,FangFang Zhou,Xiyu Gong,Songwen Tan,Yongju He

Cuproptosis is a newly identified copper-dependent cell death and holds great promise for cancer therapy. However, transporting enough copper into cancer cells is a challenge. Herein, an intelligent cupreous nanoplatform (denoted as CuO2-MSN@TA-Cu2+), consisting of in situ formation of CuO2 within mesoporous silica nanoparticles (MSN) and then deposition with a tannic acid (TA)-Cu2+ complex, is designed and developed to realize on-demand copper delivery for cuproptosis-based combination therapy. CuO2-MSN@TA-Cu2+ exhibits tumor microenvironment-triggered therapeutic activity, wherein the outer TA-Cu2+ complex is readily disassembled to release Cu2+ and liberate the internal CuO2 to produce Cu2+ and H2O2. The overloaded Cu2+ can not only directly convert endogenous H2O2 and self-supplied H2O2 into highly toxic hydroxyl radicals for chemodynamic therapy (CDT) via Cu-based Fenton-like reaction but also undergo glutathione-mediated reduction into Cu+ species to induce potent cellular cuproptosis and enhance CDT. The experimental results indicate that CuO2-MSN@TA-Cu2+ produces remarkable cytotoxicity against cancer cells and significantly suppresses tumor growth by 93.42% in mice-bearing 4T1 breast tumors. This work provides a new paradigm to boost cuproptosis-related therapy and may also inspire the design of advanced therapeutic nanoplatforms.

Bovine serum albumin-bound homologous targeted nanoparticles for breast cancer combinatorial therapy

Caili Chen,Bohong Xie,Shuming Sun,Sheng Guo,Zishan Yang,Liuzhong Yang,Yana Zhang,Shu-Ang Li,Wei Sun,Zihao Wang,Shuang Qin,Yinghua Ji

Breast cancer, the most common lethal cancer among women, is characterized by the uncontrolled growth of abnormal cells in breast tissue. Therefore, synergistic anticancer strategies are essential, particularly for maximizing drug delivery to tumor sites. Herein, bovine serum albumin (BSA)-bound nanoparticles encapsulating the photosensitizer chlorin e6 (Ce6) (BC) with a CuO 2 core (BC/CuO 2 NPs) were developed for cuproptosis-promoted cancer photodynamic therapy (PDT). The cancer cell membrane (CC) was then coated onto the surfaces to produce BC/CuO 2 @CC NPs for breast cancer combinatorial therapy. BSA serves dual functions as both a stabilizing scaffold for metal peroxide nanomaterials and a molecular connector for Ce6. The BC/CuO 2 @CC NPs group showed the stronger internalization capability than the other groups. BC/CuO 2 @CC NPs could effectively induce the greatest degree of apoptosis and death ratio (81.77 %), and lead to cuproptosis by downregulating the expression of DLAT, LIAS, and FDX1 protein in vitro . The intra-tumoral accumulation of BC/CuO 2 @CC NPs was 8.3- and 7.7-fold higher than that of Ce6 and BC/CuO 2 @CC NPs at 24 h postinjection, respectively. Moreover, synergistic efficacy of cuproptosis and PDT not only inhibited tumor growth but also prevented liver metastases. Thus, our work may be a novel approach for efficient and targeted cancer treatment.

Micro-environment-triggered chemodynamic treatment for boosting bacteria elimination at low-temperature by synergistic effect of photothermal treatment and nanozyme catalysis

Haotian Wu,Li Tian,Haijuan Qin,Xiao Zhou,Xiying Chen,Weiran Li,Jing Zhang,Shuo Wang,Yaqing Liu

An injectable hydrogel dressing, Zr/Fc-MOF@CuO 2 @FH, was constructed by combing acid-triggered chemodynamic treatment (CDT) with low-temperature photothermal treatment (LT-PTT) to effectively eliminate bacteria without harming the surrounding normal tissues. The Zr/Fc-MOF acts as both photothermal reagent and nanozyme to generate reactive oxygen species (ROS). The CuO 2 nanolayer can be decomposed by the acidic microenvironment of the bacterial infection to release Cu 2+ and H 2 O 2 , which not only induces Fenton-like reaction but also enhances the catalytic capability of the Zr/Fc-MOF. The generated heat augments ROS production, resulting in highly efficient bacterial elimination at low temperature. Precisely, injectable hydrogel dressing can match irregular wound sites, which shortens the distance of heat dissipation and ROS diffusion to bacteria, thus improving sterilization efficacy and decreasing non-specific systemic toxicity. Both in vitro and in vivo experiments validated the predominant sterilization efficiency of drug-resistant methicillin-resistant Staphylococcus aureus (MRSA) and kanamycin-resistant Escherichia coli (KREC), presenting great potential for application in clinical therapy.

GSH-depleting and H2O2-self-supplying hybrid nanozymes for intensive catalytic antibacterial therapy by photothermal-augmented co-catalysis

Junqin Li,Wenhua Yi,Yuze Luo,Ke Yang,Lidan He,Caiyun Xu,Le Deng,Dinggeng He

Nanozyme-based chemodynamic therapy (CDT) has shown tremendous potential in the treatment of bacterial infections. However, the CDT antibacterial efficacy is severely limited by the catalytic activity of nanozymes or the infection microenvironments such as insufficient hydrogen peroxide (H 2 O 2 ) and overexpressed glutathione (GSH). Herein, a versatile hybrid nanozyme (MoS 2 /CuO 2 ) is rationally constructed by simply decorating ultrasmall CuO 2 nanodots onto lamellar MoS 2 platelets of hydrangea-like MoS 2 nanocarrier via a covalent Cu-S bond. The MoS 2 /CuO 2 nanozyme exhibits the peroxidase-mimic activity for catalytically converting H 2 O 2 produced by acid-triggered decomposition of the decorated CuO 2 into hydroxyl radical (•OH). Meanwhile, the MoS 2 /CuO 2 can consume GSH overexpressed in the infection sites via redox reaction mediated by polyvalent transition metal ions (Cu 2+ and Mo 6+ ) for enhanced CDT. More importantly, MoS 2 support can promote the conversion of Cu 2+ to Cu + by a co-catalytic reaction based on the Mo 4+ /Mo 6+ redox couples, and provide photonic hyperthermia (PTT) to augment the peroxidase-mimic activity. The developed MoS 2 /CuO 2 nanozymes possesses a desirable catalytic property, as well as a remarkably improved antibacterial efficiency both in vitro and in vivo . Taken together, this study proposes a synergetic multiple enhancement strategy to successfully construct the versatile hybrid nanozymes for intensive in vivo PTT/CDT dual-mode anti-infective therapy. Statement of significance Chemodynamic therapy (CDT) has shown great potentialities in the treatment of bacterial infections, while its therapeutic efficiency is severely limited by the infection microenvironments such as insufficient hydrogen peroxide (H 2 O 2 ) and overexpressed glutathione (GSH). Here, we rationally construct a hybrid nanozyme (MoS 2 /CuO 2 ) with peroxidase-like activity that can enhance CDT by regulating local microenvironments, that is, simultaneously self-supplying H 2 O 2 and consuming GSH. Importantly, MoS 2 support can promote the conversion of Cu 2+ to Cu + by the Mo 4+ /Mo 6+ redox couples, and provide photonic hyperthermia (PTT) to augment the peroxidase-mimic activity. The developed MoS 2 /CuO 2 shows desirable PTT/CDT dual-mode antibacterial efficacy both in vitro and in vivo . This study proposes a versatile hybrid nanozyme with multiple enhancement effects for intensive in vivo anti-infective therapy.
