Curcumin, a plant-derived polyphenolic compound, exhibits diverse pharmacological activities such as antioxidant, anti-inflammatory, anticancer, neuroprotective, and cardiovascular protective effects, and is widely used in food, medicine, and other fields. However, its poor water solubility and easy oxidative degradation limit its extensive application in biomedicine. To solve these problems, a series of biomedical polyurethanes (Cur-PU) with similar molecular weights but different PEG contents were successfully synthesized using HO-PCL-OH and HO-PEG-OH as soft segments and curcumin as a chain extender. The results indicated that increasing the PEG content reduced the T1m, T1c, and H1cof Cur-PU, along with a slower crystallization rate and lower crystallinity. More importantly, a higher PEG content decreased the water contact angle but increased water solubility and water uptake, which, combined with reduced crystallinity, enhanced hydrophilicity, swelling ratio, curcumin release rate, and degradation rate in an enzymatic solution and pH 8.0 buffer. Thus, precise regulation of Cur-PU’s degradation and curcumin release was achieved by controlling the PEG content. Biocompatibility tests confirmed that Cur-PU exhibited excellent antioxidant and antibacterial activities, making it a highly promising biomedical material.

Purpose To utilize a novel type of polymer-drug conjugate micelle to enhance the delivery of low-potency curcumin. Methods Multiple curcumin molecules were conjugated to poly(lactic acid) (PLA) via tris(hydroxymethyl)aminomethane (Tris) linker producing the hydrophobic drug-binding block; methoxy-poly(ethylene glycol) (mPEG) was employed as the hydrophilic block. Micelles were characterized by size, loading capacity, stability, and critical micelle concentration (CMC). Human hepatocellular carcinoma (HepG2) cells were employed to assess cytotoxicity and intracellular targeting ability of micelles. Results mPEG-PLA-Tris-Cur micelles were within nanorange (<100 nm). CMC of such micelles (2.3 ± 0.4 μg/mL) was 10 times lower than mPEG-PLA micelles (27.4 ± 0.8 μg/mL). Curcumin loading in mPEG-PLA-Tris-Cur micelles reached 18.5 ± 1.3% (w/w), compared to traditional mPEG-PLA micelles at 3.6 ± 0.4% (w/w). IC 50 of mPEG-PLA-Tris-Cur micelles (~22 μg/mL at curcumin-equivalent dose) was similar to unmodified curcumin. Placebo and drug-encapsulated conjugate micelles could be efficiently internalized to cytoplasmic compartment of HepG2 cells. Conclusions Micelle-forming polymer-drug conjugates containing multiple drug molecules were an efficient means to increase loading and intracellular delivery of low-potency curcumin.

Acrolein (ACR) is a toxic unsaturated aldehyde that is formed during different steps of thermal food processing. Here, we explored the kinetics of curcumin and ACR and elucidated the pathway of curcumin trapping ACR by preparing a mono-adduct of ACR (CMA-1) conjugated with curcumin. The synergistic scavenging effect and mechanism of curcumin combined with quercetin on ACR was analyzed by liquid chromatography–tandem mass spectrometry (LC–MS/MS). Comparing the uses of curcumin and quercetin both individually and in combination, we found that quercetin in combination resulted in more curcumin being transformed into CMA-2, while curcumin in combination made the amount of di-ACR conjugated to quercetin (QDA) increase. We also added combined curcumin and quercetin into grilled chicken wings to demonstrate that curcumin and quercetin could scavenge ACR by forming their own ACR adducts and antioxidant activity during the process. Our results have noted a new strategy, in which some combinations of dietary polyphenols might contribute to the removal of toxic ACR produced during thermal food processing.

A facile approach was investigated to encapsulate and protect curcumin (Cur) by self-assembly of lysozyme (Ly) and carboxymethylcellulose (CMC) of different degrees of substitution (DSs). This work studied the influence of Ly–CMC coacervates on the binding, solubility and stability of Cur. The interactions of Cur with Ly–CMC coacervates were researched by UV–vis, steady-state fluorescence, synchronous fluorescence and circular dichroism spectra. These results were explained in terms of the formation of Ly–CMC coacervates with electrostatic interaction, which led to unfold the structure of Ly for providing Cur with more hydrophobic microenvironment than free Ly. Meanwhile, the CMC of higher DS provide more negative charges, and produce more attraction to Ly than that of lower DS values. Moreover, the coacervates of Ly–CMC of higher DS are found to be superior for enhancing the stability of Cur. Our work provided a new insight for understanding the biomolecule protective system based on protein/polysaccharide complexes.

Ultrasound has a recognized ability to modulate the structure and function of proteins. Discovering the influential mechanism of ultrasound on the intramolecular interactions of egg-white protein isolate–curcumin ( EPI –Cur) nanoparticles and their intermolecular interaction during freeze drying and redispersion is meaningful. In this study, under the extension of pre-sonication time, the protein solubility, surface hydrophobicity, and curcumin encapsulation rate showed an increasing trend, reaching the highest value at 12 min of treatment. However, the values decreased under the followed extension of ultrasound time. After freeze drying and redispersion were applied, the EPI –Cur sample under 12 min of ultrasound treatment exhibited minimal aggregation degree and loss of curcumin. The retention and loading rates of curcumin in the lyophilized powder reached 96 % and 33.60 mg/g EPI , respectively. However, under excessive ultrasound of >12 min, scanning electron microscopy showed distinct blocky aggregates. Overexposure of the hydrophobic region of the protein triggered protein-mediated hydrophobic aggregation after freeze drying. X-ray diffraction patterns showed the highest crystallinity, indicating that the free curcumin-mediated hydrophobic aggregation during freeze drying was enhanced by the concentration effect and intensified the formation of larger aggregates. This work has practical significance for developing the delivery of hydrophobic active substances. It provides theoretical value for the dynamic dispersity change in protein-hydrophobic active substances during freeze drying and redissolving.

Graphical In this study, the reduction responsive prodrug folic acid-polyethylene glycol-disulfide bond-curcumin (FA-PEG-SS-CUR) was synthesized. The prodrug could self-assemble into micelles in an aqueous solution. The chemical structure of the prodrug was determined using FTIR and NMR. Subsequently, the properties of the micelles were evaluated through TEM, particle size analysis, cell experiments, and in vitro drug release assessment. In order to improve the delivery performance of curcumin (CUR) in liver cancer cells, we prepared a targeted and reduction-sensitive prodrug, folic acid-polyethylene glycol-disulfide bond-curcumin (FA-PEG-SS-CUR). The synthesis involved the conjugation of CUR with NH 2 -PEG-NH 2 through a disulfide bond, followed by the connection of CUR-SS-PEG-NH 2 to FA-COOH via acylation. The resultant FA-PEG-SS-CUR prodrug self-assembled into nano micelles in aqueous solutions, displaying excellent dispersibility with an approximate particle size of 119 nm. In vitro drug release studies demonstrated the sensitivity of FA-PEG-SS-CUR to glutathione (GSH), displaying sustained release performance. After 72 h, FA-PEG-SS-CUR exhibited a drug release rate of 38% at low GSH concentration and 59% at high GSH concentration. Notably, FA-PEG-SS-CUR micelles exhibited favorable biocompatibility. The hemolysis rate resulting from co-incubation with red blood cells remained below 3%. Furthermore, cytotoxicity experiments unveiled the substantial cytotoxicity of FA-PEG-SS-CUR micelles against HepG2 tumor cells. Cellular uptake studies confirmed the greater internalization of FA-PEG-SS-CUR by HepG2 cells compared to the non-targeted PEG-SS-CUR. The findings highlight the potential of FA-PEG-SS-CUR as an ideal drug delivery system for CUR due to its facile synthesis and strong self-assembly performance.

Curcumin, a natural polyphenol, has various biological functions including antioxidant and antimicrobial properties, but the functions are limited by its low aqueous solubility. To address this, two proteins, bovine serum albumin (BSA) and gelatin were used to form curcumin–protein nanocomplexes using a charge-switch method, by mixing oppositely charged curcumin and proteins. Complexation with BSA and gelatin increased the curcumin solubility to 391.77 (±15.70) μg mL−1 and 143.64 (±2.29) μg mL−1 respectively, and the loading amount (LA) of curcumin to 21.36% and 15.57%, respectively. Moreover, enhanced bioaccessibility, and antioxidant and antimicrobial properties were observed after complexation. After complexation, the minimum inhibitory concentration (MIC) of curcumin decreased by eightfold against Vibrio parahaemolyticus, more than eightfold against Vibrio harveyi, and twofold against Bacillus cereus and Streptococcus iniae, which are food and aquaculture related pathogens. The curcumin–protein nanocomplexes presented in this work could serve as non-antibiotic additives for preventing and managing microbial diseases in agri-food applications.

Maize porous starch-curcumin microspheres were prepared by encapsulating curcumin into cross-linked porous starch and oxidized porous starch to investigate the effect of modified porous starch in embedding and protecting curcumin. The morphology and physicochemical properties of microspheres were analyzed using scanning electron microscopy, Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction, Zeta/DLS, Thermal stability, and antioxidant activity; the release of curcumin was evaluated with a simulated gastric-intestine model. The FT-IR results revealed that curcumin was amorphously encapsulated in the composite and hydrogen bond formation between starch and curcumin was one of the major driving forces for encapsulation. Microspheres increased the initial decomposition temperature of curcumin, which has a protective effect on curcumin. Modification improved the encapsulation efficiency and the scavenging free radical ability of porous starch. The release mechanism of curcumin from microspheres fits first-order and Higuchi models well in gastric and intestinal models, respectively, indicating that encapsulation of curcumin within different porous starches microspheres enables controlled release of curcumin. To recapitulate, two different modified porous starch microspheres improved the drug loading, slow release and free radical scavenging effects of curcumin. Among them, the cross-linked porous starch microspheres had higher encapsulation and slow release ability for curcumin than the oxidized porous starch microspheres. It provides theoretical significance and data basis for the encapsulation of active substances by modified porous starch.

Comprehensive Summary The intensive use of antibiotics intensifies the development of bacterial resistance, which has become a serious problem globally. Methicillin-resistant Staphylococcus aureus (MRSA) has resulted in significant morbidity and mortality. Therefore, it is an urgent need to develop new antimicrobial drugs and administration methods. Herein, we report a dual functional diblock copolymer PLL 20 - b -PBLG 20 , which was prepared by superfast and water-insensitive polymerization on N -carboxyanhydrides (NCA) initiated by tetraalkylammonium carboxylate. In addition to direct antimicrobial activity, PLL 20 - b -PBLG 20 also exerts a synergistic bactericidal effect against MRSA with curcumin, a plant extract with antibacterial property. Moreover, PLL 20 - b -PBLG 20 successfully encapsulates curcumin to form nanoparticles via self-assembly. The combination of dual functional PLL 20 - b -PBLG 20 and curcumin holds promise in combating MRSA infections.

A novel efficacious strategy for real-time monitoring of the release of hydrophobic cargo curcumin (molecule model nutraceuticals) from a lipid–curcumin-loaded silica colloidal crystal (L(Cur)-SCC) film controlled by lipase was developed. Curcumin was dispersed in a proportion of a digestible lipid complex (glycerol trioleate and glycerol tristearate) to prepare a lipid–curcumin complex and then loaded into the SCC film by a capillary to prepare an L(Cur)-SCC film. Lipase-triggered degradation of the digestible lipid complex resulted in curcumin release being tracked in real-time by ordered porous layer interferometry (OPLI). The optical thickness changes (ΔOT) of the L(Cur)-SCC film depend on the mass changes of the lipid–curcumin complex due to the migration of interference fringes caused by the lipase degradation of the digestible lipid complex. Curcumin release from the L(Cur)-SCC film was characterized and analyzed in combination with an ultraviolet–visible spectrophotometer, a nanoparticle size analyzer, and an attenuated total reflection infrared spectrometer. The introduction of a soluble dietary fiber (pectin) into the L(Cur)-SCC film delayed the release rate of curcumin. Furthermore, the real-time sustained release of curcumin from the L(Cur)-SCC film in the simulated digestive fluids was tracked. This study provides an early exploration of the real-time controlled release of lipid-soluble nutraceuticals in the gastrointestinal tract.

We developed a facile method for the fabrication of a biodegradable delivery system composed of two blocks: curdlan and curcumin. This was achieved by chemical functionalization of curdlan through tosylation, amination followed by complexation with curcumin. A comprehensive evaluation of structural characterization and component stability showed that cur-cum complex exhibited better anticancer properties with enhanced thermal properties. The cur-cum complex shows pH sensitive sustained release behaviour with higher release at acidic pH and kinetic data of drug release follows the Korsmeyer-Peppas model. The cur-cum complex has ability to block the proliferation of the MCF-7 cell line as revealed by MTT assay which showed increased toxicity of cur-cum complex against these cell lines. The results obtained from western blot analysis demonstrated that the co-administration of cur and cum effectively induced apoptosis in MCF-7 cells. This effect was observed by a considerable upregulation of the Bcl-2/Bax ratio, a decline in mRNA expression of LDHA, level of lactate and LDH activity. The results clearly depict the role of functionalized curdlan as efficient carrier for curcumin delivery with prolonged, sustained release and enhanced bioavailability, thereby improving the overall anticancer activity.

The use of polysaccharide Pickering emulsions for delivering bioactive compounds has attracted significant attention recently. However, the destabilizing effects of bioactive compounds on these emulsions remain largely unexplored, limiting the development of high-performance delivery systems. This study investigates how curcumin loading affects the stability and delivery performance of Lycium barbarum leaf polysaccharide Pickering emulsions. Curcumin loading significantly destabilizes these emulsions, reducing stability indices of mineral-containing emulsions from 92.3 %–94.8 % (unloaded) to 31.7 %–90.7 % (loaded), and demineralized emulsions from 96.4 %–97.6 % (unloaded) to 0 % (loaded). Mechanistic studies reveal that curcumin interacts with polysaccharides at the oil-water interface through hydrogen bonds and hydrophobic interactions, forming large (556.42–652.38 nm), highly wettable (contact angle θ o/w of 49.3°-63.8°) composite particles that disrupt the emulsion network. Simulated digestion studies show that demineralized emulsions have higher free fatty acid release rates (71.6 %–74.9 %) and lower curcumin bioaccessibility (33.5 %–34.2 %), compared to mineral-containing emulsions with rates of 44.2 %–62.5 % and bioaccessibility of 36.8 %–41.3 %. This research provides critical insights for improving food emulsion applications.
