Cyclocarya paliurus , as an important edible and medicinal plant, its effects and mechanisms on hyperuricemic nephropathy remain unclear. Herein, we investigated the hypouricemic and nephroprotective effects of the water extracts of C. paliurus leaves (CPE) in a hyperuricemic nephropathy rat model induced by adenine and ethambutol. The results showed that CPE could significantly decrease plasma uric acid (PUA) and urea nitrogen (PUN), as well as reduce renal fibrosis in the hyperuricemic nephropathy rat model. Plasma metabolomics indicated that CPE improved the disordered arachidonic acid metabolism. Meanwhile, CPE reduced renal inflammation via inhibition of COX--2. Moreover, CPE could improve disturbed purine metabolism by inhibiting the hepatic xanthine oxidase (XOD), reversing the renal urate transporter 1 (URAT1) and organic anion transporter 1 (OAT1). Collectively, this study demonstrated that CPE could exhibit hypouricemic effect by improving purine metabolism, and attenuate kidney injury by improving arachidonic acid metabolism and alleviating kidney inflammation.

Polymer microspheres with inter-connecting pores are widely used as microsphere materials. In the study, the ethyl cellulose microspheres (ECM) were prepared by using the solvent-evaporation method. Based on that, a method for the separation and purification of benzoic acid from crude extract ofCyclocarya paliuruswas established by the ECM and high performance liquid chromatography (HPLC). The ECM after the sorption equilibrium was desorbed by using 40% methanol as the analytical solvent. The content of benzoic acid in eluent is up to 0.0216 mg/mL, and the benzoic acid can be obtained with a high purity of 82.22%. Furthermore, the adsorption-desorption behavior of benzoic acid onto ECM was investigated. The results of adsorption kinetics of benzoic acid showed that the adsorption followed the pseudo-first-order kinetic model. The ECM was characterized by using Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), scanning electron microscope (SEM), and X-ray diffraction (XRD). The results showed that the ECM has a high adsorption property due to its more porous structure, phenolic hydroxyl group, and other oxygen-containing functional groups. This method and the ECM can be used stably, continuously, and efficiently to purify the benzoic acid from the methanol extract ofC. paliuruson a large scale. Keywords:ethyl cellulose microspheres (ECM);HPLC;adsorption-desorption;benzoic acid;Cyclocarya paliurus

Diabetes Mellitus is a complex metabolic disorder, primarily characterized by persistent high blood sugar levels, and it is becoming increasingly prevalent with numerous associated complications. The leaves ofCyclocarya paliurus(Batal.) Iljinskja, traditionally prepared as a tea beverage in China, is frequently used in folk medicine for managing metabolic syndromes, particularly diabetes and hyperlipidemia. However, the main active components responsible for its hypoglycemic effect and their underlying mechanisms remain unclear. The current study aimed to clarify the main chemical components of the aqueous extract ofC. paliurusleaves and to explore their mechanisms of action. The primary constituents from the aqueous extract ofC. paliurusleaves were isolated and identified using macroporous adsorption resin, preparative liquid chromatography, and nuclear magnetic resonance technology. The contents of these identified compounds in the leaves were quantified using HPLC. An integrated approach combining network pharmacology and molecular docking was initially used to predict the potential molecular targets and associated signaling pathways responsible for the hypoglycemic activity of the compounds, with subsequent experimental validation performed in a hyperglycemic zebrafish model. From the aqueous extract, a total of seven compounds were obtained and subsequently identified as Chlorogenic Acid (CA), Quercetin-3-O-β-D-glucuronide (Q3GA), Astragalin, 3,4-Dicaffeoylquinic Acid (3,4-DCA), Afzelin, Quercetin, and Kaempferol. Their contents inC. paliurusleaves, as determined by HPLC, were 24.88 mg/g, 30.87 mg/g, 1.21 mg/g, 1.19 mg/g, 5.24 mg/g, 2.43 mg/g, and 1.34 mg/g, respectively. Network pharmacology analysis identified AKT1, TNF, and IL1B as key targets for the hypoglycemic effects of both the aqueous extract and the seven individual compounds. These findings were further supported by RT-PCR experiments in a zebrafish model, which showed that blood glucose regulation occurs through the downregulation of TNF and IL1B and the upregulation of AKT1 protein. The aqueous extract is rich in Chlorogenic Acid, Quercetin, and their derivatives, all of which display significant hypoglycemic activity.

This research investigated the variations of triterpenoid contents in Cyclocarya paliurus leaves and their underlying anticancer activity and mechanism. Leaf samples were collected from twelve geographical locations in China for the experiments. Significant variation in the triterpenoid content and growth inhibition on HeLa cells were observed among the C. paliurus populations. Antiproliferation effects of the leaf samples were significantly affected by the pentacyclic triterpenoids contents except for oleanolic acid. Among the pentacyclic triterpenoids, hederagenin exhibited the best anticancer activity with IC 50 of 17.42 μg/mL. Based on the results, pentacyclic triterpenoids may induce HeLa cell apoptosis via mitochondrial pathway. RNA sequencing indicated that the ACTB , ATF4 , TUBA1B and TUBA1C genes might be the key ones involved in the apoptosis induction by cyclocaric acid B and hederagenin. These results provided useful information for application of C. paliurus in functional beverage and pharmaceutical industries.

Cyclocarya paliurus is an edible and medicinal plant exhibiting significant hypoglycemic effect. However, its active components are still unclear and need further elucidation. In this research, the active components of the leaves of C. paliurus responsible for the α -glucosidase inhibitory activity were screened and identified based on a spectrum–effect relationship study in combination with ultra-performance liquid chromatography tandem mass spectrometry (UPLC–MS/MS) analysis. The 70% ethanol eluate fraction of the leaves of C. paliurus with the strongest α -glucosidase inhibitory activity was obtained after extraction and purification with macroporous resin. Their chromatographic fingerprints (15 batches) were established by UPLC analysis and 32 common peaks were specified by similarity analysis. Their IC 50 values for α -glucosidase inhibition were measured by an enzymatic reaction. Several multivariate statistical analysis methods including hierarchical cluster analysis, principal component analysis, partial least square analysis and gray relational analysis were applied to explore the spectrum–effect relationship between common peaks and IC 50 values, and the chromatographic peaks making a large contribution to efficacy were screened out. To further elucidate the active components of leaves of C. paliurus , the 70% ethanol eluate fraction was characterized by UPLC–MS/MS analysis, and 10 compounds were identified. This study provides a valuable reference for further research and development of hypoglycemic active components of C. paliurus .

In this study, gelatin-chitosan (GEL-CS) composite films added with 0.1 %, 0.2 %, and 0.3 % Cyclocarya paliurus flavonoids (CPF) were prepared. Then their appearance properties, mechanical properties, barrier properties, microstructure, thermal stability properties, antioxidant activity , and antibacterial properties were investigated. As compared with GEL-CS film, the GEL-CS films with CPF were darker in color, had higher water vapor barrier, higher elongation at break, and higher thermal stability. Additionally, microstructure analysis with Fourier infrared spectroscopy, scanning electron microscopy, and X-ray diffraction demonstrated that hydrogen bonding was the main force for cross-linking CPF with other membrane substrates. Moreover, the addition of CPF strengthened the antioxidant and antimicrobial properties of the membranes. These results indicated that the CPF addition could endow membranes with more excellent functional properties and bioactivity, accompanied by environmentally friendly and edible features. The GEL-CS-CPF composite film would be a potential and prospective packing material for food preservation applications.

The escalating threat of methicillin-resistant Staphylococcus aureus (MRSA) necessitates novel therapeutic strategies. Our previous work suggested that an extract from Cyclocarya paliurus leaves (ECPL) inhibits MRSA by targeting the cell division protein FtsZ. Here, guided by anti-MRSA activity, we isolated three compounds from ECPL: asiatic acid (AA), maslinic acid (MA), and ursolic acid (UA). They exhibited antibacterial activity against MRSA and induced cell elongation, indicative of division arrest. Time-kill assays showed AA and MA are bactericides, while UA is bacteriostatic. Mechanistically, these compounds disrupt cell division by differentially affecting FtsZ dynamics: AA promotes polymerization, whereas MA and UA inhibit it. SPR analysis showed direct FtsZ binding to AA (Kd = 2.4 μM), MA (Kd = 9.8 μM), and UA (Kd = 0.7 μM). Molecular docking predicted a shared FtsZ binding pocket but revealed that AA adopts a distinct conformation driven by unique interactions, including a hydrogen bond with Arg191—an interaction not observed for MA or UA, which instead form hydrogen bonds with Thr265 and Thr309. Despite these divergent effects on polymerization and distinct binding modes, all compounds ultimately disrupted Z-ring assembly and septum formation. In a murine skin infection model, AA, selected for its bactericidal activity and unique FtsZ modulation mechanism, significantly reduced bacterial burden and accelerated wound healing. Collectively, our findings validate these compounds as direct FtsZ-targeting agents and establish AA as a promising anti-MRSA lead compound with a novel mechanism disrupting the bacterial divisome.

Cyclocarya paliurus (CP) polysaccharide (CPP) is a chemical component contained in CP, which has been reported to possess significant hypoglycemic activity. The present study aimed to investigate the radiosensitizing effect and underlying mechanisms of CPP on hypoxic A549 and H520 human non‑small cell lung carcinoma cells. Cell viability, apoptosis and proliferation were determined using Cell Counting kit‑8 assay, flow cytometry and colony formation assay, respectively. mRNA and protein expression levels were determined by reverse transcription‑quantitative PCR and western blot analysis, respectively. The results suggested that CPP markedly inhibited the viability of hypoxic A549 and H520 cells. In response to combined treatment with CPP and radiation, hypoxic A549 and H520 cells exhibited enhanced apoptosis; in addition, cell proliferation was suppressed and the expression levels of hypoxia‑inducible factor‑1α, survivin and cleaved caspase‑3 were modified. Furthermore, CPP in combination with radiation affected the mammalian target of rapamycin (mTOR)/Akt/phosphatidylinositol‑4,5‑bisphosphate 3‑kinase (PI3K) pathway. These findings indicated that CPP may enhance the radiosensitivity of hypoxic A549 and H520 cells; this effect may be associated with inhibition of the mTOR/Akt/PI3K pathway. The potential radiosensitizing effects of CPP on hypoxic A549 and H520 cells suggested that CPP may be an effective target for treatment of non‑small cell lung carcinoma.

Methicillin-resistant Staphylococcus aureus (MRSA) has evolved to tolerate almost all known β-lactam antibiotics, and been exhibiting resistance to vancomycin . Developing new drugs against the MRSA infection are urgently needed. Natural resource is enriched with antimicrobial molecules. Here, we report an extract from leaves of Cyclocarya paliurus (ECPL), a species of tree native to China and being traditionally cultivated as medicinal plant , which shows a broad spectrum of bacteriostatic and bactericidal effects at MIC 50 15.99 ± 0.55 μg/ml against methicillin-susceptible and -resistant S. aureus . The ECPL suppresses bacterial expression of FtsZ at both transcriptional and protein levels, inhibits its GTPase activity, blocks the assembly dynamics of FtsZ, and disrupts the formation of Z-ring. The bacteria cells fail to divide, enlarge initially, and lyse eventually. In mouse model, the ECPL facilitates skin wound healing with MRSA infection as effective as vancomycin. Our study unveils the potential of identifying novel molecule in ECPL for development of new class of antibiotic against both methicillin-susceptible and -resistant S. aureus , and support domestication of the C. paliurus growing from wild to cultivated, and further scale up the cultivation into industrialized farm for sustainable harvest for pharmaceutic interest.

Arjunolic acid (AA), as the most abundant triterpenoid component in Cyclocarya paliurus (Batal) Iljinskaja, previously displayed delipidating effects in free fatty acid (FFA)-induced HepG2 steatosis cells. However, whether AA still possesses the ameliorative effects on nonalcoholic fatty liver disease (NAFLD) in normal hepatocytes and in vivo remains vacant and the detailed mechanisms are not defined. In vitro , AA dose-dependently alleviated lipid accumulation in FFA-challenged primary hepatocytes without cytotoxicity. In vivo , AA showed pleiotropic benefits, including attenuating adiposity, mitigating hepatic steatosis and inflammation, improving lipid disorders and insulin resistance, and restoring the impaired gut barrier. Mechanically, we found that AA indirectly activated Sirt1/AMPK-modulated lipid metabolism through elevating NAMPT-mediated NAD + level, and triggering autophagy, together mediating the lipid-lowering effects. Collectively, our results convey that AA can substantially mitigate NAFLD via indirectly activating Sirt1/AMPK signaling, inducing autophagy and restoring gut barrier, and will be considered as a promising candidate for NAFLD therapy.
