Tetrabromobisphenol A exposure compromises antioxidant defense and mitochondrial ultrastructural damage, exacerbating hepatocellular apoptosis in Cyprinus carpio

Yimin Li,Xiaolong Gao,Chunnuan Zhang,Zihao Meng,Mengkang You

Tetrabromobisphenol A (TBBPA), widely employed as a flame retardant in electronic products and plastics, has been routinely detected in multiple environmental matrices including aquatic compartments and sediments. The common carp ( Cyprinus carpio ), an established model organism in toxicological research, was utilized in this investigation. This study aimed to evaluate the hepatotoxic effects of varying TBBPA concentrations through histological and molecular biochemical parameter analysis. Fish were administered TBBPA at concentrations of 0, 0.005, 0.05, and 0.5 mg/L for 14 days. Collectively, this investigation demonstrated that TBBPA exposure elicits multiple pathological alterations in piscine hepatic tissue, encompassing ultrastructural damage, oxidative stress, calcium homeostasis dysregulation, and apoptosis. Histopathological analysis revealed that exposure to 0.005 mg/L TBBPA induced mitochondrial and endoplasmic reticulum swelling. Biochemical analyses demonstrated TBBPA-mediated disruption of hepatic redox homeostasis, consequently triggering lipid peroxidation. Furthermore, TBBPA exposure resulted in hepatic calcium overload, characterized by diminished Ca 2+ -ATPase activity and elevated intracellular Ca 2+ concentrations. Concurrently, TUNEL assay and qRT-PCR results indicated concentration-dependent induction of apoptosis by TBBPA. This study demonstrates hepatotoxic effects induced by environmentally relevant TBBPA concentrations while elucidating potential toxicological mechanisms in the piscine liver. These findings advance the understanding of TBBPA-mediated pathological impacts on the fish liver.

Distinct responses from triglyceride and cholesterol metabolism in common carp (Cyprinus carpio) upon environmental cadmium exposure

Wenli Guo,Beibei Gao,Xiaoqian Zhang,Quanzhong Ren,Dizhi Xie,Junping Liang,Hui Li,Xianfeng Wang,Yuru Zhang,Sijin Liu,Guoxing Nie

Due to high persistence and bioavailability, Cadmium (Cd) is one of the most prevalent environmental contaminants, posing an elevating threat to the ecosystems. It has been evidenced that high-dose Cd elicits deleterious effects on aquatic organisms, but the potential toxicities of Cd at environmentally relevant concentrations remains underappreciated. In this study, we used common carp to investigate how environmental Cd exposure affects triglyceride (TG) and cholesterol metabolism and underlying mechanisms. The data indicated that Cd resulted in the shift of TG from the liver to blood and the movement of cholesterol in the opposite direction, ultimately giving rise to the storage of crude lipid in liver and muscle, especially hepatic cholesterol retention. Cholesterol, instead of TG, became the principal cause during the progression of hepatic lipid accumulation. Mechanistic investigations at transcriptional and translational levels further substantiated that Cd blocked hepatic biosynthesis of TG and enhanced TG efflux out of the liver and fatty acid β-oxidation, which collectively led to the compromised TG metabolism in the liver and accelerated TG export to the serum. Additionally, strengthened synthesis, retarded export and oxidation of cholesterol detailed the hepatic prominent cholesterol retention. Taken together, our results demonstrated that environmental exposure to Cd perturbed lipid metabolism through triggering distinct responses from hepatic TG and cholesterol homeostasis. These indicated that environmental factors (such as waterborne Cd) could be a potential contributor to the prevalence of non-alcoholic fatty-liver disease in aquaculture and more efforts should be devoted to the ecological risk assessment of pollutants under environmental scenarios.

Melatonin alleviates lead-induced fatty liver in the common carps (Cyprinus carpio) via gut-liver axis

Zhiying Miao,Zhiruo Miao,Xiaohua Teng,Shiwen Xu

As a widespread aquatic environmental contaminant, Lead (Pb) can provoke hepatic injury in various animals. Melatonin (MT) plays a crucial role in the regulation of inflammatory response. Accumulating evidence elucidates exogenous toxins can elicit hepatic lipid metabolic disorders by influencing the gut microbiome. Nevertheless, the effects of Pb on gut microbiota and hepatic lipid metabolism of the common carps, and whether MT can prevent and cure Pb-induced toxicity via regulating microbiome remains unknown. Here, metagenomic and transcriptomic analysis were subsequently implemented to identify the Pb exposure-triggered prominent alternation of gut-liver signal. In the present study the severe intestinal injury and fatty liver formation caused by Pb in common carp were preliminarily determined. Metagenomic analysis confirmed that the gut microbiome dominant phyla, family and genus of the common carps were Fusobacteria , Fusobacteriaceae and Cetobacterium . Meanwhile, lipopolysaccharide (LPS) biosynthesis pathway was regarded as one of the main responsible for Pb exposure. Subsequently, LPS was demonstrated as the Pb-triggered microbial-derived signal of the common carps by ELISA analysis, and involves in the hepatic metabolic disorders via deteriorating the intestinal barrier. Additionally, it confirmed that hepatocytes ferroptosis associated with Pb-evoked fatty liver of the common carps, and the aggravation of lysosomal dyshomeostasis as well as inhibition of AMPK phosphorylation were referred to lipid metabolic disorders. The results of the present study demonstrated microbial-derived signal induced by aquatic Pb contaminant cause fatty liver formation in the common carps, and the protective effects of MT on Pb toxicity were performed by receding LPS over-synthesis, restraining microbiota-sourced LPS transport, along with attenuation of hepatocytes ferroptosis.

MiRNA-194 targets MyD88a to regulate immune response in Cyprinus carpio after recovery from gossypol toxicity

Hui Li,Shuangfeng Li,Liming Zhao,Shuying Zhang,Jing Zhang,Xianfeng Wang,Junping Liang

Cottonseed meal has been one of the predominant protein feedstuffs used in aqua feed, which contains gossypol . It is well known that gossypol, a natural polyphenolic compound found in seeds, can be toxic to fish. The toxicity of gossypol persists even after a recovery period; however, the underlying mechanism remains largely unknown. To investigate the toxic mechanism of gossypol after the recovery period, Cyprinus carpio were administered a continuous oral dose of 20 mg/kg·d gossypol (gossypol acetate dose/fish weight·d) for one week, followed by a two-week recovery period. The spleen index, spleen morphology, lysozyme (LZM) activity, acid phosphatase (ACP) activity and nitric oxide (NO) concentration were obviously changed in the spleens of C. carpio after two weeks of recovery. Then, miRNA sequencing identified 22 differentially expressed miRNAs in the spleens of C. carpio two weeks after the gossypol treatment. The mRNA expression levels of immune-associated genes were significantly increased in the spleens after recovery. MiR-194 was chosen for future study through a search of existing literature and an analysis of miRNA and mRNA expression levels. To further characterize the potential target of miR-194, we identified putative miR-194 binding sites in the 3′UTR of myeloid differentiation factor 88a (MyD88a). Dual-luciferase reporter assays revealed that miR-194 downregulated the expression level of MyD88a through binding to seed-matching sequence in the 3′ UTR of MyD88a. Moreover, C. carpio were intraperitoneally administered with miR-194 antagomir. Furthermore, down-regulation of miR-194 increased the expression levels of MyD88a and some pro-inflammatory cytokines and anti-inflammatory cytokines. Altogether, the results demonstrate miR-194 can target MyD88a to regulate the immune response of C. carpio after a two-week recovery period following gossypol administration.

Bioaccumulation and elimination, acute toxicity analysis and risk assessment of diflubenzuron in common carp (Cyprinus carpio)

Lu Qiao,Lu Chen,Zhen Yang,Jinhua Xu,Jinlong Song,Xingyang Li,Huiwu Sun,Yingchun Mu

Diflubenzuron has been applied in agriculture and aquaculture, and its residues in ecological environment and food chain could result in chronic exposure and long-term toxicity effects for human health. However, limited information is available regarding diflubenzuron levels in fish and associated risk assessment. This study performed the analysis for dynamic bioaccumulation and elimination distribution of diflubenzuron in carp tissues. The results indicated that diflubenzuron was absorbed and enriched by fish body along with higher enrichment in lipid-rich tissues of fish. The peak concentration in carp muscle reached 6-fold of diflubenzuron concentration in aquaculture water. The median lethal concentration (LC 50 ) of diflubenzuron at 96 h was 12.29 mg/L, presented low toxicity to carp. Risk assessment results showed that the chronic risk from dietary exposure to diflubenzuron through carp consumption for Chinese residents of children and adolescents, adults and elderly people were acceptable, while posed a certain risk for young children . This study provided the reference for pollution control , risk assessment and scientific management of diflubenzuron.

Lipotoxic Effects of Gossypol Acetate Supplementation on Hepatopancreas Fat Accumulation and Fatty Acid Profile in Cyprinus carpio

Yuru Zhang,Junmei Zhang,Yanhua Ren,Mengdan Zhang,Junping Liang

Cottonseed meal is an excellent protein source with low price and abundant nutrients, but the existence of free gossypol restricts its wide application in animal feeds. To explore the toxic effects of free gossypol, the effects of gossypol acetate on the growth performance, hepatopancreatic fat accumulation, and muscle fatty acid composition of the common carp (Cyprinus carpio) were investigated in the present study. Carp were randomly divided into four groups, which were fed a diet supplemented with 0, 170, 340, and 680 mg/kg gossypol acetate, respectively. After 60 days of feeding, the gossypol residue in the tissue, growth performance, blood biochemical indices, hepatopancreas histology, fatty acid components, and expression of key genes involved in lipid metabolism pathways were measured. It was found that the concentrations of free gossypol were increased significantly in the serum, bile, and hepatopancreas of carp fed 340 mg/kg and 680 mg/kg gossypol acetate (). Dietary gossypol acetate had a limited effect on growth performance, and high concentrations of gossypol acetate induced excessive hepatopancreatic fat deposition and hepatocyte injury in C. carpio. In addition, gossypol acetate decreased total polyunsaturated fatty acids (PUFAs) and docosahexaenoic acid in muscle. In the hepatopancreas, decreased HSL, PPARα, and CD36 may result in fat accumulation. In the muscle, decreased FADS2 may be the key reason for the decrease in PUFAs. Taken together, these findings indicated that dietary gossypol acetate can cause lipid metabolism disorders, such as hepatopancreatic damage and significant changes in tissue fatty acid profile, which are regulated by the key lipid metabolic genes in common carp.

Cross-linking effects of EGCG on myofibrillar protein from common carp (Cyprinus carpio) and the action mechanism

Chong Tan,Qian-Da Xu,Nan Chen,Qiang He,Qun Sun,Wei-Cai Zeng

The cross-linking effects and action mechanism of epigallocatechin gallate (EGCG) on myofibrillar protein from common carp ( Cyprinus carpio ) were investigated. According to particle size, zeta potential, and atomic force microscopy, EGCG could cause the aggregation of myofibrillar protein, while hydrogen bonds and electrostatic interactions were the main molecular forces. With the measurement of Fourier transform infrared spectrum, surface hydrophobicity, fluorescence spectrum, circular dichroism spectrum, and molecular dynamics simulation, EGCG could make the spatial configuration of myofibrillar protein loose, enhance the exposure of amino acid residues, and further change its secondary and tertiary structures by forming intermolecular and intramolecular hydrogen bonds with myofibrillar protein. In addition, the gel properties of myofibrillar protein were improved by EGCG. All results suggested that EGCG had the cross-linking effects on myofibrillar protein in carp meat and could further improve its properties, which showed the potential to improve the qualities of fish meat in food industry. Practical applications Compared with other meat, fish meat is particularly easy to break and deteriorate during its processing and sales due to the short length and low cross-linking degree of fish myofibrillar protein, which shows some negative impacts on the quality of fish meat. In the present study, epigallocatechin gallate (EGCG) showed the significant cross-linking effects on carp myofibrillar protein and further improved its physicochemical properties. All results suggested that EGCG had the potential to increase the cross-linking degree of fish myofibrillar protein and improve its properties, so as to ameliorate the quality of fish meat during processing and storage.

Chlorpyrifos caused necroptosis via MAPK/NF-κB/TNF-α pathway in common carp (Cyprinus carpio L.) gills

Jianqing Chen,Bing Shao,Jinliang Wang,Zhiqiang Shen,Honggui Liu,Shu Li

Chlorpyrifos (CPF) is an organophosphate insecticide and can cause cell death of animals. In the study, the common carp were exposed to CPF at 0 μg/L (the control group), 1.16 μg/L (the low dose group), 11.6 μg/L (the medium dose group), and 116 μg/L (the high dose group), respectively. The carp were euthanized at the 30th day and gills were collected immediately. The ultrastructural and histopathological observations showed obvious necrosis characteristics and inflammatory injury in the CPF-treated groups. CPF exposure activated the MAPK pathway, in which the mRNA and protein expressions of extracellular signal-regulated (ERK), p38 MAP kinase (p38), and c-Jun N-terminal kinase (JNK) were increased; the mRNAs and proteins of NF-κB and TNF-α were activated; and the mRNAs and proteins of necroptosis related genes were changed (the mRNA and protein expression of RIPK1 , RIPK3 , MLKL, and FADD were increased and caspase-8 was decreased) with concentration dependency. Taken together, we concluded that CPF exposure activated the MAPK/NF-κB/TNF-α pathway, promoted inflammatory injure and evoked necroptosis in common carp gills. In addition, CPF-induced inflammation and necroptosis was concentration dependency. The toxic effects of CPF on gills provided data for both aquaculture and toxicological studies.

Dietary (−)-Epicatechin supplementation regulates myofiber development, fillet quality, and antioxidant status of Yellow River carp (Cyprinus carpio)

Jiali Mi,Dan Liu,Chaobin Qin,Xiao Yan,Peng Pang,Yinghao Yun,Luming Wang,Guoxing Nie

(−)-Epicatechin (EC) exhibits antioxidant effects and strengthens skeletal muscle growth and differentiation in mammals. However, there are no reports about the effects of EC in fish. This study evaluated the effects of EC on muscle antioxidant capacity, myofiber development, texture characteristics, and histomorphology were also investigated through feeding trial. 300 fish (16.27 ± 0.24 g) were fed four diets with different levels of EC (0, 100, 500, and 1000 mg/kg) for 60 days. The result showed that 1000 mg/kg EC supplementation enhanced antioxidant capacity and protein content in the muscle. Meanwhile, higher expression levels of proliferating cell nuclear antigen ( pcna ), myogenic differentiation antigen ( myod ), myogenin , myosin myosin heavy chain ( myhc ), and myosin light chain ( mylc ) were detected in the muscle of EC treatment, while the myostatin ( mstnb ) mRNA level was decreased in the EC 1000 group, the similar trends were observed in the protein expression levels of MyoD , Myogenin, MyLc ( P  < 0.05). Muscle hardness and pH value were increased significantly in EC 500 and 1000 groups ( P  < 0.05). Additionally, myofiber density, small-sized fiber percentage, and expression of genes related to AMP-activated protein kinase (AMPK) signaling pathway was promoted in the EC groups. These results indicated that dietary EC improved fillet quality and promoted myofiber development in Yellow River carp , which might be related to AMPK pathway.

The Effect of High Levels Carbohydrate on Intestinal Microbiota, Metabolites, and Health of Common Carp (Cyprinus carpio L.)

Jinrui Xiong,Liping Yang,Luming Wang,Shaoyang Zhi,Mengjuan Zhao,Chunchu Xu,Leya Qu,Xiaorui Guo,Xiao Yan,Chaobin Qin,Guoxing Nie

Long-term consumption of high-carbohydrate feed may adversely affect intestinal health of fish; however, the underlying roles remain ambiguous. This study examined the effects of varying carbohydrate levels on the intestinal flora of common carp and assessed how microbial metabolites influence intestinal health. Two hundred seventy common carps were chosen and distributed randomly into three groups that fed diets containing starch at levels of 15% (low-carbohydrate diet [LCD]), 28% (medium-carbohydrate diet [MCD]), and 45% (high-carbohydrate diet [HCD]) for 60 days. A significant increase in final body weight, weight gain rate, and specific growth rate within the MCD group, while feed conversion ratio exhibited a decrease in comparison to the other groups ( p < 0.05). Feeding with a HCD led to decreased activity of catalase and increased malondialdehyde content, which was consistent with reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR) analysis results ( p < 0.05). Specifically, the RT-qPCR results revealed that HCD treatment significantly upregulated il1β , il6 , and il8 transcript levels. Whereas, the il10 messenger RNA (mRNA) was markedly reduced in comparison to the LCD group. Furthermore, the HCD group exhibited an increased abundance of Proteobacteria , accompanied by a reduction in Fusobacteria abundance, and also revealed an upsurge in opportunistic pathogenic bacteria, such as Aeromonas and Shewanella . The correlation analysis demonstrated negative correlations of anti-inflammatory active substances such as fucoxanthin, (S)-reticuline, hecogenin, and uridine with Aeromonas , but positive correlations with Luteolibacter . In summary, dietary carbohydrates might mediate intestinal flora to regulate their metabolites and affect intestinal inflammatory response.

Facilitated Bioaccumulation of Perfluorooctanesulfonate in Common Carp (Cyprinus carpio) by Graphene Oxide and Remission Mechanism of Fulvic Acid

Liwen Qiang,Meng Chen,Lingyan Zhu,Wei Wu,Qiang Wang

As one of the most popular carbon-based nanomaterials, graphene oxide (GO) has the potential to be released in aquatic environment and interact with some coexistent organic pollutants, such as perfluorooctanesulfonate (PFOS), which is an emerging persistent organic pollutant. In this study, the adsorption of PFOS on GO in the presence of fulvic acid (FA), the impacts of GO and FA on PFOS toxicokinetics in carp (Cyprinus carpio), and in vitro digestion behaviors were examined. The results indicated that PFOS could be strongly adsorbed on GO with a Freundlich affinity coefficient KF of 580 ± 205 (mg/g)/(mg/L)n, while the adsorption was suppressed by FA due to competitive adsorption. GO significantly enhanced the bioaccumulation of PFOS in blood, kidney, liver, gill, intestine, and muscle of carp, and the corresponding bioaccumulation factor (BAF) was in the range of 2026–53513 L/kg. The enhancement was greatest for liver and intestine, which was 10.3 and 9.33 times of that without GO, respectively. In vivo toxicokinetic and in vitro digestion–absorption experiments indicated that GO could carry PFOS to penetrate the intestine cells. There herein, PFOS absorption, especially via intestine, and the uptake rate coefficient (ku) were greatly enhanced, leading to distinctly promoted bioaccumulation of PFOS in fish. However, FA could facilitate the flocculation of GO in the intestine and also accelerate excretion of GO–PFOS complex. Thus, in the presence of FA, PFOS absorption was reduced and the promotion effect of GO on PFOS accumulation was remitted.

Microbial dysbiosis induced in Cyprinus carpio by tetrabromobisphenol A exposure: Mediation through gut barrier impairment and oxidative stress

Yimin Li,Chunnuan Zhang,Zihao Meng,Mengkang You,Huajuan Shi

The frequent detection of Tetrabromobisphenol A (TBBPA) in environmental media has elicited considerable scientific concern. The ecotoxicological impacts of TBBPA on intestinal health in the common carp ( Cyprinus carpio ) were systematically investigated through integrated histopathological, biochemical, and metagenomic analyses. Fish were exposed to environmentally relevant TBBPA concentrations (0, 0.005, 0.05, and 0.5 mg/L) for a 14-day duration. Dose-dependent intestinal damage was induced by TBBPA exposure, manifested as villus fusion and atrophy, oxidative stress (diminished SOD/CAT activities and elevated MDA levels), and downregulated expression of tight junction proteins ( ZO-1 , Claudin-3 , Occludin ). This collectively compromised barrier integrity and triggered pro-inflammatory cytokine upregulation of TNF-α alongside anti-inflammatory IL-10 suppression. Significant intestinal microbial dysbiosis was detected via 16S rRNA sequencing. Alpha diversity indices were reduced at low concentrations (0.005 mg/L) of TBBPA but elevated at higher concentrations (0.05–0.5 mg/L). TBBPA exposure induced gut microbiota perturbations, characterized by depletion of beneficial taxa ( Cetobacterium ) and enrichment of opportunistic pathogens ( Legionella and Thermomonas ). Functional prediction analyses indicated that these microbial alterations may influence carbohydrate metabolism, and vitamin biosynthesis within the intestinal tract of common carp. Collectively, these findings demonstrated that TBBPA disrupted intestinal health via synergistic mechanisms involving oxidative stress, histopathological damage, and microbiota-mediated dysregulation. This investigation addresses a critical knowledge gap regarding the impacts of TBBPA on fish gut microbiota, while providing provided a reference for assessing the potential ecological risks of TBBPA in the environment.
