Surface display of spring viremia of carp virus glycoprotein on Lactococcus lactis and its protection efficacy in common carp (Cyprinus carpio L.)

Chen Zhang,Sheng Guo,Zhao Zhao,Zi-Rao Guo,Rui Ma,Gao-Xue Wang,Bin Zhu

Spring viremia of carp virus (SVCV) causes devastating disease in aquaculture, resulting in significant economic impact. To develop an effective means against SVCV infection, a Lactococcus lactis ( L.lactis ) based subunit vaccine (pNZ-UGA) was developed based on surface displaying of SVCV glycoprotein using anchoring motif of the cA (C terminus of the peptidoglyvsn-binding) domains of AcmA, a major autolysin from L.lactis . The surface expression of SVCV glycoprotein was verified by indirect immunofluorescence assay. The efficacy of the constructed vaccine was further evaluated in common carp . The results showed that the higher levels of specific IgM could be detected in fish vaccinated with pNZ-UGA, compared with that in PBS and L.lactis groups. Immune-related genes including TNF-α , IL-6b , IL-1β , Cxcr 1 , Cxca , IFNg2b , I-IFN , and IgM expression in pNZ-UGA group were strongly up-regulated, revealing that robust innate immune response was induced. Notably, the lowest cumulative mortality (13.46%) was observed in fish vaccinated with pNZ-UGA vaccine after SVCV challenge, whereas the cumulative mortality were 100.00% and 92.31% in PBS and L.lactis groups, respectively. This study suggests the potential use of the recombinant L.lactis with surface displaying antigen proteins as effective vaccines against SVCV and other fish virus infection .

Micro-algal astaxanthin ameliorates polystyrene microplastics-triggered necroptosis and inflammation by mediating mitochondrial Ca2+ homeostasis in carp’s head kidney lymphocytes (Cyprinus carpio L.)

Lei Diao,Meiqi Ding,Hongzhen Sun,Yawei Xu,Rui Yin,Huijie Chen

Polystyrene microplastics (PM) is a pressing global environmental concern, posing substantial risks to aquatic ecosystems . Microalgal astaxanthin (MA), a heme pigment, safeguards cells against oxidative damage induced by free radicals, which contributes to various health conditions, including aging, inflammation and chronic diseases. Herein, we investigated the potential of MA in ameliorating the immunotoxicity of PM on carp ( Cyprinus carpio L.) based on head kidney lymphocytes treated with PM (250 μM) and/or MA (100 μM). Firstly, CCK8 results showed that PM resulted in excessive death of head kidney lymphocytes. Secondly, head kidney lymphocytes treated with PM had a higher proportion of necroptosis, and the levels of necroptosis-related genes in head kidney lymphocytes were increased. Thirdly, the relative red fluorescence intensity of JC-1 and MitoSox showed decreased mitochondrial membrane potential and increased mtROS in head kidney lymphocytes treated with PM. MitoTracker® Green FM fluorescence analysis revealed enhanced mitochondrial Ca 2+ levels in PM-treated lymphocytes, corroborating the association between PM exposure and elevated intracellular Ca 2+ dynamics. PM exposure resulted in upregulation of calcium homeostasis-related gene (Orail, CAMKIIδ and SLC8A1) in lymphocytes. Subsequent investigations revealed that PM exposure reduced miR-25-5p expression while increasing levels of MCU, MICU1, and MCUR1 . Notably, these effects were counteracted by treatment with MA. Furthermore, PM led to the elevated secretion of inflammatory factors (IFN-γ, IL-1β, IL-2 and TNF-α), thereby inducing immune dysfunction in head kidney lymphocytes. Encouragingly, MA treatment effectively mitigated the immunotoxic effects induced by PM, demonstrating its potential in ameliorating necroptosis, mitochondrial dysfunction and immune impairment via regulating the miR-25-5p/MCU axis in lymphocytes. This study sheds light on safeguarding farmed fish against agrobiological threats posed by PM, highlighting the valuable applications of MA in aquaculture.

Construction of fish scale (Cyprinus carpio L.) gelatin-fatty acid conjugate for loading curcumin: Effect of alkyl chain length on the interaction and stability

Haoxin Li,Yanxi Wang,Wan Aida Wan Mustapha,Xiaoping Zhang,Fankui Zeng,Jia Liu

A fish scale (FS) gelatin-fatty acid conjugate (GFC) with alkyl chain lengths of 8–18 was constructed to increase the aqueous solubility of curcumin. The effect of alkyl chain length on the interaction between GFC and curcumin was characterized by dynamic light scattering (DLS), X -ray photoelectron spectroscopy (XPS), fluorescence spectroscopy (FS), and isothermal titration calorimetry (ITC). The surface hydrophobicity (from 4987 ± 223.79 to 9982 ± 262.78) and curcumin loading capacity (from 8.20 ± 0.54 to 31.18 ± 1.41 μg/mg) of the GFC exhibited significant enhancements through increasing alkyl chain lengths from 8 to 18. This was accompanied by a reduction in particle size (from 661.5 ± 28.9 to 329.7 ± 6.6 nm) and ζ -potential (from −2.7 ± 0.92 to −26.8 ± 0.27). FS and ITC confirmed that GOC shared an optimal binding constant ( K a , 2.40 × 10 8  L·mol −1 and 3.47 × 10 5  M −1 ) and binding site ( n , 1.45 and 2.276) with curcumin among GFCs. Increasing GFC's alkyl chain length also boosted the stability of entrapped curcumin against the thermal environment and ultraviolet radiation. These results could be beneficial for gelatin-based nanocarrier development and application.

Molecular identification of glucose transporter 4: The responsiveness to starvation, glucose, insulin and glucagon on glucose transporter 4 in common carp (Cyprinus carpio L.)

Liping Yang,Shaoyang Zhi,Guokun Yang,Chaobin Qin,Xiao Yan,Mingming Niu,Wenlei Zhang,Mingyu Liu,Mengjuan Zhao,Guoxing Nie

Glucose transporter 4 (GLUT4) is comprehensively investigated in mammals, while the comparative research of GLUT4 in common carp is deficient. To investigate the function of GLUT4, carp glut4 was first isolated. The open reading frame of carp glut4 was 1518 bp in length, encoding 505 amino acids. A high-sequence homology was identified in carp and teleost, and the phylogenetic tree displayed that the carp GLUT4 was clustered with the teleost. A high level of glut4 mRNA was analysed in fat, red muscle and white muscle. After fasting treatment, glut4 mRNA expression was increased significantly in muscle. In the oral glucose tolerance test experiment, glut4 mRNA was also significantly elevated in muscle, gut and fat. Furthermore, intraperitoneal injection of insulin resulted in the upregulation of glut4 gene expression significantly in white muscle, gut and fat. On the contrary, the glut4 mRNA level in the white muscle, gut and fat was markedly downregulated after glucagon injection. These results suggest that GLUT4 might play important roles in food intake and could be regulated by nutrient condition, insulin and glucagon in common carp. Our study is the first to report on GLUT4 in common carp. These data provide a basis for further study on fish GLUT4.

Jianxin Zhang,Mengyuan Huang,Junchang Feng,Yongyan Chen,Meng Li,Xiaolin Meng,Xulu Chang

This study is aimed at exploring the effects of Bacillus coagulans NRS 609 on the growth performance, intestinal health, antioxidant activity, and immune response in common carp (Cyprinus carpio L.) by colonizing the intestinal tract with the beneficial strain. Gavage B. coagulans with GFP-tag at 109 CFU/mL into common carp for 7 days. Results indicated that B. coagulans were maintained in the intestine for at least 14 days. Four groups fed B. coagulans at different levels of inclusion, including 0 (CK), (A), (B), and (C) CFU/g of feed for 8 weeks. Experimental groups showed significantly better growth performance than the control (). The treatment groups had higher digestive enzyme activity, villus height, and muscle thickness than the control (). Supplementation with B. coagulans also altered the composition of the intestinal flora. In addition, an enhancement of the antioxidant enzyme and a reduction of malondialdehyde in the liver were observed with the addition of B. coagulans (). Moreover, the cytokines of IL-1β and IL-12, IL-10, and TGF-β, significant upregulation in experimental groups (). Based on the results, the optimal level of B. coagulans was 107 CFU/g in diets for common carp.

Sodium gluconate increases Bacillus velezensis R-71003 growth to improve the health of the intestinal tract and growth performance in the common carp (Cyprinus carpio L.)

Xulu Chang,Meiru Kang,Lili Yun,Yihao Shen,Junchang Feng,Guokun Yang,Jianxin Zhang,Xiaolin Meng

Host-derived probiotics are widely used in aquaculture because they are able to adapt to the host intestinal environment more easily than other probiotics. However, decreased numbers and activity of live probiotics that occur during processing, storage, transportation, and consumption hinders the increased use of probiotics. In this study, the ability of sodium gluconate to promote the growth of Bacillus velezensis R7–1003, isolated from the intestinal tract of the common carp , was evaluated in vivo. The actions of dietary supplementation with B. velezensis R7–1003 and/or sodium gluconate were investigated in the common carp with respect to growth, the morphology of the intestine, digestive and immune functions, and antioxidant capacity, as well as intestinal microbiota. The results revealed that the addition of 0.3 and 0.6% sodium gluconate to the diet increased B. velezensis R7–1003 growth in the intestinal tract of the common carp. Furthermore, 0.6% sodium gluconate exhibited better results than 0.3%. A diet with host–derived B. velezensis R7–1003 supplementation improved the growth performance of the fish. It also increased the length of the intestinal villi and tight junction gene expression levels, including claudin-2, occludin , and ZO-1 . Host–derived B. velezensis R7–1003 supplementation enhanced the activities of protease, α-amylase, lipase , alkaline phosphatase , acid phosphatase , lysozyme , catalase , glutathione peroxidase , decreased the level of MDA, increased the level of the anti-inflammatory cytokine TGF-β, and decreased the level of the pro-inflammatory cytokine TNF-α. Furthermore, B. velezensis R7–1003 increased the levels of several probiotics, including Lactobacillus and Lactococcus , and bacteria that produce short-chain fatty acids, including Faecalibacterium , Bacteroides , and Clostridium . The ratio of Firmicutes/Bacteroidetes was reduced in the intestine of the common carp. Importantly, this study indicated that the combination of B. velezensis R7–1003 and sodium gluconate provided additional beneficial effects to the common carp. Thus, this study demonstrated that sodium gluconate promoted B. velezensis R7–1003 growth, which increased the growth performance and intestinal health of the common carp. The results indicated that further study is warranted concerning the use of B. velezensis R7–1003 combined with sodium gluconate as a diet supplement in other economically viable fish.
