The construction of multifunctional nano-enzymes is a feasible strategy for fighting multi-drug resistant (MDR) bacterial biofilm-associated infections. Extracellular DNA (eDNA) is an important functional part of biofilm formation, including the initial adherence of bacteria to subsequent development and eventual maturation. A nano-enzyme platform of graphene oxide-based nitrilotriacetic acid-cerium(IV) composite (GO-NTA-Ce) against bacterial biofilm infection has been developed. When located at the site of bacteria-associated infection, GO-NTA-Ce could inhibit the biofilm formation and effectively disperse the formed biofilm by degrading the eDNA. In addition to Ce-mediated deoxyribonuclease (DNase)-like activity, near-infrared laser irradiation of GO-NTA-Ce could produce local hyperthermia to kill the bacteria that lost the protection by the biofilm matrix. In addition, graphene is also a new green broad-spectrum antimicrobial material that can exert its antimicrobial effects through physical damage and chemical damage. In short, our GO-NTA-Ce nano-enzyme platform is capable of effectively eradicating drug-resistant bacterial biofilm infections through the triple action of DNase-like enzyme properties, photothermal therapy, and graphene-based antimicrobial activity, and the nano-composite has excellent potential for the treatment of MDR bacterial biofilm infections.

A visual aptasensor based on the combination of graphitic carbon nitride loaded with platinum nanoparticles (PtNPs/g-C 3 N 4 ) and deoxyribonuclease I (DNase I) was developed for detecting acetamiprid. The prepared PtNPs/g-C 3 N 4 exhibited excellent peroxidase (POD)-like activity, demonstrating the capacity to oxidize the colourless o-phenylenediamine (OPD) to produce the coloured product diaminophenazine (DAP) in the presence of hydrogen peroxide (H 2 O 2 ). The DNA aptamer of acetamiprid can adsorb onto the surface of PtNPs/g-C 3 N 4 , thereby inhibiting its POD activity. The binding of acetamiprid to its aptamer results in the aptamer desorbing from the surface of PtNPs/g-C 3 N 4 and subsequent digestion by DNase I. Ascribed to the synergistic effect of these factors, the aptasensor can achieve the rapid on-site detection of acetamiprid based on the acetamiprid-induced the colour change of the solution just with the smartphone. Furthermore, due to the remarkable fluorescence signal of DAP at 570 nm, the aptasensor under fluorescence mode enables highly sensitive and quantitative detection of acetamiprid with a linear range of 1 ng/mL to 4 µg/mL and a detection limit as low as 0.31 ng/mL. The aptasensor has successfully realized the detection of acetamiprid in river water and cucumber samples, offering a novel perspective for the rapid assessment of environmental pollution and food safety risks associated with acetamiprid residues. Patulin (PAT) is a mycotoxin that exists in a wide range and is harmful to human health, which has aroused great attention to food safety. In this study, we established a novel assay to detect food samples contaminated by PAT. First of all, we proposed a truncation protocol based on the secondary structure of aptamer. By removing the stem-loop structure one by one, an optimized truncated PAT aptamer (P-30) was obtained. To the best of our knowledge, the mechanism between PAT and P-30 was elaborated in the assisting of molecular docking and circular dichroism for the first time. Furthermore, we developed a sensitive aptasensor platform to detect PAT in food samples. The aptamer modified with gold nanoclusters (AuNCs) was used as fluorescent probe, and BSA-directed synthesized MnO 2 nanoflakes (MnO 2 NFs) as fluorescence receptor. Additionally, fluorescence signal amplification strategy assisted by DNase Ⅰ successfully improved the sensitivity of aptasensor. The developed assay had a low detection limit of 8.5 ng/L, and can be effectively used in apple and grape juice samples. Thus, the established aptasensor shows a broad application prospect in PAT detection.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized clinically by recurrent abdominal pain and bloody diarrhea, for which there is currently a lack of curative therapeutic strategies. In recent years, neutrophil extracellular traps (NETs) have been identified as playing a key role in the pathogenesis of UC, emerging as a potential therapeutic target. Although deoxyribonuclease I (DNase I) can exert therapeutic effects by degrading the DNA backbone of NETs, its clinical application is limited by inherent drawbacks, including poor in vivo stability, low bioavailability, and inefficient penetration of biological barriers. Herein, this study constructed a biomimetic membrane-based nanodelivery system. Initially, zeolitic imidazolate framework-8 (ZIF-8) was used to encapsulate DNase I, forming ZD nanoparticles to enhance enzymatic stability. Subsequently, these nanoparticles were coated with bacterial outer membrane vesicles (BOMVs), yielding the final BVZD composite system, designed to improve targeting to inflammatory sites. The BVZD system exhibited suitable particle size, good biocompatibility, and a demonstrated ability to specifically accumulate in the inflamed intestinal regions of a UC model. Both in vitro and in vivo experiments confirmed that BVZD effectively degraded NETs, suppressed endoplasmic reticulum stress (ERS) by downregulating the expression of prostaglandin-endoperoxide synthase 2 (PTGS2), and promoted the repair of the intestinal epithelial barrier. These findings highlight the promising therapeutic potential of BVZD for the treatment of UC.

A fluorometric patulin (PAT) assay is presented that is based on the use of magnetic reduced graphene oxide (rGO) and DNase I. The fluorescence of the PAT aptamer labelled with 6-carboxyfluorescein (FAM) is quenched by magnetized reduced graphene oxide (rGO-Fe 3 O 4 ) due to fluorescence resonance energy transfer (FRET). However, in the presence of PAT, the labelled aptamer is stripped off from rGO-Fe 3 O 4 . The rGO-Fe 3 O 4 is then magnetically separated so that the fluorescence of free labelled PAT aptamer is restored. DNase I cannot hydrolyze the aptamer on rGO-Fe 3 O 4 , but it can cleave the free aptamer-PAT complex. This will release FAM and PAT which can undergo a number of additional cycles to trigger the cleavage of abundant aptamer. Recycling of DNase I-assisted target therefore leads to a strong amplification of fluorescence and consequently to an assay with low limit of detection. The detection limit for PAT is as low as 0.28 μg L −1 which is about 13 times lower than that without using DNase I. The method offers a new approach towards rapid, sensitive and selective detection based on an aptamer. Conceivably, it has a wide scope in that it may be applied to numerous other analytes if appropriate aptamers are available.

Diabetic patients suffer from delayed fracture healing and impaired osteogenic function, but the underlying pathophysiological mechanisms are not fully understood. Neutrophil extracellular traps (NETs) formed by neutrophils in high glucose microenvironments affect the healing of wounds and other tissues. Some evidence supports that NETs may inhibit osteogenic processes in the microenvironment through sustained inflammatory activation. In this study, we observed that high glucose-induced NETs led to sustained inflammatory activation of macrophages. Pro-inflammatory NETs inhibited the osteogenic function of osteoblasts in vitro. A bone defect healing model based on diabetic rat animal models confirmed that bone healing was impaired in a high glucose environment, but this process could be reversed by DNase I, a NETs clearance agent. More importantly, the classic hypoglycemic drug metformin had a similar antagonistic effect as DNase I and could reverse the inhibitory effect of NETs on osteogenesis in a high-glucose environment. In summary, we found that NETs formation induced by high glucose microenvironment is a potential cause of osteogenic dysfunction in diabetic patients, and metformin can reverse this osteogenic disadvantage.

Neutrophil extracellular traps (NETs), consisting of chromatin DNA filaments coated with granule proteins, promote metastasis by enhancing tumor cell migration to distant organs. Recent studies indicate that NETs adhere to cancer cell membranes and enhance cell motility significantly to induce liver metastasis in patients with breast and colon cancers. Herein, a dynamically deformable protein delivery strategy is developed to prevent liver metastasis by disassembling NETs. Specifically, poly amino acid conjugating with polyethylene glycol (PAAP) is explored and synthesized for DNase-1 delivery. Notably, PAAP/DNase-1 degrades chromatin to induce apoptosis, followed by cell membrane rupture and remaining DNase-1 releases to the extracellular. More importantly, the released DNase-1 disassembles NET-DNA to prevent liver metastasis induced by NET. In all, PAAP/DNase-1 treatment not only suppresses tumor growth by degrading intracellular chromatin, but also prevents the liver metastasis by disassembling the NET-NDA. This strategy may provide brand-new inspiration to prevent the liver metastasis fundamentally in patients with metastatic colon and breast cancer.

A homogeneous electrochemical aptasensor was developed for sensitive detection of aflatoxin B1 (AFB1) by integrating ultrasmall silica nanochannel film (SNF)-modified electrode, two-dimensional (2D) nanoprobes, and DNase I-assisted target recycling. The 2D nanoprobes (Ru(NH 3 ) 6 3+ /AFB1 Apt@GO) were prepared by co-loading cationic electrochemical probe Ru(NH 3 ) 6 3+ and AFB1 specific aptamers (AFB1 Apt) onto graphene oxide (GO) via π–π stacking and electrostatic interaction. SNF was rapidly formed on indium tin oxide (ITO) electrodes within 10 s. The resulting structure provided 2–3 nm nanochannels, which enabled the electrostatic enrichment of Ru(NH 3 ) 6 3+ . In addition, the 2D nanoprobes were unable to enter the ultrasmall nanochannels of the SNF due to size exclusion and thus could not access the underlying electrode surface. In the presence of AFB1, aptamer–target binding caused conformational changes and aptamer detachment, releasing Ru(NH 3 ) 6 3+ . The detached aptamer was digested by DNase I, triggering further Ru(NH 3 ) 6 3+ release as AFB1 participated in additional recognition cycles. The released Ru(NH 3 ) 6 3+ was electrostatically enriched at the electrode surface, amplifying the ECL signal. DNase I-assisted recycling enhanced the ECL response by 3-fold compared to the non-enzymatic system. Under optimized conditions, the aptasensor exhibited a linear detection range for AFB1 detection from 3.12 × 10 −4 to 312 ng/mL and a limit of detection of 302 fg/mL. The method was successfully applied to detect AFB1 in corn and peanut samples, offering a simple, rapid, and highly sensitive strategy for electrochemical toxin detection in food safety analysis.

Background Absent in melanoma 2 (AIM2) inflammasome-dependent pyroptosis and neutrophil extracellular traps (NETs) have been implicated in chronic liver disease (CLD). However, the specific intrahepatic cell type that undergoes AIM2 inflammasome-dependent pyroptosis and how their interaction augments hepatic inflammation/fibrosis remains unclear. Methods The expression and correlation of AIM2 inflammasome-dependent pyroptosis-related indicators and NETs were analyzed in biopsy tissue and blood specimens from chronic hepatitis patients (CHs). Cell-based experiments were conducted to investigate their interaction. In vitro and in vivo experiments were used to analyze their effects on the progression of hepatic inflammation/fibrosis as well as their clinical importance. Results Elevated levels of AIM2 inflammasome-dependent pyroptosis indicators and NETs were detected in biopsy tissue and blood specimens. Circulating NETs were positively correlated with pyroptosis-related indicators, and both were related with disease severity. Confocal imaging revealed that AIM2 was mainly localized to hepatic macrophages, indicating that hepatic macrophages were the major cell type that underwent pyroptosis. NETs were directly engulfed by macrophages and then stimulated AIM2 inflammasome-dependent macrophage pyroptosis in vitro, which amplified the activation of hepatic stellate cells (HSCs) and increased collagen deposition. Administration of the NETs degradation agent DNase I or the AIM2 inflammasome activation inhibitor ODN A151 effectively alleviated chronic liver inflammation/fibrosis progression in vivo. Conclusion sNETs-induced AIM2 inflammasome-dependent pyroptosis in macrophages facilitates liver inflammation/fibrosis progression. The identified NET–AIM2 inflammasome cascade could serve as a novel therapeutic target for hepatic inflammation/fibrosis progression.

Self-assembled DNA nanostructures, as a novel type of nano-biomaterials, are widely used for smart and targeted drug delivery due to their precise structures and versatile functions. The binding affinity of drugs to DNA nanostructures plays an important role in drug loading, release and efficacy. The interaction of mithramycin (MTR) with AS1411-tethered DNA nanotrains (AS1411NTrs) in the presence of different divalent metal ions (Mg 2+ , Zn 2+ , and Mn 2+ ) was studied by fluorescence spectroscopy, differential scanning calorimetry and viscosity measurement. The binding mode between the MTR dimers and AS1411NTrs was determined by viscosity experiment and melting temperature measurement. The thermodynamic parameters of the binding process (binding stoichiometry, binding constant, enthalpy change, entropy change and Gibbs free energy change) were determined by fluorescence spectroscopy and differential scanning calorimetry, and the effect of different metal ions on the binding was compared. The release of the MTR dimers loaded by AS1411NTrs in the absence and presence of DNase I was analyzed. The in vitro cytotoxicity of the MTR dimers to HepG2 cancer cells and L02 normal cells and their intracellular uptake efficiency were evaluated. The binding parameters were correlated with the loading, release and cytotoxicity of the MTR dimers. These studies showed that AS1411NTrs had high drug loading and sustained release effect on (MTR) 2 Mg 2+ , (MTR) 2 Zn 2+ and (MTR) 2 Mn 2+ . This drug loading system improved the lethality to cancer cells and reduced the side effects on normal cells. This study may provide a theoretical basis for the clinical application of MTR loaded by DNA nanostructures.

Natural killer (NK) cell-mediated immunotherapy has shown a great promise for treatments of tumors, while some strategies that can improve its therapeutic efficacy and reduce potential adverse events are highly desired. We herein report a pH-sensitive second near-infrared (NIR-II) photothermal liposomal nanocomplex for enhanced NK cell-based immunotherapy of breast cancer. Such nanocomplex (LNC DS ) consists of charge-transfer nanoparticles (CTN) with pH-sensitive NIR-II photothermal effect, deoxyribonuclease I (DNase I), NK cell stimulant (SIS3) and a surface coated thermal-responsive liposome shell. Upon 1064 nm laser irradiation, LNC DS generates mild heat in a controlled manner, which results in destruction of thermal-responsive liposome shell to allow on-demand release of stimulants and DNase I in tumor sites. The released DNase I not only kills cancer cells, but also induces immunogenic cell death (ICD), which synergizes with the action of the released SIS3 to promote activation of NK cells and cytotoxic T lymphocytes, contributing to enhanced therapeutic efficacy of immunotherapy. As a result, a single treatment of LNC DS upon NIR-II photoactivation greatly inhibits the growths of subcutaneously implanted primary and distant tumors in a breast cancer murine model, and even completely prevents lung metastasis. This study thus offers a photo-controlled drug delivery nanosystem for efficacy and precise NK cell-mediated immunotherapy of cancer.

In the present work, a convenient signal-on electrochemical label-free aptasensor for 17β-estradiol (E2), a typical steroidal hormones endocrine disrupting chemicals , was proposed. 6-mercapto-1-hexanol (MCH) self-assembled monolayer (SAM) modified Au (MCH/Au) electrode was used as the substrate electrode. Graphene is used with bi-functions, not only to adsorb E2 binding aptamer, serving as the recognition element to E2, but also to be assembled onto MCH/Au electrode when sensing E2, to controllably turn on the electron transfer (eT), and further indicate the signal to E2 concentration. With the synergistic effect of DNase I enzyme, highly sensitive detection of E2 was achieved at this aptasensing system, with a linear range from 0.07 to 10 pM and a detection limit of 50 fM. An outstanding selectivity towards E2 was proven for the sensing system by simultaneously detecting 100-fold potential co-existing interferences. The stability and reproducibility were also evaluated to be satisfactory. Spiked real water analysis further indicated its reliability and potential in practical environmental monitoring.
