A fluorometric patulin (PAT) assay is presented that is based on the use of magnetic reduced graphene oxide (rGO) and DNase I. The fluorescence of the PAT aptamer labelled with 6-carboxyfluorescein (FAM) is quenched by magnetized reduced graphene oxide (rGO-Fe 3 O 4 ) due to fluorescence resonance energy transfer (FRET). However, in the presence of PAT, the labelled aptamer is stripped off from rGO-Fe 3 O 4 . The rGO-Fe 3 O 4 is then magnetically separated so that the fluorescence of free labelled PAT aptamer is restored. DNase I cannot hydrolyze the aptamer on rGO-Fe 3 O 4 , but it can cleave the free aptamer-PAT complex. This will release FAM and PAT which can undergo a number of additional cycles to trigger the cleavage of abundant aptamer. Recycling of DNase I-assisted target therefore leads to a strong amplification of fluorescence and consequently to an assay with low limit of detection. The detection limit for PAT is as low as 0.28 μg L −1 which is about 13 times lower than that without using DNase I. The method offers a new approach towards rapid, sensitive and selective detection based on an aptamer. Conceivably, it has a wide scope in that it may be applied to numerous other analytes if appropriate aptamers are available.

Diabetic patients suffer from delayed fracture healing and impaired osteogenic function, but the underlying pathophysiological mechanisms are not fully understood. Neutrophil extracellular traps (NETs) formed by neutrophils in high glucose microenvironments affect the healing of wounds and other tissues. Some evidence supports that NETs may inhibit osteogenic processes in the microenvironment through sustained inflammatory activation. In this study, we observed that high glucose-induced NETs led to sustained inflammatory activation of macrophages. Pro-inflammatory NETs inhibited the osteogenic function of osteoblasts in vitro. A bone defect healing model based on diabetic rat animal models confirmed that bone healing was impaired in a high glucose environment, but this process could be reversed by DNase I, a NETs clearance agent. More importantly, the classic hypoglycemic drug metformin had a similar antagonistic effect as DNase I and could reverse the inhibitory effect of NETs on osteogenesis in a high-glucose environment. In summary, we found that NETs formation induced by high glucose microenvironment is a potential cause of osteogenic dysfunction in diabetic patients, and metformin can reverse this osteogenic disadvantage.

A homogeneous electrochemical aptasensor was developed for sensitive detection of aflatoxin B1 (AFB1) by integrating ultrasmall silica nanochannel film (SNF)-modified electrode, two-dimensional (2D) nanoprobes, and DNase I-assisted target recycling. The 2D nanoprobes (Ru(NH 3 ) 6 3+ /AFB1 Apt@GO) were prepared by co-loading cationic electrochemical probe Ru(NH 3 ) 6 3+ and AFB1 specific aptamers (AFB1 Apt) onto graphene oxide (GO) via π–π stacking and electrostatic interaction. SNF was rapidly formed on indium tin oxide (ITO) electrodes within 10 s. The resulting structure provided 2–3 nm nanochannels, which enabled the electrostatic enrichment of Ru(NH 3 ) 6 3+ . In addition, the 2D nanoprobes were unable to enter the ultrasmall nanochannels of the SNF due to size exclusion and thus could not access the underlying electrode surface. In the presence of AFB1, aptamer–target binding caused conformational changes and aptamer detachment, releasing Ru(NH 3 ) 6 3+ . The detached aptamer was digested by DNase I, triggering further Ru(NH 3 ) 6 3+ release as AFB1 participated in additional recognition cycles. The released Ru(NH 3 ) 6 3+ was electrostatically enriched at the electrode surface, amplifying the ECL signal. DNase I-assisted recycling enhanced the ECL response by 3-fold compared to the non-enzymatic system. Under optimized conditions, the aptasensor exhibited a linear detection range for AFB1 detection from 3.12 × 10 −4 to 312 ng/mL and a limit of detection of 302 fg/mL. The method was successfully applied to detect AFB1 in corn and peanut samples, offering a simple, rapid, and highly sensitive strategy for electrochemical toxin detection in food safety analysis.

Background Absent in melanoma 2 (AIM2) inflammasome-dependent pyroptosis and neutrophil extracellular traps (NETs) have been implicated in chronic liver disease (CLD). However, the specific intrahepatic cell type that undergoes AIM2 inflammasome-dependent pyroptosis and how their interaction augments hepatic inflammation/fibrosis remains unclear. Methods The expression and correlation of AIM2 inflammasome-dependent pyroptosis-related indicators and NETs were analyzed in biopsy tissue and blood specimens from chronic hepatitis patients (CHs). Cell-based experiments were conducted to investigate their interaction. In vitro and in vivo experiments were used to analyze their effects on the progression of hepatic inflammation/fibrosis as well as their clinical importance. Results Elevated levels of AIM2 inflammasome-dependent pyroptosis indicators and NETs were detected in biopsy tissue and blood specimens. Circulating NETs were positively correlated with pyroptosis-related indicators, and both were related with disease severity. Confocal imaging revealed that AIM2 was mainly localized to hepatic macrophages, indicating that hepatic macrophages were the major cell type that underwent pyroptosis. NETs were directly engulfed by macrophages and then stimulated AIM2 inflammasome-dependent macrophage pyroptosis in vitro, which amplified the activation of hepatic stellate cells (HSCs) and increased collagen deposition. Administration of the NETs degradation agent DNase I or the AIM2 inflammasome activation inhibitor ODN A151 effectively alleviated chronic liver inflammation/fibrosis progression in vivo. Conclusion sNETs-induced AIM2 inflammasome-dependent pyroptosis in macrophages facilitates liver inflammation/fibrosis progression. The identified NET–AIM2 inflammasome cascade could serve as a novel therapeutic target for hepatic inflammation/fibrosis progression.

Self-assembled DNA nanostructures, as a novel type of nano-biomaterials, are widely used for smart and targeted drug delivery due to their precise structures and versatile functions. The binding affinity of drugs to DNA nanostructures plays an important role in drug loading, release and efficacy. The interaction of mithramycin (MTR) with AS1411-tethered DNA nanotrains (AS1411NTrs) in the presence of different divalent metal ions (Mg 2+ , Zn 2+ , and Mn 2+ ) was studied by fluorescence spectroscopy, differential scanning calorimetry and viscosity measurement. The binding mode between the MTR dimers and AS1411NTrs was determined by viscosity experiment and melting temperature measurement. The thermodynamic parameters of the binding process (binding stoichiometry, binding constant, enthalpy change, entropy change and Gibbs free energy change) were determined by fluorescence spectroscopy and differential scanning calorimetry, and the effect of different metal ions on the binding was compared. The release of the MTR dimers loaded by AS1411NTrs in the absence and presence of DNase I was analyzed. The in vitro cytotoxicity of the MTR dimers to HepG2 cancer cells and L02 normal cells and their intracellular uptake efficiency were evaluated. The binding parameters were correlated with the loading, release and cytotoxicity of the MTR dimers. These studies showed that AS1411NTrs had high drug loading and sustained release effect on (MTR) 2 Mg 2+ , (MTR) 2 Zn 2+ and (MTR) 2 Mn 2+ . This drug loading system improved the lethality to cancer cells and reduced the side effects on normal cells. This study may provide a theoretical basis for the clinical application of MTR loaded by DNA nanostructures.

Natural killer (NK) cell-mediated immunotherapy has shown a great promise for treatments of tumors, while some strategies that can improve its therapeutic efficacy and reduce potential adverse events are highly desired. We herein report a pH-sensitive second near-infrared (NIR-II) photothermal liposomal nanocomplex for enhanced NK cell-based immunotherapy of breast cancer. Such nanocomplex (LNC DS ) consists of charge-transfer nanoparticles (CTN) with pH-sensitive NIR-II photothermal effect, deoxyribonuclease I (DNase I), NK cell stimulant (SIS3) and a surface coated thermal-responsive liposome shell. Upon 1064 nm laser irradiation, LNC DS generates mild heat in a controlled manner, which results in destruction of thermal-responsive liposome shell to allow on-demand release of stimulants and DNase I in tumor sites. The released DNase I not only kills cancer cells, but also induces immunogenic cell death (ICD), which synergizes with the action of the released SIS3 to promote activation of NK cells and cytotoxic T lymphocytes, contributing to enhanced therapeutic efficacy of immunotherapy. As a result, a single treatment of LNC DS upon NIR-II photoactivation greatly inhibits the growths of subcutaneously implanted primary and distant tumors in a breast cancer murine model, and even completely prevents lung metastasis. This study thus offers a photo-controlled drug delivery nanosystem for efficacy and precise NK cell-mediated immunotherapy of cancer.

In the present work, a convenient signal-on electrochemical label-free aptasensor for 17β-estradiol (E2), a typical steroidal hormones endocrine disrupting chemicals , was proposed. 6-mercapto-1-hexanol (MCH) self-assembled monolayer (SAM) modified Au (MCH/Au) electrode was used as the substrate electrode. Graphene is used with bi-functions, not only to adsorb E2 binding aptamer, serving as the recognition element to E2, but also to be assembled onto MCH/Au electrode when sensing E2, to controllably turn on the electron transfer (eT), and further indicate the signal to E2 concentration. With the synergistic effect of DNase I enzyme, highly sensitive detection of E2 was achieved at this aptasensing system, with a linear range from 0.07 to 10 pM and a detection limit of 50 fM. An outstanding selectivity towards E2 was proven for the sensing system by simultaneously detecting 100-fold potential co-existing interferences. The stability and reproducibility were also evaluated to be satisfactory. Spiked real water analysis further indicated its reliability and potential in practical environmental monitoring.

DNA-based biomaterials, including polyelectrolyte complex (PEC) hydrogels composed of DNA and other molecules, have received considerable attention due to their programmability and good biocompatibility. However, the cost for DNA synthesis is fairly high, especially for applications where a large quantity of DNA is needed. Construction of biomaterials using biomass-derived DNA is a feasible strategy to address this challenge. In this work, we fabricated and characterized PEC hydrogels with or without covalent cross-linking using biomass-derived DNA and chitosan (CS). Characterization results revealed detailed microstructures and mechanical properties of these two hydrogels, suggesting their potential application in DNA preservation and protein immobilization. We next developed a versatile DNA preservation strategy combining DNA-CS PEC hydrogel-based DNA encapsulation and chitosanase-mediated DNA release, which successfully maintained the integrity and functionality of various types of DNA, i.e., double-stranded DNA (dsDNA), single-stranded DNA (ssDNA), plasmid DNA, and genomic DNA, during the treatment of DNase I and hydrogen peroxide (H2O2). Remarkably, this method also demonstrated good efficiency in protecting DNA containing a broadly used unnatural base pair, dNaM-dTPT3, against the attack of silver ion. Additionally, we developed a convenient and reliable strategy for protein immobilization with the covalently cross-linked PEC hydrogel. This strategy demonstrated good performance in the reusability of the immobilized enzyme, evidenced by 56.4% product yield of the initial cycle of usage achieved by the immobilized horseradish peroxidase (HRP) in the tenth cycle of usage. This work not only broadens the application scope of biomass-derived polymers, but also provides feasible strategies for the sustainable and cost-effective preservation and immobilization of biological macromolecules.

The combination of fluorophore-labelled single-strand DNA probes and nanomaterial quenchers has shown great potential in miRNA detection. The development of advanced detection systems by understanding and controlling the fluorescence quenching/recovery via nanoquenchers’ microstructures and local morphologies is an attractive area warranting further investigations. Inspired by nanopore sequencing, we present a novel miRNA sensing strategy using fluorophore-labeled DNA as probes and a type of large-pore-sized mesoporous polydopamine nanoparticles (MPDA-L, 70 nm in diameter) as fluorescence quenchers. It is revealed that the quenching efficiency of MPDA-L towards the fluorophore labelled on the probe, reached more than 99% at a relatively low particle concentration. Moreover, the mesopores effectively protected the probe DNA from cleavage by DNase I which was used for signal amplification. Sensitive detection of miRNA with a low detection limit of 32–40 pM, as well as a linear detection range of up to 5 nM, was realized by the mesopore effects via a greatly improved differential affinity of ssDNA and the probe–miRNA heteroduplex toward the surface of nanoquenchers. Interestingly, enhanced DLVO (Derjaguin–Landau–Verwey–Overbeek) repulsion generated inside the pore surface by the negative surface–curvature effect correlates with the improved duplex detachment and fluorescence recovery. The developed strategy can be successfully applied to quantify down-regulated let-7a and up-regulated miRNA-21 in different types of cancer cells by using total RNA samples from cell lysate. These findings are expected to inspire strategies and pave a way for utilizing porous nanomaterials for constructing miRNA detection systems.

Mutations in cytoplasmic DNA-degrading enzymes can lead to the accumulation of cytoplasmic DNA (cytoDNA), which excessively activates DNA-sensing pathways and exacerbates inflammatory aging. Reducing cytoDNA levels to suppress DNA-sensing mechanisms is therefore critical for treating elderly-onset rheumatoid arthritis (EORA). In this study, we constructed Trex1 mRNA loaded lipid nanoparticles (LNPs) via microfluidics and prepared DNase I loaded polydopamine (PDA) nanoparticles through oxidative polymerization. These two components were co-encapsulated into methacrylated hyaluronic acid (HAMA) microspheres using microfluidic photopolymerization. The LNPs incorporate cationic lipids to facilitate mRNA loading and promote endosomal escape, enabling efficient translation of TREX1 and subsequent recognition and degradation of cytoDNA. Meanwhile, cationic mesoporous polydopamine electrostatically adsorbs and degrades extracellular DNA. The microspheres function as a reservoir for sustained nanoparticle release, enabling synergistic inhibition of DNA sensing pathways. This microsphere based vaccine upregulates TREX1 expression in antigen-presenting cells (APCs) and reduces cytoDNA levels, thereby suppressing overactivation of the cGAS-STING signaling axis and promoting immune tolerance. It also attenuates the differentiation of CD4+ T cells into Th1, Th2, Th17, and Treg subsets. In an aged rat model of rheumatoid arthritis, the vaccine alleviated soft tissue swelling, synovitis, and cartilage and bone damage. By clearing excess cytoDNA and restraining DNA-sensing hyperactivation, this vaccine induces cellular immune tolerance and represents a promising therapeutic strategy for rheumatoid arthritis in the elderly.
