Deoxynivalenol (DON) is a globally significant mycotoxin that can contaminate grains, representing a serious hazard to human and animal health. The current research identified a DON-degrading dehydrogenase from Devosia sp. 63-57 ( Ds DDH). A Ds DDH demonstrated noticeable activation in the presence of Ca 2+ , with electron acceptors including pyrroloquinoline quinone (PQQ), phenazine methosulfate (PMS), and 2,6-dichlorophenolindophenol (DCPIP). The Ds DDH effectively degraded over 82 % of DON (10 μg/mL) within 30 min using PMS or DCPIP as electron acceptors. Additionally, the Ds DDH retained 76 % of its activity after pre-incubating for 2 h at 45 °C. Moreover, the Ds DDH-mediated degradation product of DON was confirmed to be 3-keto-DON, which significantly showed lower toxicity than DON, as verified by zebrafish larvae assays. Furthermore, DON contents in corn and wheat flour decreased from 9.58 to 9.73 to 1.36 and 1.77 μg/g, resulting in degradation rates of 85.85 and 81.76 %, respectively. These findings provide valuable insights into developing a biological detoxification strategy for DON in cereal and feed applications, laying a promising foundation for safer and more effective mycotoxin control.

This study developed a novel molecularly imprinted electrochemical cell sensor for ultrasensitive detection of deoxynivalenol (DON). Using o -phenylenediamine as the functional monomer and Caspase-3 as the template molecule, a molecularly imprinted polymer film was synthesized on screen-printed electrodes via electropolymerization. After elution, the sensor was integrated into a three-electrode system. Upon DON exposure, Caspase-3 released from HepG2 cells specifically binds to the imprinted cavities, altering the current response of the electrochemical indicator [Fe(CN)₆] 4−/3− . Using differential pulse voltammetry (DPV), the peak current showed a linear relationship with DON concentration from 0.2 to 100 pg/mL, with a detection limit of 0.193 pg/mL (R 2 = 0.9977). This work provides a new method for rapid DON detection in food and expands the application of molecular imprinting in mycotoxin analysis.

Deoxynivalenol (DON) has drawn considerable attention for its obvious pathogenicity and wide use in agro-products, which cause a potential threat to human health. In this work, an electrochemical immunosensor is developed for the highly sensitive and selective detection of DON in wheat flour using AuNPs-BP-MWCNTs-COOH and antibodies. The AuNPs-BP-MWCNTs-COOH nanocomposite was prepared via an in situ reduction reaction and ultrasonic-assisted liquid-phase exfoliation. The nanocomposite exhibits a larger surface area, decent stability, excellent electron transfer capability, good protein binding capability and prominent specificity. The plentiful carboxyl group on the nanocomposite can bind to the amino group of the antibody, and AuNPs have an affinity for the sulfhydryl group of the antibody, which makes it feasible for the nanocomposite to load the antibody. The peak currents are plotted against the logarithm of DON concentration from 0.002 to 80 ng mL−1 with a limit of detection (LOD) of 0.5 pg mL−1. This approach establishes an effective label-free immunosensor platform for the detection of DON with high sensitivity and selectivity in various food and agricultural products.

Deoxynivalenol (DON) is one of the most common mycotoxins distributed in food and feed, which causes severe liver injury in humans and animals. Cold atmospheric plasma (CAP) has received much attention in mycotoxin degradation due to the advantages of easy operation, high efficiency, and low temperature. So far, the majority of studies have focused on the degradation efficiency and mechanism of CAP on DON, while there is still little information available on the hepatotoxicity of DON after CAP treatment. Herein, this study aimed to investigate the effect of CAP on DON-induced hepatotoxicity both in vitro and in vivo and its underlying mechanisms. The results showed that 120-s CAP treatment achieved 97 % degradation of DON. The vitro hepatotoxicity of DON in L02 cells was significantly reduced with CAP treatment time. Meanwhile, CAP markedly alleviated DON-induced liver injury in mice including the balloon-like degeneration of liver tissues and elevation of AST and ALP level. The underlying mechanism for CAP detoxification of DON-induced hepatotoxicity was further elucidated. The results showed that DON caused severe oxidative stress in cells by suppressing the antioxidant signaling pathway of Nrf2/HO-1/NQO-1, consequently leading to mitochondrial dysfunction and cell apoptosis, accompanied by cellular senescence and inflammation. CAP blocked DON inhibition on the Nrf2/HO-1/NQO-1 signaling pathway through the efficient degradation of DON, accordingly alleviating the oxidative stress and liver injury induced by DON. Therefore, CAP is an effective method to eliminate DON hepatotoxicity, which can be applied in the detoxification of mycotoxin-contaminated food and feed to ensure human and animal health.

Deoxynivalenol (DON) is a secondary metabolite produced by fungi that contaminates food during the process of production and storage, and can potentially cause acute or chronic poisoning upon ingestion. This study employs surface-enhanced Raman spectroscopy (SERS) combined with density functional theory (DFT) to detect the pure DON sample. The investigation focuses on the adsorption mechanism of DON molecules on gold nanoparticles (AuNPs) and indicates that the O6 atoms in the DON molecule act as sensitive sites for nucleophilic reactions by using DFT calculations. Experimental results demonstrate that pure DON molecules exhibit a strong SERS response, displaying excellent linear and non-linear relationships across concentration ranges of 0.01–0.2 μg/mL (R 2 = 0.992) and 0.2–1 μg/mL (R 2 = 0.997), respectively. The limit of detection (LOD) and quantification (LOQ) are 0.0085 μg/mL and 0.0256 μg/mL, respectively, which show excellent detection sensitivity. These findings can provide a theoretical foundation for monitoring DON toxins in foodstuffs.

Deoxynivalenol (DON) contamination in food products significantly threatens human health, necessitating a reliable and sensitive detection method. This study aims to develop a simple, low-cost, and effective electrochemical immunoassay method for detecting DON based on the nickel‑iron bimetallic Prussian blue analog (Ni Fe PBA). The Ni Fe PBA nanozymes with high peroxidase-like activity were synthesized using an environmentally friendly chemical precipitation method. In the presence of hydrogen peroxide (H 2 O 2 ), the current change of thionine oxidation initiated by Ni Fe PBA nanozymes can be exploited to diagnose DON. Under optimal conditions, the proposed method achieved quantitative detection of DON in the range of 10–10 7 pg mL −1 with a detection limit of 4.5 pg mL −1 (S/N = 3), demonstrating excellent selectivity, reproducibility, and stability. In addition, the DON immunosensor provides satisfactory results for the detection in real samples, demonstrating the feasibility of the proposed sensor in detecting of DON in such products.

Scutellaria barbata D. Don has been used in traditional Chinese medicine for its heat-clearing and detoxifying effects to treat colitis. Polysaccharides are the key bioactive constituents of S. barbata . In this study, a heteropolysaccharide (SBP80) was extracted from S. barbata using water extraction methods. In vivo studies in mice demonstrated that SBP80 exhibited significant therapeutic effects on ulcerative colitis (UC). Specifically, SBP80 improved body weight and disease activity index (DAI) scores in UC mice, restored colon length, alleviated inflammatory cell infiltration in colonic tissues, and suppressed the levels of inflammatory cytokines and oxidative stress factors. Further purification of SBP80 yielded two polysaccharides, SBP80–1 (7.2 kDa) and SBP80–2 (292.3 kDa). Structural analysis revealed that both SBP80–1 and SBP80–2 were primarily composed of mannose, rhamnose, glucuronic acid, glucose, and galactose, with minor amounts of xylose, arabinose, and fucose. In addition, SBP80–2 contained higher amounts of glucuronic acid and xylose compared to SBP80–1. Methylation analysis further showed that the proportion of terminal residues and 1,6-linked residues in SBP80–2 was higher, while SBP80–1 exhibited more 1,4-linked residues. In vivo zebrafish experiments demonstrated that both SBP80–1 and SBP80–2 inhibited intestinal shortening and neutrophil infiltration in zebrafish with UC. They also reduced nitric oxide (NO) and reactive oxygen species (ROS) levels. Moreover, SBP80–2 exhibited superior anti-UC activity, which may be attributed to its structural characteristics. In conclusion, our findings indicate that S. barbata polysaccharides (SBP80–1 and SBP80–2) may be promising natural agents for the treatment of UC.

A ratiometric aptasensor for deoxynivalenol (DON) detection was developed based on FRET between graphene oxide (GO) and carbon dots. Blue carbon dots, functionalized with aptamers, act as energy donors, while GO functions as the energy acceptor. In the absence of DON, the addition of GO quenches the fluorescence of the blue carbon dots, while the fluorescence of red carbon dots, used as internal reference signals, remains unchanged. When DON is introduced, the aptamer selectively binds to the target, increasing the distance between the donor and acceptor, disrupting FRET, and gradually restoring the blue fluorescence. The sensor demonstrated a strong linear relationship within the concentration range of 0.05–200 ng/mL of DON, with a detection limit of 14.7 pg/mL. The superior analytical performance and high sensitivity indicate the potential of the sensor for DON detection in real sample matrices.

ABSTRACT Ethnopharmacological relevance Fritillaria cirrhosa D. Don (FCD) is a traditional Chinese medicine used to treat respiratory disorders, known for its effects in clearing heat, moistening the lungs, resolving phlegm, and relieving cough. Additionally, the total alkaloids extracted from FCD can alleviate asthma symptoms and reduce airway inflammation. However, no studies have investigated the effects of total alkaloids on lung macrophages. Aim of the study This study explored whether the total alkaloids of FCD (TAs-FCD) reduce M2 macrophage polarization and, consequently, attenuate airway remodeling in asthmatic mice. This study further elucidated its mechanism of action in treating allergic asthma. Materials and methods The extracted TAs-FCD was analyzed for its composition using UPLC-Q-TOF/MS. Network pharmacology was employed to identify the active ingredients and potential mechanisms of TAs-FCD in the treatment of allergic asthma. A mouse model of ovalbumin-induced allergic asthma was established, adopted, and validated through in vivo experiments. Hematoxylin-eosin staining (H&E), immunohistochemistry (IHC), immunofluorescence staining (IF), enzyme-linked immunosorbent assay (ELISA), Western blotting (WB), and real-time fluorescence quantitative polymerase chain reaction (q-PCR) were used to investigate the role of TAs-FCD in inhibiting M2 macrophage polarization in the context of allergic asthma. Results A total of 66 active ingredients were screened from 116 compounds using SWISSADME. The targets of these 66 compounds were predicted by SwissTargetPrediction, resulting in 808 unique drug targets after excluding duplicates. Additionally, 1,756 targets related to allergic asthma were identified from the DisGeNET, Genecard, and OMIM databases. This led to 267 cross-targets between the active ingredient targets and allergic asthma targets, including interleukin (IL)-1β, tumor necrosis factor (TNF), and STAT3. Animal experiments demonstrated that TAs-FCD improved histopathological injury in mouse lungs, reduced peri-airway collagen fiber accumulation, airway mucus secretion, and airway smooth muscle proliferation. TAs-FCD also lowered IL-1β, TNF-α and IL-4 levels in lung tissues and alleviated airway inflammation. Furthermore, TAs-FCD significantly reduced levels of Arg1 and CD206, which are closely associated with M2 macrophages, and downregulated the expression of p-STAT3 and p-JAK2. Conclusion TAs-FCD may inhibit M2 macrophage polarization by regulating the JAK2/STAT3 pathway, thereby alleviating airway remodeling and inflammation in allergic asthmatic mice.

Background Allii Macrostemonis Bulbus is also named Xiebai in China. It is an edible vegetable, and also a famous herb for treating coronary heart disease. Allium chinense G. Don (ACGD) and Allium macrostemon Bunge (AMB) are it botanical sources. The aim of this study was to explore the cardioprotective effects, and decipher the visual spatial distribution and absolute content of primary metabolites derived from these two herbs. Methods H9c2 cells were used to perform the hypoxia-reoxygenation (H/R)-induced myocardial injury model. Their protective effects were evaluated by apoptosis levels. Furthermore, matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry imaging approach (MALDI-TOF MSI) was carried out to present the spatial location of primary metabolites including fatty acids, amino acids, carotenoids, and vitamins in these two Allium herbs. Multiple analytical methods were applied to perform quantitative analysis of these primary metabolites in AMB and ACGD bulbs by liquid chromatography tandem mass spectrometry (LC–MS). Results First, AMB and ACGD extracts both could increase the cell viability in H9c2 cells, and attenuate H/R-induced injury. They markedly decreased apoptosis, accompanied by activating the BCL-2/BAX pathway. Further, MALDI-TOF MSI-based relative quantification results showed several amino acids, fatty acids, carotenoids, and vitamins were largely rich in the tunics and outside scales of fresh bulbs, while some primary metabolites were abundant in their developing flower buds. Absolute quantification results displayed total contents of amino acids in ACGD bulbs were higher than those in AMB, while total contents of fatty acids and vitamins provides opposite trends in these two Allium herbs. The total contents of carotenoids and trace elements showed no significant differences between AMB and ACGD samples. Conclusions This study would be helpful to understand the myocardial injury protection effects of these two Allium herbs, and the spatial accumulation and quantitative content levels of their main nutrients. Water-distilled essential oil from the dried bulbs of Allium chinense (Liliaceae) was analyzed by gas chromatography–mass spectrometry (GC–MS). Eighteen compounds, accounting for 98.4% of the total oil, were identified and the main components of the essential oil of A. chinense were methyl allyl trisulfide (30.7%), dimethyl trisulfide (24.1%), methyl propyl disulfide (12.8%) and dimethyl disulfide (9.6%) followed by methyl allyl disulfide (3.4%) and methyl propyl trisulfide (3.6%). The essential oil exhibited contact toxicity against the booklice ( Liposcelis bostrychophila ) with an LC 50 value of 441.8 μg/cm 2 while the two major constituents, dimethyl trisulfide and methyl propyl disulfide had LC 50 values of 153.0 μg/cm 2 and 738.0 μg/cm 2 against the booklice, respectively. The essential oil of A. chinense possessed strong fumigant toxicity against the booklice with an LC 50 value of 186.5 μg/l while methyl allyl trisulfide (LC 50 = 90.4 μg/l) and dimethyl trisulfide (LC 50 = 114.2 μg/l) exhibited stronger fumigant toxicity than methyl propyl disulfide (LC 50 = 243.4 μg/l) and dimethyl disulfide (LC 50 = 340.8 μg/l) against the booklice. The results indicated that the essential oil and its major constituents have potential for development into natural insecticides or fumigants for control of insects in stored grains.

Vomitoxin (DON) residues in grains are of great concern to public health. Herein, a label-free aptasensor was constructed to detect DON distributed in grains. Cerium-based metal-organic framework composite gold nanoparticles ( [email protected] ) were used as substrate materials to facilitate electron transfer and provided more binding sites for DNA. The separation of DON-aptamer (Apt) complex and cDNA was achieved by magnetic separation technique based on magnetic beads (MBs), ensuring the specificity of the aptasensor. Exonuclease III (Exo III)-assisted cDNA cycling process strategy would be triggered when cDNA was separated and introduced to the sensing interface for further signal amplification. Under optimal conditions, the constructed aptasensor presented a wide detection range from 1 × 10 −8 mg·mL −1 to 5 × 10 −4 mg·mL −1 for DON, and the detection limit was 1.79 × 10 −9 mg·mL −1 , including a satisfactory recovery in cornmeal sample spiked with DON. The results showed that the proposed aptasensor had high reliability and promising application potential in detecting DON.
